Reproductive Ecology of Neptunea(Barbitonia) arthritica cumingii (갈색띠매물고둥, Neptunea(Barbitonia) arthritica cumingii의 번식생태)

  • Gonad index (GI), conditon index, the reproductive cycle with gonadal development of the Neptunea (Barbitonia) arthritica cumingii, were investigated histologically, based on the samples which have been collected from the subtidal zone of Oeyeondo, Boryeong, Korea from January to December, 2006. Neptunea (Barbitonia) arthritica cumingii is dioecious and oviparous. Monthly changes in the gonad index (GI), studied for determination of spawning period, were closely associated with gonad developmental phases. The GI reached a maximum in April, and gradually decreased from May to August due to spawning. The gonadsomatic index and condition index showed similar patterns to gonad developmental phases and the spawning period. The reproductive cycle according to gonad developmental phases of this species can be classified into five successive stages in females and males: in females, early active stage (September to October), late active stage (November to February), ripe stage (February to June), partially spawned stage (May to August) and recovery stage (June to August); in males, the early active stage (September to October), late active stage (November to February), ripe stage (February to June), copulation (April to July), and recovery stage (July to August). Spawning occurred between May to August in females and April to July in males, and spawning peak in females was observed between June and July when the seawater temperature rose to above $19^{\circ}C$. Percentages of first sexual maturity of female and male snails ranging from $50.1{\sim}60.0\;mm$ in shell height were over 50%, and 100% for snails over 60.1 mm in shell height. The sex ratios of females to males were not significantly different from a 1:1 sex ratio.

  • Background : Reactive oxygen species (ROS) take center stage as executers in ventilator-induced lung injury (VILI). The protein with DNA-damage scanning activity, poly (ADP-ribose) polymerase-1 (PARP1), signals DNA rupture and participates in base-excision repair. Paradoxically,overactivation of PARP1 in response to massive genotoxic injury such as ROS can induce cell death through ${\beta}$ -nicotinamide adenine dinucleotide ($NAD^+$) depletion, resulting in inflammation. The purpose of this study is to investigate the role of PARP1 and the effect of its inhibitor in VILI. Methods : Forty-eight male C57BL/6 mice were divided into sham, lung protective ventilation(LPV), VILI, and PARP1 inhibitor (PJ34)+VILI (PJ34+VILI) groups. Mechanical ventilator setting for the LPV group was $PIP\;15cmH_2O$ + $PEEP\;3cmH_2O$ + RR 90/min + 2 hours. The VILI and PJ34+VILI groups were ventilated on a setting of $PIP\;40cmH_2O$ + $PEEP\;0cmH_2O$ + RR 90/min + 2 hours. As a PARP1 inhibitor for the PJ34+VILI group, 20 mg/Kg of PJ34 was treated intraperitoneally 2 hours before mechanical ventilation. Wet-to-dry weight ratio and acute lung injury (ALI) score were measured to determine the degree of VILI. PARP1 activity was evaluated by using an immunohistochemical method utilizing biotinylated NAD. Myeloperoxidase (MPO) activity and the concentration of inflammatory cytokines such as tumor necrosis factor $(TNF)-{\alpha}$, interleukin $(IL)-1{\beta}$, and IL-6 were measured in bronchoalveolar lavage fluid (BALF). Results : In the PJ34+VILI group, PJ34 pretreatment significantly reduced the degree of lung injury, compared with the VILI group (p<0.05). The number of cells expressing PARP1 activity was significantly increased in the VILI group, but significantly decreased in the PJ34+VILI group (p=0.001). In BALF, MPO activity, $TNF-{\alpha}$, $IL-1{\beta}$, and IL-6 were also significantly lower in the PJ34+VILI group (all, p<0.05). Conclusion : PARP1 overactivation plays a major role in the mechanism of VILI. PARP1 inhibitor prevents VILI, and decreases MPO activity and inflammatory cytokines.

Studies on the In Vitro Development of Cloned Embryos by Somatic Cell Nuclear Transfer in Korean Native Goats (재래산양의 체세포 핵이식에 의한 복제수정란의 체외발달에 관한 연구)

  • The present study was conducted to examine some factors affecting in vitro development of oocytes from somatic cell nuclear transfer (SCNT) in Korean native goats. Recipient oocytes were surgically collected after superovulation by using CIDR and FSH, PMSG, hCG and estrous synchronization in Korean Native goats. For nuclear transfer, the fibroblasts from caprine ear cells and fetal fibroblasts were surgically harvested and were cultured in vitro until cell confluency in serum-starvation condition (TCM-199 + $0.5\%$ FBS) for 3 to 5 days. The zona pellucidae of matured oocytes were partially drilled by laser irradiation. A single somatic cell was individually transferred into each enucleated oocyte. The reconstructed oocytes were then electrically fused and activated. Activated NT embryos were cultured in mSOF medium supplemented with $0.8\%\;BSA\;6\~7\;day\;at\;39^{\circ}C,\;5\%\;CO_2,\;5\%\;O_2,\;90\%\;N_2$ in air. There were no significant difference in the number of embryos cleaved and 4-cell development between the fibroblast nuclei from mature ear cells and fetal cells, but the rate of 8-cell development was higher (P<0.05) in ear cells $(40.5\%)$ than in fetal cells $(55.5\%)$. However, the embryo development to morula or blastocyst was not significantly different between both the groups$(6.7\%\;vs\;16.0\%)$, respectively. The number of embryo cleaved $(79.0\%)$ were higher (P<0.05) in the oocytes activated with ionomycin+6-DMAP than in the oocytes activated electrically $(9.5\%)$. The development of fused embryos to morula or blastocyst was found $15.6\%$ in ionomycin+6-DMAP, but no morula or blastocysts were developed in electrical stimulation. The development rate of SCNT embryos to morula or blastocyst was love. (P<0.05) in SCNT embryos $(19.0\%\;vs\;0.0\%)$ than that in parthenotes $(66.1\%\;vs\;59.1\%)$. In the parthenotes, the cleavage rate and development to morula or blastocyst were significantly higher (P<0.05) as $86.8\%\;and\;50.0\%$ in ovulated oocytes than in follicular oocytes $(69.0\%\;vs\;23.6\%)$, respectively. These results suggest that some factors Including superovulation treatment, oocyte source, maturation of follicular oocytes, activation method and culture condition may affect in vitro developmental capability of embryos produced by somatic cell nuclear transfer in Korean Native goats, and the fusion rate be greatly low compared with other species.

Early Growth of Cultured Larval Haddock, Melanogrammus aeglefinus (Haddock, Melanogrammus aeglefinus 자어의 초기성장)

  • The objective of this study was to examine the early growth of haddock, Melanogrammus aeglefinus larvae from a series of reared specimens for provide information to developmental biology and more information on the aspect of aquaculture in the larvae of this species. Larvae were reared in the laboratory and sampled periodically for developmental study until 67 days after hatching. An increase in total length of fish indicated continuous growth, described by the growth expression of the type $TL=3.5374e^{0.0536X}(r^2=0.8759$, where TL is total length and X is at days after hatching) and $BW=0.0002e^{0.1858X}(r^2=0.8671$, where BW is body weight and X is at days after hatching), respectively. Pattern of body depth and pectoral fin length are instantaneous growth which expression of the type $BD=0.3545e^{0.0778X},\;r^2=0.9563$(where BD is body depth and X is at days after hatching) for body depth growth and the type $PL=0.0111e^{0.1591X},\;r^2=0.9194$(where PL is pectoral fin length and X is at days after hatching) for pectoral fin length growth. The relationship of body depth and total length expressed as $BD=0.2397X-0.5735(r^2=0.9957$, where BD is body depth and X is total length), and pectoral fin length and total length is $PL=0.1929X-1.3767(r^2=0.9882$, where PL is pectoral fin length and X is total length) pectoral fin length against body depth simultaneously recorded for juvenile haddock(PL=0.8117BD-0.9718, $r^2=0.9814$, where PL is pectoral fin length and BD is body depth). Relationship of body depth and body weight was expressed the type of $BD=-9.4734X^2+19.046X+1.3672,\;r^2=0.941$(where BD is body depth and X is body weight), and pectoral fin length and body weight expressed the type of $PL=6.379X^2+14.023X+0.3774,\;r^2=0.9494$(where PL is pectoral fin length and X is body weight). From this point view, growth characteristics of juvenile haddock in this experiment may be useful to establish a successful culture technique for rearing larval haddock.

  • The initial growth and herbicidal responses of Scirpus planiculmis were examined under both laboratory and greenhouse condition. Initial growth of S. planiculmis at two different soils(soil A and B), was compared with the growth at normal paddy soil, and growth rates at two soils were 32% and 86% as compared to the growth rates at normal paddy soil. Two different soils were soil A(rice-uncultivable) and soil B(rice-cultivable). Transplanted(2.5 leaf stage) and direct seeded rice(Dongjinbyeo) could not grow at soil A. The growth of transplanted rice at soil B was almost same as the growth at normal paddy soil, but the growth of direct seeded rice was 50% of the growth at normal paddy soil. S. planiculmis among the perennial weeds was most tolerant to NaCl, and Cyperus serotinus, Eleocharis kuroguwai, Sagittaria pygmaea, Sagittaria trifolia, Potamogeton distinctus were followed. The intial growth rate of S. planiculmis was more rapid than C. serotinus. Cutting types and times of corm reduced 20-40% of the initial growth of S. planiculmis. During the initial growth of S. planiculmis, the growth of S. planiculmis was severly inhibited by the earlier removal of corm from seedlings, and it represents that the corm plays an important role in the initial growth of this species. Bentazone among 14 commercialized herbicides showed the best safety on direct seeded rice and the highest control effect agaist S. planiculmis.

  • To estimate the possible amount of weed-crop competitions under the various cropping patterns of two rice cultivars of different type, comparisons between weedy check and weed-free plots in plant height, tillering number, and plant weight were investigated. Fluctuations in Importance Value, competition value, and net productivities were used to discuss the crop-weed competition, respectively. As a result, differences in dominant weed species, competitive crop variety, most effective cropping pattern, and crop yielding characteristics under the different weed competitions were investigated, respectively.

  • This study has been made to make an observation regarding present status of the coal mine props which is desperately needed for coal production, despite of great shortage of the timber resources in this country, and investigate the effects of diffusion process on the decay resistances of the mine props as applied preservatives of Malenit and chromated zinc chloride. The results are as follows. 1. Present status of the coal mine props Total demand of coal mine props in the year of 1975 was approximately 456 thousand cubic meters. The main species used for mine props are conifer (mainly Pinus densiflora) and hardwood (mainly Quercus). Portions between them are half and half. With non fixed specification, wide varieties of timber in size and form are used. And volume of wood used per ton-of coal production shows also wide range from 0.017 cubic meter to 0.03 cubic meter. 2. Decay resistance test a) The oven dry weight decreased between untreated specimen and treated specimen has not shown any significantly, although it has shown some differences in average values between them. It may be caused by the shorter length of the test. b) The strength of compression test between untreated specimen and treated specimen has also shown the same results as shown in case of weight decrease. Reasons assumed are the same. c) The amounts of the extractives in one percent of sodium hydroxide (NaOH) between untreated and treated specimen have shown the large value in case of untreated specimen than that of treated. 3. The economical benifit between untreated and treated wood when applied in field has seen better in long term base in case of treated wood, although the primary cost of treated wood add a little bit more cost than that of the untreated wood.

Identification of Streptomyces scopuliridis KR-001 and Its Herbicidal Characteristics (Streptomyces scopuliridis KR-001의 분리 동정 및 잡초 방제효과)

  • With increasing environmental issues from synthetic chemical herbicides, microbe-originated herbicides could be a fascinating alternative in current agriculture. We isolated Streptomyces strains that produced herbicidal active metabolite(s) against a grass weed Digitaria sanguinalis. According to the result from 16S rDNA sequence comparison with the close strains, the best isolate (Code name MS-80673) was identified as Streptomyces scopuliridis KR-001. The closest type strain was Streptomyces scopuliridis RB72 which was previously reported as a bacteriocin producer. The optimal culture condition of S. scopuliridis KR-001 was $28^{\circ}C$, pH 7.0 and culture period 4 to7 days. Both of soil and foliar application of the crude culture broth concentrate was effective on several troublesome or noxious weed species such as a Sciyos angulatus in a greenhouse and field condition. Phytotoxic symptoms of the culture broth concentrate of S. scopuliridis KR-001 by foliar application were wilting and burndown of leaves, and stems followed by discoloration and finally plant death. In crops such as rice, wheat, barley, hot pepper and tomato, growth inhibition was observed. These results suggest that the new S. scopuliridis KR-001 strain producing herbicidal metabolites may be a new bio-herbicide candidate and/or may provide a new lead molecule for a more efficient herbicide.

Immune-modulation Effect of Ulmus macrocarpa Hance Water Extract on Balb/c Mice (왕느릅나무 껍질 열수 추출물의 마우스에서의 in vivo 면역조절 효과)

  • Traditional medicinal plants are widely used to treat many diseases, such as inflammation, infections, and even cancer. Ulmus macrocarpa Hance, a Chinese elm species, is distributed in Korea, China, and Japan. The stem bark is widely employed in Korean traditional medicine to treat dermatitis, mastitis, and edema. The aim of this study was to investigate whether water extract of U. macrocarpa Hance bark (Ulmus cortex) has a immune-modulating function in a mouse model. Three different concentrations (30 mg/kg, 100 mg/kg, and 300 mg/kg) of Ulmus cortex water extract (UCWE) were orally administered to mice for 14 days, and their immune responses were analyzed. Cytokines, such as interleukin (IL)-2, IL-12, and IFN-${\gamma}$, increased in the blood of UCWE-fed groups when compared with a control group. In contrast, the IL-4 level did not change in any of the UCWE-fed groups Cell-mediated cytotoxicity was also assayed using lymphokine-activated killer cells (LAK). LAK showed greater cytotoxicity in the UCWE-fed groups than LAK in the control group. Internal organ indices, such as liver, kidney, spleen, and thymus, were similar in all the groups, including the control group, indicating that UCWE may have been nontoxic in the experimental animals. These data suggest that UCWE has an immune-modulating function in a mouse model.

Studies on the Extending of Plywood Adhesives used Foliage Powder (낙엽분말(落葉粉末)을 이용(利用)한 합판용(合板用) 접착제(接着劑)의 증량(增量)에 관(關)한 연구(硏究))

  • It was planned and performed to study the possibility on the use of inexpensive and easily acquirable foliage powder, which processed by pulverizing after dried, instead of imported expensive wheat flour for the extending of plywood adhesives. Pine leaves of softwood trees, Poplar, Oak and Sycamore leaves of broad leaved species were selected and harvested to pulverize into the minute foliage powder. The harvested foliages from each selected species were pulverized into 40 mesh particles after dried at $100{\sim}105^{\circ}C$ condition during 24 hours in drying oven. To compare the extending effect of plywood adhesives with these foliage powders 100 mesh wheat flour using at current plywood industry was also prepared. Foliage powder and wheat flour were extended into 10, 20, 30, 50 and 100% to the urea and phenol formaldehyde resin. After plywoods were processed by the above extending method shear strength of extended plywoods were analyzed and discussed. The results obtained at this study are as follows: 1) Among 10% extensions of urea formaldehyde resin plywood, dry shear strength of plywood extended by wheat flours was the highest and that of non-extended plywood the next. Plywood extended with foliage powder showed the lowest dry shear strength. The order of dry shear strength of plywoods extended by foliage powder was that of Oak foliage powder extension, the best, that of Sycamore, that of Pine, and that of Poplar. 2) Among 20% extensions of urea formaldehyde resin plywood, plywood extended by wheat flour showed the highest dry shear strength, and the next was plywood by Poplar foliage powder. All these two showed higher dry shear strength than non-extension plywoods. Except Poplar, dry shear strength of foliage powder extension plywoods was bad, but the order of dry shear strength of plywoods extended by foliage powder was Pine, Poplar and Oak. 3) In the case of 30% extensions of urea formaldehyde resin plywood, dry shear strength of wheat flour extension was the highest and non-extension the next. Dry shear strength of foliage powder extension plywoods was poor with a rapid falling-off in strength. 4) Among 50% and 100% extensions of urea formaldehyde resin plywood, only wheat flour showed excellent dry shear strength. In the case of foliage powder extension, low dry shear strength showed at the 50% extension of Pine and Poplar, and plywoods of 50% extension of Oak foliage powder delaminated without measured strength. All plywoods of 100% foliage powder extension delaminated, and then shear strength were not measured. 5) Among wet shear strength of 10% extensions of urea formaldehyde resin plywood, wheat flour extension was the highest as in the case of dry shear strength, and non-extension plywood the next. Except Poplar foliage extension, all foliage powder extension plywoods showed low shear strength. 6) Wet shear strength of plywoods of 20% extension lowered in order of non-extension plywood, plywood of wheat flour extension and plywood of foliage powder extension, but other plywoods of foliage powder extension except plywoods of Poplar and Oak foliage powder extension delaminated. 7) Wet shear strength of 30% or more extension of urea formadehyde resin plywood were weakly measured only at 30% and 50% extension of wheat flour, and wet shear strength of plywoods extended by foliage powder were not measured because of delaminating. 8) Dry shear strength of phenol formaldehyde plywoods extended by 10% wheat flour was the best, and shear strength of plywoods extended by foliage powder were low, but the order was Oak, Poplar, and Pine. Plywood of Sycamore foliage powder extension delaminated. 9) In the case of 20% extensions of phenol formaldehyde resin, dry shear strength of plywood extended by wheat flour was the best, but plywood of Pine foliage powder extension the next, and the next order was Oak and Poplar foliage powder. Plywood of Sycamore foliage powder extension delaminated. 10) Among dry shear strength of 30% extensions of phenol formaldehyde plywood, that of Pine foliage powder extension was on the rise and more excellent than plywood of wheat flour extension, but Poplar and Oak showed the tendency of decreasing than the case of 20% extension. Plywood of Sycamore foliage powder extension delaminated. 11) While dry shear strength of 50% and 100% extension plywoods were excellent in the case of Pine foliage powder and wheat flour extension, that of hardwood such as Poplar, Oak, and Sycamore foliage powder extension were not measured because of delaminating. 12) As a filler the foliage powder extension of urea formaldehyde resin is possible up to 20% with Poplar foliage powder. And also as an extender for phenol formaldehyde resin, Pine foliage powder can be added up to the same amount as that in the case of wheat flour.
