A Study on the Conductivity of Polycrystalline Semiconductor Nickel Oxide (NiO의 Semiconductivity에 關한 硏究)

  • The conductivity of polycrystalline NiO is measured in the temperature range of $200^{\circ}C\; to\; 800^{\circ}C$ under oxygen pressures from $1.52{\times}10^2\; mmHg\; to\; 10^{-4}$ mmHg. The plots of the log ${\sigma}$ vs 1/T at constant oxygen pressure are found to be linear and the activation energies obtained from the slopes of these plots show that the energies are greater under high oxygen pressure than under low pressure. The transition points are found from the curves. The dependence of the conductivity on the $O_2$ pressure, in the above temperature range, is to be regular but it does not obey the theoretical expression, i.e.${\sigma}σ = K_{ox}P^{1/6}.$ The activation energies are calculated from the curves at the various condition.

  • Zn(II), Cd(II) and Hg(II) complexes with a general composition[$M(L)_2(X)_2$], where L=4-aminopyridine (4AP) and $X=NO_2{^-}$ were prepared under microwave irradiation. The metal complexes were characterized by elemental analyses, molar conductance, IR, Far-IR, electronic, NMR ($^1H$, $^{13}C$), XPS spectral and thermal studies. The spectroscopic studies reveal the composition, different modes of bonding, electronic transition, different chemical environment of C and H atoms and the electronic state of the metal atoms. On the basis of the characterization data, tetrahedral geometry is suggested for all the complexes. The free ligand (4-aminopyridine) and their metal complexes were screened against phytopathogenic fungi and bacteria in vitro and the activities were compared.

  • Extracellular nuclease(s) in buffalo rumen fluid were purified from strained rumen fluid by a procedure involving Seitz filtration, acetone fractionation and gel filtration on Sephadex G-100. The enzyme resolved into two peaks exhibiting both DNase and RNase activities. The molecular weight of enzyme corresponding to peaks I and II were approximately 30,000 and 12,000 respectively. The properties of enzymes from the two peaks, however, were same. Optimum temperature for both DNase and RNase activities was at $50^{\circ}C$. Whereas DNase activity was stable upto $60^{\circ}C$, RNase activity was stable only up to $50^{\circ}C$. DNase activity recorded two pH optima, one at pH 5.5 and the other at pH 7.0. RNase activity recorded a broad pH optimum between pH 6.0-8.0. pH stability of the enzyme coincided with pH optima for both the activities. DNase activity was stimulated by $Mg^{2+}$ and $Mn^{2+}$ and inhibited by $Fe^{2+}$, $Zn^{2+}$, $Hg^{2+}$ and $Ag^+$. RNase activity was also stimulated by $Mg^{2+}$ and $Mn^{2+}$ and inhibited by $Cu^{2+}$, $Fe^{2+}$, $Zn^{2+}$, $Hg^{2+}$ and $Ag^+$. Reducing agents stimulated both the activities.

  • The optimum pH and temperature for the purified extracellular enzyme activity were 8.0 and 37$^{\circ}C$, respectively. NaCl was required for the activation of the enzyme and optimum concentration was 0.5M. This enzyme activity was inhibited by HgC $l_2$, CoC $l_2$ and ZnC $l_2$ and stimulated by CaC $l_2$. The activity of enzyme was increased by L-cysteine and 2-mercaptoethanol, but decreased by ο-phenanthroline, $\rho$-CMB, EDTA and iodoacetate. The $K_{m}$ and $V_{max}$ values of extracellular enzyme appeared as 0.717% and 15.39U/mg, respectively.y.

Biosorption of Hg(II) ions from synthetic wastewater using a novel biocarbon technology

  • Mercury is a toxic pollutants present in different types of industrial effluents and is responsible for environmental pollution. Removal of Hg(II) ions from synthetic wastewater was studied using the activated biocarbon produced from the leaves of Tridax procumbens (Asteraceae). The particle size of the biocarbon (BC) is in the range of $100-120{\mu}m$. The effects of initial metal ion concentration, pH, contact time, and amount of biocarbon on the biosorption process were studied at temperature of $28{\pm}2^{\circ}C$. Batch experimental studies showed that an equilibrium time of 160 min was required for the maximum removal of Hg(II) at the optimized biocarbon dose of 2.5 g per 100 mL of synthetic wastewater. The optimum pH required for maximum removal (96.5%) of Hg(II) ions was found to be 5.5. The biosorption of metal ions onto activated biocarbon surface is probably via an ion exchange mechanism. The biocarbon can be regenerated with minimum loss. Further, it can be reused without any chemical activation. The findings of the research suggested that, the biocarbon produced from cost effective renewable resources can be utilized for the treatment of industrial wastewater.

Purification and Properties of Extracellular Protease from Streptomyces rimosus (Streptomyces rimosus가 생산하는 Protease의 정제와 특성)

  • Extracellular neutral pretense of Streptomyces rimosus producing oxytetracycline was purified by ammonium sulfate fractionation, DEAE Sephadex A-50 chromatography and Sephadex G-100 gel filteration, and was showed single band on the cathodic gel electrophoresis. The optimum pH and temperature of the enzyme were pH 8.0 and 6$0^{\circ}C$, respectively. The enzyme was activated about 80% in the presence of Co$^{2+}$ ion, and strongly inhibited by Hg$^{2+}$, Fe$^{2+}$ and chelatig agent, EDTA. Molecular weight of the enzyme was estimated to be 12, 000. The Km value of the enzyme of casein as a substrate was 2.7$\times$10$^{-4}$M.

High Performance of SWIR HgCdTe Photovoltaic Detector Passivated by ZnS

  • Short wave infrared (SWIR) photovoltaic devices have been fabricated from metal organic vapour phase epitaxy (MOVPE) grown n- on p- HgCdTe films on GaAs substrates. The MOVPE grown films were processed into mesa type discrete devices with wet chemical etching employed for meas delineation and ZnS surface passivatlon. ZnS was thermally evaporated from effusion cell in an ultra high vacuum (UHV) chamber. The main features of the ZnS deposited from effusion cell in UHV chamber are low fixed surface charge density, and small hysteresis. It was found that a negative flat band voltage with -0.6 V has been obtained for Metal Insulator Semiconductor (MIS) capacitor which was evaporated at $910^{\circ}C$ for 90 min. Current-Voltage (I-V) and temperature dependence of the I-V characteristics were measured in the temperature range 80 - 300 K. The Zero bias dynamic resistance-area product ($R_{0}A$) was about $7500{\Omega}-cm^{2}$ at room temperature. The physical mechanisms that dominate dark current properties in the HgCdTe photodiodes are examined by the dependence of the $R_{0}A$ product upon reciprocal temperature. From theoretical considerations and known current expressions for thermal and tunnelling process, the device is shown to be diffusion limited up to 180 K and g-r limited at temperature below this.

A study on the characteristics of double insulating layer (HgCdTe MIS의 이중 절연막 특성에 관한 연구)

  • The double insulating layer consisting of anodic oxide and ZnS was formed for HgCdTe metal insulator semiconductor(MIS) structure. ZnS was evaporated on the anodic oxide grown in H$_{2}$O$_{2}$ electrolyte. Recently, this insulating mechanism for HgCdTe MIS has been deeply studied for improving HgCdTe surface passivation. It was found through TEM observation that an interface layer is formed between ZnS and anodic oxide layers for the first time in the study of this area. EDS analysis of chemical compositions using by electron beam of 20.angs. in diameter and XPS depth composition profile indicated strongly that the new interface is composed of ZnO. Also TEM high resolution image showed that the structure of oxide layer has been changed from the amorphous state to the microsrystalline structure of 100.angs. in diameter after the evaporation of ZnS. The double insulating layer with the resistivity of 10$^{10}$ .ohm.cm was estimated to be proper insulating layer of HgCdTe MIS device. The optical reflectance of about 7% in the region of 5.mu.m showed anti-reflection effect of the insulating layer. The measured C-V curve showed the large shoft of flat band voltage due to the high density of fixed oxide charges about 1.2*10$^{12}$ /cm$^{2}$. The oxygen vacancies and possible cationic state of Zn in the anodic oxide layer are estimated to cause this high density of fixed oxide charges.

Purification and Characterization of Cholesterol Oxidase Produced by Soil Microorganism HSL613 (토양 미생물 HSL613이 생산하는 Cholesterol Oxidase의 정제 및 특성)

  • The extracellular cholesterol oxidase produced from a soil microorganism HSL613 was purified and partially characterized. Through a series of purification procedures including concentration with CH2 concentrator, DEAE-cellulose column chromatography and gel filtration on Superose12, the purified enzyme was shown to have a specific activity of 108 units/mg protein giving 30.8-fold purification and final yield of 66%. The molecular weight of the enzyme was estimated to be 59,500 daltons by SDS-PAGE. The optimum temperature and pH for this enzyme were $50^{\circ}C$ and 6.0, respectively. The activity of the purified cholesterol oxidase was inhibited by $Ag^{2+}$, $Hg^{2+}$ and SDS.
