방사선조사가 백서 타액선의 clusterin 발현에 미치는 영향 (Effect of irradiation on expression of clusterin in the rat salivary glands)

  • Purpose : To investigate clusterin expression in the acini and ductal cells of rat submandibular glands after Co-60 gamma irradiation. Materials and Methods : The male Sprague-Dawley rats weighing approximately 250 gm were divided into control and experimental groups. The experimental group was irradiated with a single absorbed dose of 2, 5, 10, and 15 Gy on the head and neck region. All the rats were sacrificed at 1, 3, 7, 14, 21, and 28 days after irradiation. The specimens including the submandibular gland were sectioned and observed using a immunohistochemical method. Results : In the 2 Gy group, clusterin expression was similar to that of the control group at 1 day after irradiation and it was observed in the striated ductal cells at 3 days after irradiation. In the 5 Gy group, clusterin expression was observed in the striated ductal cells at 1 day after irradiation and gradually increased in the 10 and 15 Gy groups. In the 15 Gy group, clusterin expression was prominent in the striated ductal cells at 1 day after irradiation, but it gradually decreased with the experimental period. The destruction of the striated ductal cells was observed in the 2 Gy group at 21 days after irradiation and in the 5, 10, and 15 Gy groups at 7 days after irradiation. The destruction of the acinar cells was observed in the 2 Gy group at 28 days after irradiation and in the 5, 10, and 15 Gy groups at 14 days after irradiation. Conclusion : Clusterin expression was induced by low doses of irradiation and it appeared to be involved in the regulation of cellular response to irradiation.

열플라즈마에 의한 CFC의 분해공정 (Decomposition Process of CFC by Thermal Plasma)

  • 환경문제와 관련하여 오존층 파괴의 주요 원인으로 알려진 CFC를 열플라즈마를 이용하여 완전하게 분해하였다. CFC113($C_2Cl_3F_3$)을 선정하여 열플라즈마 분해에서의 적절한 공정 조건을 검토하였다. 실험에 앞서, 상압에서 300 K~5000 K범위에서 CFC113, $H_2$, $O_2$간의 열역학적 화학평형조성을 고찰함으로써 CFC113의 분해 생성의 경향을 알 수 있었다. 실험은 상압, 상온에서 CFC113과 $H_2$, $O_2$혼합가스의 주입량, 그리고 냉각관 직경의 변화에 따른 분해생성물을 조사하였고 이를 기체크로마토그래피로 분석하였다. 그 결과, 각각 99.99%이상의 분해율을 보였다. CFC113/$H_2$=1/3에서 $O_2$비가 증가할수록 CO로의 전화율은 감소하였다. CFC113/$O_2$=1/1, 1/1.5, 1/2에서 $H_2$비가 3이상 증가될수록 CO로의 전화율이 증가하였다. 그 이유는 $H_2$첨가가 증가할수록 환원분위기에서 $H_2O$가 생성되고 $CO_2$생성량이 감소하기 때문이다. DC power가 증가하여도 CO로의 전화율 변화는 차이가 없었으며, 총유량이 증가할 경우 CO전화율이 약간 감소하는 경향을 보였다. 냉각관의 직경을 8 mm에서 4 mm로 작게 할 경우 빠른 냉각속도로 인하여 CO로의 전화율이 증가하였다.

Ethylene Oxide 처리와 Gamma가 조사가 건조 농산물의 품질에 미치는 영향 (Effects of Ethylene Oxide Fumigation and Gamma Irradiation on the Quality of Dried Agricultural Products)

  • 주요 농산 건조가공품에 오염된 미생물의 효과적인 살균법 개발을 목적으로, 현행방법인 ethylene oxide 훈증처리와 감마선 조사에 따른 미생물의 살균효과와 이화학적 품질에 미치는 영향을 검토한 결과는 다음과 같다. 시료의 미생물 오염도는 고추장 분말, 참깨, 느타리버섯 분말, 당근 flake의 순이었으며, 전세균이 $4.6{\times}10^{3}/g~1.5{\times}10^{6}/g$, 내열성 세균이 $1.3{\times}10^{3}/g~1.4{\times}10^{6}/g$, 산저항성 세균이 $8.4{\times}10^{2}.g~8.8{\times}10^{5}/g$, 내삼투압성 곰팡이를 포함한 곰팡이류는 $4.0{\times}10^{2}/g~1.5{\times}104/g$, 대장균군은 당근 flake를 제외한 나머지 시료에서 $4.0{\times}10^{3}/g~2.7{\times}10^{5}/g$ 정도로 오염되어 있어 위생적인 살균처리가 요구된다. 살균효과 실험에서는 대부분의 시료가 7~10 kGy의 감마선 조사로서 모든 미생물의 사멸이 가능하였으나 ethylene oxide 처리구에서는 전반적으로 전세균과 내열성 포자균 및 곰팡이의 살균이 불충분하였다. Ethylene oxide 처리는 시료의 색소, 아미노산, 무기질, 색도 등에 상당한 영향을 미쳤으나 적정선량의 감마선 조사는 이들에 대해 비교적 안전하였으며, 실온에서 3개월 동안 저장된 시료의 전반적인 기호도는 감마선 조사구가 ethylene oxide 처리구나 대조구보다 우수하게 나타났다.

2차세공이 형성된 모더나이트와 알루미나를 혼합한 촉매상에서 Triphenylmethane의 분해반응 (Catalytic Cracking of Triphenylmethane on Alumina Mixed with Mordenite Formed Secondary Pore)

  • 수소형 모더나이트를 불화수소산처리하여 실리카/알루미나의 비가 다른 변형된 모더나이트를 만들고 이를 $\gamma$-알루미나와 혼합한 촉매를 제조하여 미소반응기에서 triphenylmethane (TPM)의 분해반응을 실시하였다. 미세공 구조인 수소형 모더나이트를 불화수소산처리로 탈알루미늄하면 산량은 감소하지만 2차 세공인 중세공이 형성되어 TPM의 분해활성과 2차 분해생성물인 벤젠의 선택성이 향상되었다. 그러나 더욱 불화수소산처리된 모더나이트는 산량 감소와 일부 세공구조의 파괴에 의한 미세공막힘 현상에 의해 분해활성이 저하되었다. 따라서 전처리된 모더나이트상에서 TPM의 분해활성과 벤젠의 선택성은 모더나이트의 실리카/알루미나 비가 17 정도에서 가장 우수하였다. 이들 전처리된 모더나이트를 알루미나와 혼합한 촉매는 순수한 모더나이트 촉매보다 TPM의 분해활성이 더욱 우수하였다. 이는 알루미나의 큰 중세공에서 TPM이 1차 분해반응하고 다음에 모더나이트의 중세공과 미세공에서 분해되는 단계적인 분해반응이 일어나기 때문이다.

Step-wise Anion-Exchange in Layered Double Hydroxide Using Solvothermal Treatment

  • Synthetic layered double hydroxides (LDHs), [$Mg_4Al_2(OH)_{12}]CO_3{\cdot}nH_2O$, were prepared in the submicron size of plate-like polycrystals. Anion-exchange reactions with various linear dicarboxylic acids were performed to produce LDH/organic hybrid materials by solvothermal treatment in toluene. X-ray powder diffraction spectra for the products indicated that the interlayer spacings of LDHs remarkably changed, up to 20 $\AA$ when 1,10-decanedicarboxylic acid anions were intercalated as an organic guest. Dicarboxylates-LDHs samples could be also re-exchanged consecutively with other dicarboxylic acids or carbonate without serious destruction of layer structure under the scanning electron microscopic observation.

Chlorophenol류의 세포화학적 독성에 관한 연구 (Studies on Cytochemical Toxicities of Chlorophenols to the Rat)

  • Chlorination of the polluted water may produce odoriferous and objectionable-tasting chlorophenols which are hazardous to health. These studies were undertaken to investigate the hazardous effects of chlorophenols to the rat. 1. The chlorophenols such as o-chlorophenol and 2,6-dichlorophenol inhibited rat growth and caused increment of the ratio between liver weight and body weight. 2. The hemoglobin content, hamatocrit ratio and A/G of rat blood were decreased by chlorophenols administration. The activities of alkaline phosphatase, lactic dehydrogenase (LDH) and glutamate oxaloacetate transaminase (GOT) in serum as well as in liver were increased provisonally and decreased after one or two weeks adminstration. 3. The liver mitochondrial respiration ($QO_{2}$) was inhibited by chlorophenols treatment in in-vivo and in-vitro test. 4. The liver microsomal cytochrome P-450 was decreased by chlorophenols administration 5. Liver tissue was degenerated with congestion, atrophy, swelling, vacuolation, dilation of rough endoplasmic reticulum and denature of mitochondrial particle with swelling, and cristal destruction by chlorophenols adminstration. 6. After one and two weeks of adminstration of chlorophenols to rat, the aberrations of bone marrow chromosome and inhibition of its mitosis were observed respectively.

Structural and Morphological Alterations of Candida albicans Cells after Treatment with Atratoxin $B_1$ from Holothuria atra (Jaeger)

  • Atratoxin $B_1$ which was found to inhibit the growth of Candida albicans caused structural and morphological alteration of the cells. Increased accumulation of vesicles and membranous bodies in the cytoplasm, and alterations of the cell membrane and cell wall were most obvious. Sequential lytic events of the cells eventually resulted in complete disintegration of the cytoplasmic structures. These results suggested that atratoxin $B_1$ functioned by either blocking the biosynthetic step during cell wall synthesis, altering cell wall metabolism or dissolution of the cell organelles. These changes caused a progressive destruction of the cell wall leading to cell lysis.

  • 광촉매 반응이 자연유기물에 의한 나노여과막의 오염에 미치는 영향을 살펴보았다. 광촉매 분해공정은 자연유기물의 분해와 변형에 효율적이었으며 이산화티타늄과 고정화 비드를 광촉매로 사용하였다. 광촉매적 특성을 비교하기 위하여 칼슘 이온 존재 시의 휴민산의 광분해를 모델 반응으로 설정하였다. 광분해 전에는 치밀한 막오염층이 형성되어 막오염을 가속화시킨 반면, 광분해 후에는 막오염이 크게 감소하였다.

  • Dielectric thin films of Pb$\_$0.72/La$\_$0.28/Ti$\_$0.93/O$_3$(PLT(28)) have been deposited on Pt(111)/Ti/SiO$_2$/Si(100) substrates in-situ by pulsed laser deposition using different annealing and deposition processes. We have investigated the effect of hydrogen annealing on the ferroelectric properties of PLT thin films and found that the annealing process causes the diffusion of hydrogen into the ferroelectric film resulting in the destruction of polarization. Two-step process to grow PLT films was adopted and verified to be useful to enlarge the grain size of the film. Structural properties including dielectric constant, and ferroelectric characteristics of PLT thin films were shown to be strongly influenced by grain size. The film deposited by using two-step process including pre-annealing treatment has a strong (111) orientation. However, the films deposited by using single-step process with hydrogen annealing process shows the smallest grain size.

  • Extract of Acanthopanax senticosus has recently been demonstrated to possess significant antidiabetic potential, in accordance with the traditional use of this plant as an antidiabetic natural health product. The present study evaluated the effects of fermented extract (FE) of this plant on glucose-stimulated insulin secretion, glucose uptake, and streptozotocin-induced type 1 diabetes model. A 3 h pretreatment with FE prevented $IL-1{\beta}$ and $IFN-{\gamma}$ toxicity in isolated rat islets. However, it did not affect insulin-stimulated glucose uptake in C2C12 myotubes. In addition, pretreatment of mice with FE blocked the destruction of streptozotocin-induced islets and the development of type 1 diabetes. FE reduced blood glucose level, increased insulin secretion, and improved glucose tolerance in streptozotocin-treated mice, whereas nonfermented extract (NFE) had moderate effects. Immunohistochemical staining for insulin clearly showed that pretreatment with FE blocked the STZ-induced islets destruction and restored the number of islet cells that secreted insulin to the level of the control. Although the active principles and their mechanisms of action remain to be identified, FE may nevertheless represent a novel complementary therapy and a source of novel therapeutic agents against type 1 diabetes mellitus.
