Development and Preliminary Evaluation of a Leukocyte Removal Aptamer Filter (압타머를 이용한 백혈구제거필터의 개발 및 예비평가)

  • Background: Leukocyte reduction filters are widely used to prevent transfusion reactions caused by leukocytes in blood components. Commercial filters are not sufficient for removal of leukocytes for prevention of transfusion associated graft-versus-host disease; therefore, irradiation of blood components was performed using expensive equipment. Techniques using an aptamer substituted for antibody have been developed and are available in clinical areas. The purpose of this study is to develop the aptamer filter system and to evaluate its efficiency and the possibility of its clinical application. Methods: Aptamers targeted to CD45 were selected by the Postech Aptamer Initiative. The aptamer filter in which aptamers attached to beads were bound to leukocytes and removed by magnetic field was developed. Filtration of 14 units of leukoreduction-red blood provided by Korean Red Cross Blood Services was performed using aptamer filters. Leukocyte removal rate and red cell recovery rate were evaluated and bacterial culture was performed. Results: After filtration using the aptamer filters, 45.6% of leukocytes were additionally removed and the red cell recovery rate was 92.8%. No growth in the bacterial culture was observed. Conclusion: In order to apply the cell depletion technique utilizing an aptamer to blood filter system, we developed and evaluated the aptamer filter system. Through improvement of the binding efficiency of the aptamer and the filtering process, and application of the various aptamers for other different cells, we suggest that this technique can be applied in the clinical area, such as a substitution for the irradiation process for TAGVHD prevention.

Effects of Dietary Supplementation of Galla Rhois on Productivity in Broiler Chickens (사료 내 오배자 첨가가 육계 생산성에 미치는 영향)

  • This experiment was designated to investigate the effects of dietary supplementation with various concentrations of oriental herbal natural extract, Galla Rhois (GR), on growth performance and meat quality on broiler chickens. A total of 80 two-day-old broiler chicks were randomly designated to four groups, GR 0% diet (control), GR 0.10%, 0.25%, and 0.50%-treated diet, composed 20 chicks and fed a standard diet supplemented with GR and monitored the growth performance every 5 days during 30 days. Body weight gain (BWG) in all treated groups was increased compared to control group during overall period, showing significant (P<0.05) increase in GR 0.25% and 0.50% independent on concentration, though all group represented a similar level of feed intake (FI) and feed conversion rate (FCR). In analysis of the crude proteins and fatty acid composition in leg and breast meats in control, GR 0.10% and GR 0.50%, there was no significant difference for crude proteins and fatty acid composition in leg meats among 3 groups. Whereas the crude proteins and fatty acid composition in breast meat in GR 0.50% showed significantly higher than those of control (P<0.05). Furthermore, the composition of a-linoleic acid (C18:3n-3) and conjugated linoleic acid, which are known as anticancer and antioxidative fatty acids, are higher than those of control. These results demonstrate that Galla Rhois appears to improve growth performance, feed efficiency and meat quality on broiler chickens, focusing on potential use as a dietary supplement.

Age-Based Characteristics of West Syndrome in Patients with Mitochondrial Disease (미토콘드리아 질환에서 웨스트 증후군 환자의 경련 발생 연령에 따른 임상 양상 비교)

  • Purpose: West syndrome is a severe form of age-specific epilepsy that typically affects infants younger than 2 years of age with mitochondrial disease. We aimed to examine age-specific characteristics of the syndrome in these patients. Methods: We retrospectively analyzed 54 patients with West syndrome diagnosed with mitochondrial disease between March 2006 and March 2016. We compared treatment strategies and diagnostic and clinical variables between patients with early-onset (<6 months of age) and late-onset (${\geq}6$ months of age) seizures. Results: Seizure was the first symptom in 30 (90.9%) and 13 (65%) patients of the early-onset and late-onset groups, respectively (P=0.046). Delayed development was observed in 3 (9.1%) and 7 (35%) patients of the early-onset and late-onset groups, respectively (P=0.023). Lactate levels were normal in 17 patients (55%) of the early-onset group and 5 (25%) of the late-onset group (P=0.036), while initial brain magnetic resonance imaging (MRI) findings were normal in 23 (67.6%) and 8 (40%) patients of the early-onset and late-onset groups, respectively. Final MRI findings were abnormal in 32 patients (94.1%) of the early-onset group and 18 (90%) of the late-onset group (P=0.036). Although ketogenic diets reduced seizure frequency in both groups, the difference was not significant. Conclusion: There is no significant difference in epilepsy-related variables when patients are divided based on a cut-off age of 6 months. However, differences in the first symptom at onset and MRI findings were observed. Although lactate levels were not of significant diagnostic value in the early-onset group, they may be in the late-onset group.

Selection of Ground Covering Plant Applicable to Aronia Production in the Highland Rolling Plains (고랭지 경사밭 아로니아 재배시 적정 피복식물 선발)

  • This study was conducted to nominate optimal ground cover plants eventually enhancing Aronia production in the highland rolling plains. Total number of 17 weed species were observed in Aronia field when no cover plant was applied. Meanwhile, 12, 14, 15 and 16 weed species were observed when kentucky bluegrass, white clover, rattail fescue and ground ivy were used, respectively. Untreated native weed species were 73.6 cm tall before cut, and kentucky bluegrass, white clover, Rattail fescue and ground ivy were 57.5, 36.8, 48.3 and 40.9 cm, respectively. Based on plant height before cut, two shortest plants, white clover and ground ivy, were considered effective as ground cover plants in Aronia field. Coverage at $3^{rd}$ year by cover plants ranged from 85% to 100%. Coverage of uncovered Aronia field by native weed species was 95% while coverage by 4 treatments, kentucky bluegrass, white clover, rattail fescue and ground ivy were 100, 87, 85 and 100%, respectively. Aronia yield per plant at $3^{rd}$ year was 1,916 g with white clover cover followed by 1,770 g with Rattail fescue, 1,766 g with ground ivy, 1,098 g without cover plants and 931 g with Kentucky Bluegrass. Out results indicated that ground ivy was the best among all treatments based on 3 criteria, (1) short plant architecture, (2) rapid ground covering and (3) better weed control. In addition, ground ivy cover appeared to secure better yield.

Effect of Low-dose Aspirin on Implantation and Pregnancy Rates in Patients Undergoing Frozen-thawed Embryo Transfer (동결보존 배아 이식에서 저 용량 아스피린 투여가 임신율과 착상율에 미치는 영향에 관한 연구)

  • Objective: Low-dose aspirin have been proposed to improving endometrial receptivity and pregnancy rate in COH-IVF by increasing endometrial perfusion. However, the effect of low-dose aspirin in COH-IVF could be negligible because there have been large quantity of other important factors responsible for changing endometrial perfusion accompanied by COH procedure. In contrast, in frozen-thawed embryo transfer cycles which were not accompanied by COH procedure, the effects of low-dose aspirin in endometrial blood flow seems to be more certain than in COH-IVF cycles. In this study, we analyzed the effect of low-dose aspirin treatment on implantation and pregnancy rates in patients undergoing frozen-thawed embryo transfer Methods: From January 2003 to December 2003, total 264 cycles from 264 patients who attended infertility clinic at Samsung Cheil Hospital were enrolled in this study. All cases included in this study, embryos were frozen and thawed at the pronuclear stage and three days after incubation, at least 2 or more good quality embryos were transferred into uterus. In study group, low dose aspirin (100 mg/day) was administrated from the first or second date of menstrual day to 9 days after embryo transfer. On the other hand, control group did not take any medicine except estradiol valerate for endometrial priming. Several variables including implantation and pregnancy rates were compared in both groups. After then, each groups were stratified by endometrial thickness checked at embryo transfer (ET) day such as (28 mm versus <8 mm) and same variables above described were compared between study and control groups. Results: The mean age, infertility duration, endometrial thickness at embryo transfer day and mean number of transferred embryo were not significantly different in both groups. Also, implantation rates (study group: 15.8%, control group: 20.5%) and pregnancy rate (study group: 45.1%, control group: 43.5%) were not significantly different between two groups. (p>0.05) After we analyzed same variables stratified by endometrial thickness checked at embryo transfer day, we could not found any significant difference between study and control groups. Conclusions: Low-dose aspirin treatment seems to have no advantage of improving implantation and pregnancy rates in patients undergoing frozen-thawed embryo transfer.

Effects of Topical Moisturizers on the Skin of Healthy Full-term Infants and Toddlers (국소 보습제 도포가 정상 영유아 피부에 미치는 영향에 관한 연구)

  • Moisturizers are the most prescribed products in dermatology. Treatment with moisturizers aims to maintain skin integrity and overall well-being by providing a healthy appearance. Moisturizers perform very important functions in baby care; however, there are few studies on the effects of moisturizers on the skin of infants. To investigate the effects of moisturizers on the skin of healthy full-term infants and toddlers, thirty-one healthy, full-term, 6- to 36-month-old infants and toddlers without any dermatologic conditions received moisturizer applied to the whole body except the eyes and diaper area after bathing twice daily for 4 weeks. Clinical assessments were conducted before treatment, immediately after the treatment period, and 1 and 4 weeks after treatment. At all visits, skin hydration, transepidermal water loss (TEWL), skin pH, and skin roughness were measured, the skin surface was photographed, and any adverse events were recorded. After using moisturizer, skin hydration significantly increased and TEWL and roughness significantly decreased. The skin pH was modified to mildly acidic and the skin surface was visually smoother than before treatment. There were no statistical significant differences of effects of moisturizers according to age and sex, and adverse events were not observed. The results of moisturizer application on the skin were increased skin hydration, recovery of barrier function, balancing skin pH within a mildly acidic range, and increasing the smoothness of the skin surface for 4 weeks.

Among members ofcitizen movement groups,government workers,and students who are living inInchon were selected as 613 samples using a purposive sampling method. MultipleClassification Analysis (MCA) and cross-tabulation methods were used in theanalysisThe study of identity in an area is important in terms of providing the solution ofthe problem in a region and social integration of the citizens. The scores of the indexabout Inchon's identity are quite low and more than half of the respondents to allthree groups showed the middle position of the scores from the identity index. By thecharacteristics of the respondents,female,unmarried single,30 years or more,lowerincome groups showed relatively higher identity index scores than other counter-parts . Physicochemical Comparison of Two Different Shark Meats Used for Preparation of Dombaeki (돔배기용 상어육의 이화학적 성분 비교)

  • Dombaeki, a traditional salted shark meat, has been widely used as a customary religious food in the Daegu-Gyeongbuk area of Korea. Two different sharks, Sphyrna zygaena (SZ) and Isurus oxyrinchus (IO) are traditionally used to prepare Dombaeki. Chemical components, lipid classes, fatty acid levels, and sterol compositions of meats prepared from the two sharks were investigated. There were no significant differences in chemical composition between the two dried shark meats. Major amino acids in shark meat were leucine, lysine, arginine, proline, isoleucine, valine, phenylalanine, glutamic acid, and asparagine, which together accounted for about 40% of total amino acids. Levels of amino acids in IO meat were higher than in SZ meat. Major fatty acids in the two shark meats were palmitic ($C_{16:0}$), stearic ($C_{18:0}$), oleic ($C_{18:1}$), arachidonic ($C_{20:4}$), and docosahexanoic (DHA, $C_{22:6}$) acids, which accounted for about 80% of total fatty acids. Notably, IO meat showed higher amounts of DHA (31.8%) and eicosapentanoic acid (EPA, 1.8%) than did SZ meat. The two shark meats showed similar dry weight levels of total lipids, with triacylglycerols, free fatty acids, sterols, and phospholipidscomprising on average 5.0, 2.0, 13.0 and 63.0% of total lipids, respectively. The dominant classes of phospholipids were mainly phosphatidylcholine (PC) and phosphatidylethanolamine (PE). PC content in the two shark meats was higher than that of PE, although the differencewas not great. The major fatty acids in phospholipids were myristic, palmitoleic, stearic, and docosanoic acids. Total volatile basic nitrogen content and the pH of SZ meat were lower than those of IO meat, whereas the Hunter's 'a' and 'b' values of SZ meat were higher than those of IO meat. These results suggest that shark meat may be useful as a functional food to prevent several degenerative diseases.

Comparison of the Quality Characteristics of Radish by Soaking Using Sun-dried Salt and Leuconostoc starter (무 절임 제조 시 천일염과 스타터 첨가에 따른 품질특성 비교)

  • In this study, the effects of the use of a starter on radish soaked at $4^{\circ}C$ for 28 days using two kinds of domestic sun-dried salt (white and gray salt) were determined. As a result, the moisture contents of the radish soaked with white and gray salt were 7.93 and 4.50%, respectively. The salinity levels were found to be equal (90%). No significant difference was found between the two groups in terms of the mineral contents of the salts therein ($37812.41{\pm}1922.95$ and $39755.13{\pm}1205.70$ mg/100 g, respectively). However, iron and zinc contents of gray salt were higher than those of white salt while the calcium, potassium, and magnesium contents were lower than those of white salt. After the addition of the starter, the sugar content increased, the pH decreased, and the total acidity rapidly decreased compared with the non-starter group during fermentation. The total microorganism count of the two groups increased during the fermentation period of 28 days. From the initial fermentation for 7 days, the lactic acid bacteria in the non-starter group increased while those in the leuconostoc starter group steadily increased within the whole fermentation period. The Leuconostoc spp. in non-starter group increased, but that in the starter group decreased to pH 4.0 on day 14.

  • This study was conducted to find out the effect of herbicides on soil micro-organism and soil enzyme in loam soil, and on pathogenic microorganism in continuous pepper cropping soil. The result was summarized as follows: When herbicides were treated, the number of soil microorganism generally decreased at the early stage of incubation, and gradually increased at 30 days incubation. The number of fungi was significantly decreased seven times comparison with control plot. The number of actinomycetes was increased at 20 days incubation and that of pytium was increased for all days incubation in Linuron treatment plot. Although the activity of soil enzyme tended to decrease when herbicides were treated, the activities of urease and phosphatase were higher than that of control plot at the early stage of incubation in MO, Linuron and Simetryne treatment plot. In Simetryne treatment plot, the activity of protease was increased at the early stage, and suddenly decreased after 30 days incubation. There was no significant effect of herbicides on the activities of ${\beta}$-glucosidase and polygalacturonase. The activity of cellulase was inhibited at the early stage of incubation, but that of cellulase was higher than that of control plot after 20 days incubation. High significance was showed the correlation coefficient between soil microorganisms except fungi and soil respiration as herbicides were treated, and the total microorganism and soil respiration in Linuron plot. when Dicamba and Simetryne were treated, the correlation coefficient between the total microorganism and the activities of protease and urease were considerably significant.
