Dynamic responses of a riser under combined excitation of internal waves and background currents

  • In this study, the dynamic responses of a riser under the combined excitation of internal waves and background currents are studied. A modified Taylor-Goldstein equation is used to calculate the internal waves vertical structures when background currents exist. By imposing rigid-lid boundary condition, the equation is solved by Thompson-Haskell method. Based on the principle of virtual work, a nonlinear differential equation for riser motions is established combined with the modified Morison formula. Using Newmark-${\beta}$ method, the motion equation is solved in time domain. It is observed that the internal waves without currents exhibit dominated effect on dynamic response of a riser in the first two modes. With the effects of the background currents, the motion displacements of the riser will increase significantly in both cases that wave goes along and against the currents. This phenomenon is most obviously observed at the motions in the first mode.

Tidal and Nontidal Fluctuations of Currents in the Western Channel of the Korea Strait

  • We carried out simultaneous and long-time observations of currents across the western channel of the Korea Strait during the period of May, 1994 to September, 1996 in order to understand the spatial and temporal variability of tidal and nontidal currents in the Strait. Results show that currents in the Strait are quite variable in space and time, and they largely consist of mean current and diurnal and semidiurnal tidal currents of about equal magnitudes. The mean currents include the Tsushima Current and the Korea Strait Undercurrent at the center of the channel. The former occupies the upper two-thirds and the latter the lower one-third of the water column. The semidiurnal and diurnal currents are largely rectilinear in the direction of NE-SW and their amplitude variation across the channel appears to be small. However, the diurnal currents at some locations show rotational characteristics with significant nontidal effects. The station close to the Korean coast leads the phase and the phase difference of the semidiurnal current across the channel appears to be less than half an hour while that of the diurnal current is over five hours.

Acepromazine inhibits hERG potassium ion channels expressed in human embryonic kidney 293 cells

  • The effects of acepromazine on human ether-$\grave{a}$-go-go-related gene (hERG) potassium channels were investigated using whole-cell voltage-clamp technique in human embryonic kidney (HEK293) cells transfected with hERG. The hERG currents were recorded with or without acepromazine, and the steady-state and peak tail currents were analyzed for the evaluating the drug effects. Acepromazine inhibited the hERG currents in a concentration-dependent manner with an $IC_{50}$ value of $1.5{\mu}M$ and Hill coefficient of 1.1. Acepromazine blocked hERG currents in a voltage-dependent manner between -40 and +10 mV. Before and after application of acepromazine, the half activation potentials of hERG currents changed to hyperpolarizing direction. Acepromazine blocked both the steady-state hERG currents by depolarizing pulse and the peak tail currents by repolarizing pulse; however, the extent of blocking by acepromazine in the repolarizing pulse was more profound than that in the depolarizing pulse, indicating that acepromazine has a high affinity for the open state of the channels, with a relatively lower affinity for the closed state of hERG channels. A fast application of acepromazine during the tail currents inhibited the open state of hERG channels in a concentration-dependent. The steady-state inactivation of hERG currents shifted to the hyperpolarized direction by acepromazine. These results suggest that acepromazine inhibits the hERG channels probably by an open- and inactivated-channel blocking mechanism. Regarding to the fact that the hERG channels are the potential target of drug-induced long QT syndrome, our results suggest that acepromazine can possibly induce a cardiac arrhythmia through the inhibition of hERG channels.

Role of $K^+$ Channels to Resting Membrane Potential of Rabbit Middle Cerebral Arterial Smooth Muscle Cells

  • The aim of the present study is to investigate the contribution of $Ca^{2+} ?activated\;K^+\;(K_{Ca})$ channels and delayed rectifier $K^+\;(K_V)$ channels to the resting membrane potential (RMP) in rabbit middle cerebral arterial smooth muscle cells. The RMP and membrane currents were recorded using the whole-cell patch configuration and single $K_{Ca}$ channel was recorded using the outside-out patch configuration. Using the pipette solution containing 0.05 mM EGTA, the RMP was $-25.76{\pm}5.08$ mV (n=12) and showed spontaneous transient hyperpolarizations (STHPs). The membrane currents showed time- and voltage-dependent outward currents with spontaneous transient outward currents (STOCs). When we recorded the membrane potential using the pipette solution containing 10 mM EGTA, the RMP was depolarized and did not show STHPs. The membrane currents showed no STOCs but only showed slowly inactivating outward currents. External TEA (1 mM) reversibly inhibited the STHPs, depolarized the RMP, reduced the membrane currents, abolished STOCs, and decreased the open probability of single $K_{Ca}$ channel. When $K_V$ currents were isolated, the application of 4-AP (5 mM) depolarized the RMP. The important aspect of our results is that $K_{Ca}$ channel is responsible for the generation of the STHPs in the membrane potential and plays an important role in the regulation of the RMP and $K_V$ channel is also responsible for the regulation of the RMP in rabbit middle cerebral arterial smooth muscle cells.

  • A magnitude of generated harmonic currents along with the operation of traction has nonlinear characteristics. The harmonic currents generated along with the operating speed of electrical railroad traction is to analyze very difficulty. This paper therefore presents probabilistic approach for the harmonic currents evaluation about the operating speed of the arbitrary single traction. To use probabilistic method for railroad system, probability density function(PDF) using measuring data based on the realistic harmonic currents per operating speed is calculated. Mean and variance of harmonic currents of single traction also are obtained the PDF of the operating speed and electrical railroad traction model. Uncertainty of harmonic currents expects to results through mean and variance with PDF. The probability of harmonic currents generated with the operating of arbitrary many tractions is calculated by the convolution of functions. The harmonics of different number of tractions are systematically investigated. It is assessed by the total demand distortion(TDD) for the railroad system.

  • Modulatory effect of cAMP-PKA system on inhibitory- and excitatory herbs-induced ion currents were investigated by nystatin-perforated patch clamp method under voltage-clamp condition. Ion currents induced by Bupleuri radix and Coptidis rhizoma were not affected by cAMP-PKA system. Ion current induced by Ecliptae herba was partially inhibited by cAMP-PKA system. Ion currents induced by Aconiti tuber and Boshniakiae herba were inhibitory modulated and ion current induced by Zingiberis rhizoma was excitatory modulated by cAMP-PKA system. Modulation of cAMP-PKA system on ion currents induced by Ginseng radix was fluctuated. In this results, it can be seen generally that excitatory herbs-induced ion currents were modulated by cAMP-PKA system while cAMP-PKA system did not affect inhibitory herbs-induced ion currents.

수치모형을 통한 함평만과 해제반도 주변해역의 조류 및 조석잔차류 분포 (Tidal and tide-induced residual currents around Hampyung Bay and Hajae Peninsula by numerical simulation)

  • In order to understand the currents around Hampyung Bay and Haeje Peninsula, 2D numerical simulations for tidal currents and tide-induced residual currents were carried out. Dominant semidiurnal tidal currents have reversing form and flow NNE-SSW from northern Haeje Peninsula to Songi Island, E-S at northern Haeje Peninsula and NNW-SSE in Hampyung Bay. In flood, a part of currents from Imja Island~Nakwhol Island flow along the main stream flowing northeast at offshore region and the rest flow into Hampyung Bay flowing east along the northern coast of Haeje Peninsula. In ebb, currents from Hampyung Bay flow west along the northern coast of Haeje Peninsula and run together with the main stream flowing southeast at offshore region. The currents create an anticyclonic circulation in flood and a cyclonic circulation in ebb around Haeje Peninsula including Hampyung Bay. Tidal currents are accumulated on Doripo which located at the entrance of Hampyung Bay and show high current velocities. Tidal currents and tide induced residual currents are weak at the inside of Hampyung Bay which has narrow entrance, shallow water depth and wide intertidal zone. An anticyclonic eddy is formed around Gaksi Island as a result of tide induced residual currents. In northern coast of Haeje Peninsula, slow constant currents flow east. It is expected that a gradual change of sediment and an increase of flushing time for suspended materials are carried by tidal currents occurring in Hampyung Bay.

Pharmacological and electrophysiological characterization of rat P2X currents

  • Adenosine 5'-triphosphate (ATP) is an important extracellular signaling molecule which is involved in a variety of physiological responses in many different tissues and cell types, by acting at P2 receptors, either ionotropic (P2X) or G protein-coupled metabotropic receptors (P2Y). P2X receptors have seven isoforms designated as $P2X_{1^-}P2X_7$. In this study, we investigated the electrophysiological and pharmacological properties of rat $P2X_{1^-}P2X_4$ currents by using whole-cell patch clamp technique in a heterologous expression system. When ATP-induced currents were analyzed in human embryonic kidney (HEK293) cells following transient transfection of rat $P2X_{1^-}P2X_4$, the currents showed different pharmacological and electrophysiological properties. ATP evoked inward currents with fast activation and fast desensitization in $P2X_{^1-}$ or $P2X_{3^-}$ expressing HEK293 cells, but in $P2X_{2^-}$ or $P2X_{4^-}$ expressing HEK293 cells, ATP evoked inward currents with slow activation and slow desensitization. While PPADS and suramin inhibited $P2X_2$ or $P2X_3$ receptor-mediated currents, they had little effects on $P2X_4$ receptor-mediated currents. Ivermectin potentiated and prolonged $P2X_4$ receptor-mediated currents, but did not affect $P2X_2$ or $P2X_3$ receptor-mediated currents. We suggest that distinct pharmacological and electrophysiological properties among P2X receptor subtypes would be a useful tool to determine expression patterns of P2X receptors in the nervous system including trigeminal sensory neurons and microglia.

  • The effects of wall charge and bias voltage on exo-electron emission currents were examined. In addition, the effects of doping elements on the currents were investigated. These results indicated that the statistical delay is inversely proportional to the exo-electron emission currents measured.

Effects of Pine Needle Extract on Pacemaker Currents in Interstitial Cells of Cajal from the Murine Small Intestine

  • Extracts of pine needles (Pinus densiflora Sieb. et Zucc.) have diverse physiological and pharmacological actions. In this study we show that pine needle extract alters pacemaker currents in interstitial cells of Cajal (ICC) by modulating ATP-sensitive $K^+$ channels and that this effect is mediated by prostaglandins. In whole cell patches at $30^{\circ}C$, ICC generated spontaneous pacemaker potentials in the current clamp mode (I = 0), and inward currents (pacemaker currents) in the voltage clamp mode at a holding potential of -70 mV. Pine needle extract hyperpolarized the membrane potential, and in voltage clamp mode decreased both the frequency and amplitude of the pacemaker currents, and increased the resting currents in the outward direction. It also inhibited the pacemaker currents in a dose-dependent manner. Because the effects of pine needle extract on pacemaker currents were the same as those of pinacidil (an ATP-sensitive $K^+$ channel opener) we tested the effect of glibenclamide (an ATP-sensitive $K^+$ channels blocker) on ICC exposed to pine needle extract. The effects of pine needle extract on pacemaker currents were blocked by glibenclamide. To see whether production of prostaglandins (PGs) is involved in the inhibitory effect of pine needle extract on pacemaker currents, we tested the effects of naproxen, a non-selective cyclooxygenase (COX-1 and COX-2) inhibitor, and AH6809, a prostaglandin EP1 and EP2 receptor antagonist. Naproxen and AH6809 blocked the inhibitory effects of pine needle extract on ICC. These results indicate that pine needle extract inhibits the pacemaker currents of ICC by activating ATP-sensitive $K^+$ channels via the production of PGs.
