보조 미생물전기화학적 혐기성소화의 적용적합성 검증을 위한 bench scale에서의 성능향상 연구 (Study of performance improvement in bench scale auxiliary bioelectrochemical anaerobic digestion for application suitability verification)

  • 오늘날 급격한 인구증가 및 도시화로 인해 음식물류 폐기물 발생량이 크게 증가하였으며, 음식물류 폐기물에서 발생되는 음폐수의 적절한 처리방안에 대한 관심이 증가하였다. 혐기성 소화(Anaerobic digestion; AD)는 음폐수의 바람직한 처리방법으로 알려졌지만, 긴 처리기간 및 공정 불안정 등의 문제로 개선이 필요하며, 그 중 기존 AD에 보조 반응조를 추가한 보조 미생물전기화학적 혐기성소화(Auxiliary bioelectrochemical anaerobic digestion; ABEAD)가 적절한 개선방안으로 제시되었다. 하지만 아직 20 L 이상 용량에서의 연구는 이뤄지지 않았으며, 따라서 본 연구에서는 100 L의 용량에서 ABEAD의 성능향상을 평가하고 규모증가에 따른 성능변화를 비교하였다. 반응조는 AD와 ABEAD로 구성되었다. 유효용량 100 L, 유기물부하율 4 kg/m3/d, HRT 20 days 및 중온소화(35℃) 조건으로 운전하였으며, AD는 기계적 교반, ABEAD는 기계적 교반 및 펌프를 통한 bulk 용액 순환이 이뤄졌다. ABEAD의 전극재질은 SUS304를 사용하였고, 0.4V의 전압을 공급하였다. 성능비교는 pH, 휘발성지방산(Volatile fatty acids; VFAs), 유기물제거율 및 메탄 생성량을 비교해 수행하였다. 실험결과 AD는 pH 및 VFAs가 각각 평균 7.37 및 3,880 mg/L, ABEAD는 각각 평균 7.5 및 2,870 mg/L로 VFAs의 빠른 처리를 통해 공정안전성 향상되었고, 유기물제거율 및 메탄생성량의 경우 AD는 각각 평균 65.8 % 및 85.1 L/d, ABEAD는 각각 평균 76.1 % 및 108.1 L/d로 유기물의 빠른 처리 및 메탄전환이 이루어져 비교적 큰 규모에서도 ABEAD의 성능향상이 나타남을 확인하였다. 또한 이전 소규모 연구들과 비교를 통해 규모에 따른 성능향상폭을 비교했을 때에도 큰 차이가 나지 않는 것으로 판단되며, 따라서 ABEAD는 BEAD 기술의 상용화 및 적용에 적합한 것으로 사료된다.

Effects of Concentrate to Roughage Ratio on the Formation of cis-9, trans-11 CLA and trans-11-Octadecenoic Acid in Rumen Fluid and Plasma of Sheep When Fed High Oleic or High Linoleic Acid Oils

  • A metabolism trial with four ruminally fistulated sheep was conducted in a $4{\times}4$ Latin square design to examine the effect of concentrate to roughage ratio (70:30 vs. 85:15) and oil source (soybean oil vs. rapeseed oil) on the ruminal fermentation pattern and $C_{18}$-fatty acids composition including trans11-$C_{18:1}$ (trans11-ODA) and cis9, trans11-18:2 (cis9, trans11-CLA) in the rumen fluid and plasma. Oil was added to the concentrate at 5% level of the total diet (DM basis) and chopped rye grass hay was fed as roughage. An increased level of concentrate (85%) within supplemented oil slightly lowered pH but increased ammonia concentration. Supplementation of rapeseed oil relatively increased pH and ammonia concentration. Higher concentrate level resulted in increased tendencies of total VFA concentration while oil source did not affect the total VFA concentration and VFA proportion. Whole tract digestibilities of DM, CP, EE, NDF and OM in diets slightly increased at higher concentrate level. Proportions of oleic acid ($C_{18:1}$) and linoleic acid ($C_{18:2}$) in the rumen fluid were influenced by the fatty acid composition of oil source but oil source did not affect the in vitro formations of trans11-ODA and cis9, trans11-CLA. Slightly increased trans11-ODA and cis9, trans11-CLA proportions, however, were observed from the sheep fed high roughage diet supplemented with both soybean oil and rapeseed oil. The $C_{18:1}$ and $C_{18:2}$ composition in supplemented oils responded to those in plasma of sheep. Effects of concentrate to roughage ratio and oil source on trans11-ODA and cis9, trans11-CLA proportions in plasma were found to be small. Proportion of cis9, trans11-CLA in plasma tended to be increased from the sheep fed high roughage diet and collection time at 9h post feeding.

Effects of Monensin and Fish Oil on Conjugated Linoleic Acid Production by Rumen Microbes in Holstein Cows Fed Diets Supplemented with Soybean Oil and Sodium Bicarbonate

  • The present study was conducted with four ruminally canulated Holstein cows to observe the effects of monensin or fish oil on diet fermentation and production of conjugated linoleic acids (CLAs) in the rumen when fed diets supplemented with soybean oil and sodium bicarbonate. Cows of the control treatment were fed a basal diet (CON) consisting of 60% commercial concentrate and 40% chopped rye grass hay. Cows of other treatments were fed the same diet as CON, but the concentrate was supplemented with 7% of soybean oil and 0.5% of sodium bicarbonate (SO-B), SO-B supplemented with monensin (30 ppm, SO-BM) or concentrate supplemented with 6.3% of soybean oil, 0.5% of sodium-bicarbonate, 30 ppm of monensin and 0.7% of fish oil (SO-BMF). Dry matter (DM) intake of the cows was significantly (p<0.011) reduced by feeding the SO-BMF diet compared to the other diets which did not differ in DM intake. Whole tract digestibility of major dietary components was significantly (p<0.004-0.027) higher for SO-BMF than the other supplement-containing diets. Dietary supplements did not clearly affect rumen pH and ammonia concentrations compared to the CON diet. Significantly reduced (p<0.05) total VFA concentration was obtained by the addition of fish oil to the diet (SO-BMF) compared to other diets. No differences, however, were obtained in major VFA proportions as well as in total VFA between the supplemented diets. The SO-BM diet increased (p<0.01-0.05) the concentrations of trans-11 $C_{18:1}$ and linoleic acid in rumen fluid. Total CLA concentration was also increased by the feeding of SO-B and SO-BM diets during early fermentation times (up to 3 h) post-feeding. Cis-9, trans-11 CLA concentration in rumen fluid was highest (p<0.05) for SO-B up to 1 h while the highest (p<0.01) value for SO-BM occurred at 3 h post-feeding. An increased trans-10, cis-12 CLA concentration was obtained from the SO-B and SO-BM diets at 1 and 3 h post feeding compared to the other diets. Supplementation of oils with monensin and sodium bicarbonate increased (p<0.05) the proportions of $C_{18:1}$ and CLA in the plasma of cows, but the effect of monensin and/or fish oil was limited to trans-10, cis-12 CLA.

음식물쓰레기 산발효공정에서 우점하는 세균의 동정 및 효율적인 VFA 생산을 유도하는 세균의 선별 (Identification of Bacteria Occurred Dominantly and Screening of Best VFAs Producing Bacteria in Food Waste Fermentation Process)

  • 본 연구에서는 산발효 공정에 관여하는 세균들을 조사 분리하고 분리된 세균들을 이용한 VFAs의 생산효율 증가 효과를 검토하였다. 연구 결과는 다음과 같다. 음식물쓰레기 산발효 공정에서 혐기성배지를 이용하여 분리된 주요 세균은 Escherichia coli, Clostridium formicoaceticum, Clostridium butyricum, Clostridium acetobutyricum 등 12 종이며 Escherichia coli와 Clostridium spp.가 우점종을 차지하고 있는 것으로 나타났으며 VFAs 생산에 주도적 역할을 하는 것으로 판단되었다. 아세트산의 생성 효율 향상에 관한 실험에서 E. coli를 $6{\times}10^8\;cells/gTS$의 농도로 접종하였을 때 287 mg/gTS(8,176 mg/L)이 생산되었고 Clostridium formicoaceticum을 $5{\times}10^4\;cells/gTS$의 농도로 접종하였을 때 551 mg/gTS(15,715 mg/L)이 생산되어 대조군과 비교하여 3배의 높은 생산성을 보였다. 부틸산의 생산에 대한 실험에서 Clostridium butyricum을 $2.5{\times}10^5\;cells/gTS$의 농도로 접종하였을 때 214 mg/gTS(6,106 mg/L)이 생성되었고 Clostridium acetobutyricum을 $1.5{\times}10^5\;cells/gTS$의 농도로 접종하였을 때 254 mg/gTS(7,261 mg/L)이 생성되어 대조군과 비교하여 2배의 생산성을 나타내었다.

수소발효의 유기성 폐수 분해 특성 (Characteristics of Organic Wastewater Degradation on Hydrogen Fermentation)

  • 연속형 혐기성처리 반응조에서 배양된 수소발생 슬러지를 이용하여 증온 조건에서 회분식 혐기성 처리방법으로 유기성 폐수로부터 전환되는 수소가스 및 대사산물들에 대한 연구를 수행하였다. 수소발생에 대한 기질로는 sucrose를 이용하였다. 처리과정에서 발생된 누적수소가스, 휘발성지방산(VFAs) 및 solvents는 Gompertz equation을 이용한 비선형회귀분석을 통하여 계산하였다. 처리과정 중 수소가스는 반응초기에 발생하였고, 발생된 가스내 수소가스가 차지하는 비율은 약 20%이었다. 반응 전과정에서 메탄가스는 발생하지 않았다. 비수소가스발생율은 sucrose 농도가 40 g/l일 때 0.956 ml/g VSs/h이었으며, sucrose 농도가 300g/l의 경우는 0.011 ml/g VSS/h이었다. 수소가 발생하는 기간 동안 VFAs의 생성은 acetate, butyrate의 순으로 높게 생성되었으나, propionate로의 전환은 발견되지 않았다. solvents의 경우 butanol이 가장 높게 발생하였다.

Influence of Sunflower Whole Seeds or Oil on Ruminal Fermentation, Milk Production, Composition, and Fatty Acid Profile in Lactating Goats

  • This study aimed to investigate the effect of sunflower seeds, either as whole or as oil, on rumen fermentation, milk production, milk composition and fatty acids profile in dairy goats. Fifteen lactating Damascus goats were divided randomly into three groups (n = 5) fed a basal diet of concentrate feed mixture and fresh Trifolium alexandrinum at 50:50 on dry matter basis (Control) in addition to 50 g/head/d sunflower seeds whole (SS) or 20 mL/head/d sunflower seeds oil (SO) in a complete randomized design. Milk was sampled every two weeks during 90 days of experimental period for chemical analysis and rumen was sampled at 30, 60, and 90 days of the experiment for ruminal pH, volatile fatty acids (tVFA), and ammonia-N determination. Addition of SO decreased (p = 0.017) ruminal pH, whereas SO and SS increased tVFA (p<0.001) and acetate (p = 0.034) concentrations. Serum glucose increased (p = 0.013) in SO and SS goats vs Control. The SO and SS treated goats had improved milk yield (p = 0.007) and milk fat content (p = 0.002). Moreover, SO increased milk lactose content (p = 0.048) and feed efficiency (p = 0.046) compared to Control. Both of SS and SO increased (p<0.05) milk unsaturated fatty acids content specially conjugated linolenic acid (CLA) vs Control. Addition of SS and SO increased (p = 0. 021) C18:3N3 fatty acid compared to Control diet. Data suggested that addition of either SS or SO to lactating goats ration had beneficial effects on milk yield and milk composition with enhancing milk content of healthy fatty acids (CLA and omega 3), without detrimental effects on animal performance.

Effects of Sorghum Hybrid and Grain Supplementation of Silage-Based Diets on Nutrient Digestibilities and Passage Rates and Ruminal Metabolism in Growing Steers

  • Six medium-framed steers, fitted with ruminal cannulas, were utilized in a $6{\times}6$ Latin square design with a $3{\times}2$ arrangement of treatments to determine the effects of sorghum hybrid and grain supplementation on nutrient digestibilities and passage rates and ruminal metabolism of silage-based diets fed to growing steers. The diets consisted of three wholes-plant silages (a high grain-containing, grain sorghum and middle-season, moderate grain-containing, and late-season, low grain-containing forage sorghums), each fed with or without 25% rolled grain sorghum. No significant interactions occurred between sorghum hybrid and grain supplementation for the digestion or passage rate criteria measured. Ruminal butyrate concentration was the only fermentation characteristic affected by a hybrid ${\times}$ grain supplementation interaction. The grain sorghum silage diets had the highest DM, OM, and ADF digestibilities; the late-season silage diets, the lowest. Digestibility of NDF tended to be highest (p < 0.10) for the grain sorghum silage, whereas starch digestibility was not affected by sorghum hybrid. Ruminal ammonia, acetate, propionate, butyrate, and total VFA concentrations were highest for the grain sorghum silage diets. Grain supplementation increased DM and OM digestibilities, but had no effect on digestibilities of NDF, ADF, and starch. Ruminal pH was decreased, but total VFA concentration and acetate : propionate ratio were not affected by grain supplementation.

Use of Nitrate-nitrogen as a Sole Dietary Nitrogen Source to Inhibit Ruminal Methanogenesis and to Improve Microbial Nitrogen Synthesis In vitro

  • An in vitro study was conducted to determine the effect of nitrate-nitrogen used as a sole dietary nitrogen source on ruminal fermentation characteristics and microbial nitrogen (MN) synthesis. Three treatment diets were formulated with different nitrogen sources to contain 13% CP and termed i) nitrate-N diet (NND), ii) urea-N diet (UND), used as negative control, and iii) tryptone-N diet (TND), used as positive control. The results of 24-h incubations showed that nitrate-N disappeared to background concentrations and was not detectable in microbial cells. The NND treatment decreased net $CH_4$ production, but also decreased net $CO_2$ production and increased net $H_2$ production. Total VFA concentration was lower (p<0.05) for NND than TND. Suppression of $CO_2$ production and total VFA concentration may be linked to increased concentration of $H_2$. The MN synthesis was greater (p<0.001) for NND than UND or TND (5.74 vs. 3.31 or 3.34 mg/40 ml, respectively). Nitrate addition diminished methane production as expected, but also increased MN synthesis.

Comparisons of In vitro Nitrate Reduction, Methanogenesis, and Fermentation Acid Profile among Rumen Bacterial, Protozoal and Fungal Fractions

  • The objectives were to compare the ability of various rumen microbial fractions to reduce nitrate and to assess the effect of nitrate on in vitro fermentation characteristics. Physical and chemical methods were used to differentiate the rumen microbial population into the following fractions: whole rumen fluid (WRF), protozoa (Pr), bacteria (Ba), and fungi (Fu). The three nitrogen substrate treatments were as follows: no supplemental nitrogen source, nitrate or urea, with the latter two being isonitrogenous additions. The results showed that during 24 h incubation, WRF, Pr and Ba fractions had an ability to reduce nitrate, and the rate of nitrate disappearance for the Pr fraction was similar to the WRF fraction, while the Ba fraction needed an adaptation period of 12 h before rapid nitrate disappearance. The WRF fraction had the greatest methane ($CH_4$) production and the Pr fraction had the greatest prevailing $H_2$ concentration (p<0.05). Compared to the urea treatment, nitrate diminished net gas and $CH_4$ production during incubation (p<0.05), and ammonia-N ($NH_3$-N) concentration (p<0.01). Nitrate also increased acetate, decreased propionate and decreased butyrate molar proportions (p<0.05). The Pr fraction had the highest acetate to propionate ratio (p<0.05). The Pr fraction as well as the Ba fraction appears to have an important role in nitrate reduction. Nitrate did not consistently alter total VFA concentration, but it did shift the VFA profile to higher acetate, lower propionate and lower butyrate molar proportions, consistent with less $CH_4$ production by all microbial fractions.

  • In spite of the well confirmed advantages of wilting on the fermentation quality, wilting silages are difficult to make at optimum dry matter content. The prolonged wilting, especially in humid condition, result in lower water soluble carbohydrate (WSC) contents, extensive protein breakdown and sometimes higher total volatile fatty acid (VFA) during wilting an ensilage. Apart from difficulty of controlling moisture content, another problem associated with wilted silages is different evaluation on the potential productivity. In temperature climate of Asia, therefore further researches on the effects of wilting on the silage fermentation and potential productivity seen to be necessary.
