Fractionation and Availability of Cu and Zn in Paddy Soils Following a Long-Term Applications of Soil Amendments (토양개량제를 장기연용한 논토양에서 구리와 아연의 분획화 및 유효도)

  • We investigated the effects of a long term application of soil amendments such as lime, silicate, compost, or combinations of these materials on the contents of Cu and Zn in paddy soil and brown rice. The results obtained from a sequential extraction of Cu and Zn in paddy soils and brown rice, using $H_2O$, $KNO_3$, $Na_2$-EDTA and $HNO_3$ and showed that the most of Cu and Zn were NaOH-extractable (organically bound form) and $HNO_3$-extractable (sulfide and residual form). Cu contents of NaOH and EDTA extractable increased with the long term application of compost while the contents of Zn extracted by $KNO_3$ was decreased even though $HNO_3$ extractable-Zn was prominent chemical form in paddy soils. The percentage and contents of Zn, extracted by $KNO_3$ for each combination treatment of soil amendments, was decreased but the contents of Cu was not affected. The content of NaOH extractable-Cu was proportionally increased with increase in organic matter content irrespective of the extractants used in this experiment. The contents of Zn and exchangeable K were also increased with increase in organic matter content. However, we could not find any relationship between the extractable forms of Cu and Zn, and CEC, OM. while increase in CEC, contents of cations, and organic matter decreased the content of Cu in brown rice.

Inabenfide-Induced Alleviation of Salt Stress in Rice as Linked to Changes in Salicylic Acid Content and Catalase Activity

  • The effect of inabenfide was investigated in rice seedlings subjected to salt stress in relation to changes in chlorophyll fluorescence(${\Delta}F/Fm'$), lipid peroxidation, salicylic acid(SA) content, and catalase(CAT) activity. A reduction in shoot growth of rice seedlings by 120 mM NaCl treatment was significantly alleviated by pretreatment with 30 ${\mu}M$ inabenfide. Sodium ion content was not affected by pretreatment with inabenfide, suggesting that alleviation was not due to a reduction in sodium ion uptake by the rice seedlings. At three days after NaCl treatment, the rice seedlings pretreated with inabenfide showed a higher ${\Delta}F/Fm'$(30%) and lower lipid peroxidation(28%) compared with the rice seedlings treated with NaCl alone. After NaCl treatment, CAT activity in the third leaf of rice seedlings decreased significantly but alleviated by pretreatment with inabenfide. Furthermore, pretreatment with inabenfide also reduced the level of SA which accumulated drastically in the third leaf of rice seedlings within a day after exposure to salt stress. These results suggest that inabenfide prevents SA accumulation in rice seedlings under salt stress which eventually induces the alleviation of salt stress damage.

  • In this study, PTMSP[poly(1-trimethylsilyl-1-propyne)]/PDMS[poly(dimethylsioxane)]-NaY zeolite and PTMSP/PDMS-NaA zeolite composite membranes were made to incorporate zeolite into PTMSP/PDMS graft copolymer in order to improve the selectivity and thermal stability, the drop of permeability by physical aging effect during long period of time for the PTMSP membrane. To investigate the physico-chemical characteristics of composite membranes, the analytical methods such as FT-IR, $^1H$-NMR, TGA, SEM, and GPC have been utilized. The gas permeability and selectivity properties of $H_2$ and $N_2$ were evaluated. The permeability of the PTMSP/PDMS-NaY zeolite and PTMSP/PDMS-NaA zeolite composite membranes than PTMSP/PDMS graft copolymer membrane increased, increased as zeolite content increased. On the contrary, the selectivity ($H_2/N_2$) of the composite membranes decreased as zeolite content increased. PTMSP/PDMS-NaA zeolite composite membrane showed better permeability and separation factor than PTMSP/PDMS-NaY zeolite composite membrane.

Salinity Tolerance of Progenies between Korean Cultivars and IRRI's New Plant Type Lines in Rice

  • To select new germplasm for salinity tolerance from new plant type (NPT) breeding lines, the sixty F$_4$ lines selected from the crosses between Korean cultivars and IRRI's NPT lines were evaluated for salinity tolerance at the seedling stage with salinized culture solution (EC=12 dS/m) in the controlled conditions. Two NTP lines derived from a cross between 'Ilmibyeo' and 'IR66152-AC5-1', 'HR15258-7-1' and 'HR15258-27-1', were found to have good tolerance. The salinity tolerance of the lines was compared to their parents and the sensitive ('IR29') and tolerant ('Pokkali') checks in three salinity levels, no salinity (control) and an EC of 12 and 16 dS/m. Visual salinity score, shoot Na+ and Na-K ratio in two NPT lines was significantly low compared with the parents and IR29. Indicating that salinity tolerance of the lines might be derived from a transgressive segregation. The relative water content of the lines was higher than Pokkali, and the dry weight of shoot and root was proportionally decreased to salinity score and salinizing concentration. The visual salinity scores were significantly correlated with shoot Na concentration, Na-K ratio, relative water content, and reduction of dry weight (P<0.01). Their tolerance was attributed to root and shoot characteristics that led to high shoot water content, thus diluting the toxic effect of salts.

  • In order to determine the stability of ginseng components in this salt concentration when used to ginseng as additive ingredient of sauces or seasonings, we study on the content and charactristic of ginsenosides and changes in pH and color, ginseng tail and ginseng extract were treated with various concentration of NaCl solution. In this experiment, extract of ginseng tail were increased in pH as NaCl concentration were increased, but ginseng extract have not changed evidently. The both solution were decreased in color as the salt concentration were increased. Yield of n-butanol extract was decreased in 5% NaCl concentration, while it was increased in the above concentration, and ginseng extract was changed higher than ginseng tail. Ginsenosides content were increased in 5% NaCl concentration, both $ginsenosied-Rb_1$, $-Rb_2$, -Rc, -Rd of diol line and ginsenoside-Re of triol line and increased in above NaCl concentration. Especially ginsenoside-Re showed to sensitive response to the changes of the salt concentration.

Water Deficit in Salt- and Drought- stressed Rice (Oryza sativa L.) Seedlings (염과 건조처리에 따른 벼 유묘의 수분결핍)

  • Plants are often subjected to periods of soil and atmospheric water deficit during their life cycle. To find critical levels for identification of tolerant rice variety to salt- and drought-stresses, we investigated the water deficiency in the leaf of a Dongjinbyeo (DJ) cultivar, identified as intolerant variety, subjected to NaCl- and Polyethylene glycol 6000 (PEG)- treatments. The relative water content and water potential in leaf of DJ plant sharply declined along the high concentration and time after treatment in NaCl- and PEG-treated rice plants. To elucidate the method of simple screening of tolerant variety to salt- and drought-stresses, we examined the relationship between relative water content and water potential of leaves in NaCl- and PEG-treated rice plants. The relationship between relative water content and water potential in leaf of DJ plant showed the highest correlation in 80 mM NaCl-treatment, and showed high correlation only 8% PEG treatment. These results indicate that the critical level of salt stress for screening of tolerant rice was 80 mM NaCl at 48 h after NaCl treatment, and the critical concentration of drought stress for screening of tolerant rice was 8% PEG at 96 h after PEG treatment.

Determination of the Effect of Feeding Different Sodium Chloride Regimes on Growth Performance, Viscera Organ Weights and Meat Quality Parameters of Broilers from Hatch to 35 Days of Age

  • This study intended to investigate the growth performance and meat quality traits in broilers fed different regimens of NaCl over a period of 35 days. Seven hundred and fifty 1-day-old chicks (Cobb 500) were randomly assigned to five NaCl feeding regimens to obtain three replicates for each regimen. Broilers in CON group were provided with concentrations of 0.35% NaCl for the duration of the experimental period. The other groups were provided with two concentrations of NaCl (0.45% and 0.55%) under two durations, namely: 1) 1~28 d: 0.45% or 0.55% NaCl for 28 days then CON; and 2) 1~35 d: 0.45% or 0.55% NaCl for 35 days. Broilers fed 0.55% NaCl for 1~35 d showed improved (P<0.05) growth performance compared with broilers fed the CON regimen. Moreover, higher water holding capacity of breast meat was shown by the broilers fed 0.55% NaCl for 1~28 d when compared with that in broilers fed the CON diet (P<0.05). Broilers fed 0.45% NaCl showed an increase (P<0.05) in the lightness of the meat over CON diet and the 0.55% NaCl diet. In addition, broilers offered CON had the highest redness value and highest crude ash content in the breast meat (P<0.05). Feeding 0.45% NaCl for 1~35 d and 0.55% NaCl for 1~28 d resulted in a higher (P<0.05) protein content in breast meat. In conclusion, broilers fed diets with 0.55% NaCl for 1~35 d showed improved growth performance as well as water holding capacity and protein content of the breast meat.

Characterization of the Functional Properties of Soy Milk Cake Fermented by Bacillus sp.

  • The mucilage production and tyrosine content in soy milk cake (SMC) fermented by Bacillus firmus NA-1, Bacillus subtilis GT-D, and B. subtilis KU-A was improved by fortification with 10% defatted soybean flour. The fibrinolytic activity and consistency of the SMC were drastically increased by solid-state fermentation for 1 day. However, the consistency of the fermented SMC gradually decreased during fermentation for 3 days. Furthermore, the tyrosine content of the freeze-dried powder of SMC fermented by three Bacillus sp. was 9 times higher than that of unfermented SMC. The soybean proteins, including the 7S and 11S subunits, were partially digested during alkaline fermentation, producing lower molecular-weight peptides. The fibrinolytic enzyme produced in SMC fermented by B. firmus NA-l and B. subtilis KU-A exhibited higher thermal stability than that of B. subtilis GT-D fermentation. The powder obtained from B. subtilis GT-D fermentation had an ${\alpha}$-amylase activity and lower consistency compared to those of B. firmus NA-1 and B. subtilis KU-A. In addition, this powder contained 6.3% moisture content, 27% crude protein content and 9 units of fibrinolytic activity and proteolytic activity.

Optimization of the Sucrose and Ion Concentrations for Saikosaponin Production in Hairy Root Culture of Bupleurum falcatum

  • Saikosaponin productivity was examined in a Bupleurum falcatum L. BFHR2 hairy root culture in response to changes in the sucrose content $(2{\sim}8%)$, nitrogen content $(0{\sim}250mM\;NH_4NO_3)$, phosphate content $(0{\sim}12mM\;NaH_2PO_4)$, and the potassium content $(0{\sim}87.2 mM\; KCl)$ of the culture media. We found that the conditions for maximal saikosaponin production differed from those for optimal root growth. Highest saikosaponin yield was achieved for 8% sucrose, 62mM $NH_4NO_3$ 1.2 mM $NaH_2PO_4$, and 0.5mM KCl.

Corrosion behavior of Mg-(0~6)%Zn Casting Alloys in 1M NaCl Solution (1M NaCl 용액에서 Mg-(0~6)%Zn 주조 합금의 부식 거동)

  • The effects of the Zn content on the microstructure and corrosion behavior in 1M NaCl solution were investigated in Mg-(0~6)%Zn casting alloys. The MgZn phase was scarcely observed in the Mg-1%Zn alloy, while the Mg-(2~6)%Zn alloy consisted of ${\alpha}$-(Mg) and MgZn phases. With an increase in the Zn content, the amount of the MgZn phase was gradually increased. Immersion and electrochemical corrosion tests indicated that the Mg-1%Zn alloy had the lowest corrosion rate among the alloys, and a further increase in the Zn content resulted in the deterioration of the corrosion resistance. Microstructural examinations of the corroded surfaces and EIS analyses of surface corrosion films revealed that the best corrosion resistance at 1%Zn was associated with the absence of MgZn phase particles in the microstructure and the contribution of Zn element to the formation of a protective film on the surface. A micro-galvanic effect by the MgZn particles led to the increased rate of corrosion at a higher Zn content.
