Effect of Priming, Temperature and Light Quality on Germination of Pokeweed(Phytolacca americana) Seed (Priming, 온도 및 광질이 미국자리공 종자의 발아에 미치는 영향)

  • Pokeweed, a polycarpic plant, has been used as herbage medicine, vegetable or dye. It, however, is known as an aggressive plant in the vicinity of the industrial area evolving air pollutants. The experiment was done to determine the effect of priming using nitrates, germination temperature and light quality on germination of its seed to get information on the optimum germination process as well as its establishment. The daily percent germination was measured to 12 days after sowing since its seeds were treated by two different nitrates [KNO$_3$, Ca(NO$_3$)$_2$]. their different concentrations (0, 50, 150, 300mM), then treatment duration (1, 3, 6 days), different germination temperature (day /night; 30/30, 30/20, 20/30, 20/2$0^{\circ}C$) and light quality (red, white, dark) before or during germination. The percent germination was greater in the KNO$_3$ treatment than in the Ca(NO$_3$)$_2$ but in the priming treatment with KNO$_3$ in comparison with no-priming. In the priming treatment with KNO$_3$, the percent germination was increased with its increased concentration to 150mM although decreased with delayed duration to 6 days. Regardless of light quality, the greater percent germination was shown in the order of 2$0^{\circ}C$ constant and 20/3$0^{\circ}C$ alternative, 3$0^{\circ}C$ constant, 30/2$0^{\circ}C$ alternative temperature. The germination was less in the dark during germination than in the illumination in which the red light had greater percent germination compared to white light. The seeds primed with KNO$_3$ were germinated under the alternative temperature even in the dark condition.

Effect of Bioluminescence Stimulating Agent of the Genetically Engineered Strain KG1206 on the Monitoring of the Petroleum Hydrocarbon Contaminated Groundwater Samples (발광유전자 재조합 균주 활성 촉진 조건이 석유계 탄화수소 오염지하수 모니터링에 미치는 영향)

  • This paper describes the application of bioluminescence stimulating agents on a genetically engineered microorganism, Pseudomonas putida mt-2 KG1206, to monitor toluene analogs using in groundwater samples from petroleum hydrocarbon contaminated sites. The maximum bioluminescent response with pure chemicals followed in the order: m-methyl benzyl alchohol > m-toluate > toluene > m-xylene > benzoate > p-xylene > o-xylene. Generally, the bioluminescence production of strain mixed with groundwater samples was dependent on the contaminated total inducer concentrations. However, few samples showed opposite results, where these phenomena may be caused by the complexicity of environmental samples. Two chemicals, SL(sodium lactate) and KNO$_3$, were tested to determine a better bioluminescence stimulant. Both chemicals stimulate the bioluminescence activity of strain KG1206, however, a slightly high bioluminescence was observed with nitrogen chemical. This selected stimulant was then tested on samples collected from contaminated groundwater samples. The bioluminescence activity of all samples mixed with the strain was stimulated with KNO$_3$ amendment. This suggests that the low bioluminescence activity exhibited by the environmental groundwater samples can be stimulated by amending the culture with a proper agent, such as nitrogen compound. These findings would be useful, especially, when strain was used to monitor the groundwater samples contaminated with low inducer contaminants. Overall, the results of this study found the ability of bioluminescence producing bacteria to biosensor a specific group of environmental contaminants, and suggest the potential for more efficient preliminary application of this engineered strain in a field-ready bioassay.

Sing1e or Combined Treatment Effect of Priming, Gibberellin and Prechilling on Germination of Gourd Seeds

  • The present experiment was done to determine the effect of priming, $GA_3$ and prechilling treatment alone and in their combination on germination of gourd (Lagenaria siceraria Standl.) seeds. Priming using $KNO_3$ and $GA_3$ were treated with different concentrations and periods whereas prechilling was done at only different periods. FR-yongjadaemok and FR-kunghap were used as test cultivars and their daily germination rates were measured at treatment levels. The best germination rate of each treatment was shown in a week prechilling at $3^{\circ}C$, a day priming with 100 mM $KNO_3$ and a day treatment with 0.01 mM $GA_3$ although the prechilling had the highest rate among them. The rate of priming treated after the prechilling was continuously decreased with its longer treatment periods and was less than no-priming. In addition, the rate of $GA_3$ treatment after the prechilling was not affected by its concentrations and treatment periods. Priming and $GA_3$ treatments before the prechilling, however, showed similar germiabilities between their treatment levels including no-treatment. The results mean that sequential order of available presowing treatments is important to enhancing the rate.

  • To establish the mass propagation method of Typha orientatis Presl, several factors influencing seed germination were investigated. The best seed germination of T. orientalis was achieved under the light and 25~3$0^{\circ}C$ temperature. GA$_3$ tended to enhance seed germination, and 500 mg.L$^{-1}$ GA$_3$ showed 90.7% germination, which was 1.2 times of control. Compared to control, BA treatment prohibit seed germination. Especially, seeds were not germinated under 500 mg.L$^{-1}$ BA treatment. In case of KNO$_3$ and Ca(NO$_3$)$_2$ treatment, the germination rate was decreased by all concentrations, except 100 mM Ca(NO$_3$)$_2$.

Effect of Preservation Period, Light, Temperature, and Priming on the Seed Germination of Lysimachia mauritiana (자생 갯까치 수영(Lysimachia mauritiana)의 종자발아에 미치는 저장기간, 광, 온도 및 Priming 처리의 영향)

  • Because Lysimachia mauritiana is not able to maintain the seed viablity for a long time, it has difficulties in using for the ornamental plants or landscape gardening. This study was conducted to elucidate the optimal germination conditions of Lysimachia mauritiana seeds. The germination rate of the seeds was higer in light condition than in dark one. In experment determining the proper germination temperature, the germination rates were 2.2%, 96.9% and 91.2% under 35$\circ $C, 25$\circ $C, and 20$\circ $C, respectively in light condition. The approprate temperature for the germination was 25$\circ $C in light. The decreases of germination rate with the increase of the preservation periods was able to recover by priming pretreated with NaCl solution and KNO$_{3}$ + K$_{3}$H$_{2}$PO mixed solutions. When the storage period was 300 days, the germination rate was 75% without priming treatments. But the seeds primed with NaCl 0.1M and KNO$_{3}$ + K$_{3}$H$_{2}$PO mixed solutions 0.1M, showed germination rate above 90%. T$_{50}$ also was shorten from 5.25 without priming to 3.78 with NaCl 0.1M. Seed priming may induce earlier sprouts and increase germination rate of long-preserved seeds of native wild plants.

Studies on the Promoting of Seed Germination of Adenophora triphylla var. Japanica $H_{ARA}$ (잔대종자 발아촉진에 관한 연구)

  • The objective of this study is to promote germination for mass cultivation of Adenophora triphylla. After the seed soaking for I day in BA and $GA_3$ solution, seed germination was effectively promoted in the treatment of 500mg/L $GA_3$ and the highest germination rate was 94%.The average germination days were 1.5days earlier than those of non-treated seeds. In a chemicals treatment, seed germination rate was 54% in 1% $KNO_3$ treatment but KOH treatment was no significantly effected. The durations of chilling treatment for breaking of dormancy were longer than 2weeks. The optimal temperature for germination was $25^{\circ}C$. The promotion of seed germination is presumed to be due to the breaking of dormancy by $GA_3$, chilling treatment rather than seed coat maceration by KOH or $KNO_3$ treatments.

  • The interfacial Properties of ferrosoferric oxide suspended in aqueous KNO3 solution are investigated by studying the zeta potentials and surface charge densities at $25^{\circ}C$. The zeta potentials are obtained by measuring the electrophoretic mobility and the surface charge densities by potentiometric titrations in the aqueous KNO3 solutions of different concentrations from 10-3 to 10-1M. The data are interpreted by the surface dissociation and complexation model of Davis, et als. and the modified model.

  • This study was carried out to determine the effect of medium solidity, salt strength, sugar and nitrogen sources, and pH levels on in vitro multiplication of pathogen-free yam (Dioscorea opposita Thunb.). Liquid medium was more effective in the growth of plant height, fresh weight, and formation of microbulb than the solid medium. Optimal condition for plant fresh weight, growth, and multiplication axillary bud was in 1MS salt strength with 60 $g{\cdot}L^{-1}$ sucrose and half strength of $KNO_3$. Optimal condition for microbulb formation was $\frac{1}{2}$ MS salt strength supplemented with glucose 60 $g{\cdot}L^{-1}$ and half strength of $KNO_3$. The number of leaves and nodes were sharply increased from 2 to 5 weeks, whereas plant fresh weight was steadily increased from 3 to 11 weeks after inoculation. Microbulbs were formed at 2 weeks after inoculation and continuously increased until 12 weeks.

Evaluating the Performance of Draw Solutions in Forward Osmosis Desalination Using Fertilizer as Draw Solution (유도용액으로 비료를 사용한 정삼투 해수담수화에서 유도용액의 성능 평가)

  • This study is to evaluate the performance of draw solutions in forward osmosis desalination using fertilizer as draw solution. Considering osmotic pressure, solubility, and pH, $NH_4NO_3$, $NH_4H_2PO_4$, $(NH_4)_2HPO_4$, KCl, $KNO_3$, and $KHCO_3$ were screened from a comprehensive lists of fertilizer. Their performance were evaluated in terms of pure water permeate flux, reverse solute flux, and specific reverse solute flux for nitrogen and phosphorus. KCl showed the highest pure water permeate flux among the selected fertilizers while $(NH_4)_2HPO_4$ draw solution had the lowest flux. $NH_4H_2PO_4$ showed the lowest reverse solute flux and specific reverse solute flux for nitrogen followed by $(NH_4)_2HPO_4$, $KNO_3$, and $NH_4NO_3$. Although the pure water permeate flux of $NH_4H_2PO_4$ is lower than the other draw solutions, because it contains both nitrogen and phosphorus, and have the lowest reverse solute flux and specific reverse solute flux, it is a promising candidate as draw solution for forward osmosis desalination.

Transfer of Genetic Substance Through the Cell Wall of Geranium (Pelargonium zonale hybrids, 'Pinto Scarlet') Callus (제라니움 세포벽을 통한 유전물질의 전이)

  • The possibility that DNA could move out of the single cells isolated from geranium (Pelargonium zonale hybrids, 'PintoScarlet') callus was determined by the elechophoretic DNA analysis after treatment of low pH, various concentrations of KNO$_3$, 2,4-D, and GA$_3$ followed by the centrifugal force, all of which are hewn to and the physico-chemical properties of the cell wall. The centrifugal force of l,800 xg was need for DNA migration after the above treatment, but 7k300 xg was required without the treatments. In this experiment the optimum concentration (300 mg/L) of sodium dodecyl sulfate (SDS) used as an anion detergent to collect the negatively charged DNA was very critical not to damage the cell wall It can be concluded that the centrifugal force played a key role for the DNA migration through the cell wall, and the treatments of low pH (4.0), 0.5% KNO$_3$, 1.5 mg/L GA$_3$and 1mg/L 2,4-D further increased the DNA migration.
