대청호에서 수환경 요인과 식물플랑크톤의 변동 (Dynamics of Water Environmental Factors and Phytoplankton in Taechong Reservoir)

  • 대청호의 중.하류 주요지점에서 수질특성을 파악하기 위하여 환경요인과 식물플랑크톤의 변동을 98년 6월부터 99년 6월까지 매월 조사하였다. 환경요인 중 수온, DO및 pH는 각각 5.3~27.7$^{\circ}C$, 6.2~13.8 mgO$_2$/1 및 6.4~9.5 범위로서 계절적 영향이 컸다. 무기영양염 중 NH$_4$, NO$_3$, SRP 및 SRSi 농도는 각각 18~104 $\mu$g N/1, 79~1,111 $\mu$g N/1, 0~16 $\mu$g P/1 및 0~3.0 mg Si/l 범위였다. 무기질소 중 NH$_4$ 는 NO$_3$의 5.5~7.2%수준이었다. NO$_3$는 하계에만 다소 낮았고 다른 시기에는 거의 일정하였다. SRP와 SRSi농도는 남조류가 우점하는 하계에 증가하였고 수온이 감소된 10월 이후에 규조류의 증가와 함께 SRP와 SRSi농도가 감소하였다. 따라서 SRSi는 SRP와 더불어 조류 생장에 밀접한 관련성이 있는 영양염으로 평가되었다. Chl-$\alpha$ 평균농도는 8~12 $\mu$g/1 범위로서 하류(댐)에서 가장 낮았다. 또한 하절기에 TN/TP비가 감소할수록 chi-$\alpha$ 농도는 증가하였고 TN/TP비의 평균값은 110으로서 조류의 생장에 대해 P 제한성이 강하였다. 식물플랑크톤의 변동은 계절에 따라 소수종이 우점하는 특성이 관찰되었다. 하계에는 남조류 Anabaena, Microcystis 및 Oscillatoria속 조류가 주로 초여름부터 늦가을까지 발생하여 번무하였다. 이 중에서 A. spiroides v. crassa는 0.3~2.0$\times$$10^4$cells/ml, M. aeruginosa는 6.4$\times$$10^2$~1.0$\times$$10^4$cells/m1 및 O. limosu는 4.6$\times$$10^3$~1.6$\times$$10^4$ cells/ml 범위로서 변화 폭이 가장 컸다. 반면 동계에는 규조류 Stephanodiscus속 조류가 11월~4월에 평균 4.9$\times$$10^2$cells/m1수준이었고 1월에 평균 세포수가 1.3$\times$$10^3$ cells/ml로 많아 중요종으로 볼 수 있었다.

충남탄전, 대동누층군의 셰일과 탄질암에 관한 암석화학 및 환경지구화학적 특성 (Petrochemistry and Environmental Geochemistry of Shale and Coal from the Daedong Supergroup, Chungnam Coal Field, Korea)

  • Characteristics of sedimentary rocks and enrichment of toxic elements in shale and coal from the Chungnam coal field were investigated based upon geochemistry of major, trace and rare earth elements. Shale and coal of the area are interbedded along the Traissic to the Jurassic Daedong Supergroup, which can be subdivided into grey shale, black shale and coal. The coal had been mined, however all the mines are abandonded due to the economic problems. The shale and coal are characterized by relatively low contents of $SiO_2$, and $Al_2O_3$ and high levels of loss-on-ignition (LOI), CaO and $Na_2O$ in comparison with the North American Shale Composite (NASC). Light rare earth elements (La, Ce, Yb and Lu) are highly enriched with the coal. Ratios of $Al_2O_3/Na_2O$ and $K_2O/Na_2O$ in shale and coal range from 30.0 to 351.8 and from 4.2 to 106.8, which have partly negative correlations against $SiO_2/Al_2O_3$ (1.24 to 6.06), respectively. Those are suggested that controls of mineral compositions in shale and coal can be due to substitution and migration of those elements by diagenesis and metamorphism. Shale and coal of the area may be deposited in terrestrial basin deduced from high C/S (39 to 895) and variable composition of organic carbon (0.39 to 18.40 wt.%) and low contents of reduced sulfur (0.01 to 0.05 wt.%). These shale and coal were originated from the high grade metamorphic and/or igneous rocks, and the rare earth elements of those rocks are slightly influenced with diagenesis and metamorphism on the basis of $Al_2O_3$ versus La, La against Ce, Zr versus Yb, the ratios of La/Ce (0.38 to 0.85) and Th/U (3.6 to 14.6). Characteristics of trace and rare earth elements as Co/Th (0.07 to 0.86), La/Sc (0.31 to 11.05), Se/Th (0.28 to 1.06), V/Ni (1.14 to 3.97), Cr/V (1.4 to 28.3), Ni/Co (2.12 to 8.00) and Zr/Hf (22.6~45.1) in the shale and coal argue for inefficient mixing of the simple source lithologies during sedimentation. These rocks also show much variation in $La_N/Yb_N$ (1.36 to 21.68), Th/Yb (3.5 to 20.0) and La/Th (0.31 to 7.89), and their origin is explained by derivation from a mixture of mainly acidic igneous and metamorphic rocks. Average concentrations in the shale and coal are As=7.2 and 7.5, Ba=913 and 974, Cr=500 and 145, Cu=20 and 26, Ni=38 and 35, Pb=30 and 36, and Zn=77 and 92 ppm, respectively, which are similar to those in the NASC. Average enrichment indices for major elements in the shale (0.79) and coal (0.77) are lower than those in the NASC. In addition, average enrichment index for rare earth elements in coal (2.39) is enriched rather than the shale (1.55). On the basis of the NASC, concentrations of minor and/or environmental toxic elements in the shale and coal were depleted of all the elements examined, excepting Cr, Pb, Rb and Th. Average enrichment indices of trace and/or potentially toxic elements (As, Cr, Cu, Ni, Pb, U and Zn) are 1.23 to 1.24 for shale and 1.06 to 1.22 for coal, respectively.

고체상미량추출법을 이용한 BTEX와 MTBE의 경쟁적 추출효과 및 미량분석에 관한 연구 (Competitive Extraction and Trace Analysis of BTEX and MTBE by Solid-Phase Microextraction (SPME))

  • In this study, Solid-phase microextraction (SPME) with GC/FID was studied as a possible alternative to liquid-liquid extraction for the analysis of BTEX and MTBE. Experimental parameters affecting the SPME process (such as kind of fibers, adsorption time, desorption time, volume ratio of sample to headspace, salt addition, and magnetic stirring) were optimized. Experimental parameters such as CAR/PDMS, adsorption time of 20 min, desorption time of 5 min at $250^{\circ}C$, headspace volume of 50 mL, sodium chloride (NaCl) concentration of 25% combined with magnetic stirring were selected in optimal experimental conditions for analysis of BTEX and MTBE. The general affinity of analytes to CAR/PDMS fiber was high in the order p-Xylene>Toluene>Ethylbenzene>MTBE>Benzene. The linearity of $R^2$ for BTEX and MTBE was from 0.970 to 0.999 when analyte concentration ranges from $30{\mu}g/L$ to $500{\mu}g/L$, respectively. The relative standard deviation (% RSD) were from 2.5% to 3.2% for concentration of $100{\mu}g/L$ (n=5), respectively. Finally, the limited of detection (LOD) observed in our study for BTEX and MTBE were from $7.5{\mu}g/L$ to $15{\mu}g/L$, respectively.

Anticancer Activity of Novel Daphnane Diterpenoids from Daphne genkwa through Cell-Cycle Arrest and Suppression of Akt/STAT/Src Signalings in Human Lung Cancer Cells

  • Although the immense efforts have been made for cancer prevention, early diagnosis, and treatment, cancer morbidity and mortality has not been decreased during last forty years. Especially, lung cancer is top-ranked in cancer-associated human death. Therefore, effective strategy is strongly required for the management of lung cancer. In the present study, we found that novel daphnane diterpenoids, yuanhualine (YL), yuanhuahine (YH) and yuanhuagine (YG) isolated from the flower of Daphne genkwa (Thymelaeaceae), exhibited potent anti-proliferative activities against human lung A549 cells with the $IC_{50}$ values of 7.0, 15.2 and 24.7 nM, respectively. Flow cytometric analysis revealed that the daphnane diterpenoids induced cell-cycle arrest in the G0/G1 as well as G2/M phase in A549 cells. The cell-cycle arrests were well correlated with the expression of checkpoint proteins including the up-regulation of cyclin-dependent kinase inhibitor p21 and p53 and down-regulation of cyclin A, cyclin B1, cyclin E, cyclin dependent kinase 4, cdc2, phosphorylation of Rb and cMyc expression. In the analysis of signal transduction molecules, the daphnane diterpenoids suppressed the activation of Akt, STAT3 and Src in human lung cancer cells. The daphnane diterpenoids also exerted the potent anti-proliferative activity against anticancer-drug resistant cancer cells including gemcitabine-resistant A549, gefitinib-, erlotinib-resistant H292 cells. Synergistic effects in the growth inhibition were also observed when yuanhualine was combined with gemcitabine, gefitinib or erlotinib in A549 cells. Taken together, these findings suggest that the novel daphnane diterpenoids might provide lead candidates for the development of therapeutic agents for human lung cancers.

Nano TiNx를 함유한 Ti복합체의 제조 (Fabrication of Titanium Composites Containing nano-sized TiNx)

  • In this research we tried to make nano-sized TiNx by using planetary milling, and we made the composites double layered of titanium and nano-sized TiNx by using spark plasma sintering apparatus after mixing with the different ratio of pure titanium powder, and they were heat treated at $850^{\circ}C$ for 30 minutes. The crystal structures of nano-sized TiNx powders and the composites were analyzed by X-ray diffraction (XRD). The microstructures of the powders were analyzed by using scanning electron microscopy (FESEM) and the 40-50 nm size of nano-sized TiNx particle on the surface of agglomerated particles was investigated. With increasing the ratio of nano-sized TiNx of the composites, the microvickers hardness of the composites was increased.

Evaluation of photon radiation attenuation and buildup factors for energy absorption and exposure in some soils using EPICS2017 library

  • In this paper, the EPICS2017 photoatomic database was used to evaluate the photon mass attenuation coefficients and buildup factors of soils collected at different depths in the Philippine islands. The extraction and interpolation of the library was accomplished at the recommended linear-linear scales to obtain the incoherent and total cross section and mass attenuation coefficient. The buildup factors were evaluated using the G-P fitting method in ANSI/ANS-6.4.3. An agreement was achieved between XCOM, MCNP5, and EPICS2017 for the calculated mass attenuation coefficient values. The buildup factors were reported at several penetration depths within the standard energy grid. The highest values of both buildup factor classifications were found in the energy range between 100 and 400 keV where incoherent scattering interaction probabilities are predominant, and least at the region of predominant photoionization events. The buildup factors were examined as a function of different soil silica contents. The soil samples with larger silica concentrations were found to have higher buildup factor values and hence lower shielding characteristics, while conversely, those with the least silica contents have increased shielding characteristics brought by the increased proportions of the abundant heavier oxides.

한국산 그물베도라치(Dictyosoma burgeri)의 산란행동 유도 및 난 발생과 자어의 형태발달 (Spawning Behavior and Morphological Development of the Eggs and Larvae of the Blennioid Fish, Dictyosoma burgeri from Korea (Pisces: Stichaeidae))

  • 이 연구는 우리나라에 서식하는 그물베도라치의 산란행동과 난 발생 및 자어의 형태발달을 조사하여 분류학적 연구의 기초자료로 이용하고자 실시하였다. 수정란은 구형으로 백색의 불투명한 침성 점착란이었고, 유구의 수는 3~4개였으며, 난의 크기는 1.31~1.47mm(평균 $1.38{\pm}0.05mm$, n=10) 범위였다. 유구의 크기는 0.12~0.38 mm (평균 $0.25{\pm}0.10mm$, n=10)였다. 사육수온은 $14.5{\sim}16.5^{\circ}C$ (평균 $15.0{\pm}0.05^{\circ}C$)를 유지하였고, 난 발생 과정은 64세포기 후 30시간이 경과하여 상실기에 달하였으며, 64세포기 후 744시간이 경과하여 머리 윗부분이 난막을 뚫고 나오면서 부화가 시작되었다. 부화직후의 자어는 전장 7.10~7.60mm(평균 $7.34{\pm}0.18mm$)로 입은 열려 있었고, 부화 후 6일째의 후기 자어는 전장 9.06~9.13 mm (평균 $9.10{\pm}0.02mm$)로 꼬리지느러미의 윗부분에 줄기가 분화하기 시작하였고, 난황은 모두 흡수되었으며, 먹이 섭식활동을 시작하였다.

Ginseng-derived type I rhamnogalacturonan polysaccharide binds to galectin-8 and antagonizes its function

  • Background: Panax ginseng Meyer polysaccharides exhibit various biological functions, like antagonizing galectin-3-mediated cell adhesion and migration. Galectin-8 (Gal-8), with its linker-joined N- and C-terminal carbohydrate recognition domains (CRDs), is also crucial to these biological processes, and thus plays a role in various pathological disorders. Yet the effect of ginseng-derived polysaccharides in modulating Gal-8 function has remained unclear. Methods: P. ginseng-derived pectin was chromatographically isolated and enzymatically digested to obtain a series of polysaccharides. Biolayer Interferometry (BLI) quantified their binding affinity to Gal-8, and their inhibitory effects on Gal-8 was assessed by hemagglutination, cell migration and T-cell apoptosis. Results: Our ginseng-derived pectin polysaccharides consist mostly of rhamnogalacturonan-I (RG-I) and homogalacturonan (HG). BLI shows that Gal-8 binding rests primarily in RG-I and its β-1,4-galactan side chains, with sub-micromolar KD values. Both N- and C-terminal Gal-8 CRDs bind RG-I, with binding correlated with Gal-8-mediated function. Conclusion: P. ginseng RG-I pectin β-1,4-galactan side chains are crucial to binding Gal-8 and antagonizing its function. This study enhances our understanding of galectin-sugar interactions, information that may be used in the development of pharmaceutical agents targeting Gal-8.

Hot Wall Epitaxy(HWE)법에 의한 $AgInS_2$단결성 박막의 성장과 가전자대 갈라짐에대한 광전류 연구 (Photocurrent Study on the Splitting of the Valence Band and Growth of $AgInS_2$GaAs Single Crystal Thin Film by Hot Wall Epitaxy)

  • 수평 전기로에서 AgInS₂ 다결정을 합성하여 HWE(Hot Wall Epitaxy)방법으로 AgInS₂ 단결정 박막을 반절연성 GaAs(100)기판에 성장시켰다. AgInS₂ 단결정 박막의 성장 조건은 증발원의 온도 680℃, 기판의 온도 410℃였고 성장 속도는 0.5㎛/hr였다. AgInS₂ 단결정 박막의 결정성의 조사에서 10 K에서 광발광(photoluminescence)스펙트럼이 597.8 nm(2.0741 eV)에서 exciton emission스펙트럼이 가장 강하게 나타났으며, 또한 이중결정 X-선 요동곡선(DCRC)의 박폭치(FWHM)도 121 arcsec로 가장 작아 최적 성장 조건임을 알수 있었다. Hall 효과는 van der Pauw 방법에 의해 측정되었으며, 온도에 의존하는 운반자 농도와 이동도는 293K에서 각각 9.35×10/sup 16/㎤, 294㎠/V·s 였다. AgInS₂ /SI(SEmi-Insulated) GaAs(100) 단결정 박막의 광흡수와 광전류 spectra를 293K에서 10K까지 측정하였다. 광흡수 스펙트럼으로부터 band gap E/sub g/(T)는 Varshni 공식에 따라 계산한 결과 2.1365eV-(9.89×10/sup-3/eV/K/)T²(T+2930K)이었으며 광전류 스펙트럼으로부터 Hamiltopn matrix(Hopfield quasicubic mode)법으로 계산한 결과 crystal field splitting Δcr값이 0.1541eV이며 spin-orbit Δso 값은 0.0129eV임을 확인하였다. 10K일때 광전류 봉우리들은 n=1 일때 A₁-, B-₁와 C₁-exction 봉우림을 알았다.

유한요소 분석을 통한 심미적 교정 브라켓의 응력 및 구조분석에 관한 연구 (STRESS DISTRIBUTION IN ESTHETIC ORTHODONTIC BRACKETS : AN ANALYSIS USING THE FINITE ELEMENT METHOD)

  • 본연구는 유한요소 분석법의 사용으로 세가지 형태의 브라켓에 tipping과 torquing force를 부여하여 응력분포를 알아보고 또한 스테인레스 스틸과 세라믹의 재질을 임의 설정하여 응력 분포를 알아보아 생 역학적으로 우수한 브라켓을 디자인 하는 데 기초가 되기위해 수행되었다. 선정된 세가지 브라켓은 A:Transcend(Unitek) B:Signature(RMO) C:PAW(Plane Arch Wire appliance) 이었다. 철선은 0.0175x0.025 스테인레스 스틸과 C형은 새롭게 고안된 마름모꼴 철선이 이용되었다. tipping force는 4.27N을 브라켓의 wire slot의 gingival wall의 mesial surface와 대각선으로 대칭되는 면에 각각의 node에 분력의 형태로 작용시켰다. torquing force는 couple force의 형태로 32.858N을 19.7도 회전하였고 bracket C형은 동량의 힘을 11.3도 회전하여 X와 Y의 방향으로 분력의 형태로 나누어 해석하였고 von Mises stress(최대상당응력)가 각 브라켓에서 기록되었다. 이에 다음과 같은 결론을 얻었다. 첫째, 유한요소 해석을 통해 브라켓과 wire slot을 실제로 제작하지 않고도 각각의 재료에 따른 응력분포를 예측할 수 있었다. 둘째, 응력의 집중부위는 Tipping시 브라켓 A, B, C모두 유사하게 나타났는데 브라켓 wire slot의 gingival wall과 mesial surface가 만나는 부위와 wire slot의 incisal wall과 distal surface가 만나는 부위였다. Torquing시 wire slot의 길이 방향을 따라 넓게 분포되었고 gingival wall과 협면으로 이행되는 모서리 그리고 incisal wall내면에 집중되었다. 셋째, 브라켓에 작용하는 응력은 Tipping에 의한 것보다는 Torquing에 의한 영향이 더 큼을 알수 있었으며 Torquing force를 기준으로 브라켓을 설계하는 것이 바람직한 것으로 판단되었다. 넷째, 브라켓의 형상과 함께 재료의 변화가 응력감소에 영향을 미치는 것으로 사료되므로 브라켓 제작시 한가지 재료보다 응력이 많이 받는 부위는 다른 재료를 사용하여 응력의 분포를 줄일수도 있음을 알수있었다. 다섯째, 사용하기에 용이하고 운용이 효율적일 것으로 판단되는 브라켓C형(PAW브라켓)은 응력집중현상이 다른 두 브라켓보다 크게 나타났으므로 이의 제작시 기계적 성질이 우수한 재료를 사용해야 할 것으로 사료되었다.
