  • Three workers, field operators in lubricating oil processing of petroleum refinery industry were found unconscious by other worker. One of them who were exposed to an high concentration of H2S was presented with Glasgow Coma Score of 5, severe hypoxemia on arterial blood gas analysis, normal chest radiography, and normal blood pressure. On hospital day 7, his mental state became clear, and neurologic examination showed quadriparesis, profound spasticity, increased tendon reflexes, abnormal Babinski response, and bradykinesia. He was also found to have decreased memory, attention deficits and blunted affect which suggest general cognitive dysfunction, which improved soon. MRI scan showed abnormal signals in both basal ganglia and motor cortex, compatible with clinical findings of motor dysfunction. Neuropsychologic testing showed deficits of cognitive functions. SPECT showed markedly decreased cortical perfusion in frontotemporoparietal area with deep white matter. Another case was recovered completely, but the other expired the next day.

  • We were trying to study the validity of Puffer fish's poison(Tetrodotoxin- TTX) to make a traditional Korean Medical treatment. The following conclusions were made after literary studies. 1. The first record of the puffer fish dates back 2000 years ago in the Chinese text Book of Mountain and Sea and other texts from the similar period. 2. Puffer fish's poison IS known as tetrodotoxin which is an amino perhydroquinazoline compound. It has a chemical formula of $C_{11}H_{17}N_3O_8$ in the hemiacetal structure and has the molecular weight of 319. 3. Tetrodotoxin (TTX) plays a role as potent neurotransmitter blocker by blocking the $Na^+$ -gate channel which hinders the influx of $Na^+$ ion into the cell. 4. Symptoms of the puffer fish poisoning ranges from blunted sense in the lips and tongue, occasional vomiting in the first degree to sudden descending of the blood pressure, apnea, and other critical conditions in the fourth degree. Intoxication of the puffer fish poison progresses at a rapid pace as death may occur after an hour and half up to eight hours in maximum. Typical death occurs after four to six hours. 5. Ways to treat the puffer fish poisoning include gastric irrigation, induce vomiting, purgation, intravenous fluid injection, and correcting electrolytic imbalance and acidosis. In cases of dyspnea, apply oxygen inhalation and conduct artificial respiration. 6. Tetrodotoxin (TTX) may be applied in treating brain disorders, ocular pain, excess pain in the large intestine and ileum, and relieving tension of the skeletal museles, neuralgia, rheumatism, arthritis, and etc. 7. In terms of Oriental medicine, the puffer fish poison has characteristics of sweet, warm, and poisonous. It's known efficacies are to tonify weakness, dispel damp, benefit the lower back, relieve hemorrhoid, kills parasites, remove edema, and so forth. And the puffer fish eggs processed with ginger are said to be effective against tuberculosis and lung cancer, thus, it's validity must be investigated and further research should be followed.

Dyeing Properties and Ultraviolet-cut Ability of Dyed Fabrics with Petasites japonicus Extract (머위 추출액에 의한 직물의 염색성과 자외선 차단성)

  • It is well known that the Petasites japonicus has been used for a long time medicine for the treatment of allergic diseases such as lacquer poisoning. However, the exact components and dyeing properties of its effects is still not known. Therefore, the purpose of this study was to investigate the dyeing property and ultraviolet-cut ability of silk and nylon fabrics that was dyed variously with the Petasites japonicus. The Petasites japonicus extract was done by boiling with distilled water at $100^{\circ}C$ for 1 hour. As mordanting agent, we used Aluminum potassium sulfate ($AlK(SO_4)_2{\cdot}12H_2O$), Copper(II) sulfate pentahydrate ($CuSO_5{\cdot}5H_2O$), Iron(II)Chloride ($FeCl_2{\cdot}4H_2O$). The best K/S value of dyeing temperature and time, all the fabrics were $100^{\circ}C$, 90min. Silk fabric was dyed yellow(0.8Y 7.6/2.2) and nylon fabric was dyed reddish yellow(10.1 YR 7.4/3.0). Silk fabric and nylon fabric was changed greenish yellow on mordanting with $CuSO_5{\cdot}5H_2O$ and $FeCl_2{\cdot}4H_2O$ respectively. And the colorfastness of washing and dry-cleaning was improved by using mordanting agent(4~5 grade). Ultraviolet-cut ability(UV-B) was showed more 90% in dyed nylon fabrics.

  • The paralytic shellfish poisoning was occurred among 25 laborers who worked at breaking-up of ships in Pusan for 5 days from March 29 to April 2 of 1956. For the purpose of accurately defining the paralytic shellfish poison(PSP) , the authors carried out mouse bioassay and chemical analysis. The results were summarized as follows: 1. The mean amount of Paralytic shellfish toxin was $1,207.8{\mu}g$ Per 100gm meat, and the mean death time of mouse was 5 minutes 16 second. 2. The properties of the PSP were mainly gonyautoxin group by chemical analysis(TLC, IR, $^{1}H-NMR$).

Isolation of Major foodborne Pathogenic Bacteria from Ready-to-Eat Seafoods and Its Reduction Strategy (해산물식품 중 식중독원인균의 오염패턴 및 저감화 방안)

  • The contamination frequency of major foodborne pathogenic bacteria was investigated from 213 seafood samples including sliced raw fish and shellfish in Busan and CyeongNam province area. Tested microorganisms were Salmonella spp. Staphyloroccus aureus, Vibrio parahaemolyticus, Escherichia coli O157:H7, Bncillus cereus, Listeria monocytogenes and Campylobacter jejuni. The frequency of isolated microorganisms was V. parahaemolyticus (30.5%), B. cereus (9.9%), S. aureus (3.8%) and other pathogenic bacteria (1.4%). from July to October, total isolation rates were greater than 50% and V. parahaemolyticus was dominant among the microorganisms isolated. The bacteria isolation rate (49.2%) in raw seafoods including shellfishes was higher than one (28.9%) in sliced raw fish. V. parahaemelyticus isolates were resistant to ampicillin (96.9%), amikacin (29.2%) and tetracycline (27.7%), and B. cereus isolates were resistant to ampicillin (100%), Penicillin G (100%), rifampicin (71.4%) and tetracycline (14.3%). The growth of V. parahaemolyticus and B. rereus was greatly inhibited below $10^{\circ}C$, but increased at ambient temperature. Washing seafood with tap water showed to reduce total count of remaining V. parahaemolyticus. Thus temperature control under $10^{\circ}C$, sufficient washing and prompt eating appeared to reduce the risk of food poisoning by these bacteria in seafoods.

  • This study was carried out to get a industrial information about a possibility of IgY antibody production, antimicrobial activity and properties of IgY antibody in egg yolk. The residual antibody activities of IgY were 91.5% after heating for 30min at 6$0^{\circ}C$. At the same time. these activities, were 73.2% after heating 15min at $65^{\circ}C$ and decreased vapidly at 7$0^{\circ}C$ and little antibody activity was left after heating for 15min at 8$0^{\circ}C$. When the prepared IgY was incubated at various pH ranges from 7 to 2 for 5hr at 37$^{\circ}C$., the antibody activity was stable from pH 7 to 4 and remained to 69.8% at pH 3.

Identification of 19 Species of Poisonous Plants from Jeju Island and Construction of a Database Using DNA-barcoding (DNA-barcoding을 이용한 제주도 자생 독성 식물 19종의 종 식별 및 데이터베이스 구축)

  • Food poisoning accidents caused by poisonous plants occur every year. As certain poisonous plants are mistaken for edible plants causing food poisoning, accurate species identification of poisonous plants is required. DNA barcodes suitable for species identification of poisonous plants and database that can be used for accurate species identification are necessary for their use in forensic cases. In this study, species identification of 19 poisonous plants native to Jeju Island using seven DNA barcodes (trnH-psbA, trnL-trnF, trnL intron, rbcL, matK, ITS1-ITS4, 18S rRNA) was performed to construct a database containing sequence information and DNA barcode universality. trnL-trnF barcode and ITS1-ITS4 barcode were the easiest markers for PCR amplification and sequence retrieval, and the combination of the two barcodes enabled single species identification in 18 out of 19 plants. Therefore, when an investigation of unknown poisonous plants is requested, combination of trnL-trnF and ITS1-ITS4 barcodes is considered as a primary marker for species identification. The database of recommended DNA barcodes for each poisonous plant presented in this study will be helpful in plants poisoning cases.

Bactericidal Effects of Food-borne Bacteria using Chlorine Dioxide and Electrolyzed Water (이산화염소수와 전해수를 이용한 식중독균의 살균효과)

  • The present study investigated the bactericidal effects of chlorine dioxide (CD) and electrolyzed water (EW) on pathogenic bacteria, such as Bacillus cereus, Staphylococcus aureus, Salmonella Typhimurium, and Escherichia coli O157:H7, by treatment them with CD and EW, respectively, for 0, 2, 4, 6, 8, and 10 min. Additionally, the sensitivities of Gram-positive (B. cereus and S. aureus) and Gram-negative (S. Typhimurium and E. coli O157:H7) to CD and EW were compared, respectively. In CD, the D-values for B. cereus, S. aureus, S. Typhimurium, and E. coli O157:H7 were 1.85±0.64, 2.06±0.85, 2.26±0.89, and 2.59±0.40 min, respectively. In EW, the D-values for B. cereus, S. aureus, S. Typhimurium, and E. coli O157:H7 were 2.13±0.32, 1.64±0.64, 1.71±0.32, and 1.86±0.36 min, respectively. All strains decreased consistently for 10 min in both CD and EW. However, the D-values of each bacterial species did not differ significantly between CD and EW (P>0.05). When comparing the bactericidal effect of CD and EW, no difference in D-value was observed, even though the pH and available chlorine concentration of CD were significantly lower than those of EW. These data could be used for the application of CD and EW in the food industry, considering characteristics such as the selection of optimal disinfectants, determination of optimal concentrations, and sensitivity to disinfection targets.

Distribution of Hemolytic Vibrio sp. in Sea Water of the Beaches of Busan During Mid-summer (여름철 부산시 소재 해수욕장 해수의 용혈성 비브리오균 분포)

  • The distribution of hemolytic Vibrio sp. from sea water of three main beaches located in Busan (Gwangan(G), Haeundae(H) and Songjeong(S) beaches) was investigated from June to September 2006 ; this is mid-summer and the main season for bathing. The monthly detection ratio from each beach was 29.2% (7 of 24 samples, G), 33.3% (8 of 24 samples, H), and 16.7% (4 of 24 samples, S). The most probable number(MPN) of strains detected ranged from 1.8-36(G), 1.8-180(H) and 1.8-18(S) MPN/100mL. Of the isolated strains, 24 strains showed definite hemolytic activity. These 24 strains were identified as Vibrio fluvialis, Vibrio vulnificus, Aeromonas hydrophila, Actinobacillus ureae and Eikenella corrodens. Vibrio fluvialis was detected from all three beaches investigated. Vibrio vulnificus was detected from Haeundae and Gwangan beaches. Gwangan beach had a higher detection ratio of Vibrio sp. than Haeundae and Songjeong beaches. These results suggest that seafood harvested from the vicinity of theses beaches may cause food poisoning and risk management to prevent Vibrio septicemia is required, especially for Haeundae and Gwangan beaches.

A Study on the Mortality in Oxygen and Toxic Gas Concentration According using Experimental Animals (실험동물을 이용 산소 및 유해가스 농도에 따른 치사율 연구)

  • It may occur health hazards or death by suffocation or acute poisoning in case of oxygen deficiency in ambient or exposure to harmful gas. As a part of accident prevention, we studied the change of activity and lethal dose by changing the concentration of several hazardous gas with inhalation exposure chamber and laboratory animals. We investigated the lethality and motility change during either the 4 hrs whole body exposure to oxygen, nitrogen, toluene, $H_2S$, CO and 48 recovery. As results, it is estimated that 5% oxygen concentration as lethal concentration and 5.5% as $LC_{50}$ (rat, 4 hrs) with statistics for dose-response. The results of lethality in oxygen deficient condition (approximately 6%), the lethalities were 40%, 20% with 20 ppm $H_2S$, 600 ppm CO respectively, and was not increased the lethality with 8% CO. Thus, it was confirmed that the $H_2S$, CO had influence to lethal dose, while toluene had low fluence.
