Negligible Effect of Ginkgo Biloba Extract on the Pharmacokinetics of Cilostazol

  • Ginkgo biloba (G. biloba) extract is a widely used phytomedicine for the oral treatment of peripheral vascular disease. Cilostazol is a synthetic antiplatelet and vasodilating agent for the treatment of intermittent claudication resulting from peripheral arterial disease. It is likely to use concomitantly G. biloba extract and cilostazol for the treatment of peripheral arterial disease, which raises a concern of increasing their adverse effects of herbal-drug interactions. To clarify any possible herbal-drug interaction between G. biloba extract and cilostazol, the effect of the G. biloba extract on the pharmacokinetics of cilostazol was investigated. As cilostazol is known to be eliminated mainly by cytochrome P450 (CYP)-mediated metabolism, we investigated the effects of G. biloba extract on the human CYP enzyme activities and the effect of G. biloba extract on the pharmacokinetics of cilostazol after co-administration of the two agents to male beagle dogs. The G. biloba extract inhibited more or less CYP2C8, CYP2C9, and CYP2C19 enzyme activities in the in vitro microsomal study with $IC_{50}$ values of 30.8, 60.5, and $25.2{\mu}g/ml$, respectively. In the pharmacokinetic study, co-administration with the G. biloba extract had no significant effect on the pharmacokinetics of cilostazol in dogs, although CYP2C has been reported to be responsible for the metabolism of cilostazol. In conclusion, these results suggest that there may not be a pharmacokinetic interaction between G. biloba extract and cilostazol.

L-Phenylalanine Production by Regulatory Mutants of Excherichia coli K-12 (Escherichia coli K-12 대사조절 변이주에 의한 L-페닐알라닌 생산)

  • In order to overproduce L-phenylalanine, various kind of regulatory mutants were isolated from parental Escherichia coli K-12. MWEC 83 Producing 7.4g/l of L-phenylalanine has been derived as a tyrosine and tryptophan double auxotrophic mutant. To produce L-phenylalanine without adding L-tyrosine and L-tryptophan, revertant strain MWEC 101 was isolated from MWEC 83. Further various analogues and valine resistant mutants were isolated from MWEC 101. MWEC 101-5 was the most excellent strain that produced 17.9g/l of L-phenylalanine after having been cultivated for 54 hours in 15% glucose medium. It was disclosed that activities of rate-limiting enzymes including chorismate mutase and prephenate dehydratase in MWEC 101-5 were desensitized to 2mM L-phenylalanine in the enzyme reaction mixture and that activities level of 3-deoxy-D-arabino-heptulosonic acid-7-phosphate synthase and prephenate dehydratase were increased more than 20 times over those of the parental strain.

Molecular Analysis of the UGT1A1 Gene in Korean Patients with Crigler-Najjar Syndrome Type II

  • Purpose: Crigler-Najjar syndrome type II (CN-2) is characterized by moderate non-hemolytic unconjugated hyperbilirubinemia as a result of severe deficiency of bilirubin uridine diphosphate-glucuronosyltransferase (UGT1A1). The study investigated the mutation spectrum of UGT1A1 gene in Korean children with CN-2. Methods: Five Korean CN-2 patients from five unrelated families and 50 healthy controls were enrolled. All five exons and flanking introns of the UGT1A1 gene were amplified by polymerase chain reaction (PCR) and the PCR products were directly sequenced. Results: All children initially presented with neonatal jaundice and had persistent indirect hyperbilirubinemia. Homozygous p.Y486D was identified in all five patients. Three patients had an associated homozygous p.G71R and two a heterozygous p.G71R. The allele frequency of p.Y486D and p.G71R in healthy controls was 0 and 0.16, respectively. No significant difference in mean serum bilirubin levels was found between homozygous carriers of p.G71R and heterozygous carriers. Conclusion: The combination of homozygous p.Y486D and homozygous or heterozygous p.G71R is identified. The p.Y486D and p.G71R can be screened for the mutation analysis of UGT1A1 in Korean CN-2 patients.

Effects of Noise and Vibration on Oxygen Consumption and Ammonia Excretion in Cultured Catfish (Silurus asotus) (양식장에서 사육하는 메기 (Silurus asotus)의 산소소비 및 암모니아 배설에 미치는 소음진동 영향)

  • The main objective of this investigation was to examine oxygen consumption and ammonia excretion within cultured catfish, Silurus asotus ($22.9{\pm}0.9cm$, $100.7{\pm}11.7g$ n = 30) by noise and vibration stress in aquaculture farm. The vibration of 48, 58 and 68 dB (V) and noise of $77.6{\pm}1.8dB$ (A) from an electric vibrator was turned on for 15 minutes during each hour each day(0800-1800) for 11 days experimental period. The oxygen consumption (OC) of S. asotus the beginning of the experiment (0 day) in 58 dB group after 1, 5, 9 and 11 days was decreased 21.8, 30.2, 36.0 and 53.2%, respectively. In 68 group after 1, 7 and 11 days was decreased 22.7, 35.1 and 57.7%, respectively. The OC decreased exponentially and the relationship between them was expressed as OC = 0.374D + 90.762 ($r^2=0.048$) at 48 dB, OC = -3.581D + 89.520 ($r^2=0.831$) at 58 dB and OC = -4.109D + 90.907 ($r^2=0.884$) at 68 dB. Ammonia excretion (AE) of the beginning of the experiment in 48, 58 and 68 groups after 1 day was increased by 34.8, 51.8 and 63.2%, respectively, but it was decreased significantly from 3 to 11 days. The AE increased exponentially and the relationship between them was expressed as AE = -1.646D + 115.915 ($r^2=0.265$) at 48 dB, AE = -8.230D + 122.132 ($r^2=0.750$) at 58 dB and AE = -7.086D + 123.690 ($r^2=0.614$) at 68 dB.

Activity Profiles of Linear, Cyclic Monomer and Cyclic Dimer of Enkephalin

  • The cyclic dimers of enkephalin were isolated as minor components during the solution synthesis of the corresponding cyclic monomers. The ratio of cyclic dimer to monomer was approximately 1:4 from the percent of yields. In the receptor binding assay of two cyclic dimmers, ($Tyr_2-C[D-Glu-Phe-gPhe]_2$ 6, $Tyr_2-C[D-Asp-Phe-gPhe-rLeu]_2$ 8), both analogs exhibited the high preference for ${\delta}$ receptor compared to monocyclic counterparts. In the nociceptive activity, both showed about 5 times less potent than the cyclic monomers. The repeated synthesis of 14-membered cyclic analog, Tyr-C[D-Glu-Phe-gPhe-D-rLeu] 14, which was known as having three distinct cis-trans isomers, gave rise to apparently different conformational analog arousing only trans isomer. In the receptor binding assay, it showed tremendously high selectivity toward ${\mu}$ receptor $({\delta}/{\mu}=160)$.

On the Decomposition of Cyclic G-Brauer's Centralizer Algebras

  • In this paper, we define the G-Brauer algebras $D^G_f(x)$, where G is a cyclic group, called cyclic G-Brauer algebras, as the linear span of r-signed 1-factors and the generalized m, k signed partial 1-factors is to analyse the multiplication of basis elements in the quotient $^{\rightarrow}_{I_f}^G(x,2k)$. Also, we define certain symmetric matrices $^{\rightarrow}_T_{m,k}^{[\lambda]}(x)$ whose entries are indexed by generalized m, k signed partial 1-factor. We analyse the irreducible representations of $D^G_f(x)$ by determining the quotient $^{\rightarrow}_{I_f}^G(x,2k)$ of $D^G_f(x)$ by its radical. We also find the eigenvalues and eigenspaces of $^{\rightarrow}_T_{m,k}^{[\lambda]}(x)$ for some values of m and k using the representation theory of the generalised symmetric group. The matrices $T_{m,k}^{[\lambda]}(x)$ whose entries are indexed by generalised m, k signed partial 1-factors, which helps in determining the non semisimplicity of these cyclic G-Brauer algebras $D^G_f(x)$, where G = ℤr.

  • Amadori compounds(1-deoxy-1-amino-2-ketoses) are important precursors of color, flavor and aroma produced in foods. Amadori compounds occur naturally in tobacco. The contribution of amadori compounds to smoke quality has been (of) interest because of their roles of the Maillard reaction in the leaf chemistry. The amounts of these compound in tobacco are affected by the processes of aging, drying and storage conditions. In this study, eight compounds were chemically synthesized because amadori compounds (have not been sold commercially these days.) were not available for obtaining commercially. The aim of this study was to develop the analytical method of amadori compounds in tobacco leaf by the liquid chromatography mass spectrometry using triple quadrupole analyzer(LC-MS/MS). This method was simple, rapid, selective and sensitive, and eight amadori compounds were simultaneously and quantitatively analyzed within 20 minutes. This method showed excellent accuracy and precision. Recovery rates of amadori compounds ranged from 86% to 102%, with relative standard deviation(RSD) ranged from 2.6% to 5.9%. This method was applied to analysis of amadori compounds contents of tobacco leaves in different varieties. Furthermore, it was expected that the method could be extended to the analysis of other amadori compounds.

  • This research is to build the foundation of systematic application of color in cosmetology by analyzing color attributes in women's makeup presentation. The result were as follows. 1. The most popular color series in make up were R then RP and YR. The most popular color tone is 'd' and 'lt'. 2. Colors in make up according to age was as follows. For eye shadow, people aged 18 to 24 used 'lt' tone of the R color series; people aged 25 to 34 used 'lt', 's', 'sf tone of the R color series, 'lt' tone of the PB color series, 'lt' tone of the YR color series; people over 35 'g' tone of the YR color series, 'sf' tone of the P color series. For lipstick, people aged 18 to 24 used 'd' tone of the R color series; people aged 25 to 34 used 'd', 'sf' tone of the R color series; people over 35 used 'd' tone of the R color series. For lip-gloss, people aged 18 to 24 used 'v', 'lt', 'b', 's' tone of the R color series; people aged 25 to 34 used 's' 'd' 'dp' 'sf' tone of the R color series; people over 35 used 'b' tone of the R color series. 3. Make up colors according to marital status was as follows. For eye shadow, while married interviewees used 's', 'dk' tone of the R color series, single interviewees used 'lt', 'sf' tone of the R color series. For lipstick, while married interviewees used 'd', 'g' tone of the R color series, single interviewees preferred to use madder 'd', 'sf' tone of the R color series. For lip-gross, while married interviewees used 'd' tone of the R color series, single interviewees used 'b' tone of the R color series the most.

Hydrodynamic Characteristics in a Hexagonal Inverse Fluidized Bed (장방형 역유동층의 동력학적 특성)

  • Hydrodynamic characteristics such as gas holdup, liquid circulation velocity and bed expansion in a hexagonal inverse fluidized bed were investigated using air-water system by changing the ratio ($A_d$/$A_r$) of cross-sectional area between the riser and the downcomer, the liquid level($H_1$/H), and the superficial gas velocity($U_g$). The gas holdup and the liquid circulation velocity were steadily increased with the superficial gas velocity increasing, but at high superficial gas velocity, some of gas bubbles were carried over to a downcomer and circulated through the column. When the superficial gas velocity was high, the $A_d$/$A_r$ ratio in the range of 1 to 2.4 did not affect the liquid circulation velocity, but the maximum bed expansion was obtained at $A_d$/$A_r$ ratio of 1.25. The liquid circulation velocity was expressed as a model equation below with variables of the cross-sectional area ratio($A_d$/$A_r$) between riser to downcomer, the liquid level($H_1$/H), the superficial gas velocity($U_g$), the sparser height[(H-$H_s$)/H], and the draft Plate level($H_b$/H). $U_{ld}$ = 11.62U_g^{0.75}$${(\frac{H_1}{H})}^{10.30}$${(\frac{A_d}{A_r})}^{-0.52}$${(\frac({H-H_s}{H})}^{0.91}$${(\frac{H_b}{H})}^{0.13}$

  • This study aims to investigate the application possibility on natural gas in relation to the catalytic combustion of methane on Pd/cordierite catalyst which is currently used as an automobile converter catalyst. The surface area of the catalyst tested was determined to be about 18.7㎡/g and to keep stable condition in structure at mid-high temperatures. The activation energy for methane combustion reaction was estimated to be 19.2 kcal/mol and a hysterisis on the catalyst activity was observed in terms of the catalyst deactivation as the reaction temperature was varied for the methane combustion. On Pd/cordierite catalyst, The characteristics of methane combustion were studied as functions of space velocity and air/fuel ratios below 700$^{\circ}C$.
