밀기울을 첨가한 토하젓의 숙성과정 중 영양성분의 변화 (Changes in Nutritional Components of Toha-jeot with Wheat Bran during Fermentation)

  • In this study, to activate the industrialization and to improve the quality of Toha-jeot by shortening the fermentation period, we investigated the changes in the nutritional components of Toha-jeot. salt-fermented Toha shrimp( Caridina denticulata denticulata $D_{E}$ $H_{AAN}$) which was salted with a low-salt group and high-salt group during fermentation. In this experiment. there are four groups of Toha-jeot which were manufactured with 15% ratio of common salt: the first group containing 2% wheat bran (w2%-L). the second high-salt group containing 2% wheat bran( w2%-H) , the third low-salt group containing 4% wheat bran (w4%-L) and the last high-salt group containing 4% wheat bran(w4%-H). These four groups were refrigerated at 4${\pm}$1$^{\circ}C$ and then taken out for analysis at three month intervals during 9 month. Among the free amino acid contents in Toha-jeot, 22 kinds were detected. 6 month after the fermentation when the quantity of the amino acid contents in Toha-jeot is highest, ornitine, glutamic acid, leucine. alanine. lysine and valine occupy the majority, in the order of abundance. In cases of nucleotides. 6 month after the fermentation. from the groups w2%-L, w2%-H and w4%-L, inosine and IMP were not detected. and hypoxanthine, AMP, ADP were detected but 9 month after the fermentation ADP was not detected. The main constituents of fatty acid were as follows : (a) from w2%-L, w2%-H, 6 month after the fermentation. $C16:0$, $C12:0$, $C18:1$, $C18:3$, and $C16:1$. (b) from w4%-L. 6 month after the fermentation, $C18:3$, $C16:0$, $C12:0$ and $C18:1$. (c) from W4%-H, $C16:0$, $C12:0$, $C18:3$ and $C18:1$. In case of mineral contents. Na, Ca. K. Mg, Fe. Zn, Mn and Cu were detected according to the magnitude of the quantity. From the group w4%-H, high quantity of Na was detected during the total fermentation period. In case of color value, from the groups w2%. the values of L. a. b were highest after 6 month fermentation and were decreased after 9 month fermentation, while from groups w4%, the values of L, a, b were gradually decreased after 3 month fermentation.ion.

全南地方의 一部 人工湖水의 富營養化에 관한 조사연구 (A Study on the Eutrophication in Artificial Lakes in Chonnam Area)

  • In order to analyze the water quality in artificial lakes in Chonnam area, a chemical and biological examination of Dongbock Lake and Changsung Lake was conducted from September to December 1983 and May 1984. A summary of the surveyed results is as follows 1. In Dongbock Lake, pH ranged from 7.2-8.1, D.O.: 8.2-12.6mg/l, B.O.D.: 4.4-22.1 mg/l, C.O.D.: 1.0-3.4rag/l, Cl$^-$: 5.9-11.9mg/l, Total-P: 0.001-0.071 mg/l, and Total -N: 0.016-0.697 mg/l, respectively. 2. In Changsung Lake, pH ranged from 7.2-8.1, D.O.: 8.1-9.8mg/l, B.O.D.: 0.9-2.9mg/l, C.O.D.: 1.9-3.4mg/l. Total- P: 0.006-0.016mg/l and Total -N:0.006-0.033mg/l, respectively. 3. The Phytoplankton identified in this investigation were distributed in a total of 46 genera and 76 spedes in Dongbock Lake 37 genera and 45 species in Changsung Lake. 4. In Dongbock Lake, it was found that the dominant algae were Melosira sp., Microcystis sp. and Synedra sp. in September Melosira sp. and Microcytis sp. in October, but Cymbella sp., Naviculla sp. and Nitzschis sp. were also observed in OctoberAsterionella sp., Melosira sp. and Microsystis sp. in November and Melosira sp., Asterionella sp sp. and Synedra sp. in December 1983. 5. In Changsung Lake, it was found that the dominant algae were Melosira sp., Lyngrbya sp. and Microcystis in September Melosira sp. and Synedra sp. in October and November and Melosira sp., Lyngbya sp. and Asterionella sp. in December 1983. The dominant algae were Melosira sp., Lyngbya sp. and Euglena sp. in May 1984. 6. It was found that the dominant algae in both Dongbock and Changsung Lakes were Microcystis sp., Melosira sp. and Asterionella sp.. Which are strongly related with water-bloom. Therefore, it could be suggested that the eutrophication phenomena is going to occur very easily in Dongbock Lake and possibly in Changsung Lake.

Genetic Organization of the hrp Genes Cluster in Erwinia pyrifoliae and Characterization of HR Active Domains in HrpNEp Protein by Mutational Analysis

  • The disease-specific (dsp) region and the hypersensitive response and pathogenicity (hrp) genes, including the hrpW, $hrpN_{Ep}$, and hrpC operons have previously been sequenced in Erwinia pyrifoliae WT3 [Shrestha et al. (2005a)]. In this study, the remaining hrp genes, including the hrpC, hrpA, hrpS, hrpXY, hrpL and hrpJ operons, were determined. The hrp genes cluster (ca. 38 kb) was comprised of eight transcriptional units and contained nine hrc (hrp conserved) genes. The genetic organization of the hrp/hrc genes and their orientation for the transcriptions were also similar to and collinear with those of E. amylovora, showing ${\geq}80%$ homologies. However, ORFU1 and ORFU2 of unknown functions, present between the hrpA and hrpS operons of E. amylovora, were absent in E. pyrifoliae. To determine the HR active domains, several proteins were prepared from truncated fragments of the N-terminal and the C-terminal regions of $HrpN_{Ep}$ protein of E. pyrifoliae. The proteins prepared from the N-terminal region elicited HR, but not from those of the C-terminal region indicating that HR active domains are located in only N-terminal region of the $HrpN_{Ep}$ protein. Two synthetic oligopeptides produced HR on tobacco confirming presence of two HR active domains in the $HrpN_{Ep}$. The HR positive N-terminal fragment ($HN{\Delta}C187$) was further narrowed down by deleting C-terminal amino acids and internal amino acids to investigate whether amino acid insertion region have role in faster and stronger HR activity in $HrpN_{Ep}$ than $HrpN_{Ea}$. The $HrpN_{Ep}$ mutant proteins $HN{\Delta}C187$ (D1AIR), $HN{\Delta}C187$ (D2AIR) and $HN{\Delta}C187$ (DM41) retained similar HR activation to that of wild-type $HrpN_{Ep}$. However, the $HrpN_{Ep}$ mutant protein $HN{\Delta}C187$ (D3AIR) lacking third amino acid insertion region (102 to 113 aa) reduced HR when compared to that of wild-type $HrpN_{Ep}$. Reduction in HR elicitation could not be observed when single amino acids at different positions were substituted at third amino acids insertion region. But, substitution of amino acids at L103R, L106K and L110R showed reduction in HR activity on tobacco suggesting their importance in activation of HR faster in the $HrpN_{Ep}$ although it requires further detailed analysis.

Fe/Au 이중층의 응집현상을 이용한 FePd 나노 점 형성에 관한 연구 (Study on Formation of FePd Nano-dot Using Agglomeration of Fe/Au Bilayer)

  • 자기조립화된 Fe/Au 이중층 위에 $L1_0$형 구조를 갖는 FePd 나노 점을 성공적으로 제작하였다. AFM를 이용하여 초기에 편평한 Fe/Au 이중층 박막이 온도가 증가함에 따라서 응집되어 나노 점 구조로 변형되는 것을 확인하였다. 또한 형성된 이중층위에 FePd 다층막을 $300^{\circ}C$, $350^{\circ}C$, $400^{\circ}C$, $450^{\circ}C$에서 각각 증착하였다. 초격자 구조를 갖는 FePd 다층막의 표면형상은 응집현상에 의하여 자기조립화된 이중층의 형상과 유사하였다. XRD 측정결과, $350^{\circ}C$ 이상에서 열처리된 FePd 다층막은 $L1_0$형 구조를 갖는다는 것을 확인하였다. 그리고 박막두께에 따른 XPS 측정결과는 전체 박막의 화학적 조성이 증착순서와 일치하는 것을 보여주었다. 결과적으로 추가적인 식각공정 없이 화학적으로 규칙화된 FePd 초격자 나노 점의 제작에 성공하였다.

D-xylose 발효효모의 분리 및 성질 (Isolation and properties of D-xylose fermenting yeast)

  • D-xylose로부터 직접 ethanol을 생산할 목적으로 D-xylose를 유일한 탄소원과 에너지원으로 이용하는 효모들을 토양, 톱밥, 썩은 나무들로부터 분리하였다. 이 균주들 가운데서 ethanol 생산력이 가장 우수한 균주는 썩은 나무로부터 분리한 Candida sp. L-16으로 동정되었다. 이균주를 이용한 ethanol 발효의 최적 조건은 진탕속도 60rpm, 배양온도 28C, 초기 pH 4.5 및 D-xylose 농도 5%이었다. 알코올 생산은 배양 4일째에 최대치에 도달했고 이때의 ethanol 농도는 2.4%(v/v)로서 이론적인 ethanol 수율은 총당에 대해 74.4%이었다.

The Effects of Dietary Turkish Propolis and Vitamin C on Performance, Digestibility, Egg Production and Egg Quality in Laying Hens under Different Environmental Temperatures

  • In this study, the effects of propolis and vitamin C (L-ascorbic acid) supplementation in diets were investigated on feed intake (FI), body weight (BW), body weight gain (BWG), feed conversion rate (FCR) and digestibility and on egg production and qualities (weight, mortality, shell thickness) in laying hens exposed to heat stress. A total of 150 Hyline White Leghorn, aged 42 weeks, hens was divided into five groups of 30 hens. Chicks were randomly divided into 1 positive control, 1 control and 3 treatment groups. The chicks were kept in cages in temperature-controlled rooms at $22^{\circ}C$ for 24 h/d (positive control, Thermoneutral, TN group) or $34^{\circ}C$ for 9 h/d from 08.00-17.00 h followed by $22^{\circ}C$ for 15 h (control, heat stress, HS group) and fed a basal diet or basal diet supplemented with vitamin C (250 mg/kg of L- ascorbic acid/kg of diet) or two levels of propolis (2 and 5 g of ethanol extracted propolis/kg of diet). Increased FI (p<0.05) and improvement in FCR (p<0.05), hen day egg (p<0.05) and egg weight (p<0.05) were found in Vitamin C and propolis-supplemented laying hens reared under heat stress conditions. Mortality rate was higher in the control group than TN, vitamin C and propolis groups (p<0.05). Digestibility of dry matter, organic matter, crude protein and ether extract improved with increasing of both dietary vitamin C and propolis (p<0.05). Vitamin C or propolis supplementation did not affect either the percentage shape index, yolk index or haugh unit and albumen index (p>0.05). However, the egg shell thickness and egg shell weight appeared to be increased in Vitamin C and propolis groups in comparison to HS group birds (p<0.05). In conclusion, dietary supplementation of laying hens with anti-oxidants (vitamin C and propolis) can attenuate heat stress-induced oxidative damage. These positive effects were evidenced by increased growth performance and digestibility, improvement of egg shell thickness and egg weight in comparison to non-supplemented birds. Moreover, supplementation with propolis (5 g/kg diet) was the most efficient treatment.

알릴아민 항진균제의 합성과 생물학적 평가 (Synthesis and Biological Evaluation as a Potential Allylamine Type Antimycotics)

  • Structure-activity relationship studies of allylamine type of antimycotics were carried out to evaluate the effect of naphthyl and methyl portion of naftifine. Compounds with 4-fluorophenyl(2a-5a), 2-fluorophenyl(2b-5b), 2,4-dichlorophenyl(2c-5c), 2,6-dichlorophenyl(2d-5d), 4-nitrophenyl(2e-5e), and 2,3-dihydro-benzo[1,4]dioxan-6-yl( 2f-5f) instead of naphthyl group with hydrogen(3a-3f), methyl(4a-4f), and ethyl(5a-5f) in the place of methyl in naftifine were synthesized and tested their in vitro anti-fungal activity against five different fungi. Eight compounds(3a, 5a, 3c, 4c, 4d, 5d, 5e, and 4f) showed significant antifungal activity against T. mentagrophytes. (E)-N-Ethyl-(3-phenyl-2-propenyl)-4-nitro-benzenemethaneamine(5e) displayed moderate antifungal activity against all five different fungi.

L-폴리락타이드, D-폴리락타이드의 활성과 입체복합체 폴리락타이드의 제조 및 특성연구 (Synthesis and Characterization of Poly(L-lactide)(L-PLA), Poly(D-lactide)(D-PLA) and Stereocomplex-poly(lactide)(PLA))

  • 본 연구에서는 L-lactide, D-lactide를 단량체로 하여 tin 2-octoate, lauryl alcohol을 각각 촉매와 개시제로 사용하여 촉매량(0.5~1.0 wt%)과 개시제의 양(0.5~0.1 wt%)을 달리하여 $140^{\circ}C$에서 lactide의 고리개환중합으로 고분자량의 L-PLA와 D-PLA를 합성하였다. 합성된 PLA는 $M_n$ : 30000~90000 g/mol의 다양한 분자량이 나타났으며 합성된 L-PLA와 D-PLA를 이용해 조성비를 다르게 하여 stereocomplex-PTA를 제조하였다. Stereocomplex-PLA의 용융점, 열분해온도, 열적 안정성은 단열고분자 L-PLA, D-PLA보다 높아짐을 확인하였다. 이러한 결과로 보아 이성질체 L-PLA, D-PLA 분자간 인력이 작용한다는 사실을 예상한 수 있었고 XRD를 통해 모폴로지의 변화를 살펴본 결과 단일고분자와 stereocomplex-PLA는 상이하게 변하는 사실을 확인하였다. 또한 기계적 강도를 측정하여 L-PLA, D-PLA보다 stereocomplex-PLA의 최대응력과 변형률이 증가함을 확인하였다.

Heat Shock Stress에 의한 Lactobacillus acidophilus 30SC의 생리적 특성 (Physiological Properties of Lactobacillus acidophilus 30SC Exposed to Heat Shock Stress)

  • Probiotics로서의 활성이 높은 Lactobacillus acidophilus 30SC의 생존성을 증진시키기 위한 기초 자료를 얻고자, heat shock stress를 가한 후 생균수를 측정하고, 생존율의 변화를 통해 고온 처리에 의한 고온 및 냉동 내성의 증진 효과를 평가하였다. 또한 열처리 동안 새로이 발현되는 단백질을 1차원 및 2차원 전기영동을 이용하여 확인하였으며, 주사전자현미경을 사용하여 세포 모양을 관찰하였다. L. acidophilus 30SC는 $55^{\circ}C$의 heat shock stress를 받았을 때 생존 균수가 감소하는 것으로 나타났다. 나머지 처리구는 $37^{\circ}C$에서 계속 배양한 것과 별다른 차이를 나타내지 않았다. 특히 $45^{\circ}C$로 heat shock stress를 준 경우 $37^{\circ}C$에서 배양한 것과 거의 동일하였다. L. acidophilus 30SC에 $45^{\circ}C$로 heat shock stress를 가한 뒤 추가로 55 및 $60^{\circ}C$에 노출시켰을 때 가장 높은 생존율을 나타냈고, 치사 수준인 $55^{\circ}C$의 heat shock stress를 받은 후 $55^{\circ}C$$60^{\circ}C$에 노출되었을 때 생존율이 급격히 감소하는 경향을 보였다. L. acidophilus 30SC에 $55^{\circ}C$로 15분 Heat shock stress를 준 경우 약 22와 25 kDa의 단백질들이 새로이 발현된 것으로 나타났으나, 24와 27 kDa로 추정되는 단백질의 발현 정도는 낮았음을 확인하였다. 2차원 전기영동을 실시한 결과, $37^{\circ}C$에서 배양한 대조구와 비교할 때 $55^{\circ}C$로 heat shock stress를 준 경우 새로이 5개의 protein spot을 발견할 수 있었다. 주사전자현미경으로 세포의 형태를 관찰한 결과 heat shock stress를 준 경우에는 세포의 길이가 신장되는 경향을 나타내었다.

Formation of DNA-protein Cross-links Mediated by C1'-oxidized Abasic Lesion in Mouse Embryonic Fibroblast Cell-free Extracts

  • Oxidized abasic residues arise as a major class of DNA damage by a variety of agents involving free radical attack and oxidation of deoxyribose sugar components. 2-deoxyribonolactone (dL) is a C1'-oxidized abasic lesion implicated in DNA strand scission, mutagenesis, and covalent DNA-protein cross-link (DPC). We show here that mammalian cell-free extract give rise to stable DPC formation that is specifically mediated by dL residue. When a duplex DNA containing dL at the site-specific position was incubated with cell-free extracts of Po ${\beta}-proficient$ and -deficient mouse embryonic fibroblast cells, the formation of major dL-mediated DPC was dependent on the presence of DNA polymerase (Pol) ${\beta}$. Formation of dL-specific DPC was also observed with histones and FEN1 nuclease, although the reactivity in forming dL-mediated DPC was significantly higher with Pol ${\beta}$ than with histones or FEN1. DNA repair assay with a defined DPC revealed that the dL lesion once cross-linked with Pol ${\beta}$ was resistant to nucleotide excision repair activity of cell-free extract. Analysis of nucleotide excision repair utilizing a model DNA substrate containing a (6-4) photoproduct suggested that excision process for DPC was inhibited because of DNA single-strand incision at 5' of the lesion. Consequently DPC mediated by dL lesion may not be readily repaired by DNA excision repair pathway but instead function as unusual DNA damage causing a prolonged DNA strand break and trapping of the major base excision repair enzyme.
