담배 워형질체의 전기융합을 위하 pH 및 Ca2+ 농도 최적조건 설정 (Optimum Conditions of pH and Ca2+ Concentration for Electrofusion of Tobacco Protoplasts)

  • This study was carried out to optimize the concentration of Ca2+ and pH of fusion medium which affected electrofusion frequency of protoplasts isolated from Nicotiana tabacum L. (cv. BY4) mesophyll cells and callus. The protoplasts were electrofused in the fusion media containing two different Ca2+ concentrations and three different pH regions. Fusion frequency was lower in the fusion medium containing only 13% mannitol as osmotic stabilizer. However, higher degree of fusion frequency (47.3%) was observed in the fusion medium containing 50mM CaCl2 at pH 10.5 than any other conditions. Cell viability was decreased by Ca2+ and high pH treatment in the fusion media, while fusion frequency was increased. It is concluded that Ca2+ is involved in electrofusion of protoplasts.

Preparation and in Vitro Release of Melatonin-Loaded Multivalent Cationic Alginate Beads

  • The sustained release dosage form which delivers melatonin (MT) in a circadian fashion over 8 h is of clinical value for those who have disordered circadian rhythms because of its short halflife. The purpose of this study was to evaluate the gelling properties and release characteristics of alginate beads varying multivalent cationic species $(Al^{+++}, \; Ba^{++}, \; Ca^{++}, \; Mg^{++}, \; Fe^{+++}, \; Zn^{++})$. The surface morphologies of Ca- and Ba-alginate beads were also studied using scanning electron microscope (SEM). MT, an indole amide pineal hormone was used as a model drug. The $Ca^{++}, \; Ba^{++}, \; Zn^{++}, \; Al^{++}\; and\; Fe^{+++}\; ions\; except\; Mg^{++}$ induced gelling of sodium alginate. The strength of multivalent cationic alginate beads was as follows: $Al^{+++}\llFe^{+++} the induced hydrogel beads were very fragile and less spherical. Fe-alginate beads were also fragile but stronger compared to Al-alginate beads. Ba-alginate beads had a similar gelling strength but was less spherical when compared to Ca-alginate beads. Zn-alginate beads were weaker than Ca- and Ba-alginate beads. Very crude and rough crystals of Ba- and Ca-alginate beads at higher magnifications were observed. However, the type and shape of rough crystals of Ba- and Ca-alginate beads were quite different. No significant differences in release profiles from MT-loaded multivalent cationic alginate beads were observed in the gastric fluid. Most drugs were continuously released upto 80% for 5 h, mainly governed by the passive diffusion without swelling and disintegrating the alginate beads. In the intestinal fluid, there was a significant difference iq the release profiles of MT-loaded multivalent cationic alginate beads. The release rate of Ca-alginate beads was faster when compared to other multivalent cationic alginate beads and was completed for 3 h. Ba-alginate beads had a very long lag time (7 h) and then rapidly released thereafter. MT was continuously released from Feand Zn-alginate beads with initial burstout release. It is assumed that the different release rofiles of multivalent cationic alginate beads resulted from forces of swelling and disintegration of alginate beads in addition to passive diffusion, depending on types of multivalent ions, gelling strength and drug solubility. It was estimated that 0.2M $CaCl_2$ concentration was optimal in terms of trapping efficiency of MT and gelling strength of Ca-alginate beads. In the gastric fluid, Ca-alginate beads gelled at 0.2 M $CaCl_2$ concentration had higher bead strength, resulting in the most retarded release when compared to other concentrations. In the intestinal fluid, the decreased release of Ca-alginate beads prepared at 0.2 M $CaCl_2$ concentration was also observed. However, release profiles of Ca-alginate beads were quite similar regardless of $CaCl_2$ concentration. Either too low or high $CaCl_2$ concentrations may not be useful for gelling and curing of alginate beads. Optimal $CaCl_2$ concentrations must be decided in terms of trapping efficiency and release and profiles of drug followed by curing time and gelling strength of alginate beads.

Ca2+ Sensitivity of Anoctamin 6/TMEM16F Is Regulated by the Putative Ca2+-Binding Reservoir at the N-Terminal Domain

  • Anoctamin 6/TMEM16F (ANO6) is a dual-function protein with Ca2+-activated ion channel and Ca2+-activated phospholipid scramblase activities, requiring a high intracellular Ca2+ concentration (e.g., half-maximal effective Ca2+ concentration [EC50] of [Ca2+]i > 10 μM), and strong and sustained depolarization above 0 mV. Structural comparison with Anoctamin 1/TMEM16A (ANO1), a canonical Ca2+-activated chloride channel exhibiting higher Ca2+ sensitivity (EC50 of 1 μM) than ANO6, suggested that a homologous Ca2+-transferring site in the N-terminal domain (Nt) might be responsible for the differential Ca2+ sensitivity and kinetics of activation between ANO6 and ANO1. To elucidate the role of the putative Ca2+-transferring reservoir in the Nt (Nt-CaRes), we constructed an ANO6-1-6 chimera in which Nt-CaRes was replaced with the corresponding domain of ANO1. ANO6-1-6 showed higher sensitivity to Ca2+ than ANO6. However, neither the speed of activation nor the voltage-dependence differed between ANO6 and ANO6-1-6. Molecular dynamics simulation revealed a reduced Ca2+ interaction with Nt-CaRes in ANO6 than ANO6-1-6. Moreover, mutations on potentially Ca2+-interacting acidic amino acids in ANO6 Nt-CaRes resulted in reduced Ca2+ sensitivity, implying direct interactions of Ca2+ with these residues. Based on these results, we cautiously suggest that the net charge of Nt-CaRes is responsible for the difference in Ca2+ sensitivity between ANO1 and ANO6.

에스트로겐이 생쥐 초기배의 $\textrm{Ca}^{2+}$ 전류에 미치는 영향 (17 beta-Estradiol Increases Peak of $\textrm{Ca}^{2+}$ Current in Mouse Early Embryo)

  • 배분화과정시 나타나는 $Ca^{2+}$ 변화에 미치는 $E_2$의 영향을 알아보고자 whole cell voltage clamp 기법, 방사선 등위원소 면역측정법, 그리고 공초점 현미경을 통하여 $E_2$처리 후 나타나는 $Ca^{2+}$ 전류 변화 및 세포내 $Ca^{2+}$ 농도 변화를 조사하였다. 생쥐의 미성숙 난자는 난소의 난포를 천자하고, 배란난자는 과배란 처리 후 난관에서 회수하였다. 수정란은 과배란 처리 후 수컷 생쥐와 교미를 유도한 후 각각의 단계에 맞는 수정란을 채란하였다. 혈중 $E_2$의 농도는 심장을 천자하여 혈액을 채취한 후 배발달 단계와 호르몬 처리 시간이 일치하는 혈액만을 사용하였다. 본 실험의 결과를 요약하면 다음과 같다. 1. $E_2$처리시 미성숙난자의 제 1극체 형성률 (성숙의 지표)은 $E_2$를 처리하지 않은 난자(83% : 83/100)보다 $E_2$를 처리한 난자 (94%, 94/100)에서 유의적 (P<0.05)으로 높게 나타났다. 2. $E_2$를 처리하였을 때 $Ca^{2+}$ 내향전류의 변화는 -10 mV에서 -1.23$\pm$0.01 nA (n=15)에서 -1.50$\pm$0.03 nA (n=15)로 122% 상승함으로써 유의한 (P<0.05) 변화를 보였다. 3. $E_2$를 처리하지 않은 난자 및 수정란을 1로 한 후 $E_2$를 처리한 난자 및 수정란의 변화를 상대적인 값으로 표시하였다. $E_2$처리한 난자는 1.22$\pm$0.17 (n=10), $E_2$처리한 전핵배는 1.20$\pm$0.14 (n=10), $E_2$처리한 2세포기배는 1.07$\pm$0.01 (n=10), 4세포기배는 1.05$\pm$0.09 (n=10)를 나타냄으로써 수정란의 단계마다 $E_2$의 반응 결과가 차이가 남을 알 수 있었다. 4. $E_2$농도 곡선에서 PMSG 처리 후 $E_2$의 혈중농도는 계속적인 상승을 보이다가 배란시기에 최고치를 나타내었으며, 배란 후 다시 감소하여 8세포기에서는 급격한 감소현상이 나타났다. 이후 다시 상실기를 거쳐 배반포기 임신기간동안 $E_2$의 농도가 상승하였다. 5. $E_2$처리 후 세포내 $Ca^{2+}$ 농도변화의 결과로, $E_2$를 처리하지 않은 난자들의 세포내 $Ca^{2+}$ 농도는 836.4$\pm$131.2 (n=10), $E_2$를 처리한 난자들은 1736.4$\pm$192.0 (n=10)로써 유의한 (P<0.05) 차이를 보였다. 이상의 결과로부터 $E_2$처리에 의한 세포내 $Ca^{2+}$ 농도 상승은 $E_2$$Ca^{2+}$ 통로를 자극함으로써 세포바깥의 $Ca^{2+}$이 세포안으로 이동하여 나타나는 변화로 생각된다.

  • B. Stearothermophilus 단백질 분해 효소는 2가 금속 이온인 $Ca^{2+}$에 의하여 내열성이 향상되며 $75^{\circ}C$에서 최대 역가를 나타내었다. 2mM $Ca^{2+}$를 첨가하여 B. Stearothermophilus를 이용한 호열 ${\cdot}$ 호기성 소화공정 운전할 경우 각각 최대량 대비 51%와 27%의 DOC 및 세포외단백질의 분해가 이루어지는 것을 확인하였다.

응고제 종류와 농도에 따른 전지대두분 두부의 품질 (Quality Attributes of Whole Soybean flour Tofu Affected by Coagulant and Theirs Concentration)

  • 전지대두분(WSF) 두부의 응고제 종류와 농도별 수율은 $CaCl_2$, $CaSO_4$, GDL, $MgCl_2$, $Ca-gluconate+CaSO_4$, $GDL+CaSO_4$ 첨가구는 0.3%에서, $Ca-gluconate+MgCl_2$, $GDL+CaCl_2$ 첨가구는 0.4%에서, 그리고 $Ca-gluconate+CaCl_2$ 첨가구는 0.5%에서 각각 가장 높았으며 0.3% $GDL+CaSO_4$ 혼합응고제를 첨가하였을 때 4.48(g/g WSF)로 최고치를 나타내었다. 응고제 농도에 따른 WSF 두부의 색도는 응고제 농도가 증가함에 따라 L과 b값은 점차 증가한 후 감소하는 경향이었고, 일반두부와 시판두부에 비해 L값이 대체로 낮았으며 b값은 현저히 높았으나 a값은 비슷하였다. WSF두부의 각 응고제 농도에 따른 견고성(hardness)은 응고제 농도가 높을수록 증가하는 경향이었으며, 응고제 종류에 따른 견고성(hardness)은 전반적으로 $CaCl_2$를 사용한 경우 비교적 높았으며 $CaSO_4$와 GDL 응고제를 사용한 첨가구에서는 낮게 나타났다. 접착성(adhesiveness)은 응고제 농도가 높아질수록 대체로 감소하는 경향이었으나 $CaSO_4$, GDL, $Ca-gluconate+CaSO_4$$GDL+CaSO_4$ 첨가구가 다른 첨가구에 비하여 비교적 낮은 경향을 보였다. 응집성(cohesiveness)은 응고제 종류와 농도에 따른 통계적인 유의적 차이가 없었고, 탄력성(springiness)은 $GDL+CaSO_4$ 혼합 첨가구에서는 응고제 농도가 높을수록 높게 나타났다. 씹힘성(chewiness)은 혼합 응고제 첨가구에서 대체로 낮은 경향을 보였으며 뭉치는 성질(gumminess)은 견고성(hardness)이 증가함에 따라 증가하였다. WSF 두부의 각 응고제 농도에 따른 관능평가 중 전체적기호도에서는 $CaCl_2$, $Ca-gluconate+CaCl_2$, $GDL+CaCl_2$ 첨가구는 0.2%에서, $CaSO_4$, GDL, $MgCl_2$, $Ca-gluconate+MgCl_2$, $GDL+CaSO_4$ 첨가구는 0.3%에서, 그리고 $Ca-gluconate+CaSO_4$ 첨가구는 0.4%에서 높게 나타났으며 그 중 0.3% $GDL+CaSO_4$ 첨가구가 적당한 경도를 가지면서도 부드럽고 고소한 맛이 강하여 가장 높은 기호도를 나타내었다.

  • The purpose of this study was to evaluate the effects of $Ca(OH)_2$ and $CO_2$ additions on the corrosion of metal coupons(ductile iron, galvanized steel, copper and stainless steel). Corrosion rate and released metal ion concentration of ductile iron and galvanized steel decreased by adjusting alkalinity, calcium hardness and pH with $Ca(OH)_2$ & $CO_2$ additions on copper and stainless steel were less than those on ductile iron and galvanized steel. When ductile iron coupon was exposed to water treated with Ca(OH)$_2$&$CO_2$, additions, the main components of corrosion product formed on its surface were $CaCO_3$ and $Fe_2 O_3 or Fe_2 O_4$ which often reduce the corrosion rate by prohibiting oxygen transport to the metal surface.

Effects of $Cd^{2+}$ on the Contractility in the Antral Circular Muscle of Guinea-pig Stomach

  • The effects of $Cd^{2+}$ on spontaneous contraction, and the contractures induced by $0mM\;Na^+,\;60mM\;K^+\;and\;10^{-6}\;M$ acetylcholine, 1mM caffeine were studied in order to elucidate diverse actions of $Cd^{2+}$ on the $Ca^{2+}$ mobilization related with contractility in the antral circular muscle of guinea pig stomach. $Cd^{2+}$ inhibited the spontaneous contraction in a does dependent manner $(10^{-6}\;M\;10^{-4}\;M).\;Cd^{2+}\;(3{\times}10^{-5}M)$ suppressed 60 mM $K^+$ induced contracture composed or a phasic and a tonic response and the increased tonic response by the increased external $Ca^{2+}$ concentration. $Cd^{2+}$ also suppressed acetylcholine induced contracture composed of repetitive phasic and a tonic component and the increased tonic response by the increased external $Ca^{2+}$ concentration. Caffeine in the concentration of 1mM evoked contracture but $Cd^{2+}$ suppressed the contracture. $Cd^{2+}$ suppressed the amplitude of the $Na^+$ tee contracture dose dependently and the amplitude of $Na^+$ free contracture almost decreased to 20% of control amplitude in the concentration of $10^{-4}\;M\;Cd^{2+}$. From the above results, it is suggested that $Cd^{2+}$ may inhibit not only $Ca^{2+}$ influx via voltage sensitive, receptor operated $Ca^{2+}$ channel and Na/ca exchange but also intracellular $Ca^{2+}$ release from the sarcoplasmic reticulum in the antral circular muscle of guinea pig stomach.

筋小胞體의 ATPase 活性과 칼슘吸收能에 미치는 Caffeine의 영향 (The Effects of Caffeine on the ATPase Activity and the Calcium Uptake of the Fragmented Sarcoplasmic Reticulum of Rabbit Skeletal Muscle)

  • 토끼의 골격근 小胞體의 ATPase 活性과 Ca吸收能에 미치는 caffeine의 영향을 조사 하였다. 遠心分離로 分劃된 小胞體에서 $2,000 \\sim 8,000 \\times G$ 分劃의 ATPase 活性은 caffeine에 의하여 增大되지만 $8,000 \\times G$ 以上의 分劃에서는 아무 영향도 받지 않았다. Caffeine에 의한 이 活性增大는 이 分劃에 混在하는 mitochondria의 ATPase 活性이 增大 된 結果라고 해석된다. 小胞體의 $2,000 \\sim 10,000 \\times G$ 分劃과 $10,000 \\sim 20,000 \\times G$ 分劃의 Ca吸收能도은 反應液內 Ca의 농도가 200 nmoles/mg protein 정도 이상일 때는 caffeine에 의하여 현저히 阻害되지만, Ca의 농도가 이 以上이 때는 2,000$\\sim$10,000 分劃에서만 이 阻害現象을 볼 수 있다. 低農度 Ca에서의 이 阻害現象은 caffeine에 의하여 mitochondira의 Ca吸收도 阻害되기 때문에 나타나는 것으로 해석된다. Caffiene에 의한 筋收縮의 誘發 및 逕縮現象은 筋小胞體의 Ca 吸收가 이 特質에 의하여 阻害되고 또 蓄積된 Ca이 放出되기 때문에 일어나는 것으로 해석된다.

Interactive effect of dietary levels of calcium and 25-hydroxy vitamin D3 on the performance, serum biochemical concentration and digestibility of laying hens from 61 to 70 weeks of age

  • Objective: The present research was conducted to evaluate the interactive effect of dietary concentration of calcium (Ca) and 25-hydroxy vitamin D3 (25OHD3) on the performance, blood composition and digestibility of laying hens. Methods: A total of 540 Hy-line brown laying hens aged 61 to 70 weeks were randomly allotted in a 3×3 factorial arrangement, consisting of three levels of 25OHD3 (0, 25, and 50 ㎍/kg) and three levels of Ca (3.5%, 4.0%, and 4.5%). All diets had basal concentration of 3,000 IU/kg of vitamin D3 including the 2,800 kcal/kg of metabolic energy and 16% of crude protein. Results: The results showed that interactive effect (p<0.05) between Ca and 25OHD3 was such that dietary 25OHD3 linearly increased interleukin-6 at all levels of Ca inclusion. Interaction (p<0.05) occurred with the highest parathyroid hormone in laying hens that received dietary concentration of Ca (3.5%) with 25OHD3 (50 ㎍/kg), and Ca (4.0%) with 25OHD3 (50 ㎍/kg). Egg production and egg weight significantly (p<0.05) increased in the 4.5% Ca group compared to the 3.5% to 4.0% Ca groups. Egg shell thickness and tibia bone length also increased (p<0.05) in groups fed a high-Ca diet (4.0% to 4.5%). Phosphorus digestibility significantly (p<0.05) increased along with dietary Ca level. Among the tested 25OHD3 groups, higher (p<0.05) egg production and tibia thickness were present in hens fed 50 ㎍/kg of 25OHD3. Furthermore, Ca digestibility serum Ca and 25OHD3 were significantly increased in group offered 50 ㎍/kg of 25OHD3. Conclusion: The results gathered in this study indicate that dietary concentrations of 4.0% to 4.5% Ca and 50 ㎍/kg 25OHD3 improve the performance of hens from 61 to 70 weeks of age.
