Toxic Evaluation and Chromatographic Analysis of Cucurbitacin D and F from Sorbaria sorbifolia (쉬땅나무(Sorbaria sorbifolia) 성분으로서 cucurbitacin D, F의 독성평가 및 정량)

  • Cucurbitacin D and F, the protostane type triterpenoid of S. sorbifolia, were isolated with chromatograpic method and used as the standard substances for quantitative analysis. The compounds were identified with $^1H$-NMR, FAB-MS and UV spectrophotometer. They were separated on YMC-Pack ODS-AQ(303)[$250{\times}4.6mm$ I.D., $S-5{\mu}m$, 120A] column by HPLC. Cucurbitacin F was detected at 10.73mg/kg in cortex of S. sorbifolia, but cucurbitacin D was not. The compounds were shown to exihibit significant cytotoxicity($ED_{50}$<$0.1{\mu}g/mL$) against several tumor cell lines and acute toxicity(cucurbitacin D: 4.7mg/kg/day, cucurbitacin F: 2.5mg/kg/day) against BDF-1 mouse.

Antioxidative Activity and Component Analysis of Prunella vulgaris L. Extract/Fractions (하고초 추출물의 항산화 활성 및 성분 분석)

  • In this study, the antioxidative effects and active component analysis of 50% ethanol extract, ethyl acetate fraction and aglycone fraction obtained from Prunella vulgaris L. were investigated. The free radical scavenging activities ($FSC_{50}$) was investigated at 50% ethanol extract ($15.25{\mu}g/mL$), ethyl acetate fraction ($8.68{\mu}g/mL$), and aglycone fraction ($8.25{\mu}g/mL$) respectively. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) in $Fe^{3+}-EDTA/H_2O_2$ system using the luminol-dependent chemiluminescence assay was investigated at 50% ethanol extract ($4.68{\mu}g/mL$), ethyl acetate fraction ($1.00{\mu}g/mL$), and aglycone fraction($1.02{\mu}g/mL$) respectively. In the cellular protective effect against $^1O_2$ induced cellular damage of human erythrocytes, extract/fractions of P. vulgaris L. were increased in a concentration dependent manner($1{\sim}25{\mu}g/mL$). Especially, ${\tau}_{50}$ of aglycone fraction at concentrations of $25{\mu}g/mL$ showed the most protective effects at 337.9 min. It's showed nine times higher (+)-${\alpha}$-tocopherol (${\tau}_{50}=38.7min$) as typical antioxidant in the $^1O_2$-induced photohemolysis of human erythrocytes. TLC and HPLC were used to analyse active components in the ethyl acetate fraction and aglycone fraction of P. vulgaris L. In ethyl acetate fraction, caffeic acid, rosmarinic acid, quercetin 3-${\beta}$-D-glucoside, rutin, kaempferol-3-O-rutinoside, astragalin (kaempferol-3-O-glucoside) were identified. In aglycone fraction, caffeic acid, rosmarinic acid, quercetin, kaempferol were identified. These results indicated that extract/fraction of P. vulgaris L. is may be used in cosmetics industry as natural antioxidants by quenching and/or scavenging $^1O_2$ and other ROS, and protecting cellular membranes.

Antioxidant Activities of Fractions Obtained from Dryopteris crassirhizoma, D. nipponensis and Polystichum lepidocaulon (관중, 참지네고사리 및 더부살이고사리 분획물의 항산화 활성)

  • This study was carried out to develop ferns as the natural antioxidant materials by graduating and extracting fronds of $Dryopteris$ $crassirhizoma$, $Dryopteris$ $nipponensis$, and $Polystichum$ $lepidocaulon$, which belong to Dryopteridaceae, using solvent, and analyzing the antioxidant effect of each fraction. The n-butanol fraction of $D.$ $crassirhizoma$ (550.0 $mg{\cdot}g^{-1}$), the ethyl acetate fraction of $D.$ $nipponensis$ (374.8 $mg{\cdot}g^{-1}$), and the n-butanol fraction of $P.$ $lepidocaulon$ (781.8 $mg{\cdot}g^{-1}$) showed relatively higher total contents of polyphenol. The chloroform fraction of $D.$ $crassirhizoma$ (72.9 $mg{\cdot}g^{-1}$), and the n-hexane fraction of $D.$ $nipponensis$ (72.9 $mg{\cdot}g^{-1}$) and $P.$ $lepidocaulon$ (154.5 $mg{\cdot}g^{-1}$) contained relatively higher total contents of flavonoids. DPPH radical scavenging activity was most excellent in the n-butanol fraction of $D.$ $crassirhizoma$ ($RC_{50}=0.02mg{\cdot}mL^{-1}$) and $P.$ $lepidocaulon$ ($RC_{50}=0.04mg{\cdot}mL^{-1}$), and the water fractions of $D.$ $nipponensis$ ($RC_{50}=0.01mg{\cdot}mL^{-1}$). ABTS radical scavenging activity was potent in the n-hexane and n-butanol fractions of $D.$ $crassirhizoma$ (each $RC_{50}=0.02mg{\cdot}mL^{-1}$), the ethyl acetate fraction of $D.$ $nipponensis$ ($RC_{50}=0.03mg{\cdot}mL^{-1}$), and the n-butanol fraction of $P.$ $lepidocaulon$ ($RC_{50}=0.06mg{\cdot}mL^{-1}$). There was the large amount of total polyphenol content in the n-butanol fraction of $D.$ $crassirhizoma$ and $P.$ $lepidocaulon$, and their radical scavenging activities were potent. Therefore, it was thought that biologically active substances of each fraction layer are required to be analyzed and used.

Total Polyphenol Content and Antioxidative Activity of Wild Grape (Vitis coignetiae) Extracts Depending on Ethanol Concentrations (에탄올 농도에 따른 머루(Wild Grape, Vitis coignetiae) 추출물의 폴리페놀 함량 및 항산화 활성)

  • As an oriental medicinal plant, wild grape (WG, Vitis coignetiae) has been known to contain abundant nutrients compared to grape (Vitis vinifera L.) and as a source of stilbenes, a kind of polyphenol. This study was performed to investigate the antioxidative activity of WG extracts by measuring electron donating ability (EDA), nitrite scavenging ability (NSA), super oxide dismutase (SOD)-like activity and total polyphenol content (TPC). The extracts were obtained using microwave-assisted extraction (microwave power: 90 W, extraction time: 5 min, extractant: water, 50 and 100% ethanol). EDAs, SOD-like activities and TPCs were the highest at 50% ethanol extracts while conversely lowest at 100% ethanol extracts. EDAs of 50% and 100% ethanol extracts at 1.6 g/dL concentration, $97.70{\pm}0.55\;and\;98.05{\pm}0.36%$, were higher than those of 0.1 and 1% L-ascorbic acids, $101.44{\pm}0.98%\;and\;99.43{\pm}0.78%$, respectively. At the concentration of 1.6 g/dL, 50% ethanol extract showed lower NSA (pH 1.2) than water extract unlike EDA, TPC and SOD-like activity. Regarding TPCs of WG extracts, the activities were the highest at 50% ethanol extracts (1.6 g/dL: $38.76{\pm}0.23$ mM gallic acid equiv.) followed by water and 100% ethanol extracts. The results suggest the usefulness of developing functional foods using antioxidative active compounds of WG with high polyphenol contents.

Antioxidative Activities of Whole Plant Extracts of Solanum nigrum L. (까마중(Solanum nigrum L.) 전초 추출물의 항산화 활성)

  • In this study, the antioxidative effects of 50% ethanol extract, ethylacetate fraction and aglycone fraction obtained from dried whole plant of Solanum nigrum L. were investigated. The free radical scavenging activities ($FSC_{50}$) were $215.46{\mu}g/mL$, $42.43{\mu}g/mL$ and $52.28{\mu}g/mL$, respectively. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) in $Fe^{3+}-EDTA/H_2O_2$ system were $25.25{\mu}g/mL$, $7.05{\mu}g/mL$ and $6.25{\mu}g/mL$, respectively. 50% ethanol extract and aglycone fraction showed the cellular protective effect against $^1O_2$ induced cellular damage of rabbit erythrocytes at $5{\sim}25{\mu}g/mL$, but not at high concentrations. These results indicated that S. nigrum extract/fractions could be used as an antioxidative agent. However, it could induce cellular damage at high concentrations. In conclusion, a special caution is required to use S. nigrum extracts as a cosmetic ingredient.

  • This paper proposes a dual band Voltage Controlled Oscillator(VCO) with a standard 0.3${\mu}{\textrm}{m}$ CMOS process to generate 1.07GHz and 2.07GHz. The proposed VCO architecture with 50% duty cycle circuit and a half adder(HA) was capable of producing a frequency two times higher than that of the conventional VCOs. The measurement results demonstrate that the gain of VCO and power dissipation are 561MHz/V and 14.6mW, respectively. The phase noises of the dual band VCO are measured to be -102.55dBc/Hz and -95.88dBc/Hz at 2MHz offset from 1.07GHz and 2.07GHz, respectively.

A Study on the ZnO Piezoelectric Thin Film SAW Filter for High Frequency (ZnO 압전 박막을 이용한 고주파 SAW 필터 연구)

  • ZnO thin films on glass substrate were deposited by RF magnetron reactive sputtering at 100 W, 1.33 Pa, Ar/O2=50 : 50, 200$^{\circ}C$, and a target/substrate distance of 4 cm. Crystallinities, surface morphologies, chemical compositions, and electrical properties of the films were investigated by XRD, SEM, AFM, RBS, and electrometer. All films showed a strong preferred c-axis orientation and the chemical stoichiometry. The propagation velocity of ZnO/IDT/glass of single electrode and double electrode types SAW filter was about 2,589 m/sec, 2,533 m/sec and insertion loss was a minimum value of about -11 dB and -21 dB, respectively.

  • This paper describes a visual object detection approach utilizing ensemble based machine learning. Object detection methods employing 1D features have the benefit of fast calculation speed. However, for real image with complex background, detection accuracy and performance are degraded. In this paper, we propose an ensemble learning algorithm that combines a 1D feature classifier and 2D DNF (Disjunctive Normal Form) classifier to improve the object detection performance in a single input image. Also, to improve the computing efficiency and accuracy, we propose a feature selecting method to reduce the computing time and ensemble algorithm by combining the 1D features and 2D DNF features. In the verification experiments, we selected the Haar-like feature as the 1D image descriptor, and demonstrated the performance of the algorithm on a few datasets such as face and vehicle.

Toxicity and Uptake of 2,4,6-Trinitrotoluene (TNT) in Contaminated Soils to Eisenia fetida (토양내 오염된 2,4,6-trinitrotoluene (TNT)의 Eisenia fetida에 대한 독성 및 생물흡수)

  • Toxicity and uptake of 2,4,6-trinitrotoluene (TNT) in three different soils (OECD soil, natural soil and loess) to earthworm (Eisenia fetida) were investigated at several different spiked concentrations of TNT (0 to 200 mg/kg for OECD and natural soils, and 0 to 35 mg/kg for loess) and for different exposure periods (7, 14, 21, and 28 d). The LC50 values for 7 d exposure were 160.1, 159.4, and 28.81 mg/kg for OECD soil, natural soil, and loess, respectively. The LC50 values for 14, 21, and 28 d exposure were almost the same as those for 7 d exposure, showing that 7 d exposure time was enough to decide the toxicity (LC50) of TNT to Eisenia fetida, because the highest concentration of TNT in earthworm body was observed within around 5 d. The LC50 and uptake of TNT in loess were higher than those in OECD and natural soil. The uptake of TNT to the earthworm were correlated well with the initial concentration of TNT in the soil and TNT porewater concentration (R2> 0.9 in OECD, natural, and loess). The concentration of TNT in earthworm body decreased after 5 d, possibly caused by natural degradation of TNT by soil bacteria as well as earthworm.

Development of Split-beam Acoustic Transducer for a 50 kHz Fish Sizing Echo Sounder (50 kHz 체장어군탐지기용 분할 빔 음향 변환기의 개발)

  • An improved split-beam transducer for a 50 kHz fish-sizing echo sounder was developed. The main objective of this study was to minimize the side lobe level in the beam pattern and the distance between acoustic centers for adjacent transducer quadrants in the geometrical arrangement of array elements while maintaining a given number of transducer elements and beam width. To achieve these goals, a 32-element planar array transducer ($6{\times}6$ array with one element in each corner missing) was designed using the Dolph-Chebyshev shading function to suppress side lobes in the array beam pattern and fabricated by arranging the inter-element spacing to be substantially equal to half the wavelength using the transducer element of 0.4 times the wavelength in diameter. The performance characteristics of this split-beam transducer were evaluated in the experimental water tank of $5m{\times}5m{\times}6m$ (length${\times}$height${\times}$width). In this study, the design goal of the beam width and side lobe level for transmitting a beam pattern was initially set at $21^{\circ}$ and -30 dB, respectively. However, the measured beam width at 3 dB was $21^{\circ}$ in both directions with side lobe levels of -24.7 dB in the horizontal plane and -25.6 dB in the vertical plane. The averaged beam width at -3 dB of the receiving beam patterns for four receiving quadrants was $31.4^{\circ}$. The transmitting voltage response was 161.5 dB (re $1{\mu}Pa$/V at 1 m) at 50.23 kHz with a bandwidth of 2.16 kHz, and the averaged receiving sensitivity for four receiving quadrants was -178.13 dB (re 1 V/${\mu}Pa$) at 49.8 kHz with a bandwidth of 2.64 kHz.
