  • Objectives : Jasminum officinale L. var. grandiflorum is used as a traditional or folk remedy in China to treat arthritis, hepatitis, duodenitis, conjunctivitis, gastritis, and diarrhea. In this study, we aimed to study the hepatocyte protective activity and molecular mechanism of Jasminum officinale L. var. grandiflorum aqueous extract (JGW) using HepG2 hepatocyte cell lines. Methods : HepG2 cells were pretreated with diverse concentrations of JGW, and then the cells were exposed to tert-butyl hydroperoxide (tBHP) for inducing oxidative stress. Hydrogen peroxide (H2O2) production, glutathione (GSH) concentration, mitochondrial membrane potential (MMP) and cell viability were measured to investigate hepato-protective effects of JGW. Phosphorylation of AMP-activated protein kinases (AMPK), acetyl coenzyme A carboxylase (ACC) and effects of compound C on cell viability were examined to observe the role of AMPK on JGW-mediated cytoprotection. Results : Pretreatment with JGW (10-300 ㎍/mL) significantly suppressed cytotoxicity induced by tBHP in a concentration dependent manner and reduced the expression of cleaved PARP and cleaved caspase-3 proteins related to apoptosis in HepG2 cells. In addition, pretreatment with JGW significantly prevented the increase in H2O2 production, GSH depletion, and lower MMP induced by tBHP. Treatment with JGW (30 minutes of incubation and concentrations of 100 and 300 ㎍/mL) increased the phosphorylation of AMPK and ACC and treatment with compound C, a chemical inhibitor of AMPK, inhibited the cytoprotective effect of JGW. Conclusions : Our results demonstrated that JGW may protect hepatocytes from oxidative stress via activation of AMPK.

  • $\beta-SiC $powder with or without the addition of 1 wt% of $\alpha-SiC$ particles (seeds) was pressureless-sintered at $1950^{\circ}C$ for 0.5, 2 and 4 h using $Y_3Al_5O_{12}$ (yttrium aluminum garnet, YAG) as a sintering aid. The introduction of $\alpha-SiC$ seeds into $\beta-SiC$ accelerated :he grain growth of elongated large grains during sintering, resulting in the coarser microstructure. The fracture toughnesses of materials with $\alpha$-SiC seeds and without $\alpha-SiC$ seeds sintered for 4 h were 7.5 and 6.1 $MPa\cdot \textrm m^{1/2}$, respectively. Higher fracture toughness of the material with seeds was due to the enhanced bridging by elongated grains, resulting from coarser microstructure.

  • We investigated a flexible transparent film using the spinning multi-walled carbon nanotubes (MWCNTs). Spin-capable MWCNTs on iron catalyzed on a SiO2 wafer was grown by chemical vapor deposition, which was performed at $780^{\circ}C$ using C2H2 and H2 gas. The average diameter and length of MWCNTs grown on the substrate were ~15 nm and $250{\sim}300{\mu}m$, respectively. The MWCNT sheets were produced by continuously pulling out from well-aligned MWCNTs on a substrate. The MWCNT sheet films were produced simply by direct coating on the flexible film or grass. The thickness of sheet film was remarkably decreased by alcohol spraying on the surface of sheet. The alcohol splay increased transmittance and decreased electrical resistance of MWCNT sheet films. Single and double sheets were produced with sheet resistance of ~699 and ${\sim}349{\Omega}/sq$, respectively, transmittance of 81~85 % and 67~72%, respectively. The MWCNT sheet films were heated through the application of direct current power. The flexible transparent heaters showed a rapid thermal response and uniform distribution of temperature. In addition, MWCNT yarns were prepared by spinning a bundle of MWCNTs from vertically super-aligned MWCNTs on a substrate, and field emission from the tip and side of the yarns was induced in a scanning electron microscope. We found that the field emission behavior from the tip of the yarn was better than the field emission from the side. The field emission turn-on voltages from the tip and side of MWCNT yarns were 1.6 and $1.7V/{\mu}m$, respectively, after the yarn was subjected to an aging process. Both the configuration of the tip end and the body of the yarn were changed remarkably during the field emission. We also performed the field emission of the sheet films. The sheet films showed the turn on voltage of ${\sim}1.45V/{\mu}m$ during the field emission.

  • Background : To evaluate the efficacy of two methods of obtaining lung recruitment to reduce ventilator-induced lung injury(VILI). Methods : Fifteen New-Zealand white rabbits were ventilated in the pressure-controlled mode while maintaining constant tidal volume(10 ml/kg) and fixed respiration rate. Lung injury was induced by repeated saline lavage (PaO2<100 mmHg), and the pressure-volume curve was drawn to obtain Pflex. The animals were then randomly assigned to three groups and ventilated for 4 hours. In the control group(n=5), positive end-expiratory pressure(PEEP) less than that of Pflex by 3 mmHg was applied throughout the study. In the recruitment maneuver(RM) group(n=5), RM(CPAP of 22.5 mmHg, for 45 seconds) was performed every 15 minutes in addition to PEEP level less than Pflex by 3 mmHg This phrase is unclear. In the Pflex group, PEEP of Pflex was given without RM. Gas exchange, lung mechanics, and hemodynamics parameters as well as pathology were examined. Results : 1) Both the control and RM groups showed decreasing tendency in PaO2 with time. There was significantly decreased PaO2 at 4 hr compared to Ihr(p<0.05). But in the Pflex group, PaO2 did not decrease with time(p<0.05 vs other groups at 3, 4 hr). PaCO2 did not show significant difference among the three groups. 2) There was no significant difference in static compliance and plateau pressure. Mean blood pressure and heart rate also did not show any significant difference among the three groups. 3) The pathologic exam showed significantly less neutrophil infiltration in the Pflex group than in the control group(p<0.05). There was borderline significant difference in hyaline membrane score among the groups (p= 0.0532). Conclusion : Although recruitment maneuver of the injured lung may be important in decreasing VILI, it alone may not be sufficient to minimize VILI.

  • In this study physicochemical characteristics and stability of various manganese coated sands (MCS) prepared with different methods were evaluated. In addition, removal efficiencies of As(III) by each MCS were compared. Four different MCSs were used; B-MCS prepared by baking method, W&D-MCS prepared by wetting and dry method, NMCS prepared during the water treatment process and Birm which is a commercial MCS widely used for the removal iron and manganese. The manganese content in each MCS was following order: Birm (63,120 mg/kg) > N-MCS (10,400 mg/kg) >W&D-MCS (5,080 mg/kg) > B-MCS (2,220 mg/kg). Birm showed the least solubility (% basis) in acidic conditions. As(III) oxidation efficiency of B-MCS was continuously increased as the solution pH decreased. While As(III) oxidation efficiency of N-MCS and Birm was minimum around neutral pH. The increased As(III) oxidation efficiency above neutral pH for N-MCS and Birm could be due to the competitive adsorption of $Mn^{2+}$, which was produced from reduction of $MnO_2$, onto the surface of aluminum and manganese oxides.

  • In order to prepare SPI tofu without compression step, amounts of water added to SPI suspension was studied for textural properties of uncompressed SPI tofu prepared by first heating at $100^{\circ}C$ for 6 minutes and second heating at $75^{\circ}C$ for 25 minutes and use of $CaSO_{4}-GDL$(0.07g, 0.0075 g/g SPI) as coagulants. The hardness and uniformity were gradually increased as the water addition ratio $(gH_{2}O/g\;SPI)$ raised from 6.0 to 8.0 and cohesiveness was rather decreased. The increase in second heating time increased the hardness and gumminess and relativity higher values in hardness were measured for those tofu heated at $85^{\circ}C$ than those at $75^{\circ}C$ or $95^{\circ}C$. A multiple regression equation calculated and RSM figure showed that the effects of water addition ratio was become to be less as the heating time and temperature increased. Addition of 8 g of water per g SPI and second heating at $85^{\circ}C$ for $30{\sim}60$ minutes were found as optimal conditions to prepare uncompressed SPI tofu.

  • In order to obtain the basic information for seedling production of surf clam, Spisula sachalinensis, spawning induction and egg development were investigated. $NH_4OH$ addition and serotonin injection could induce the spawning in surf clam. Water temperature rising, sperm suspension immersion, UV-ray irradiated seawater and $H_2O_2$ addition less affected on induction of spawning than $NH_4OH$ and serotonin did. On the other hand, males were more sensitive to the treatments than females. The response time to initial spawning in the case of $NH_4OH$ addition was $3\~4$ hours. However in the case of serotonin injection, it was within 5 minutes. The number of eggs released by $NH_4OH$ addition were significantly more than those released by serotonin injection. The serotonin injection induced higher rates of germinal vesicle breakdown than the $NH_4OH$ addition. Fertilizing and hatching rates of the eggs also were the similar results. Eggs of surf clam were demersal isolated eggs and averaging $65.2{\pm}±1.8\;{\mu}m$ in diameter after spawning. Optimum range of water temperature for the development of egg was $15\~20^{\circ}C$, The required time for development of D-shaped larvae was 42 hours at $15^{\circ}C$ and 27 hours at $20^{\circ}C$, respectively.

  • Reactive oxygen species (ROS) are produced by oxidative stresses which cause various chronic diseases such as diabetes and obesity. Ginseng (Panax ginseng C.A. Mayer) has been reported to contain various biological activities such as anti-cancer, anti-diabetic, neuroprotective, radioprotective, anti-amnestic and anti-aging effects. In this study, we investigated the effects of Panax ginseng, treated with high temperatures and high pressures, on oxidative stress in C2C12 myoblasts and 3T3-L1 adipocytes. Oxidative stress was induced in the C2C12 cells through the introduction of $H_2O_2$ (1 mM), and cells were then treated with various ginseng preparations: dried white ginseng (DG), steamed ginseng (SG) and high temperature and high pressure treated ginseng (HG). In addition, 3T3-L1 preadipocytes were treated with various ginsengs for up to 8 days following standard induction of differentiation. Our results show that HG treatment significantly protected oxidative stress in both cell lines and enhanced gene expression of antioxidant enzymes. Therefore, in this study, we investigated the protective effects of ginseng on the oxidative stress of adipocytes and muscle cells.

  • Objectives : The aim of this study was to evaluate the effect of aqueous extracts of Mume Fructus(MF) on the 6-n-propyl-2-thiouracil(PTU)-induced rat hypothyroidism. Methods : Aqueous extracts of MF(yield = 19.38%) were administered, once per day for 42 days from 2 weeks before starting of PTU treatment as an oral dose of 300 and 150 ㎎/㎏(body weight), and hypothyroidism was induced by daily subcutaneous treatment of PTU 10 ㎎/㎏ for 28 days. The changes in the body weight, thyroid gland weights, liver weight, serum levels of thyroid hormone-thyroid stimulating hormone(TSH), tri-iodothyronine(T3) and thyroxine(T4), total cholesterol, low density lipoprotein(LDL), high density lipoprotein(HDL) and triglyceride (TG), aspartate aminotransferase(AST), alanine aminotransferase(ALT), liver antioxidant defense system-lipid peroxidation, H2O2, superoxide dismutase(SOD) and catalase(CAT) were examined with histopathology of thyroid glands and liver. Results : Results were compared with LevoT40.5 ㎎/㎏ treated rats. MF extracts recovered from the decreases in the body weight, liver weight, T3and T4, TG, liver CAT activities as results of PTU treatment. And MF extracts recovered from the increases of thyroid gland weights, TSH, HDL contents, liver H2O2, AST as results of PTU treatment. In addition, these PTU-induced histopathological changes in thyroid glands and liver related to hypothyroidism were dramatically decreased by treatment of both different dosages of MF extract, respectively. Conclusions : According to the above results, it is suggested that MF extracts have advantageous effects on the thyroid hormone productions with beneficial effects on the hypothyroidism related liver injuries mediated by the modulation on the antioxidant system.

  • The objective of this research was to investigate the effect of immersion in Rhus verniciflua Stokes extract (RVSE) on the total reducing ability (TRA), protein oxidation and myoglobin oxidation of beef steak (Hanwoo longissimus muscle) stored with high oxygen-modified-atmosphere packaging (MAP) (HOMAP, 75% $O_2$+20% $CO_2$+5% $N_2$) and low oxygen-MAP (LOMAP, 0% $O_2$+20% $CO_2$+80% $N_2$) at $4^{\circ}C$ for nine days. RVSE induced TRA (p<0.05), metmyoglobin (MetMb) formation, and the CIE $H^{\circ}$ value but reduced the carbonyl content and R630-R580, as an index of the intensity of redness by oxymyoglobin, and the CIE $L^*$,$a^*$, and $C^*$ values. HOMAP maintained a lower TRA, MetMb concentration, and CIE $H^{\circ}$ value but had higher R630-R580 and CIE $L^*$, $a^*$, and $C^*$ values compared to LOMAP. Therefore, RVSE induced TRA and protein oxidation stability but reduced myoglobin stability in Hanwoo beef steak. In addition, the effects of HOMAP were opposite those of RVSE.
