LIFETIME PRODUCTION PERFORMANCE OF HOLSTEIN FRIESIAN × SAHIWAL CROSSBREDS

  • The performance records of 410 Holstein Friesian crossbred cows belonging to seven genetic groups (Fl, 3/4, 1/4, 5/8, 3/8, triple cross and miscellaneous cross) maintained at Livestock Production Research Institute, Bahadurnagar, Okara were analyzed for various parameters of lifetime traits. For the analysis 2 data sets were made. Data set I included all the cows disposed off from the herd which have completed at least one lactation while for data set II performance traits for only first five lactations were considered. The data was analyzed by Mixed Model Least squares and Maximum Likelihood computer programme PC-I version. The least squares means ${\times}$ standard errors for data set I (periods are in days and milk yield is in litres) were $994.5{\pm}15.5$, $1,877.0{\pm}70.9$, $1,651.9{\pm}19.3$, $2,533.7{\pm}36.5$, $3,530.0{\pm}40.5$, $15,785.2{\pm}320.0$, $8.46{\pm}0.19$, $5.66{\pm}0.16$ and $3.79{\pm}0.08$, respectively for age at first calving (APC), Ist lactation milk yield (FLMY), productive life (PL), herd life (HL), total life (TL), lifetime milk yield (LTMY), milk yield per day of productive life (MY/PL), milk yield per day of herd life (MY/HL) and milk yield per day of total life (MY/TL). For data set II these values were $1,004.2{\pm}21.2$, $2,220.5{\pm}113.1$, $1,429.1{\pm}40.8$, $2,302.1{\pm}73.3$, $3,307.2{\pm}77.3$, $13,189.7{\pm}667.4$, $9.10{\pm}0.34$, $5.66{\pm}0.25$ and $4.02{\pm}0.18$ in the same order. For data set I the effect of year of first calving was significant for AFC, FLMY, PL, HL, LTMY and MY/PL. The season of Ist calving was significant only for MY/PL. The effect of genetic group was significant for AFC, FLMY, MY/PL and MY/TL while the effect of parity was significant for all the traits. For data set II the effect of year of Ist calving was significant only for AFC, FLMY and PL while the season of Ist calving was significant for FLMY and PL while the effect of genetic groups was significant for MY/HL only. The lifetime production performance is in general close to the various estimates reported in the literature.

Effects of EGF, $\beta-ME$, Glucose, $O_2$ Concentrations and Fibroblasts Subculture on the Development of Porcine NT Embryos (EGF, $\beta-ME$, Glucose, $O_2$ 농도 및 Fibroblast Subculture가 핵이식 배의 체외발생에 미치는 영향에 관한 연구)

  • 본 연구는 NCSU-23과 PZM-3 배양액에 EGF, $\beta-ME$와 glucose의 첨가가 돼지 난자의 체외성숙에 미치는 영향과 배양조건을 다르게 하여 계대배양한 섬유아세포를 이용한 핵이식배를 다른 배양액과 산소조건에서 배양하였을 때 체외발생율에 미치는 영향을 조사하였다. 핵이식 배를 20ng/ml EGF를 첨가 또는 첨가하지 않은 NCSU-23 및 PZM-3 배양액에서 배양하였을 때 배반포로의 체외 발생율은 각각 $12.0\pm1.3\%,\;9.6\pm1.9\%,\;10.9\pm2.1\%,\;9.1\pm2.3\%$였다. 핵이식 배를 $25{\mu}M\;\beta-ME$를 첨가 또는 첨가하지 않은 NCSU-23 및 PZM-3 배양액에서 144시간 배양하였을 때 배반포로의 체외 발생율은 각각 $9.6\pm1.7\%,\;7.3\pm2.3\%,\;11.9\pm1.8\%$$7.4\pm2.1\%$였다. $\beta-ME$를 첨가한 PZM-3 배양액에서 배양하였을 때 배반포로의 체외 발생율은 $\beta-ME$를 첨가하지 않은 배양액에서 배양한 배보다 높은 체외발생율을 나타냈다. (p<0.05). 핵이식 배를 1.5mM glucose를 첨가 또는 첨가하지 않은 NCSU-23 및 PZM-3 배양액에서 배양하였을 때 배반포로의 체외 발생율은 각각 $9.4\pm2.2\%,\;6.8\pm2.7\%,\;10.9\pm2.4\%$$8.9\pm2.6\%$였다. Glucose를 첨가한 NCSU-23과 PZM-3 배양액에서 배양하였을 때 배반포로의 체외 발생율은 glucose를 첨가하지 않은 배양액에서 배양한 배보다 높은 체외 발생율을 나타냈다. 핵이식 배를 NCSU-23 및 PZM-3 배양액과 $5\%$$20\%$ 산소 조건에서 배양하였을 때 배반포로의 체외 발생율은 각각 $11.1\pm1.8\%,\;9.8\pm1.4\%,\;12.5\pm1.6\%$$10.9\pml.5\%$였다. NUSU-23과 PZM-3 배양액에서 $5\%$ 산소 조건에서 배양하였을 때 $20\%$산소 조건에서 배양한 난자보다 높은 체외 발생율을 나타났다. 섬유아세포를 NCSU-23 배양액에서 배양하여 공여자세포로 이용하여 10 및 $11\~15\;passage$ 이내로 계대배양하였을 때의 융합율은 $60.0\~73.3\%,\;52.5\%$였다. 섬유아세포를 PZM-3 배양액에서 배양하여 공여자세포로 이용하여 10 및 $11\~15passage$ 이내의 계대배양시의 융합율은 $60.4\~75.1\%$$58.7\%$였다.

Monitoring of Particulate Matter Concentration for Forage Crop Cultivation during Winter Season in Saemangeum (새만금 내 동계 사료작물 재배에 따른 미세먼지 농도 변화 모니터링)

  • The Saemangeum has a dry surface characteristic with a low moisture content ratio due to the saline and silt soil, so the vegetation cover is low compared to other areas. In areas with low vegetation cover, wind erosion has a high probability of scattering dust. If the vegetation cover is increased by cultivating crops that can withstand the Saemangeum reclaimed environment, scattering dust can be reduced by reducing the flow rate at the bottom. Thus, the purpose of this study is to analyze the effect of suppressing the generation of fine dust and scattering dust by cultivating winter forage crops on the Saemangeum reclaimed land. While growing 0.5 ha of barley and 0.5 ha of triticale in Saemangeum reclaimed land, the concentration of fine dust was monitored according to agricultural work and growth stage. Changes in the concentrations of PM-10, PM-2.5, and PM-1.0 were monitored on the leeward, the windward and centering on the crop field. As a result of monitoring, PM-1.0 had little effect on crop cultivation. the concentration of PM-10 and PM-2.5 increased according to tillage and harvesting, and tillage had a higher increasing the concentration of PM-10 and PM-2.5 than that of harvesting. According to the growth stage of crops, the effect of suppressing scattering dust was shown, and the effect of suppressing scattering dust was higher in the heading stage than in the seedling stage. So, it was found that there was an effect of suppressing scattering dust other than the effect of land covering. Through this study, it was possible to know about the generation and suppression effect of scattering dust according to crop cultivation.

The Effects of Nitric Oxide Inhibitor on Hyperdynamic Circulation in Portal Hypertensive Rats (산화 질소 억제제가 문맥 고혈압 쥐의 혈역학 변화에 미치는 영향)

  • Background: Nitric oxide, a vasodilator synthesized from L-arginine by vascular endothelial cells, accounts for the biological activity of endothelium derived relaxing factor. Previous studies demonstrated that nitric oxide inhibitor, $N^{\omega}$-Nitro-L-Arginine(NNA) diminished the hyperdynamic splanchnic and systemic circulation in portal hypertensive rats The present study was done to determine the role of nitric oxide in the development of hyperdynamic circulations in the prehepatic portal hypertensive rat model produced by partial portal vein ligation. Methods: The portal hypertensive rats were divided into water ingestion group and NNA ingestion group. After partial portal vein ligation, NNA ingestion group and water ingestion group received NNA 1mg/kg/day and plain water through the mouth for 14 days, respectively. Cardiac output, mean arterial pressure, organ blood flow and porto-systemic shunting were measured by radioisotope labeled microsphere methods. Vascular resistances were calculated by standard equation. Results: There were significant decreases in mean arterial pressure, increases in cardiac output and cardiac index, and decreases in total systemic and splanchnic vascular resistance in portal hypertensive rats compared to normal control group (p<0.01). Compared to the water ingestion group, significantly increased mean arterial pressure with decreased cardiac output and cardiac index were developed in the NNA ingestion group. Total systemic and splanchnic vascular resistance were significantly increased in the NNA ingestion group compared to water ingestion group (p<0.05). But, there was no significant difference in portal pressure between the two groups. Conclusion: The hemodynamic results of this study indicate that hyperdynamic circulation in prehepatic portal hypertensive rat mode1 was attenuated by ingestion of NNA. Nitric oxide may play an important role in the development of hyperdynamic circulation with splanchnic vasodilation in chronic portal hypertension.

  • In this paper, we measured the biosignal using hot-water system(STYX ford202) on foot. The biosignal transition data is observed from hot-water thermotherapy on foot. pre and post demonstration conditions under 43$^{\circ}C$ and 45$^{\circ}C$ are checked about 9 physiological factors for 10 persons and 10 days. (Checking Time: pre-test, post-test(5, 10, 15, 20 minutes)). The biosignal transition of demonstration's results showed as belows; Forehead Temperature($^{\circ}C$): -0.69 $\pm$ 0.01 dec, Leg Temperature($^{\circ}C$): 1.51 $\pm$ 0.22 inc, Blood Flow($m\ell/min$): 1.18 $\pm$ 0.50 inc, Blood Pressure(mmHg): (max) -1.49$\pm$ 2.81, (min) -0.06 $\pm$ 0.13 dec, Heart Rate(bpm): 6.97 $\pm$ 0.72 inc, Blood Sugar($mg/d\ell$) : -2.41 $\pm$ 1.55 dec, Oxygen Saturation(%): 1.34 $\pm$ 0.28 inc, Body Fat(%) -1.75 $\pm$ 0.15 dec, Weight(kg): -0.10 $\pm$ 0.04 dec. Effects of temperature on pharmacokinetics of oxolinic acid in black rockfish, Sebastes schlegeli following oral administration (조피볼락, Sebastes schlegeli에 경구투여된 oxolinic acid의 약물동태에 미치는 수온의 영향)

  • Effects of temperature ($13{\pm}1.5^{\circ}C$, $23{\pm}1.5^{\circ}C$) on the pharmacokinetic properties of oxolinic acid (OA) were studied after oral administration to cultured black rockfish, Sebastes schlegeli. Serum concentrations of OA were determined using HPLC-UV detector after a single dosage of 60 mg/kg body weight (average about 500 g). The peak serum concentrations of OA at $23{\pm}1.5^{\circ}C$ and $13{\pm}1.5^{\circ}C$ were $0.60{\mu}/ml$ at 30 h and $2.22{\mu}g/ml$ at 10 h post-dose, respectively. Better absorption of OA was noted at $13{\pm}1.5^{\circ}C$ compared to $23{\pm}1.5^{\circ}C$. The elimination of OA from serum was considerably faster at $23{\pm}1.5^{\circ}C$ than at $13{\pm}1.5^{\circ}C$. Both absorption and elimination of OA were affected significantly by temperature. The kinetic profile of absorption, distribution and elimination of OA in serum was analyzed by fitting to a two compartment model, with WinNonlin program. The AUC, Tmax and Cmax at $23{\pm}1.5^{\circ}C$ were $42.16{\mu}g{\cdot}h/m\ell$, 26.13 h and $0.43{\mu}g/ml$, respectively. The AUC, Tmax and Cmax at $13{\pm}1.5^{\circ}C$ were $131.98{\mu}g{\cdot}h/ml$, 8.81 h and $2.04{\mu}g/ml$, respectively.

Studies on the Serum Total Activities and Isoenzyme PAtterns of LDH in Non-Human Primates Reared in Korea (國內詞育 원숭이의 血淸 LDH의 總活性値와 isoenzyme에 관한 硏究)

  • Non-human primates have been increasing in demand as important experimental animals and companion animals, domestically and internationally. The number of non-human primates for these purposes will be much enhanced in the near future. Despite this trend, basic physiological data are scarcely available in these animal species, leading to the difficulty to diagnose diseases when necessary, due to the absence of reference values. Particularly, there is not any report on the total activity of LDH of non-human primates, let alone LDH isoenzyme patterns, in Korea. LDH isoenzymes have a high level of efficaciousness as diagnostic and prognostic aids in various diseases. In this study, total activities and isoenzyme patterns of LDH were measured to obtain their reference values in domestically reared common marmosets, crab-eating macaques and Japanese macaques. There were widespread different values of serum total LDH among the non-human primate species experimented in this study. Serum LDH values of common marmosets and crab-eating macaques were 597.5$\pm$243.1 IU/l and 605.3$\pm$312.6 IU/l, respectively, whereas those of Japanese macaque showed 1,209$\pm$473.8 IU/l. Five isoenzyme fractions of LDH were observed in all experimented non-human primates but their ranks and proportions represented different patterns one another. In common marmosets, the percent of fraction for serum LDH1, LDH$_2$, LDH$_3$, LDH$_4$, and LDH$_{5}$ was 13.7$\pm$6.4%, 23.3$\pm$3.6%, 29.2$\pm$5.0%, 9.4$\pm$1.4% and 24.4$\pm$7.5%, respectively. The rank of LDH isoenzymes was LDH$_3$>LDH$_{5}$>LDH$_2$>LDH$_1$>LDH$_4$, in the descending order. For crab-eating macaques, the fraction of serum LDH$_1$, LDH$_2$, LDH$_3$, LDH$_4$, and LDH$_{5}$ occupied 19.5$\pm$12.7%, 25.3$\pm$9.3%, 23.8$\pm$8.1%, 10.2$\pm$2.8% and 21.3$\pm$14.2%, respectively. The order of LDH isoenzymes was LDH$_2$>LDH$_3$>LDH$_{5}$>LDH$_1$>LDH$_4$, from top to down. On the while, in Japanese macaques, the fraction of serum LDH$_1$ to LDH$_{5}$ showed 23.4$\pm$11.8%, 30.5$\pm$4.1%, 17.4$\pm$3.9%, 11.3$\pm$3.7% and 13.8$\pm$5.6%, respectively. The decreasing order indicated LDH$_2$>LDH$_1$>LDH$_3$>LDH$_{5}$>LDH$_4$. In conclusion, values such as LDH and LDH isoenzyme patterns of investigated for the first time from non-human primates reaared in Korea, could be reference values for the optimal diagnosis and therapy of diseases of the corresponding animal species. Other parameters of hematology and blood biochemistry are urgently needed to study for the benefit of our intimate non-human primates.an primates.

Change of the Size-Resolved Aerosol Concentration Due to Relative Humidity (습도 변화에 따른 에어로졸의 농도 및 크기의 변화경향 파악)

  • In this study, the atmospheric aerosol concentration measured at different relative humidity levels was analyzed. Using an optical particle counter, PM10 and PM2.5 concentration as well as particle size distribution were measured and the relation between these size resolved data and relative humidity was studied. The results showed that mass concentration increases as relative humidity increases. The comparison between PM1, PM2.5 and PM10 showed that the fine particles grow more than coarse particles as relative humidity increases. The results also showed that PM10-2.5 and relative humidity do not show close correlation, which means that the mass increase of PM10 at high relative humidity is mainly due to the growth of PM2.5.

Yield and Quality Characteristics of Ginseng's First Byproducts (인삼 1차 부산물의 생산량 및 기능성 성분 특성)

  • This study was carried out to utilize the byproducts (flower, immature and mature berry, leaf and stem) of ginseng. Yield of byproducts were $32.7{\pm}9.8g$ in flower, $68.2{\pm}2.2g$ in immature berry, $48.5{\pm}4.3g$ in mature berry, $316.2{\pm}20.5g$ in leaf, and $296.6{\pm}15.4g$ in stem per $3.3m^2$ ($180{\times}90cm$, ginseng root $675.5{\pm}35.7g$/drybasis. The total saponin contents of ginseng byproducts and root are $52.36{\pm}1.24$, $68.71{\pm}1.98$, $168.89{\pm}0.57$, $68.26{\pm}1.32$, $7.85{\pm}0.61$ and $35.08{\pm}0.96$ mg/g, respectively. The main ginsenoside of all byproducts was Re and the highest content was $132.23{\pm}1.56$ mg/g in mature berry. But flower and berry was not detected Rf and Rh1, respectively. Total polyphenolic compound content on mature berry was the highest, $2.242{\pm}0.140%$, after, immature berry > leaf > flower > root > stem order. The DPPH radical scavenging activity on mature berry was the highest, $0.115{\pm}0.004$ mg/mL($IC_{50}$), and the others were the same order of polyphenolic compound and ginsenoside content on byproducts.

Long-Term Effects of ACE Inhibitors in Post-Tuberculosis Emphysema

  • Background: Little is known about the long-term effects of angiotensin-converting enzyme (ACE) treatment on post-tuberculosis emphysema. This study evaluated the effects of ACE inhibition on cardiac function and gas exchange in patients with post-tuberculosis emphysema. Methods: At baseline and at 6 months after initiation of ACE inhibition therapy, patients underwent pulmonary function testing, arterial blood gas analysis, and echocardiography, both at rest and post exercise. Cardiac output (CO) and right ventricular ejection fraction (RVEF) were measured at those time points as well. Results: After ACE inhibition; resting and post-exercise RVEF ($Mean{\pm}SEM,\;61.5{\pm}1.0,\;67.6{\pm}1.2%$, respectively) were higher than at baseline ($56.9{\pm}1.2,\;53.5{\pm}1.7%$). Resting and post-exercise CO ($6.37{\pm}0.24,\;8.27{\pm}0.34L/min$) were higher than at baseline ($5.42{\pm}0.22,\;6.72{\pm}0.24L/min$). Resting and post-exercise $PaO_2$ ($83.8{\pm}1.6,\;74.0{\pm}1.2mmHg$, respectively) were also higher than at baseline ($74.2{\pm}1.9,\;66.6{\pm}1.6mmHg$). Post-exercise $PaCO_2$($46.3{\pm}1.1mmHg$) was higher than at baseline ($44.9{\pm}1.1;\; Resting\;42.8{\pm}0.8\;vs.\;42.4{\pm}0.9mmHg$). Resting and post-exercise A-a $O_2$ gradient ($12.4{\pm}1.4,\;17.8{\pm}1.5 mmHg$) were lower than at baseline ($22.5{\pm}1.5,\;26.9{\pm}1.6mmHg$). Conclusion: In post-tuberculosis emphysema, RVEF and CO were augmented with a resultant increase in peripheral oxygen delivery after ACE inhibition. These findings suggest that an ACE inhibitor may have the potential to alleviate co-morbid cardiac conditions and benefit the patients with post-tuberculosis emphysema.
