본 연구에서는 kaempferol 및 그 배당체의 항산화 및 항염증 효능을 평가하기 위해 free radical 소거활성, ROS inhibition assay, TARC 생성 억제 관련 실험을 수행하였다. 또한 kaempferol과 그들의 rhamnosides (${\alpha}$-rhamnoisorobin, afzelin, kaempferitrin)의 구조에 따른 생리적 활성의 상관관계를 조사하였다. Kaempferol과 ${\alpha}$-rhamnoisorobin은 free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) 소거활성($FSC_{50}$)에서 각각 62.5, 50.0 ${\mu}M$이 측정되었으며 afzelin과 kaempferitrin은 농도와 상관없이 free radical 소거활성을 나타내지 않았다. ROS inhibition assay에서는 시료 농도를 10, 50, 100 ${\mu}M$을 처리한 결과 kaempferol은 대조군(100 %)에 비하여 ROS 발현 농도를 각각 97.5, 57.8, 47.8 %로 감소하였으며 ${\alpha}$-rhamnoisorobin은 93.1, 59.1, 41.4 %의 감소를 나타내었다. TARC (thymus and activation regualted chemokine) 생성 억제능 실험을 통해 시료의 항염증 효능을 평가한 결과 kaempferol은 10, 50, 100 ${\mu}M$ 농도에서 대조군(100 %)에 대비하여 각각 48.8, 5.5, 4.4 %로 농도 의존적으로 TARC 생성을 감소시켰으며, ${\alpha}$-rhamnoisorobin은 88.1, 19.0, 1.0 %로 TARC 생성의 감소 효능을 나타내었다. 결론적으로 kaempferol과 그들의 rhamnoside 중 kaempferol과 ${\alpha}$-rhamnoisorobin은 항산화, 항염증에 대한 우수한 효능을 나타내는 것으로 사료되며 더 나아가서는 항노화 및 항염증에 효과가 있는 기능성 화장품에 응용 가능성이 있음을 시사한다.
Flavonoid는 거의 모든 야채와 과일에 함유된 주요한 구성 성분이며 flavonol은 flavonoid 그룹 중 하나로 우리가 식품으로 섭취하는 대표적인 성분으로는 kaempferol과 quercetin이 알려져 있다. kaempferol과 auercetin은 퇴행성질병의 예방에 효과가 있다고 알려져 있어 식품 첨가물로 사용되고 있다. 본 연구의 목적은 항산화제로도 알려진 kaempferol과 quercetin을 이용하여 다양한 농도에서의 소의 폐혈관내피 세포주에 대한 각각의 산화스트레스 보호효과와 두 가지 물질의 동시 처리 시에 나타날 수 있는 상승효과를 밝히기 위함이다. 1 mM의 $H_{2}O_{2}$를 처리하여 산화스트레스를 가한 세포나 가하지 아니한 세포에서나 모두 kaempferol과 quercetin의 처리로 세포의 활성도가 높았다. 그러나 특히 산화스트레스를 가한 세포에서 항산화물의 산화스트레스 극복효과가 현저하게 나타났다. 항산화물의 농도가 고농도의 경우 오히려 증가된 세포활성도가 저하되는 반응을 보여 너무 높은 농도의 처리는 오히려 항산화효과를 억제할 수 있음을 알 수 있었다. 일반적으로 우리가 섭취하는 식품에는 kaempferol과 quercetin이 함께 포함되어 있으므로 이 두 가지 물질을 일정 비율로 서로 섞은 다음 산화스트레스 처리와 함에 처리하여 조사한 결과 단독으로 처리한 결과와 달리 고농도 조건에서도 세포활성도가 아주 높게 나타났다. 이상의 결과에서 항산화물은 생체에서 일어날 수 있는 많은 산화스트레스 상태를 완화시킬 수 있으나 가장 우수한 효능을 나타낼 수 있는 적정 농도의 조사가 중요하며 특히 각각의 항산화물을 단독으로 사용하는 것 보다 함께 사용하는 것이 상승효과를 나타낼 수 있음을 알 수 있었다.
From the whole plants of Bothriospermum tenellum (Boraginaceae) four phenolic compounds, kaempferol, caffeic acid, $kaempferol-3-O-{\beta}-D-glucopyranoside$ and $kaempferol-3-O-{\alpha}-L-rhamnopyranosyl-(1{\rightarrow}6)-{\beta}-D-glucopyranoside$ have been isolated and characterized by physicochemical and spectral means.
The seasonal variations of the major six flavonol glycosides(kaempferol 2,6-dirhamnosyl glucoside, quercetin 3-O-rutinoside, kaempferol 3-O-rutinoside, isorhamnetin 3-O-rutinoside, quercetin 3-O-coumaroyl glucorhamnoside and kaempferol 3-O-coumaroyl glucorhamnoside) in Ginkgo biloba leaves were investigated. The contents were determined by HPLC on reversed phase $C_{18}$ column. This result showed that the percentage of six flavonol glycosides decreased during the season from 1.57% in May to 0.39% in November. The content of each flavonol glycoside indicated a similar tendency to decrease. However, the contents of rutinosides of kaempferol, quercetin and isorhamnetin fluctuated markedly than those of coumaroyl glucorhamnosides of kaempferol and quercetin and kaempferol 2,6-dirhamnosyl glucoside.
The drug consists of the dried entire plant of Viola diamantica (family Violaceae). It is used for the treatment of acute pyogenic diseases such as boil and carbuncles; also as tumor, high fever, tuberculosis and astringent hemostatic. Two flavonol glycosides have been isolated from the aerial parts of Viola diamantica and could be identifed as kaempferol 7-rhamnoside and kaempferol 3,7-dirhamnoside (bright yellow needle crystal, mp $225^{\circ}$, $C_{27}\;H_{30}\;O_4\;4H_2O$). Kaempferol 7-rhamnoside and kaempferol 3,7-dirhamnoside were first isolated from Viola diamantica.
The transcription factor, GATA-1, plays an important role in the $Fc{\varepsilon}RI$${\alpha}$ chain expression in mast cells and basophils. This study was conducted to investigate the downregulation of the transcription factor GATA-1 by kaempferol isolated from Nelumbo nucifera stamens in $Fc{\varepsilon}RI$-mediated allergic reactions. Kaempferol inhibited $Fc{\varepsilon}RI$-mediated histamine release. Western blotting analysis and RT-PCR showed that the protein and mRNA expression of GATA-1 was suppressed by kaempferol in a dose-dependent manner. These results suggest that kaempferol may inactivate basophils by downregulating the $Fc{\varepsilon}RI$${\alpha}$ chain expression via the inhibition of the GATA-1 expression.
Objectives : The present study was performed to clarify the antioxidant and anticancer effects of extract of stamens of Nelumbo nucifera and Kaempferol. Methods : Antioxidant effect was measured by DPPH-radical scavenging activity for electron donating ability (EDA), superoxide dismutase (SOD)-like activity for SOD and lipid peroxidation. Anticancer effect was assessed by MTT absorbance for cytotoxicity. Results : Vitamin E, Kaempferol and ethyl acetate extract of stamens of Nelumbo nucifera increased SOD-like activity and DPPH-radical scavenging activity dose-dependently. On the contrary, lipid peroxidation was time-dependently decreased. Furthermore, Kaempferol and ethyl acetate extract of stamens of Nelumbo nucifera significantly decreased the growth rate of C6 glioma cells. Conclusions : These results suggest that ethyl acetate extract of stamens of Nelumbo nucifera may be a putative antioxidant or anticancer substance.
Three flavonoid glycosides have been isolated from the aerial part of Armoracia rusticana P. (Cruciferae) in Korea and identified by means of spectral analysis as $kaempferol-3-O-{\beta}-D-xylofuranoside(l)$, $kaempferol-3-O-{\beta}-D-galactopyranoside(2)$ and $kaempferol-3-O-{\beta}-D-xylofuranosyl(1\rightarrow2)-b{\beta}-D-galactopyranoside(3)$. When 1 mg/ml of the methanol extract from the aerial part of this plant was added, lipid peroxide formation in the bromobenzene-treated rat liver decreased by 64%. Among the components isolated from title plant, compounds 2 and 3 reduced the formation of lipid peroxide by 16% and 39% respectively at the concentration of ${10}^{-1}$ mg/ml.
Microbial UDP-glycosyltransferases can convert many small lipophilic compounds into glycons using uridine-diphosphate-activated sugars. The glycosylation of flavonoids affects solubility, stability, and bioavailability. The gene encoding the UDP-glycosyltransferase from Bacillus cereus, BcGT-3, was cloned by PCR and sequenced. BcGT-3 was expressed in Escherichia coli BL21(DE3) with a glutathione S-transferase tag and purified using a glutathione S-transferase affinity column. BcGT-3 was tested for activity on several substrates including genistein, kaempferol, luteolin, naringenin, and quercetin. Flavonols were the best substrates for BcGT-3. The enzyme dominantly glycosylated the 3-hydroxyl group, but the 7-hydroxyl group was glycosylated when the 3-hydroxyl group was not available. The kaempferol reaction products were identified as kaempferol-3-O-glucoside and kaempferol-3,7-O-diglucoside. Kaempferol was the most effective substrate tested. Based on HPLC, LC/MS, and NMR analyses of the reaction products, we conclude that BcGT-3 can be used for the synthesis of kaempferol 3,7-O-diglucose.
In order to evaluate on the effect of kaempferol on the cytotoxicity of oxygen tree radicals, XTT assay was performed to determine the cell viability after skin fibroblasts derived from human (Detroit 51) that were treated with various concentrations of hydrogen peroxide $(H_2O_2)$. And also, the effect of kaempferol on the cytotoxicity induced by H202 that was examined by cell viability, lactate dehydrogenase (LDH) activity and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity in these cultures. $H_2O_2$ decreased cell viability in dose-dependent manner in these cultures and the $XTT_{90}\;and\;XTT_{50}$ values were determined at concentration of $35{\mu}M\;and\;90{\mu}M$ of $H_2O_2$ after skin fibroblasts derived from human were treated with $15{\sim}90{\mu}M$ of $H_2O_2$ for 6 hours, respectively. $H_2O_2$ was highly toxic on cultured skin fibroblasts derived from human by toxic criteria of Brenfreund and Puerner (1984). In the protective effect of kaempferol on $H_2O_2$-induced cytotoxicity, kaempferol increased DPPH radical scavenging activity and significantly decreased LDH activity. From these results, it is suggested that oxygen tree radical, $H_2O_2$, was highly toxic on cultured skin fibroblasts derived from human, and also kaempferol of flavonoid showed the protection on $H_2O_2$-induced cytotoxicity.
