Bioequivalence of Cefaclor 375 mg SR Tablet (세파클러 375 mg서방정의 생물학적 동등성)

  • This study was carried out to compare the bioavailability of $Ceclex^{(R)}$ SR TAB (test drug, cefaclor 375 mg/Tablet) with that of Ceclor $MR^{(R)}$ SR IAB (reference drug) and to estimate the pharmacokinetic parameters of cefaclor in healthy Korean volunteers. The bioavailability was examined on 24 healthy volunteers who received a single dose (375 mg) of each drug in the fasting state in a randomized balanced 2-way crossover design. After dosing, blood samples were collected for a period of 7 hours. Plasma concentrations of cefaclor were determined using HPLC with UV detection. The pharmacokinetic parameters $(AUC_{0-7h},\;C_{max},\;T_{max},\;AUC_{inf},\;K_e,\;t_{1/2},\;V_d/F,\;and\;CL/F)$ were calculated with non-compartmental pharmacokinetic analysis. The ANOVA test was utilized for the statistical analysis of the $T_{max}$, log-transformed $AUC_{0-7h$}$, log-transformed $C_{max},\;t_{1/2},\;V_d/F$, and $CL/F$. The ratios of geometric means of $AUC_{0-7h}\;and\;C_{max}$ between test drug End reference drug were $95.67\%\;(8.55\;vs\;8.18{\mu}g{\cdot}hr/ml)\;and\;103.86\%\;(2.85\;vs\;2.96{\mu}g/ml)$, respectively. The $T_{max}$ of test drug and reference drug was $2.56\pm0.15\;and\;2.23\pm0.13\;hrs,\;respectively.\;The\;90\%$ confidence intervals of mean difference of logarithmic transformed $AUC_{0-7h}\;and\;C_{max}$ were log0.90-log1.04 and log0.91-log1.13, respectively. It shows that the bioavailability of test drug is equivalent with that of reference drug.

A Study on Developing Customized Bolus using 3D Printers (3D 프린터를 이용한 Customized Bolus 제작에 관한 연구)

  • Purpose : 3D Printers are used to create three-dimensional models based on blueprints. Based on this characteristic, it is feasible to develop a bolus that can minimize the air gap between skin and bolus in radiotherapy. This study aims to compare and analyze air gap and target dose at the branded 1 cm bolus with the developed customized bolus using 3D printers. Materials and Methods : RANDO phantom with a protruded tumor was used to procure images using CT simulator. CT DICOM file was transferred into the STL file, equivalent to 3D printers. Using this, customized bolus molding box (maintaining the 1 cm width) was created by processing 3D printers, and paraffin was melted to develop the customized bolus. The air gap of customized bolus and the branded 1 cm bolus was checked, and the differences in air gap was used to compare $D_{max}$, $D_{min}$, $D_{mean}$, $D_{95%}$ and $V_{95%}$ in treatment plan through Eclipse. Results : Customized bolus production period took about 3 days. The total volume of air gap was average $3.9cm^3$ at the customized bolus. And it was average $29.6cm^3$ at the branded 1 cm bolus. The customized bolus developed by the 3D printer was more useful in minimizing the air gap than the branded 1 cm bolus. In the 6 MV photon, at the customized bolus, $D_{max}$, $D_{min}$, $D_{mean}$, $D_{95%}$, $V_{95%}$ of GTV were 102.8%, 88.1%, 99.1%, 95.0%, 94.4% and the $D_{max}$, $D_{min}$, $D_{mean}$, $D_{95%}$, $V_{95%}$ of branded 1cm bolus were 101.4%, 92.0%, 98.2%, 95.2%, 95.7%, respectively. In the proton, at the customized bolus, $D_{max}$, $D_{min}$, $D_{mean}$, $D_{95%}$, $V_{95%}$ of GTV were 104.1%, 84.0%, 101.2%, 95.1%, 99.8% and the $D_{max}$, $D_{min}$, $D_{mean}$, $D_{95%}$, $V_{95%}$ of branded 1cm bolus were 104.8%, 87.9%, 101.5%, 94.9%, 99.9%, respectively. Thus, in treatment plan, there was no significant difference between the customized bolus and 1 cm bolus. However, the normal tissue nearby the GTV showed relatively lower radiation dose. Conclusion : The customized bolus developed by 3D printers was effective in minimizing the air gap, especially when it is used against the treatment area with irregular surface. However, the air gap between branded bolus and skin was not enough to cause a change in target dose. On the other hand, in the chest wall could confirm that dose decrease for small the air gap. Customized bolus production period took about 3 days and the development cost was quite expensive. Therefore, the commercialization of customized bolus developed by 3D printers requires low-cost 3D printer materials, adequate for the use of bolus.

Purification and Characterization of Two Thermostable Xylanases from Paenibacillus sp. DG-22

  • Two thermostable xylanases, designated XynA and XynB, were purified to homogeneity from the culture supernatant of Paenibacillus sp. DG-22 by ion-exchange and gel-filtration chromatography. The molecular masses of xylanases A and B were 20 and 30 kDa, respectively, as determined by SDS-PAGE, and their isoelectric points were 9.1 and 8.9, respectively. Both enzymes had similar pH and temperature optima (pH 5.0-6.5 and $70^{\circ}C$), but their stability at various temperatures differed. Xylanase B was comparatively more stable than xylanase A at higher temperatures. Xylanases A and B differed in their $K_m$ and $V_{max}$ values. XynA had a $K_m$ of 2.0 mg/ml and a $V_{max}$ of 2,553 U/mg, whereas XynB had a K_m$ of 1.2 mg/ml and a $V_{max}$, of 754 U/mg. Both enzymes were endo-acting, as revealed by their hydrolysis product profiles on birchwood xylan, but showed different modes of action. Xylotriose was the major product of XynA activity, whereas XynB produced mainly xylobiose. These enzymes utilized small oligosaccharides such as xylotriose and xylotetraose as substrates, but did not hydrolyzed xylobiose. The amino terminal sequences of XynA and XynB were determined. Xylanase A showed high similarity with low molecular mass xylanases of family 11.

  • A simplified mathematical model for the production of high-purity fructo-oligosaccharides by the mixed-enzyme system of fructosyl transferees and glucose oxidase was proposed and compared with the experimental results. The kinetic parameters including $K_m,\;V_{max}\;and\;K_{iG}$ were estimated at $40^{\circ}C$, in which $K_m$, values decreased and $K_{iG}$ and $V_{max}$ values increased compared with those of fructosyl transferees alone. The kinetics of the mixed-enzyme system was successfully described in the form of Michaelis-Menten equations. At the reasonable sucrose concentrations tested, the simulated sugar profiles were of good agreement with the experimental ones.

Effects of Ethanol on $Na^+-dependent$ Solute Uptake in Rabbit Renal Brush-Border Membrane Vesicles

  • This study was undertaken to examine the effect of ethanol on $Na^+-dependent$ transport systems (glucose, phosphate, and dicarboxylate) in renal brush-border membrane vesicles (BBMV). Ethanol inhibited $Na^+-dependent$ uptakes of glucose, phosphate, and succinate in a dose-dependent manner, but not the uptakes of $Na^+-dependent.$ The $H^+/TEA$ antiport was reduced by 8% ethanol. Kinetic analysis showed that ethanol caused a decrease in $V_{max}$ of three transport systems, leaving $K_m$ values unchanged. Ethanol decreased phlorizin binding, which was closely correlated with the decrease in $V_{max}$ of $Na^+-glucose$ uptake. These results indicate that ethanol inhibits $Na^+-dependent$ uptakes of glucose, phosphate, and dicaboxylate and that the reduction in $V_{max}$ of $Na^+-glucose$ uptake is caused by a decrease in the number of active carrier proteins in the membrane.

Proposals of Reliable Shapes of Supplementary V-ties for Section Jacketing Method of Columns (기둥의 단면 확대보강을 위한 V-타이 보조 띠철근의 형상 제시)

  • The objective of the present study is to propose a reliable shape of supplementary V-ties in the section jacketing approach for seismic strengthening of reinforced concrete columns. A total of 24 pull-out specimens were prepared. The test parameters selected with regard to bond strength of V-ties were the shape of V-ties, embedment length of V-tie legs, and compressive strength of concrete. The measured bond strength of V-ties with different shapes were compared with that of the conventional V-ties and predictions using CEB-FIP equation. Ultimately, V-ties with pressed end-details at their legs could be recommended for the supplementary lateral reinforcement of strengthening columns with jacketing thickness less than the embedment length [= max (75mm, $6d_b$)] of conventional V-ties, where $d_b$ is the diameter of the reinforcing bar used for V-ties.

  • When group pile supporting structures are to be subjected to large lateral loads, generally, hatter piles are used in group pile with vertical piles. It is well known that batter piles resist lateral static loads which are acted upon the piles as axial farces quite well but, they show a poor performance under seismic loads. However, it is not yet known how the batter piles behave under dynamic loading and how to strengthen the batter piles to improve the seismic performance. Shaking table tests were performed to investigate the seismic behavior of the batter pile and to bring up the countermeasures to improve the seismic performance. As the result of the shaking table tests, batter piles failed due to not only the excessive increase of compressive force near the pile head but also that of tensile force. In case that the pile head was connected with pile cap by rubber joint, the max. acceleration at the pile cap was reduced due to the high damping ratio of rubber and the max. moment and max. axial farce at the pile head was decreased remarkably. When the inclinations(V:H) of the batter pile were 8:3 and 8:4, max. moment, max. shear force, and max. axial farce were reduced notably and max. acceleration and max. displacement at the pile cap was diminished, too.

TWO-COLOR VR CCD PHOTOMETRY OF OLD NOVA V603 AQUILAE

  • Results of 6 nights of CCD VR photometry of the nova-like variable V603 Aquilae (Nova Aquilae 1918) obtained at the Mallorcian 35-cm telescope in July 2004 are reported. The ephemeris for the superhump maximum is Max.HJD=2453213.60546(96)+0.14813(10)E. The waves with $3.^d9,\;1.^d4,\;0.^d135$ are statistically significant, which may be interpreted as the negative superhump-orbital, the beat periods (negative superhump- positive superhump) and the negative superhump with low amplitude, respectively. Another possible time-scale is $0.^d8,$ which has no coincidence with the beat periods. Quasi-periodic oscillations with an effective period of 18 minutes have been detected, which are close to 15.6 minutes reported by some authors. Their effective semi-amplitudes are $^m.045\;and\;0^m.051$ for V and R, respectively. This corresponds to the 0.12 mag excess in the color index V-R as compared with the mean color, which can be understood as the pulsed emission in the hotter inner parts of the accretion disk, similar to that observed in TT Ari and MV Lyr.

Effects of Percutaneous Balloon Mitral Valvuloplasty on Static Lung Function and Exercise Performance (승모판협착증 환자에서 경피적 풍선확장판막성형술의 폐기능 및 운동부하 검사에 대한 효과)

  • Background: Patients with mitral stenosis(MS) have been demonstrated to have a variable degree of pulmonary dysfunction and exercise impairment. The hemodynamic changes of MS can be reversed after percutaneous mitral balloon valvuloplasty(PMV), but the extent and time course of the imporvement in pulmonary function and exercise capacity are not defined. Methods: In order to investigate the early(3 weeks or less)and late(3 months or more) effects of PMV on pulmonary function and determine if the pulmonary dysfunction is reversible even in patients with moderate to severe pulmonary hypertension, we performed the spirometry, measurements of diffusing capacity and lung volumes, and incremental exercise tests in patients with MS before and after PMV. Results: In 46 patients with MS(age: $40{\pm}12$years, male to female ratio: 1:2, mitral valve area: $0.8{\pm}0.2cm^2$) there was a significant increase in FVC(P<0.0025), $FEV_1$(P<0.001), $FEF_{25-75%}$(P<0.001, $FEF_{50%}$(P<0.001), PEF(P<0.0005), MVV(P<0.005), $\dot{V}O_2$max (P<0.0001), and AT(P<0.0001) after average 10 days of PMV. Also there was a significant decrease in DLco(P<0.0001) and DL/VA(P<0.0001). At later($5{\pm}2$months) follow-up in 11 patients, there was no further improvement in any parameters of pulmonary function and exercise test. Twenty nine patients with sinus rhythm were divided into 16 patients with pulmonary arterial pressure(PAP) more than 35mmHg and/or tricuspid regurgitation grade n or more(group A) and 13 patients with PAP less than 35mmHg(group B). Group A Patients had significantly lower FVC(P<0.001), $FEV_1$(P<0.001), DLco(P<0.05), $\dot{V}O_2$ max(P<0.025) and mitral valve area(P<0.025) than group B patients. Group A patients after PMV, showed significant increase in FVC(P<0.001), maximum $O_2$ pulse(P<0.00001) and $\dot{V}O_2$ max(P<0.00025). Both group showed an increase in AT(P<0.0001, P<0.005), but group A showed greater decrease in $\dot{V}E/\dot{V}O_2$ and $\dot{V}E/\dot{V}CO_2$ both at AT(P<0.001, P<0.001) and $\dot{V}O_2$ max(P<0.0001, P<0.0001) after PMV compared with group B. Conclusion: These data suggest that patients with MS can show increased pulmonary function and exercise performance within 1 month after PMV. Patients with moderate to severe pulmonary hypertension had a significant increase in exercise performance compared with those with mild to no pulmonary hypertension and it is thought to be related to a significat decrease of ventilation for a given oxygen consumption at maximum exercise.

Analysis of Endotoxin Using Analytical Conditions of Optical Density in Metalworking Fluid Sample (금속가공유 시료에서 일부 Optical Density 설정값에 따른 엔도톡신의 정량)

  • This study was performed to identify the proper analytical conditions of endotoxins regarding optical density in the workplace where metalworking fluids are used. This study found that "onset time method" was more accurate than "time to $V_{max}$ method". Reproducibility and accuracy analyzed by "onset time method" was greatly higher than the "time to $V_{max}$". The optical density of "0.03" was the most appropriate analytical condition among "onset time method". In this analytical condition, linearity of 0.998 was obtained and recovery rate ranged from 88% to 105% at the endotoxin concentrations below 5 EU/mL. No significant difference of endotoxins was observed between the optical densities of "0.03" and "0.05". However, correlation coefficients were different with statistical significance (p<0.01). This study confirmed that either the optical densities "0.03" or "0.05" should be used to analyze endotoxin. Of these optical density values, OD with correlation coefficient higher than 0.98 should be used to analyze endotoxin in environmental samples.
