Gene expression in HIV-1 is regulated by the promoters in 5' long-terminal repeat (LTR) element, which contain multiple DNA regulatory elements that serve as binding sites for cellular transcription factors. YY1 could repress HIV-1 gene expression and latent infection. Here, however, we observed that virus production can be increased by YY1 over-expression and decreased under YY1 depleted condition by siRNA treatment. To identify functional domain(s) of YY1 activation, we constructed a number of YY1 truncated mutants. Our data show that full-length YY1 enhances the viral transcription both through U3 and U3RU5 promoters. Moreover, the C-terminal region (296-414 residues) of YY1 is responsible for the transcriptional upregulation, which could be enhanced further in the presence of the viral Tat protein. The central domain of YY1 (155-295 residues) does not affect LTR activity but has a negative effect on HIV-1 gene expression. Taken together, our study shows that YY1 could act as a transcriptional activator in HIV-1 replication, at least in the early stages of infection.
We developed an evolutionary optimization process involving a genetic algorithm and combinatorial chemistry (combi-chem), which was tailored exclusively for tile development of LED phosphors with a high luminescent efficiency, when excited by soft ultra violet irradiation. The ultimate goal of our study was to develop oxide red phosphors, which are suitable for three-band white Light Emitting Diodes (LED). To accomplish this, a computational evolutionary optimization process was adopted to screen a Eu$^{3+}$-doped alkali earth borosilicate system. The genetic algorithm is a well-known, very efficient heuristic optimization method and combi-chem is also a powerful tool for use in an actual experimental optimization process. Therefore the combination of a genetic algorithm and combi-chem would enhance the searching efficiency when applied to phosphor screening. Vertical simulations and an actual synthesis were carried out and promising red phosphors for three-band white LED applications, such as Eu$_{0.14}$Mg$_{0.18}$Ca$_{0.07}$Ba$_{0.12}$B$_{0.17}$Si$_{0.32}$O$_{\delta}$, were obtained.

The prototype Chlorella virus PBCV-l encodes 11 tRNA genes and over 350 protein-encoding genes in its 330 kbp genome. Initial attempts to overexpress the recombinant A189/192R protein, a putative virus attachment protein, in E. coli strain BL21(DE3) SI were unsuccessful, and multiple protein bands were detected on Western blots. However, the full-length A189/192R recombinant protein or fragments derived from it were detected when they were expressed in E. coli BL21 CodonPlus (DE3) RIL, which contains extra tRNAs. Codon usage analysis of the a189/192r gene showed highly biased usage of the AGA and AVA codons compared to genes encoded by E. coli and Chlorella. In addition, there were biases of XXA/U($56\%$) and XXG/ C($44\%$) in the codons recognized by the viral tRNAs, which correspond to the codon usage bias in the PBCV-1 genome of XXA/U ($63\%$) over those ending in XXC/G ($37\%$). Analysis of the codon usage in the major capsid protein and DNA polymerase showed preferential usage of codons that can be recognized by the viral tRNAs. The Asn (AAC) and Lys (AAG) codons whose corresponding tRNA genes are duplicated in the tRNA gene cluster were the most abundant (i.e., preferred) codons in these two proteins. The tRNA genes encoded in the PBCV-l genome seem to play a very important role during the synthesis of viral proteins through supplementing the tRNAs that are frequently used in viral proteins, but are rare in the host cells. In addition, these tRNAs would help the virus to adapt to a wide range of hosts by providing tRNAs that are rare in the host cells.

Si (111) 기판 위에 polystyrene (PS) bead를 사용하여 만들어진 약 100 nm 나노 구멍에 GaN나노선을 molecular beam epitaxy 법으로 성장하였다. 성장 온도와 III/V 비율 변화에 대하여 성장된 GaN 나노선의 모양과 광학적 특성은 scanning electron microscopy (SEM)와 photoluminescence (PL) 등으로 조사하였으며, InN/GaN 이종접합 및 InGaN p-n 다이오드구조를 성장하여 atomic force microscopy의 tip 접촉방법으로 전기적 특성을 조사하였다. PL 측정 결과 성장온도가 높아지면 Ga 빈자리와 관계된 3.28 eV의 donor acceptor pair (DAP) 신호와 3.42 eV의 stacking faults (SF) 결함에 기인된 발광 신호세기가 감소하는 결과를 SEM으로부터 나노선 폭 및 길이는 좁아지면서 짧아지는 것을 관측하였다. 또한 nitrogen 원자양이 증가하면서 Ga 빈자리와 관련된 3.28 eV DAP 신호가 증가하는 것을 관측하였다. 이들 결과로부터 GaN 나노선의 SF 발광 신호관련 원인에 대하여 논의 하였다. AFM을 이용한 I-V 측정으로부터 성장조건 변화에 따른 GaN 나노선 및 p-n 접합 나노선의 전도 특성을 조사하여 나노선의 소자 응용에 대한 기본적인 물리특성을 규명하였다.
Chemical analysis and metallographic observations of the iron slag were carried out in an attempt to estimate the old iron-making process. The slag containing $9.3\%\;TiO_2$ without Cu indicates that the ore used for smelting was sand iron, not rock ore. The phases identified in the slag were $ulv\ddot{o}spinel$, magnetite, $w\ddot{u}stite$, fayalite etc. This also supports the fact that the smelted ore was iron sand. The total amount of Fe and slag-making components$(=SiO_2+Al_2O_3+MgO+CaO)$ were $40.7\%\;and\;36.1\%$, respectively. These values were average ones found in the old slags formed in the ancient iron-making process. Assuming that $TiO_2$ in the ore combines with FeO, resulting in the formation of $ulv\ddot{o}spinel$, the estimated temperature of smelting was found to be about $1200^{\circ}C$.

Effect of volcanic ash on cell growth of Aspergillus sp. and production of exopolymers by Agrobacterium sp. and Aureobasidium pullualns was investigated. The volcanic ash contained various mineral salts such as $SiO_2$, $Al_2O_3$, CaO, $K_2O$. Maximal cell growth of Aspergillus sp. was obtained when 0.3% volcanic ash was added to medium. Cell growth of Aspergillus sp. increased with higher concentration of volcanic ash in medium. Amount of cell growth with 0.3 % volcanic ash was 6.7 times higher than that without volcanic ash. Volcanic ash also stimulated production of exopolymer as well as cell growth. Production of curdlan with 0.1% volcanic ash was 12.40 g/l whereas that without volcanic ash was 9.15 g/l. Production of pullulan with volcanic ash was also higher than that without volcanic ash.

염료 감응형 태양전지는 기존 Si 기반 PN접합 무기 태양전지에 비해서 경제적이다. 하지만 그 에너지 변환 효율은 아직까지 세계 최고 수준이 10%밖에 도달하지 못하였다. 그래서 다양한 방식의 효율개선 연구가 활발히 진행되고 있는 실정이다. 본 연구에서는 진공원자층증착(ALD)를 이용하여 Core-shell 구조의 $TiO_2$층 위에 아주 얇고 균일한 $Al_2O_3$ (알루미나) 산화막을 입혔다. 이를 통해서 염료감응형 태양전지의 에너지 변환 효율을 향상시켰다. 본 연구에서는 진공원자층증착(ALD)기술을 이용한 $Al_2O_3$ (알루미나) 산화막의 증착조건에 따른 염료감응태양전지의 효율 개선 매커니즘에 대해서 고찰하였다.

In vitro 상에서 골조직을 원활하게 재생하기 위해서는 3차원 지지체를 이용한 세포 배양과 세포 배양 시 세포의 형태와 기능을 유지/향상시키기 위한 인체 내 미세 환경 재현은 필수적이다. 따라서 본 연구에서는 뼈 성분과 유사한 생체 활성 물질인 hydroxyapatite (HA)와 생분해성 고분자인 poly $\varepsilon$-caprolactone (PCL)를 복합재료로 이용하여 내부 연결성이 우수한 골조직 재생용 3차원 지지체를 제작하였으며, 골 재생 능력 향상을 위하여 인체내 골조직의 기계적 미세 환경을 체외에서 구현한 새로운 형태의 perfusion bioreactor system을 개발/적용하였다. 또한 본 연구에서 개발된 perfusion bioreactor system의 생물학적 평가를 위해 MG63 (osteoblast like cell, 한국 세포주 은행)과 New Zealand White Rabbit에서 분리한 중간엽 줄기세포를 골조직 재생용 3차원 지지체에 파종하였다. 48시간 동안 안정화 후 perfusion bioreactor system을 이용하여 기계적 자극을 파종된 세포에 인가하였으며, 배양 기간 동안 세포의 증식 확인 및 형태학적 관찰을 실시하였다. 본 연구 결과, perfusion bioreactor system을 이용하여 기계적 자극을 인가한 실험군에서 세포의 증식 및 활성도가 대조군에 비해 우수함을 확인 할 수 있었다. 따라서, perfusion bioreactor를 이용한 세포 배양은 세포의 활성 향상 및 골조직 재생에 도움이 될 것으로 사료된다. 차후 perfusion bioreactor를 이용한 다양한 패턴의 자극이 골재생 능력 및 중간엽 줄기세포의 골 분화능에 미치는 영향에 대한 연구가 필요할 것으로 사료된다.
This study was undertaken to determine if Plantaginis Semen Herbal Acupuncture(PSA) has a protective effect against glycerol-induced acute renal failure in rats. Rats were dehydrated for 24hr and then injected with 8 ml/kg of $50\%$ glycerol, one-half of dose in each hindlimb muscle. In experiments for PSA effect, rats received 0.1 ml of PSA extraction in both sides of corresponding Shenso($BL_{23}$) of human body for 3 days after injection of glycerol. The experimental group were di vided into the Normal group, the Control group, the PSA group. Glycerol injection decreased glomerular filtration rate and increased urine volume, serum creatinine, BUN level and fractional excretion of glucose, $Na^+$, $K^+$ and $CI^-$. These result show that glycerol injection result in acute renal failure. PSA significantly increased glomerular filtration rate and significantly decreased serum creatinine, BUN level and fractional excretion of glucose, $Na^+$ and $CI^-$ as compared Control group. This suggests that PSA could be used in prevention and treatment of acute renalfailure. However, the precise mechanisms of PSA protection remain to be determined.
Cordyceps militaris mycelium extracts containing high amounts of cordycepin were evaluated in vitro for their anti-inflammatory and tumor cell growth-inhibitory activities. All extracts dose dependently inhibited the increased production of inflammatory mediators including reactive oxygen species (ROS), nitric oxide (NO), and 𝛽-hexosaminidase in lipopolysaccharide (LPS)-stimulated inflammatory cells. All extracts were evaluated for anti-proliferative activity against normal RBL-2H3 cells and diverse types of cancer cell lines, including HCT, MC5-7, U-87MG, AGS, and A549 cells. The extract showed the strongest growth inhibition (IC50 = 28.13 ㎍/mL) relative to vehicle-treated control cells against fibrosarcoma (MC5-7). We have demonstrated anti-inflammatory activity of C. militaris via inhibition of NO, ROS production, and 𝛽-hexosaminidase release in activated cells. C. militaris mycelium extract was also evaluated mechanistically and found to exert six types of anti-cancer activity, confirming its pharmacological potential. Our study suggests C. militaris use as a potential source of anti-inflammatory and anti-cancer agents. C. militaris may also be considered a functional food.
