Biodegradation of Azo and Reactive Dyes with Pseudomonas strains (Pseudomonas 속의 균주를 이용한 Azo계와 Reactive계의 Dye의 생분해)

  • This study describes biodegradation of dyes which are used in textile industries. Dyes released into the environment from industrial waste water are considered to be a serious pollution problem because of the wide spread into environment with a variety of colors. The microorganisms used in this experiment were Pseudomonas species, which had been screened from aeration tank of waste water treatment. It was found that optimum concentrations for culture media were 14g of glucose, 6g of peptone, 160 mg of Na2HPO4, 200mg of KCl, 140mg of MgSO4,.7H2O,1.0g of KH2PO4, 400mg of NaCl, 200mg of CaCl2 and dye 10ppm per litre of distilled water. The high efficiency of dye degradation was obtained at pH 7-8 and $30-35^{\circ}C$. Strains screened are excellent for removal of azo and reactive dyes, which are relatively stable and difficult to degrade. Dyes of 10ppm such as mono-azo (Lot No. 180), di-azo (Lot No. 138) and reactive red(Lot No. 2) were mostly decolorlzed within 2 days and di-azo (Lot No. 151) and reactive red(Lot No. 34, No. 00166) were decolorized within 5 days in the controlled fermenter. In the case of reactive dyes, oxygen supplies showed lower biodegradability compared to anaerobic culture.

Enhanced Production of Cellulase-Free Thermoactive Xylanase Using Corncob by a Black Yeast, Aureobasidium pullulans CBS 135684

  • Our aim was to optimize the production of cellulase-free thermoactive xylanase by Aureobasidium pullulans CBS 135684 with statistical methodology based on experimental designs. Among eleven variables, the nutrient sources that had significant effect on xylanase production were corncob, $(NH_4)_2SO_4$, xylose, $KH_2PO_4$ and tween 80, identified by the initial screening method of Plackett-Burman. The optimum concentrations of these five components were subsequently investigated using response surface methodology. The optimal concentrations ($g{\cdot}l^{-1}$) for maximum production of xylanase were corncob, 39.0; $(NH_4)_2SO_4$, 3.0; xylose, 1.8; $KH_2PO_4$ 1.4; and tween 80, 1.4, respectively. An improved xylanase yield of $8.74{\pm}0.84U{\cdot}ml^{-1}$ was obtained with optimized medium which is 2.1-fold higher production than previously obtained results ($4.10{\pm}0.10U{\cdot}ml^{-1}$) after 48 h of cultivation. In addition, the xylanase production under optimal condition reached $10.09{\pm}0.27U{\cdot}ml^{-1}$ after 72 h of cultivation.

Studies on the Bacterial Production of L-Glutamate from Acetate Part II. Cultural Conditon (초산을 이용한 글루타민산의 발효생산에 관한 연구 (제2보) 글루타민산 생성을 위한 발효조건)

  • The cultural conditions for L-glutamate production were investigated using Brevibacterium flavum nov. sp. D2209B, the most productive strain among 5 strains reported in preceeding paper. A temperature of 3$0^{\circ}C$ and a medium volume of 30 ml per 500-flask were selected as standard culture conditions. And the following results were obtained. 1. When the concentration of acetate in the medium was below 30 g per litre, the maximum amount of L-glutamate was accumulated. 2. KH$_2$PO$_4$, MgSO$_4$, FeCI$_3$ and MnCI$_2$ were required for the L-glutamate poduction, but the concentration of those inorganic salts little effected. 3. Signifcant amount of L-glutamate was accutnulated in the limited biotin concentration less than 0.3 ug per litre. 4. The addition of malic acid or succinic acid enhanced the accumulation. 5. The L-glutamate accumulation was related to the incubation time of seed; the amount of L-glutamate accumulated was maximum by inoculating 16-20 hour incubated seed. 6. In the medium containing sufficient amount of biotin for growth, L-glutamate accumulation was stimulated by the addition of penicillin at appropreate time during incubation.

  • With the increase of the use of antibiotics and invasive procedures, infections caused by multidrug-resistant Acinetobacter baumannii(MRAB) are increasing. We screened the antibiotic producing strain B-51 for antibacterial activity against MRAB from the soils and studied the effects of culture medium on the antibiotic production of B-51. The medium conditions for maximum antibiotic productivity of B-51 was 2% glycerol, 0.5% soybean meal, 0.01% $CaCl_2$, 0.01% $MgSO_4{\cdot}7H_2O$ and 0.01% $KH_2PO_4$ at an initial pH of 6.0, at $30^{\circ}C$ for 76 h.

Production of an Antihyperlipemial HMG-CoA Reductase Inhibitor from Bacillus cereus D-3 (Bacillus cereus D-3로부터 항고지혈증 HMG-CoA Reductase 저해제의 생산)

  • For the purpose of production of a novel antihyperlipemial HMG-CoA reductase inhibitor from bacteria, a bacterium which showed the highest HMG-CoA reductase inhibitory activity was isolated from traditional Doenjang. This strain was identified as Bacillus cereus (D-3) based on its microbiological characteristics and 165 rRNA sequence analysis. The maximal HMG-CoA reductase inhibitor production from Bacillus cereus D-3 was obtained by cultivation in a Glucose-CSL broth containing 2% glucose, 0.6% corn steep liquor, $0.04%\;K_{2}HPO_4$ and $0.05%\;KH_{2}PO_4$ at $30^{\circ}C$ for 36 h. The final HMG-CoA reductase inhibitory activity under the above conditions was 39.4%.

Optimization of Media Composition and Cultivation for the Mycelial Growth of Agrocybe Cylindracea (Agrocybe Cylindracea의 영양배지 조성 및 배양조건의 최적화)

  • Studies were made to optimize the meida composition and cultural conditions for the mycelial growth of Agrocybe cylindracea. Media composition for optimal growth was found to be starch 20.0g/l, bacto-soytone 4.0g/l. The media supplemented with $KH_2PO_4\;0.46g/l,\;K_2HPO_4\;1.0g/l,\;MgSO_47H_2O\;0.5g/l$ supported mycelial growth better than the media without mineral salts. Optimum temperature and pH for the growth was $28^{\circ}C$, and 6.0 respectively. Temperature range for the mycelial growth appeared to be $10-35^{\circ}C$ and the mycelium evidently lost the vitality at $40^{\circ}C$.

  • Acid mine drainage occurs when sulfide minerals are exposed to an oxidizing environment. The objective of this study was to inhibit the oxidation of pyrite by applying various coating agent such as $KH_2PO_4$, MgO and $KMnO_4$ over its surface as an oxidation inhibitors. Experiments were conducted for 8 days to test the feasibility of oxidation inhibitors. The optimal condition of coating agent for standard pyrite and IK mine was the combination of 0.01M $KH_2PO_4$, 0.01M NaOAc and 0.01M NaClO. Otherwise, for YD mine the combination of 0.01M $KMnO_4$, 0.01M NaOAc and 0.01M NaClO. The $SO_4^{2-}$ reduction efficiency of pyrite, IK and YD mine samples was 70, 92 and 84%, respectively. For 8 days, no significant increase of $SO_4^{2-}$ from pyrite sample coated with inhibitor was observed. The pH of solution remains in between 4 to 6 for the reaction conditions.

Melanoidin decolorization by immobilized cells of Aspergillus awamori, B-2 (Aspergillus awamori B-2의 균사체 고정화 의한 Melanoidin 탈색)

  • Aspergillus awamori, B-2 which has a high ability to decolorize melanoidin was selected among various fungi. Aspergillus awamori, B-2 showed the highest decolorization activity when it was cultivated in a melanoidin medium containing 3.0% glucose, 0.5% yeast extract, 0.1 % $KH_2PO_4\;and\;0.05%\;MgSO_4{\cdot}7H_2O$ at an initial pH 7.0 at $37^{\circ}C$ for 5 days. Mycelia immobilized system with. Ca-alginate was more effective on melanoidin decolorization activity showed approximately 70% in 10 days under the optimal conditions. Continuous decolorization of melanoidin using, reuse of immobilized mycelia showed an almost constant decolorization of abort 60-70% for 15 days.

Optimization of Bioflocculant Production Conditions for Organic Wastewater Treatment with Aeromonas hydrophila KH-54 (Aeromonas hydrophila KH-54가 분비하는 유기폐수처리용 생물응집제 생산조건의 최적화)

  • For the need of bio-degradable flocculant in stage of wastewater treatment, some cultural conditions of bioflocculant production were optimized with Aeromonas hydrophila KH-54. About 260 strains of type culture and bacteria isolated from marsh, pond, activated sludge, etc were examined for their ability to flocculate kaolin particles and swine wastewater. Among them, KH-54 showed the highest flocculating activity and was identified as Aeromonas hydrophila according to the cultural, morphological and physiological properties. The maximum production of the flocculant secreted by Aeromonas hydrophila KH-54 was observed in culture medium containing 2.0% mannitol, 0.05% ammonium chloride, 0.02% potassium phosphate dibasic, 0.01% $MgSO_4{\cdot}7H_2O$ and 0.05% yeast extract at initial pH 7.0 when cultured on rotary shaker controlled at $25^{\circ}C$ and 150 rpm. Under the optimized condition, the flocculating ability reached to 770 units/ml of kaolin flocculating activity and 81% of NTU removal efficiency against swine wastewater after 4 days cultivation. The bioflocculant was also effective on various organic wastewaters other than swine wastewater, showing NTU removal rate ranging from 92% to 34%.

Production and Characterization of Cholesterol Oxidase from Streptomyces sp. No.4 (방선균으로부터 Cholesterol Oxidase의 생산 및 특성)

  • An actinomycetes strain No.4 which produce the cholesterol oxidase(EC 1.1.3.6), was isolated from soil and identified as Streptomyces sp. based on taxonomic studies. The conditions of cholesterol oxidase production and enzymatic properties were investigated. The optimum composition of medium for production of the enzyme was 1% soluble starch, 2% corn steep liquor, 0.1% $KH_2PO_4$, 0.1% $NaNO_3$ and 0.05% $MgSO_4$ (pH 7.0). The optimum pH and temperature of the cholesterol oxidase were pH 6.0~7.5 and $37^{\circ}C$, respectively. The enzyme was stable in the range of pH 6.0~9.0. The isoelectric point determined by multichambered electrofocusing unit was in the range of pH 6.0~6.5.
