Purification and Characterization of Quercitrin-Hydrolyzing ${\alpha}$-L-Rhamnosidase from Fusobacterium K-60, a Human Intestinal Bacterium

  • An ${\alpha}$-L-rhamnosidase (EC 3.2.1.40.), which transforms quercitrin to quercetin, was purified from Fusobacterium K-60, a human intestinal anaerobic bacterium. The specific activity of the purified ${\alpha}$-L-rhamnosidase was 2.89 mol/min/mg protein. ${\alpha}$-L-Rhamnosidase, whose molecular size was 170 kDa by gel filtration, was composed of four subunits ($M_r$ 41,000 Da) with pI and optimal pH values of 5.2 and 5.5-7.0, respectively. The apparent $K_m$ and $V_{max}$ values for p-nitrophenyl-${\alpha}$-L-rhamnopyranoside and quercitrin were determined to be 0.057 mM and 3.4 mol/min/mg, and 0.077 mM and 5.0 mol/min/mg, respectively. This enzyme was strongly inhibited by $Cu^{2+},\;Mn^{2+}$, L-rhamnose, and p-chlormercuriphenylsulfonic acid. These findings suggest that the biochemical properties and substrate specificity of the purified enzyme are different from those of the previously purified ${\alpha}$-L-rhamnosidase. This is the first reported purification of quercitrin-hydrolyzing ${\alpha}$-L-rhamnosidase from intestinal bacteria.

Study on Film Boiling Heat Transfer of Spray Cooling in Air-Water Full Cone Spray System (물-공기 원추형 분무시스템에 있어서 분무냉각 막비등 열전달에 관한 연구)

  • The local heat flux of spray cooling in the film boiling region were experimentally investigated for the spray region of $D_{max}$ = $0.005{\sim}0.03m^3/(m^2s)$. A twin-fluid full cone spray nozzle was employed for the experiment and the distributions of droplet flow rates were obtained for air-water full cone sprays. A stainless steel block was cooled down from initial temperature of about $800^{\circ}C$ by full cone spray. In the region near the stagnation point, it was found that the experimental data are in good agreement with the results predicted from the correlations between the local heat transfer and the local droplet flow rate proposed in the previous report. However, it was found that the experimental data of $D_r$ > $0.01m^3/(m^2s)$ are a little smaller than the results predicted from the correlations.

Effect of Ba Stearate Addition on Magnetic Properties of Ba-system W-type Ferrite Magnets

  • An experiment was carried out to investigate the effect of Ba Stearate as a reducing agent on the magnetic and physical properties of anisotropic $BaFe_2-W$ type ferrite magnets. It was found that the magnetic properties of $BaO{\cdot}8.5Fe_2O_3$ were improved by adding 0.3 wt% of Ba Stearate, 0.5 wt% of $SiO_2$, and 0.5 wt% of CaO together. The optimum conditions for making magnets were as follows; semisintering condition: $1350^{\circ}C{\times}4.0$ h in nitrogen gas atmosphere, drying condition: $180^{\circ}C{\times}2.0$ h in air, sintering condition: $1160^{\circ}C{\times}1.5$ h in nitrogen gas atmosphere. Magnetic and physical properties of a typical sample were $J_m$ = 0.46 T, $J_r$ = 0.43 T, $H_{cJ}$ = 182.3 kA/m, $H_{cB}$ = 177.2 kA/m, $(BH)_{max}$ = 33.8 kJ/$m^3$, $T_C$ = $495^{\circ}C$ and $K_A$ = $2.65{\times}10^5\;J/m^3$ and $H_A$ = 1332 kA/m.

한국 여성의 생리학적 작업능력(PWC)에 관한 연구

  • 인간공학을 적용하는데 있어 가장 기본적으로 다루어져야 할 것은 인간의 신체적, 심리적능력에 관한 연구이다. 특히 산업 현장에서 흔히 이루어지는 인간의 힘을 그 동력원으로 하는 수작업의 경우에 있어 서는 작업자가 직업적인 요인에 의해 피로가 누적되거나 신체적인 무리를 가져오지 않고 수행할 수 있는 안전한 작업기준의 설정이 필요하며 이를 위해서는 인간의 신체적 능격에관한 연구가 필수적이다. 특히 최근 들어 여성의 사회진출 기회가 다양해지고 있으며 많은 여성들이 육체적 노동을 필요로 하는 직업에 종사하고 있는 현실에서 한국여성들의 신체적 능력에 관한 자료가 절대적으로 부족한 상황이다. 따라서 본 연구에서는 현재 한국의 여성들이 활동하고 육체적 작업조건들이 적절하게 설정되어 있는지를 분석하기 위한 기초 자료로서 한국 여성들의 생리학적 작업능력(PWC:physiological work capacity)을 최대산소소모 능력(Maximal Oxygen Consumption: V0$_{2}$Max)의 관점에서 파악하고자 하였다. 또한 외국 여성의 생리 학적 작업능력과 한구 여성의 생리학적 작업능력을 비교 평가하여한국 여성에게 외국여성의 기준치를 적용 시키는 것이 적절한지를 분석해 보았다. 본 연구는 피실험자에게 위험부담을 주지 않는 Submaximal test의 연속적인 방법(continuous mathod)중 하나인 램프 테스트(Ramp test)를 사용하여 심박수(H.R)와 산소 소모 량(V0$_{2}$) 측정하여 최대심박수에서의 PWC를 추정하였다. 작업 부하는 자전거 에르고노미터와 트레드 밀을 사용하여부과하였다. 실험 결과로서 자전거 에르고미터의 경우 치대산소 소모량의 추정치 평균이 1818.79ml/min으로 나타 났고 트레드밀의 경우 2076.33ml/min으로 나타났다. 이 결과는 자전거 에르고 노미터의 결과는 자전거 에르고노미터의 결과가 트레드밀의 결과에 87.60%정도 나타났다.

Design and implementation of electromagnetic band-gap embedded antenna for vehicle-to-everything communications in vehicular systems

  • We proposed a novel electromagnetic band-gap (EBG) cell-embedded antenna structure for reducing the interference that radiates at the antenna edge in wireless access in vehicular environment (WAVE) communication systems for vehicle-to-everything communications. To suppress the radiation of surface waves from the ground plane and vehicle, EBG cells were inserted between micropatch arrays. A simulation was also performed to determine the optimum EBG cell structure located above the ground plane in a conformal linear microstrip patch array antenna. The characteristics such as return loss, peak gain, and radiation patterns obtained using the fabricated EBG cell-embedded antenna were superior to those obtained without the EBG cells. A return loss of 35.14 dB, peak gain of 10.15 dBi at 80°, and improvement of 2.037 dB max at the field of view in the radiation beam patterns were obtained using the proposed WAVE antenna.

Kinetic Studies of Alkaline Protease from Bacillus licheniformis NCIM-2042

  • An extensive investigation was carried out to describe the kinetics of cell growth, substrate consumption, and product formation in the batch fermentation using starch as substrate. Evaluation of intrinsic kinetic parameters was carried out using a best-fit unstructured model. A nonlinear regression technique was applied for computational purpose. The Andrew's model showed a comparatively better $R^2$ value among all tested models. The values of specific growth rate (${\mu}_{max}$), saturation constant ($K_S$), inhibition constant ($K_I$), and $Y_{X/S}$ were found to be 0.109 $h^{-1}$, 11.1 g/l, 0.012 g/l, and 1.003, respectively. The Leudeking-Piret model was used to study the product formation kinetics and the process was found to be growth-associated. The growth-associated constant (${\alpha}$) for protease production was sensitive to substrate concentration. Its value was fairly constant up to a substrate concentration of 30.8 g/l, and then decreased.

  • In order to increase the cost effectiveness of solar cells, module production should be treated more comprehensively. Back contact cells offer distinct advantage in the interconnection of cells to modules. Thereby Mo thin film were prepared in order to clarify optimum conditions for growth of the thin film depending upon process, and then by changing a number of deposition conditions and substrate temperature conditions variously, structural and electrical characteristics were measured. For the manufacture of the Mo were vapor-deposited in the named order. Among them, Mo were vapor-deposited by using the sputtering method in consideration of their adhesive force to the substrate, and the DC power was controlled so that the composition of Mo, while the surface temperature having an effect on the quality of the thin film was changed from R.T$[^{\circ}C]$ to $200[^{\circ}C]$ at intervals of $50[^{\circ}C]$. Micro-structural studies were carried out by XRD (D/MAX-1200, Rigaku Co.) and SEM (JSM-5400, Jeol Co.). Electrical properties were measured by CMT-SR3000 Measurement System.

  • Barium Titanate ($BaTiO_3$) is one of the few titanateds which is cubic at room temperature. It has the perovskite structure, high dielectric constant (${\varepsilon}_r=300$) and a small temperature coefficient of resistance due to it's Low transition temperature ($Tc=120^{\circ}c$). PTCR (Positive Temperature Coefficient of Resistivity) thermistor in thin film $BaTiO_3$ system was prepared by using radio frequency (13.56MHz) and BC magnetron sputter equipment. Polycrystalline, and surface structure characteristics of the specimens were measured by X-ray diffraction (D-Max3, Rigaku, Japan), SEM(Scanning Electron Microscopy: M. JSM84 01, Japan), respectively. Temperature at below $600^{\circ}C$, $1000^{\circ}C$ to $700^{\circ}C$, and above $1100^{\circ}C$ for spotted $BaTiO_3$ thin films showed the amorphous, degree of crystal growth, and polycrystalline, respectively.

  • 초미세립 복합상 영구자석은 연 경자성상의 크기와 분포에 큰 영향을 받는 자성재료이다. 또한, 결정립간 교환상호작용력은 초미세립 복합상 영구자석의 잔류자속밀도에 큰 영향을 미친다. 이에 $Nd_8Fe_{86-x}Nb_xB_6$(x=0, 1, 2, 3) 조성으로 급냉자성 분말을 제조하고, 열처리 후 자기특성을 평가하였다. Nb 첨가에 따른 $Nd_2Fe_{14}B$ 상과 $\alpha$-Fe 상과의 교환상호작용력을 Henkel(${\delta}M$) plot을 통해 평가하고, 각 상의 크기와 분포를 조사하여 자기특성 변화를 합리화하였다. 본 연구에서 얻은 최대 자기특성은 $B_r$ = 9.791 kG, $_iH_c$ = 5.565 kOe, 그리고 $(BH)_{max}$ = 14.61 MGOe였고, Nb를 첨가함으로서 보자력이 증가하였다.

Measuring Preferences of University Students for Family Restaurants in the Eastern Part of Chonnam (전남 동부권 패밀리레스토랑에 대한 대학생들의 선호도 평가)

  • The purpose of this study was to identify the combinations of factors combinations conferring the highest utility of family restaurants to university students, and establish the relative importance of these factors in terms of their contribution to total utility. 196 of 200 questionnaires were utilized for the analysis. (Eds note: to whom were the questionnaires administered) Frequencies, crosstabs, and the conjoint, max. utility, BTL and Logit models, K-means cluster and one-way ANOVA analysesis, and the Friedman test were the statistical methods used for this study. The findings from this study were as follows: 1) the Pearson's R and Kendall's tau statistics (Eds note: these were not mentioned earlier) show that the model (Eds note: which model is this) fits the data well. 2) it was found that of all the respondents, especially the first and third clusters, regarded both the type of food and the price as very important factors. 3) it was found that all the respondents, especially the third cluster, most preferred a family restaurant (design and simulation) that provided less than 6 fusion and traditional foods. The first cluster most preferred family restaurant (design) that provided over 10 traditional and less than 6 ethnic foods. The second cluster most preferred a family restaurant (design and simulation) that provided over 10 traditional foods. 4) the results of the study have provided some insights into the effective types of family restaurant designs that can be successfully developed by those who manage menu variety, quality and type of food, price, and quality of service to university students dining at family restaurants.
