Tolerance mechanism to simazine (6-chloro-N,N'-diethyl-1,3,5-triazine-2,4-diamine) in Coix lacryma-jobi was investigated with respect to herbicide detoxification via glutathione conjugation. Simazine was initially absorbed by seedlings of C. lacryma-jobi and corn, but after 12 hours of treatment, no significant difference in simazine absorption was found in both species. Simazine absorbed was rapidly metabolized to glutathione-simazine conjugate. One to six hours after treatment, metabolism was approximately 2-fold faster in C. lacryma-jobi than in corn. Glutathione content was found 1.5- and 2.3-fold higher in coleoptile and root of C. lacryma-jobi, respectively, compared with corn. In both species, the highest concentration of glutathione was found in coleoptile tissue. Glutathione S-transferase that exhibits activity with 1-chloro-2,4-dinitrobenzene was not significantly different between two species. However, glutathione S-transferase activity with simazine was approximately 2-fold greater in C. lacryma-jobi than in corn. The glutathione S-transferase activity was 20 to 30% greater in shoot of either species than in root. Fast protein liquid chromatography-anion exchange column was used to separate glutathione S-transferase isozymes in coleoptiles of C. lacryma-jobi and corn. A peak of glutathione S-transferase activity with 1-chloro-2,4-dinitrobenzene and two peaks of glutathione S-transferase activity with simazine from C. lacryma-jobi were coeluted with those from corn, but showed greater activity than in the case of corn. Another glutathione S-transferase isozyme that exhibits activity with simazine was detected in the elution of C. lacryma-jobi extract, but not in corn. Electron transport in chloroplast thylakoids isolated from leaves of both species was equally sensitive to simazine applied at 1 to 100 nM. These results indicate that the simazine tolerance in C. lacryma-jobi is due to its capacity to detoxify the herbicide via glutathione conjugation, which is positively correlated with the level of glutathione content and glutathione S-transferase activity.

Impact is one of the major cause of damage to fruits druing varios processes from the production on the farm to the consumer. The tissue of fruits are ruptured in a very short period time less than 10ms by impact loading. Mechanical behavior of fruits under impact loading can be analyzed better with high speed sampling data acquisition system and one of them is a digital storage oscilloscope. A impact test system was developed to test the physical properties of fruits including apple, pear, and peach which may lead to a better understanding of the physical laws. The test system consisted of a digital storage oscilloscope and simple mechanism which can apply impact force to fresh produce. Rupture force, energy, and deffrmation were measured at the five levels of drop heights from 4 to 24cm fur each internal and external tissues. Rupture forces for apple and pear were in the range of 72.9 to 87.7 N and 70.8 to 84.1 N for external and internal tissues, respectively. Rupture forces far peach external tissues were in the range of 43.4 to 65.0 N.
$\alpha$-Cyclodextrin glucanotransferase excreted from a newly isolated Geobacillus thermosacchalytycus was purified through the ultrafiltraion, hydrophobic Sepharose CD-4B affinity chromatography, and gel filtration on Sephadex G-75, respectively. The molecular weight of the purified CGTase was 69 kDa and its N-terminal amino acid sequence was determined to be Asn-Leu-Asn-Lys-Val-Asn-Phe-Val-Ser-Asp-Val-Val-Val-Gln-Ile. The optimum pH and temperature were pH 6.0 and$ 60^{\circ}C$, respectively, and stably at the pH range of 6.0-8.0 and $60^{\circ}C$ in the presence of $Ca^{++}$. The excreted CGTase from the thermophilic G. thermosacchalytycus was $\alpha$-type showing a high coupling activity for the transglycosylation on various glucosides. The coupling reaction was carried out according to the random ternary complex mechanism.m.

Stephania delavayi Diels. (S. delavayi Diels.) has been used as a drug for pain-relieving and acute gastroenteritis treatment in China. Because the major therapeutic mechanism of anti-inflammatory drug is to inhibit the cyclooxygenase (COX)-2 and because COX-2 proteins inhibit apoptosis, COX-2 inhibitor has been thought as the anticancer drug candidate. For this reason, we examined S. delavayi Diels. as an anticancer drug. S. delavayi Diels. was fractionated with methanol and then partitioned with ethyl acetate, n-butanol and water. The antioxidant activity was evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and reducing power. DPPH radical scavenging activities of the crude fractions at the concentration of $1,000{\mu}g/mL$ were 75.23% (n-butanol), 68.11% (methanol), 63.58% (ethyl acetate), and 50.13% (water). The reducing power increased according to the concentration in dose-dependent manner. Also, when the antiproliferation effects of each fraction against human breast cancer cell-lines MDA-MB-231 and MCF-7 were examined, methanol extract, n-butanol fraction and ethyl acetate fraction exhibited cell proliferative inhibition effects in both cell-lines whereas water fraction did not. Among the crude fractions, the n-butanol fraction exhibited the most potent anti-proliferation effect. In conclusion, fractions from S. delavayi Diels. are promising anticancer drug candidates.
The induction of phase II enzymes including quinone reductase [NAD(P)H dehydrogenase(quinone): NAD(P)H : (quinone acceptor) oxidoreductase, EC 1.6.99.2] is a major mechanism of whereby a large group of heterogeneous compounds prevent the toxic, mutagenic, and neoplastic effects of carcinogen. Using murine hepatoma cells(Hepalclc7 cells), quinone reductase(QR) inducers as the possible chemopreventive agents were screened from rice bran, wheat bran, soymilk residue, defatted soybean cake, defatted sesame and perilla residues. The 80% methanol extracts of defatted sesame and perilla residues induced quinone reductase significantly while the others did have little effect on the enzyme induction. Thin layer chromatography of the extracts showed that the fastest moving band(Rf=0.70) in the developing solvent of n-butanol : n-propanol : 2N ammonia(10 : 60 : 30) was responsible for the enzyme induction by the 80% methanol extracts of defatted sesame and perilla residues. Further identification of active component(s) is in progress.
We have examined consequences of strong tidal encounters between a neutron star and a normal star using SPH as a possible formation mechanism of isolated recycled pulsars in globular clusters. We have made a number of SPH simulations for close encounters between a main-sequence star of mass ranging from 0.2 to 0.7 $M_\bigodot$ represented by an n=3/2 poly trope and a neutron star represented by a point mass. The outcomes of the first encounters are found to be dependent only on the dimensionless parameter $\eta'{\equiv}(m/(m+ M))^{1/2}(\gamma_{min}/R_{MS})^{3/2}(m/M)^{{1/6)}$, where m and M are the mass of the main-sequence star and the neutron star, respectively, $\gamma_{min}$ the minimum separation between two stars, and $R_{MS}$ the size of the main-sequence star. The material from the (at least partially) disrupted star forms a disk around the neutron star. If all material in the disk is to be acctreted onto the neutron star's surface, the mass of the disk is enough to spin up the neutron star to spin period of 1 ms.
The PTMSP/PDMS graft copolymer were synthesized from the PTMSP[poly(1-trimethylsilyl-1-propyne)] and the PDMS[poly(dimethylsiloxane)] and then the PTMSP/PDMS-borosilicate composite membranes were prepared by adding the porous borosilicates to the PTMSP/PDMS graft copolymer. The number-average molecular weight (${\bar{M}}_n$) and the weight-average molecular weight (${\bar{M}}_w$) of PTMSP/PDMS graft copolymer were 460,000 and 570,000 respectively, and glass transition temperature ($T_g$) of PTMSP/PDMS graft copolymer appeared at $33.53^{\circ}C$ according to DSC analysis. According to the TGA measurements, the addition of borosilicate to the PTMSP/PDMS graft copolymer leaded the decreased weight loss and the completed weight loss temperature went down. SEM observation showed that borosilicate was dispersed in the PTMSP/PDMS-borosilicate composite membranes with the size of $1{\sim}5{\mu}m$. Gas permeation experiment indicated that the addition of borosilicate to PTMSP/PDMS graft copolymer resulted in the increase in free volume, cavity and porosity resulting in the gradual shift of the mechanism of the gas permeation from solution diffusion to molecular sieving surface diffusion, and Knudsen diffusion. Consequently, the permeability of $H_2$ and $N_2$ increased and selectivity ($H_2/N_2$) decreased as the contents of borosilicate increased.

tRNA (m¹ G37) methyltransferase (TrmD) catalyze s the trans for of a methyl group from S-adenosyl-L-methionine (AdoMet) to G/sup 37/ within a subset of bacterial tRNA species, which have a residue G at 36th position. The modified guanosine is adjacent to and 3' of the anticodon and is essential for the maintenance of the correct reading frame during translation. We have determined the first crystal structure of TrmD from Haemophilus influenzae, as a binary complex with either AdoMet or S-adenosyl-L-homocysteine (AdoHcy), as a ternary complex with AdoHcy/phosphate, and as an apo form. The structure indicates that TrmD functions as a dimer (Figure 1). It also suggests the binding mode of G/sup 36/G/sup 37/ in the active site of TrmD and catalytic mechanism. The N-terminal domain has a trefoil knot, in which AdoMet or AdoHcy is bound in a novel, bent conformation. The C-terminal domain shows a structural similarity to DNA binding domain of trp or tot repressor. We propose a plausible model for the TrmD₂-tRNA₂ complex, which provides insights into recognition of the general tRNA structure by TrmD (Figure 2).

We have presented a classic two-ribbon filament eruption occurred in the east side of NOAA active region 11850 at 21:00 UT on 29 September 2013. Interestingly, this filament eruption was not accompanied by any flares and just there was a slight brightening in X-rays, C1.2, associated with the eruption. An accompanying huge CME was appeared at 22:12 UT in the LASCO C2 field of view and it propagates into the interplanetary space with a speed of about 440 km/s. And the related solar proton event (S2) started at 05:05 UT and peaked at 20:05 UT on 30 September 2013. The CME arrival was recorded by the ACE spacecraft around 01:30 UT on 2 October 2013. Around the CME arrival time, the solar-wind speed reached at about 640 km/s and IMF Bz showed southward component (-27 nT). Finally, the filament eruption and the CME cause geomagnetic storm (G2) at 03:00 UT on 2 October 2013. We described the detailed evolution of the filament eruption and its related phenomena such as CME, proton event, geomegnetic storm and so on. In addition, we will discuss about the activation mechanism of the filament eruption without flares.

The current study focuses on the assessment and interface response of reinforced concrete elements with composite materials (carbon fiber reinforced polymers-CFRPs, glass fiber reinforced polymers-GFRPs, textile reinforced mortars-TRM's, near surface mounted bars-NSMs). A description of the transfer mechanisms from concrete elements to the strengthening materials is conducted through analytical models based on failure modes: plate end interfacial debonding and intermediate flexural crack induced interfacial debonding. A database of 55 in total reinforced concrete columns (scale 1:1) is assembled containing elements rehabilitated with various techniques (29 wrapped with CFRP's, 5 wrapped with GFRP's, 4 containing NSM and 4 strengthened with TRM). The failure modes are discussed together with the performance level of each technique as well as the efficiency level in terms of ductility and bearing/ bending capacity. The analytical models' results are in acceptable agreement with the experimental data and can predict the failure modes. Despite the heterogeneity of the elements contained in the aforementioned database the results are of high interest and point out the need to incorporate the analytical expressions in design codes in order to predict the failure mechanisms and the limit states of bearing capacities of each technique.
