Effects of Gas Composition in the Modified Atmosphere Packaging on the Shelf-life of Longissimus dorsi of Korean Native Black Pigs-Duroc Crossbred during Refrigerated Storage

  • This study was conducted to observe the effects of gas composition in modified atmosphere packaging (MAP) on the shelf-life of Longissimus dorsi of Korean Native Black Pigs-Duroc Crossbred ($KNP{\times}D$) during refrigerated storage. Muscle sample was obtained from the left side of carcass of seven months old of $KNP{\times}D$ barrow. The sample was sliced into 1 cm in thickness, placed on trays (two slices/tray) and filled with different gas composition, i.e. 0:20:80/$O_2:CO_2:N_2$ (MAP1), 30:20:50/$O_2:CO_2:N_2$ (MAP2) and 70:20:10/$O_2:CO_2:N_2$ (MAP3). Other slices of sample were vacuum packed (VP) as a control. All packs were stored at $5{\pm}1^{\circ}C$. At 12 d of storage, pH value of MAP2 and MAP3 were higher (p<0.05) than that of MAP1 and pH value of MAP1 was higher (p<0.05) than that of VP. At 6 d of storage, redness ($a^*$) value of MAP2 and MAP3 were higher (p<0.05) than that of VP and MAP1 and, at 9 and 12 d of storage, redness value of MAP3 was higher (p<0.05) than that of VP, MAP1, and MAP2. At 3, 6, 9, and 12 d of storage, the 2- thiobarbituric acid reactive substances (TBARS) value of MAP3 was higher than that of MAP2 and TBARS value of MAP2 was higher than that of VP and MAP1. At 3, 6, 9, and 12 d of storage, volatile basic nitrogen values of MAP2 and MAP3 were higher (p<0.05) than those of VP and MAP1. At 3 d of storage, total aerobic plate counts of MAP2 and MAP3 were higher (p<0.05) than those of VP and MAP1 and, at 6 d of storage, total aerobic plate counts of MAP3 was higher (p<0.05) than that of MAP1 and MAP2. However, there was no significant different total aerobic plate count among MAP1, MAP2, and MAP3 at 9 and 12 d of storage. There was no significant different total anaerobic plate count among MAP1, MAP2, and MAP3 during storage. It is concluded that the MAP containing 30:20:50/$O_2:CO_2:N_2$ gas composition (MAP2) might be ideal for better meat quality for $KNP{\times}D$ meat.

AtMAP65-1 Binds to Tubulin Dimers to Promote Tubulin Assembly

  • In Arabidopsis thaliana, the microtubule-associated protein AtMAP65-1 shows various functions on microtubule dynamics and organizations. However, it is still an open question about whether AtMAP65-1 binds to tubulin dimers and how it regulates microtubule dynamics. In present study, the tubulin-binding activity of AtMAP65-1 was investigated. Pull-down and co-sedimentation exp eriments demonstrated that AtMAP65-1 bound to tubulin dimers,at a molar ratio of 1 : 1. Cross-linking experiments showed that AtMAP65-1 bound to tubulin dimers by interacting with $\alpha$-tubulin of the tubulin heterodimer. Interfering the bundling effect of AtMAP65-1 by addition of salt and monitoring the tubulin assembly, the experiment results indicated that AtMAP65-1 promoted tubulin assembly by interacting with tubulin dimers. In addition, five truncated versions of AtMAP65-1, namely AtMAP65-1 $\Delta$N339 (amino acids 340-587); AtMAP65-1 $\Delta$N494 (amino acids 495-587); AtMAP65-1 340-494 (amino acids 340-494); AtMAP65-1 $\Delta$C495 (amino acids 1-494) and AtMAP65-1 $\Delta$C340 (amino acids 1-339), were tested for their binding activities and roles in tubulin polymerization in vitro. Four (AtMAP65-1 $\Delta$N339, $\Delta$N494, AtMAP65-1 340-494 and $\Delta$C495) from the five truncated proteins were able to co-sediment with microtubules, and three (AtMAP65-1 $\Delta$N339, $\Delta$N494 and AtMAP65-1 340-494) of them could bind to tubulin dimers in vitro. Among the three truncated proteins, AtMAP65-1 $\Delta$N339 showed the greatest activity to promote tubulin polymerization, AtMAP65-1 $\Delta$N494 exhibited almost the same activity as the full length protein in promoting tubulin assembly, and AtMAP65-1 340-494 had minor activity to promote tubulin assembly. On the contrast, AtMAP65-1 $\Delta$C495, which bound to microtubules but not to tubulin dimers, did not affect tubulin assembly. Our study suggested that AtMAP65-1 might promote tubulin assembly by binding to tubulin dimers in vivo.

  • The digital map version 2.0 is national base map which is used for frame work data, paper map making as well as geographic information system. National Geographic Institude has been research to make small scale digital map by using large scale digital map. NGI made from 25 1/5,000 digital maps to one 1/25,000 digital map ver 2.0 with map generalization system in 2003. However, they could not make 1/10,000 and l/50,000 digital map version 2.0 because of There is no portrayal on the scale 1/10,000 and 1/50,000 digital map in the existing regulations. therefore. We create the specification of the digital map on scale in order to make small scale digital map version 2.0 such as 1/10,000 and 1/50,000 scale.

  • A Cellular embedding of a graph G into an orientable surface S can be considered as a cellular decomposition of S into 0-cells, 1-cells and 2-cells and vise versa, in which 0-cells and 1-cells form a graph G and this decomposition of S is called a map in S with underlying graph G. For a map M with underlying graph G, we define a natural rotation on the line graph of the graph G and we introduce the line map for M. we find that genus of the supporting surface of the line map for a map and we give a characterization for the line map to be embedded in the sphere. Moreover we show that the line map for any life of a map M is map-isomorphic to a lift of the line map for M.

Microtubule-associated Protein 1B Interacts with Glyceraldehydes 3-Phosphate Dehydrogenase in Bovine Follicles (한우 난포에서 발현하는 MAP1B와 GAPDH의 상호 결합)

  • Microtubule-associated protein 1B (MAP1B), a member of MAP1 family, plays a key role in neuronal development. MAP1B binds to many kinds of proteins directly or indirectly. This study was performed to investigate whether MAP1B interacts with GAPDH in bovine follicles using immunoprecipitation (IP) with Western blot analysis and immunohistochemisty. The mRNA expressions of MAP1B and glyceraldehydes 3-phosphate dehydrogenase (GAPDH) were down-regulated in bovine follicular cystic follicles (FCF). In parallel with the mRNA levels, their protein levels were also down-regulated in FCFs. In addition, MAP1B and GAPDH were co-localized at the cytoplasm of follicles. IP with Western blot analysis showed that MAP1B bound to GAPDH in normal follicles, but their binding was absent in FCFs, suggesting a low level of MAP1B and/or GAPDH expressions in FCFs. Taken together, these results suggest that MAP1B interacted with GAPDH may play a role in bovine follicle development, and that GAPDH does not function always as a loading control in bovine follicles.

The Map Generalization Methodology for Korean Cadastral Map using Topographic Map (수치지형도를 이용한 연속지적도의 지도 일반화 기법 연구)

  • Recently, demand for the use of cadastral map is increasing in both public and private area. To use cadastral map in web or mobile environment, construction of the multi-representation database(MRDB) that is the compressed into multiple scale from the original map data is recommended. In this study, the map generalization methodology for the cadastral map by applying overlay with topographic map and polygon generalization technique is suggested. This process is composed of three steps, re-constructing the network data of topographic map, polygon merging of parcel lines according to network degree, and applying line simplification techniques. Proposed methodologies are applied to the cadastral map in Suwon area. The result map was generalized into 1:5,000, 1:20,000, 1:100,000 scale, and data compression ratio was shown in 15% 8% 1% level respectively.

  • The helicoidal surfaces with pointwise 1-type or harmonic gauss map in Euclidean 3-space are studied. The notion of pointwise 1-type Gauss map is a generalization of usual sense of 1-type Gauss map. In particular, we prove that an ordinary helicoid is the only genuine helicoidal surface of polynomial kind with pointwise 1-type Gauss map of the first kind and a right cone is the only rational helicoidal surface with pointwise 1-type Gauss map of the second kind. Also, we give a characterization of rational helicoidal surface with harmonic or pointwise 1-type Gauss map.

Automated Generation of Multi-Scale Map Database for Web Map Services (웹 지도서비스를 위한 다축척 지도 데이터셋 자동생성 기법 연구)

  • Although the multi-scale map database should be constructed for the web map services and location-based services, much part of generation process is based on the manual editing. In this study, the map generalization methodology for automatic construction of multi-scale database from the primary data is proposed. Moreover, the generalization methodology is applied to the real map data and the prototype of multi-scale map dataset is generated. Among the generalization operators, selection/elimination, simplification and amalgamation/aggregation is applied in organized manner. The algorithm and parameters for generalization is determined experimentally considering T$\ddot{o}$pfer's radical law, minimum drawable object of map and visual aspect. The target scale level is five(1:1,000, 1:5,000, 1:25,000, 1:100,000, 1:500,000) and for the target data, new address data and digital topographic map is used.

A Study on the Consecutive Renewal of Road and Building Information in the Multi-scale Digital Maps (다축척 수치지도의 도로 및 건물정보 일괄갱신 연구)

  • In the existing digital map of the Ver.1.0, it is impossible to make a small scale digital map, which is under the 1/5000 scale map, by using the 1/1000 digital map which is the most large scale one. Because of this reason, the existing digital maps are produced into a 1/1000 and a 1/5000 map by means of two different scale aerial photos. The next generation digital map should be successively related to a small scale digital map based on the most large scale digital one. This is so important from the aspects of data share and the consecutive renewal. Ever since the development of the digital map of the Ver. 2.0, the possibility of making a multi-scale consecutive digital map has been presented and the related research has been done again. The most basic thing in the multi-scale digital maps is to decide the criteria of the generalization between the two scales. In this study, I try to formulate the criteria of the generalization required to make the 1/5000 digital map by using the 111000 digital one. In addition, I by to explore the application possibility of the consecutive renewal by carrying out auto-generalization.

Geological Map Database Construction Using GIS (GIS를 이용한 지질도 데이터베이스 구축에 관한 연구)

  • Geological map and data are needed for land use planning, resources development, geological hazard prevention, environment protection and education, Since the nationwide geological database in Korea has not been constructed yet, there are many problems in using the geological map and data. There are many problems such a stratigraphy unestablishment, map conservation and edge matching in geological paper map. Therefore it is difficult to construct the geological map database, but the geological map database must be constructed as soon as possible as one of national thematic map. In this study, geological maps of pilot area such as Ansung geological map on a scale of 1:50,000, Busan on a scale of 1:250,000, Namchang on a scale of 1:25,000 and the whole Korean peninsula on a scale of 1:1,000,000 were designed and constructed to database using Geographic Information System(GIS). In addition the geological map management program was developed by GIS program. The digital geological maps were produced using the constructed geological database. The database could be of access through Internet World Wide Web(WWW) environment and be distributed in Compact Disk(CD).
