백삼성분이 마우스 복강 탐식세포의 증식 및 Nitric Oxide 생성에 미치는 영향 (Effect of White Ginseng on the Proliferation of Mouse Peritoneal Macrophages and Their Nitric Oxide Synthesis)

  • 백삼성분이 탐식세포를 통한 면역반응조절에 어떠한 영향을 미치는가를 알아보기 위해 total saponin이나 ginsenoside Rb$_2$성분이 마우스 복강 탐식 세포의 증식과 nitric oxide(NO)분비 및 NOS 유전자 발현에 미치는 영향을 조사해 보았다. Total saponin 또는 ginsenoside Rb$_2$성분을 탐식세포 배양액에 0-256$\mu\textrm{g}$/$m\ell$ 농도로 첨가하여 배양한 후 $^3$H-thymidine incorporation법으로 분석을 실시한 결과 total saponin은 64$\mu\textrm{g}$/$m\ell$ 농도에서 세포내 DNA 합성을 증가시켰으며, ginsenoside Rb$_2$ 성분의 경우 16$\mu\textrm{g}$/$m\ell$ 또는 64$\mu\textrm{g}$/$m\ell$ 농도에서 DNA 합성이 증가되었다. 256$\mu\textrm{g}$/$m\ell$의 높은 농도에서 세포의 증식이 약간 저해되었지만 대조군에 비해 유의적인 감소는 보이지 않았다. NO 분비에 미치는 영향을 조사해 본 결과 20$\mu\textrm{g}$/$m\ell$ 농도의 백삼성분을 투여했을 때 NO분비가 대조군에 비해 유의적으로 증가하였다. Reverse transcription polymerase chain reaction (RT-PCR)을 이용하여 백삼성분에 의한NOS 유전자 발현의 정도를 조사한 결과 20$\mu\textrm{g}$/$m\ell$ 농도에서 nos gene발현이 대조군보다 증가되었다

염산/에탄올로 유도된 급성 위염 동물모델에서 증숙시간에 따른 홍삼의 보호 효과 (Protective effects of red ginseng according to steaming time on HCl/ethanol-induced acute gastritis)

  • 본 연구에서는 150 mM HCl/60 % ethanol로 급성 위염을 유발한 마우스에서 증숙 시간에 따른 홍삼의 위염 보효 효과에 대해 살펴보고자 하였다. 백삼과 홍삼의 증숙 시간에 따른 성분을 분석한 결과 사포닌, total polyphenol과 total flavonoid의 총 함량이 증숙 시간에 따라 증가하였고 6시간 증숙한 홍삼에서 가장 높은 함량을 보였다. 또한 1,1-diphenyl-2-picrylhydrazyl와 2,2'-azinobis-3-ethyl-benzothiazoline-6-sulfonic acid radical 소거능 실험을 통해 항산화 활성을 측정한 결과 RG 6에서 가장 높은 활성을 나타냈다. In vitro 실험 결과를 바탕으로 시료를 선택하였고 in vivo 실험을 진행하였다. 급성 위염 마우스 모델에 백삼과 6시간 증숙한 홍삼을 투여하였을 때, RG 6에서 위 점막 손상의 개선을 육안적으로 확인할 수 있었으며, 혈액에서 측정한 ROS 수치도 대조군에 비해 유의적인 감소를 보였다. 또한 염증성 사이토카인을 확인한 결과 대조군에 비해 RG6에서 감소하는 경향을 보였다. 이러한 결과들을 종합해 볼 때 증숙 시간에 따른 홍삼은 급성 위염 유발 마우스 모델에서 위염 보호 효과가 있는 것으로 사료된다.

Effects of Sodium Sulfite and Extrusion on the Nutritional Value of Soybean Products for Nursery Pigs

  • Three hundred nursery pigs were used in two growth assays (avg initial BW of 6.5 and 6.0 kg, respectively) to determine the effects of sodium sulfite ($Na_2SO_3$) as an extrusion aid for soy products used in diets for weanling pigs. In Exp. 1, treatments were arranged as $3{\times}2$ factorial with main effects of soy product [soybean meal (SBM), extruded SBM, and dry-extruded whole soybeans (DEWS)] and concentration of $Na_2SO_3$ (0 and 10 g/kg of soy product). The extruded SBM and DEWS treatments were processed in a dry extruder ($Insta-Pro^{(R)} $, Triple F Nutrition, Des Moines, IA) with barrel temperatures and throughputs of $169^{\circ}C$ and 578 kg/h, and $147^{\circ}C$ and 598 kg/h, respectively. All diets were formulated to 3.5 Mcal/kg DE, with 0.92% lysine for d 0 to 14, and 0.76% lysine for d 14 to 28. For d 0 to 14, there was a tendency for pigs fed diets with $Na_2SO_3$ to have greater ADG (p<0.08), and pigs fed SBM to have greater ADFI (p<0.02), thus pigs fed the extruded soy products has 15% greater gain/feed than those fed SBM (p<0.007). For d 14 to 28, there were no differences in ADG or gain/feed among pigs fed diets with SBM and those fed diets with the extruded soy products (p>0.15). However, pigs fed DEWS had greater ADG than pigs fed extruded SBM, and pigs fed $Na_2SO_3$ had greater ADG and ADFI compared to those not fed $Na_2SO_3$ (p<0.02 and 0.08, respectively). The positive response in ADG and gain/feed to the addition of $Na_2SO_3$ resulted with SBM and extruded SBM treatments, and not with DEWS (interaction effect, p<0.04). Overall (d 0 to 28), pigs fed DEWS had greater ADG (p<0.01) and gain/feed (p<0.08) than pigs fed extruded SBM. Also pigs fed diets with $Na_2SO_3$ had greater ADG, ADFI, and gain/feed compared to those fed diets without $Na_2SO_3$ (p<0.002, 0.04, and 0.04, respectively). Exp. 2 was designed as a $2{\times}3$ factorial with main effects of soy product (SBM and DEWS) and concentration of $Na_2SO_3$ (none, 7.5, and 15.0 g/kg of soy product). As in Exp. 1, all diets were formulated to 3.5 Mcal/kg DE, with 0.92% lysine for d 0 to 13, and 0.76% lysine for d 13 to 53. At a constant processing temperature (148 to $149^{\circ}C$, $Na_2SO_3$ increased throughput of the extruder (578, 595, and 602 kg/h for the 0, 7.5, and 15.0 g/kg additions, respectively). For d 0 to 13, treatment had no effect on ADG or ADFI, but gain/feed decreased for pigs fed SBM with increasing concentrations of $Na_2SO_3$, and increased for pigs fed DEWS with increasing concentrations of $Na_2SO_3$ (SBM vs DEWS sulfite quadratic interaction, p<0.03). For d 13 to 35, pigs fed DEWS had greater ADG (p<0.01) and gain/feed (p<0.001) than pigs fed SBM. Also, ADFI decreased and gain/feed increased with increasing concentrations of $Na_2SO_3$ (linear effects, p<0.04 and 0.01, respectively). Overall, pigs fed the diets with DEWS had greater ADG and gain/feed than pigs fed SBM (p<0.003 and 0.002, respectively), and $Na_2SO_3$ tended to decrease ADFI and increase gain/feed (linear effects, p<0.07 and 0.06, respectively). In conclusion, pigs fed DEWS had greater rate and efficiency of gain than pigs fed SBM. Also, adding $Na_2SO_3$ prior to extrusion increased yield and feed efficiency.

Effects of Amylase and Cellulase Supplementation in Sorghum-based Diets for Finishing Pigs

  • Three experiments were conducted to determine the effects of a sorghum-specific enzyme system, derived from an Aspergillus niger and Bacillus subtilis fermentation extract (carbohydrase activity of 1,650 $\alpha$-amylase units and cellulase activity of 30 fibrinolytic units/mL), on growth performance of finishing pigs. In Exp. 1,192 pigs (average initial BW of 46.1 kg) were fed sorghum-based diets without or with 360 mL of enzyme system per ton of sorghum in a 78 d growth assay. For d 0 to 39, gain/feed was improved (p<0.03) with enzyme supplementation, but ADG was not affected (p>0.15). For d 39 to 78 and overall (d 0 to 78), ADG, gain/feed, and digestibilities of DM and N were not affected (p>0.13) by enzyme supplementation. Backfat thickness, fat-free lean index, and scores for stomach keratinization and ulcers also were not affected (p>0.15) by the dietary treatments. In Exp. 2,168 pigs (average initial BW of 58.4 kg) were fed diets without or with 150, 300, or 450 mL/ton of the same enzyme system used in Exp. 1. Adding as much as 450 mL enzyme system / ton of sorghum did not affect (p>0.15) ADG or gain/feed for d 0 to 29 of the growth assay. However, during d 29 to 63, ADG increased by 11% (linear effect, p<0.02) and gain/feed increased by 10% (linear effect, p<0.06) as enzyme concentration was increased from none to 450 mL/ton of sorghum. For the overall period (d 0 to 63), ADG tended to increase (p<0.08) with enzyme supplementation, but gain/feed and digestibilities of DM and N were not affected (p>0.14). Carcass characteristics (dressing percentage, backfat thickness, and fat free lean index) also were not affected (p>0.20) by addition of the enzyme system. In Exp. 3,176 pigs (average initial BW of 46.7 kg) were fed diets without or with 450, 900, or 1,350 mL/ton of the same enzyme system used in Exp. 1 and 2 in a 71 d growth assay. Adding up to 1,350 mL/ton of enzyme had no effects (p>0.15) on ADG, gain/feed, digestibilities of DM and N, and carcass characteristics (dressing percentage, backfat thickness, and fat-free lean index). In conclusion, finishing pigs fed diets with a sorghum-specific enzyme system showed some positive trends for improved growth performance, but those effects were not large and (or) consistent.

  • [$PGF_{2{\alpha}}$]의 rat적출심방에 대한 작용을 ouabain의 작용과 비교하여 다음과 같은 결과를 얻었다. $PGF_{2{\alpha}}$의 양성변력성작용은 ouabain의 작용에 비해 potency가 강하고 efficacy는 낮았다. Ouabain은 $PGF_{2{\alpha}}$의 양성변시성작용과 유사한 양성변시성 경향을 나타내었는데 이 점에 대해 추후의 탐구가 요구된다. $PGF_{2{\alpha}}(3{\pm}10^{-8}M)$의 작용은 저칼슘농도(1.4mM)에서 ouabain$(3{\pm}10^{-3}M)$보다 현저히 $({\leq}0.05{\sim}p{\leq}0.00)$강하게 나타났고, medium내에 첨가되는 $Ca^{++}$에 대하여도 보다 예민하게 반응하였다. 저칼륨 medium(2.8mM) 또는 medium내 $K^+$첨가는 $PGF_{2{\alpha}}$보다 ouabain의 작용에 더 큰 영향을 미쳤다. $PGF_{2{\alpha}}$와 ouabain의 작용에 미치는 lidocaine ($1{\pm}10^{-5}M$이상의 고농도)의 영향은 매우 흡사하여 별다른 차이점을 볼 수 없었다. Propranolol $(3{\pm}10^{-6}M)$로써 전처치하여 아드레나린성 ${\beta}$수용체를 봉쇄한 적출심방에서 $PGF_{2{\alpha}}$ 양성변력성 및 양성변시성 작용은 방해를 받지 않았다. 이상의 결과로 보아 $PGF_{2{\alpha}}$는 ouabain의 $Na^+$, $K^+$-ATPase 억제기전파는 달리 $PGF_{2{\alpha}}$ 고유의 막수용체에 작용하여 $Ca^{++}$의 세포내 유입을 촉진시키는 기전으로 작용하는 것으로 추측되며, 그 수용체의 등정(identification)은 추후의 연구과제로 남는다.

제조방법이 다른 두 종류의 머루즙 첨가가 빵의 이화학적 및 관능적 특성에 미치는 영향 (Preparation and Characterization of Physicochemical and Sensory Properties of Bread Enriched with Two Types of Wild Grape Extract)

  • 강원도 삼척의 특산물인 머루의 활용도를 높이고 아울러 지역을 대표하는 향토식품을 개발하기 위한 일환으로, 본 연구에서는 머루즙을 첨가한 빵의 개발이 가능한지를 검토하였다. 이에 따라, 제조방법이 다른 두 종류의 머루즙(WGJ 머루즙과 WGS 머루즙)을 농도(머루즙 무첨가빵의 수분함량의 5, 10, 15, 20%)를 달리하여 첨가하여 빵을 제조한 후, 이화학적 및 관능적 특성을 머루즙 무첨가 빵과 비교함으로써, 머루의 기능성이 빵의 물성에 부정적 영향 없이 부가될 수 있는지를 평가하였다. 실험에 사용된 2종류의 머루즙은 머루를 고압 하에서 끓인 후 착즙한 머루 100%인 착즙액(wild grape juice, WGJ 머루즙)과 머루와 설탕을 동량으로 하고 100여 일 동안 저장함으로써 삼투압의 원리로 추출한 wild grape sugar mixture (WGS) 머루즙이었다. 본 연구의 결과는 크게 다음과 같이 요약할 수 있다. $\bullet$ WGJ 머루즙 vs. WGS 머루즙. WGJ 머루즙은 WGS 머루즙에 비해 수분, 조단백질, 조회분, 산도가 유의적으로 더 높았으며(p<0.05), 당도와 pH는 유의적으로 더 낮아(p<0.001), 제조방법이 다른 두 종류의 머루즙이 화학적 조성에 있어 차이가 있음을 확인하였다. $\bullet$ 머루즙 무첨가군 vs. 첨가군. 머루즙이 첨가된 반죽은 머루즙 농도에 비례하여 색은 짙어지고 pH는 감소하였다. 이는, 반죽의 색과 pH에 영향을 주는 머루즙 내의 안토시안과 유기산(주석산)에서 기인한 결과로 볼 수 있다. 머루즙 첨가는 빵 반죽의 경도(hardness), 점착성(gumminess), 저작성(chewiness)을 감소시켰고, 복원성(resilience)과 응집성(cohesivenss)은 증가시키는 결과를 보임으로써, 머루즙 첨가에 의해 빵 반죽의 내부적 결합력은 증가되고, 동시에 부드럽고 복원력이 개선된 반죽이 만들어졌음을 시사하였다. 그러나, 반죽의 이러한 물성은 제빵이 완료된 단계까지는 유지되지 않았다. 즉, 머루즙 첨가는 빵의 경도, 점착성, 저작성을 다소 증가시키는 경향을 나타내었으며, 이는 첫째, 제빵 이후 측정된 빵의 굽기 손실이 머루즙 첨가빵이 무첨가빵에 비해 상대적으로 높았다는 사실과 관련 있을 것으로 보인다. 또한 둘째, 머루즙 첨가에 의해 낮아진 pH가 반죽 단계에서는 발효속도 및 가스발생량에 영향을 주어 반죽의 텍스쳐를 개선시켰으나, 빵을 굽는 단계에서는 오히려 전분의 호화 및 팽윤을 일부 방해함으로써 빵의 경도를 증가시킨 것으로 일부 해석될 수 있다. 그러나, 제빵 상태에서 관찰된 이러한 경향은 무첨가빵과 비교할 때 유의적인 수준은 아니었으며, 머루즙 첨가는 오히려 빵의 탄력성(springiness)을 개선시킨 것으로 평가되었다. $\bullet$ WGJ 첨가군 vs. WGS 첨가군. 머루즙의 종류에 따른 반죽 및 제빵 특성을 비교하면, 머루즙 첨가에 의한 빵반죽의 색도 강화 및 pH 감소 경향은 동일 첨가농도에서 WGJ 첨가 반죽이 WGS 첨가 반죽에 비해 더욱 현저하게 나타났다. 이는, WGJ 머루즙이 WGS 머루즙에 비해 순수 머루의 함량이 높았으므로, 이에 따라 반죽의 색과 pH에 영향을 주는 머루즙 내의 안토시안과 주석산의 함량도 WGJ 첨가 반죽에서 상대적으로 더 높았기 때문이다. 머루즙 종류 및 첨가농도 의한 색과 pH의 이러한 변화 경향은 제빵 이후에까지 일관되게 유지되었다. 한편, 제조방법이 다른 두 종류의 머루즙에 의한 차이는 제빵 완료 이후 관능평가에서 비교적 뚜렷이 관찰되었다. 즉 WGJ 머루즙 첨가빵은 머루즙 첨가에 의해 빵의 부드러운 정도(consistency)와 촉촉한 정도(moistness)가 유의적으로 감소되어 무첨가빵에 비해 전반적 기호도(overall acceptability)가 낮게 평가되었으나, WGS 머루즙 첨가빵은 무첨가빵과 유의적인 차이를 나타내지 않았다. 따라서, 머루즙 종류에 의한 유의적 차이가 관찰된 관능 평가 결과를 고려한다면, WGJ 머루즙보다는 WGS 머루즙의 첨가가 더 바람직할 것으로 보인다. 또한, WGS 머루즙을 5% 첨가했을 때, 빵의 물성 및 관능적 특성이 무첨가빵과 유의적인 차이를 나타내지 않거나 혹은 개선된 현상이 관찰되었으므로, 빵의 품질 및 기호도의 유의적 저하없이 머루즙의 기능성이 제빵에 부가될 수 있을 것으로 사료되었다.

서비스 종업원의 표면행위가 반생산적 과업행동에 미치는 효과에 관한 연구: 감정소모의 매개효과를 중심으로 (The Effects of Service Employee's Surface Acting on Counterproductive Work Behavior: The Mediating Roles of Emotional Exhaustion)

  • Purpose - Counterproductive work behavior(CWB) was typically categorized according to the behavior whether it targets other people(i.e., interpersonal CWB: I-CWB). Employing organizations(i.e., organizational CWB: O-CWB) has emerged as major concerns among researchers, managers, and the general public. An abundance of researches has informed us about the understanding for the antecedents of CWB, whereas little is known about the antecedents of CWB directed distribution service in employee's emotional labor. Therefore, the purpose of this research is to propose a research model in which surface acting enhances emotional exhaustion as an emotional labor strategy, which eventually increases counterproductive work behavior(including I-CWM and O-CWB). Research design, data, and methodology - This empirical research data were gathered from the samples of full time frontline hotel employees(including front office, call center, food/beverage, concierge, and room service) in South Korea. Six hotels were selected ranged from four to five stars, including privately owned and joint-venture properties. A convenience sampling method was used to select hotels. Full time frontline hotel employees from the six hotels were surveyed using a self-administered instrument for data collection. With the strong support of hotel managers, a total of 300 questionnaires were distributed, and 252 responses were collected indicating a response rate of 84.0%. In the process of working with the 252 samples, structural equation modeling is employed to test research hypotheses(H1: The relationship between surface acting and Interpersonal counterproductive work behavior(I-CWB) is mediated by emotional exhaustion, H2: The relationship between surface acting and organizational counterproductive work behavior(O-CWB) is mediated by emotional exhaustion). SPSS 18.0 and M-Plus 7.31 software were used for the data analysis. Descriptive statistics were used to assess the distribution of the employee profiles and correlations between factors. M-Plus 7.31 software was used to test the model fit, validity, and reliability of the factors, significance of the relationship between factors, and the effects of factors in the model. Results - To test our mediation hypotheses, we used an analytical strategy suggested by Preacher & Hayes (2008) and Shrout & Bolger (2002). This mediation approach directly tests the indirect effect between the predictor and the criterion variables through the mediator via a bootstrapping procedure. Thus, it addresses some weaknesses associated with the Sobel test. We found that surface acting was positively related to emotional exhaustion. Furthermore, emotional exhaustion was a significant predictor from the two kinds of counterproductive work behavior. In addition, surface acting was not significantly associated with the two kinds of counterproductive work behavior. These results indicated that the surface acting by frontline hotel employees was associated with higher emotional exhaustion, which is related with higher interpersonal counterproductive work behavior(I-CWB) and organizational counterproductive work behavior(O-CWB). In sum, we confirmed that the positive relationship between surface acting and the two kinds of counterproductive work behavior was fully mediated by emotional exhaustion. Conclusions - The current research broadens the conceptual work and empirical studies in counterproductive work behavior literature by representing a fundamental mechanism that how surface acting affects counterproductive work behavior.

황칠나무 잎 추출물의 세포 항산화 활성과 미백활성 측정 (Cellular Antioxidant Activity and Whitening Effects of Dendropanax morbifera Leaf Extracts)

  • 본 연구에서는 황칠나무 잎 추출물의 HaCaT 세포에서의 항산화 활성과 B16F1 melanoma 세포에서의 미백활성을 측정하였다. HaCaT 세포를 이용한 실험에서, 황칠나무 잎 추출물의 에틸아세테이트 분획과 아글리콘 분획은 각각 50 및 $25{\mu}g/ml$의 농도 이하에서 독성을 나타내지 않았다. UVB $800mJ/cm^2$를 HaCaT 세포에 조사하였을 때, 황칠나무 잎 추출물은 농도 의존적으로 자외선으로부터 세포를 보호하였다. 10 mM의 $H_2O_2$$4{\mu}M$의 rose bengal을 HaCaT 세포에 처리하였을 때, 에틸아세테이트 분획($6.25{\sim}50{\mu}g/ml$) 및 아글리콘 분획($6.25{\sim}25{\mu}g/ml$)은 우수한 세포 보호 효과를 나타내었다. 황칠나무 잎 추출물의 B16F1 melanoma 세포에서의 미백활성을 측정한 결과, 미백제로 알려진 알부틴 보다 더 우수한 멜라닌 합성 저해 활성을 보였다. 이상의 결과들은 황칠나무 잎 추출물이 ROS에 대항하여 세포를 보호함으로써 생체계, 특히 태양 자외선에 노출된 피부에서 세포보호제 및 천연항산화제로서 작용할 수 있음을 가르키며, ${\alpha}$-MSH로 유도된 멜라닌 생합성 저해 효과로부터 황칠나무 잎 추출물이 새로운 미백 화장품의 원료로써 이용 가능함을 알 수 있다.

Optimization of aircraft fuel consumption and reduction of pollutant emissions: Environmental impact assessment

  • Environmental impact of aircraft emissions can be addressed in two ways. Air quality impact occurs during landings and takeoffs while in-flight impact during climbs and cruises influences climate change, ozone and UV-radiation. The aim of this paper is to investigate airports related local emissions and fuel consumption (FC). It gives flight path optimization model linked to a dispersion model as well as numerical methods. Operational factors are considered and the cost function integrates objectives taking into account FC and induced pollutant concentrations. We have compared pollutants emitted and their reduction during LTO cycles, optimized flight path and with analysis by Dopelheuer. Pollutants appearing from incomplete and complete combustion processes have been discussed. Because of calculation difficulties, no assessment has been made for the soot, $H_2O$ and $PM_{2.5}$. In addition, because of the low reliability of models quantifying pollutant emissions of the APU, an empirical evaluation has been done. This is based on Benson's fuel flow method. A new model, giving FC and predicting the in-flight emissions, has been developed. It fits with the Boeing FC model. We confirm that FC can be reduced by 3% for takeoffs and 27% for landings. This contributes to analyze the intelligent fuel gauge computing the in-flight fuel flow. Further research is needed to define the role of $NO_x$ which is emitted during the combustion process derived from the ambient air, not the fuel. Models are needed for analyzing the effects of fleet composition and engine combinations on emission factors and fuel flow assessment.

Performance of carbon fiber added to anodes of conductive cement-graphite pastes used in electrochemical chloride extraction in concretes

  • Pollution of chloride ion-reinforced concrete can trigger active corrosion processes that reduce the useful life of structures. Multifunctional materials used as a counter-electrode by electrochemical techniques have been used to rehabilitate contaminated concrete. Cement-based pastes added to carbonaceous material, fibers or dust, have been used as an anode in the non-destructive Electrochemical Chloride Extraction (ECE) technique. We studied the performance of the addition of Carbon Fiber (CF) in a cement-graphite powder base paste used as an anode in ECE of concretes contaminated with chlorides from the preparation of the mixture. The experimental parameters were: 2.3% of free chlorides, 21 days of ECE application, a Carbon Fiber Volume Fraction (CFVF) of 0.1, 0.3, 0.6, 0.9%, a lithium borate alkaline electrolyte, a current density of $4.0A/m^2$ and a cement/graphite ratio of 1.0 for the paste. The efficiency of the ECE in the traditional technique using metal mesh as an anode was 77.6% and for CFVF of 0.9% it was 90.4%, with a tendency to increase to higher percentages of the CFVF in the conductive cement-graphite paste, keeping the pH stable and achieving a homogeneous ECE in the mass of the concrete contaminated with chlorides.
