A Kinetic Study on Nucleophilic Substitution Reactions of Phenyl Y-Substituted-Phenyl Carbonates with Z-Substituted-Phenoxides: Effect of Modification of Nonleaving Group from Benzoyl to Phenyloxycarbonyl on Reactivity and Reaction Mechanism

  • Second-order rate constants for the reactions of phenyl Y-substituted-phenyl carbonates 5a-g with Z-substituted-phenoxides ($k_{Z-PhO^-}$) have been measured spectrophotometrically in 80 mol % $H_2O$/20 mol % DMSO at $25.0{\pm}0.1^{\circ}C$. 4-Nitrophenyl phenyl carbonate (5e) is up to 235 times more reactive than 4-nitrophenyl benzoate (4e). The Br$\o$nsted-type plot for the reactions of 5e with Z-substituted-phenoxides is linear with ${\beta}_{nuc}=0.54$, which is typical for reactions reported previously to proceed through a concerted mechanism. Hammett plots correlated with ${\sigma}^o$ and ${\sigma}^-$ constants for the reactions of 5a-f with 4-chlorophenoxide exhibit highly scattered points. In contrast, the Yukawa-Tsuno plot results in an excellent linear correlation with ${\rho}_Y=1.51$ and r = 0.52, indicating that the leaving-group departure occurs at the rate-determining step (RDS). A stepwise mechanism, in which leaving-group departure occurs at RDS, has been excluded since the incoming 4-$ClPhO^-$ is more basic and a poorer nucleofuge than the leaving Y-substituted-phenoxides. Thus, the reaction has been concluded to proceed through a concerted mechanism. Our study has shown that the modification of the nonleaving group from benzoyl to phenyloxycarbonyl causes a change in the reaction mechanism (i.e., from a stepwise mechanism to a concerted pathway) as well as an increase in the reactivity.

Genetic analysis of polyketide biosynthetic genes isolated from Streptomyces albus, a salinomycin producer.

  • Sequence analysis of a DNA region encompassing the site of hybridization to actl, the gene for type II minimal polyketide synthase (PKS) for actinorhodin biosynthesis, from Streptomyces ablus revealed three more complete open reading frames additional to the already found two genes, plausibly encoding ${\beta}-ketoacyl$ synthase/acyl transferase (KS/AT) and chain length determining factor (ClF). The open reading frames (ORFs) were named salA, salD, and salE, from the upstream. In the homology analysis of the deduced amino acid sequences, SalA resembles the Streptomyces glaucescens Tcml, decaketide cyclase, SalD resembles acyl carrier protein in type II PKS, and SalE resembles the Actlll ketoreductase, The whole 4.4 kb of DNA sequence obeys the same conservation pattern as other type II PKSs. Therefore, we suggest that the 4.4 kb DNA from Streptomyces albus encompasses genes encoding enzymes for polyketide biogenesis in the organism and its organization is type II. The exsitence of SaIA, an analogue of the aromatic cyclase, revealed a relatedness of the 4.4 kb DNA with the aromatic PKS.

치과용 Ti-6Al-4V 합금 골 고정판 표면에 형성된 나노튜브의 부식거동 (Corrosion Behavior of Nanotube Formed on the Bone Plate of Ti-6Al-4V Alloy for Dental Use)

  • Titanium and titanium alloys are widely used for orthopedic and dental implants for their superior mechanical properties, low modulus, excellent corrosion resistance and good biocompatibility. In this study, corrosion behaviors of nanotube formed on the bone plate of Ti-6Al-4V alloy for dental use have been investigated. $TiO_2$ nanotubes were formed on the dental bone plates by anodization in $H_3PO_4$ containing 0.6 wt % NaF solution at $25^{\circ}C$. Electrochemical experiments were performed using a conventional three-electrode configuration with a platinum counter electrode and a saturated calomel reference electrode. Anodization was carried out using a scanning potentiostat (EG&G Co, Model 263A USA), and all experiments were conducted at room temperature. The surface morphology was observed using field emission scanning electron microscopy (FE-SEM) and energy dispersive x-ray spectroscopy(EDS). The corrosion behavior of the dental bone plates was examined using potentiodynamic test(potential range of -1500~2000 mV) in a 0.9% NaCl solution by potentiostat (EG&G Co, PARSTAT 2273. USA). The inner diameter of nanotube was about 150~180 nm with wall thickness of about 20 nm. The interspace of nanotube to nanotube was 50 nm. The passive region of the nanotube formed bone plates showed the broad range compared to non-nanotube formed bone plates. The corrosion surface of sample was covered with corrosion products.

매향 딸기로부터 anthocyanin 합성 유전자의 분리 및 과실발달 과정에서의 발현 분석 (A Set of Anthocyanin Biosynthetic Genes are Differentially Expressed in Strawberry (Fragaria x ananassa cv Maehyang) during the Fruit Development Process)

  • 매향' 딸기의 안토시아닌 생합성은 개화 후 26일째 시작되어 과실의 성숙기 동안 계속된다. 딸기로부터 안토시아닌의 생합성에 관여하는 주요 유전자를 분리하였다. 각각의 유전자에 대해, 다양한 식물체의 유사 유전자의 염기서열을 비교하여 PCR (polymerase chain reaciton) primer를 제작하였다. 숙기의 딸기에서 분리된 total RNA로부터 합성된 CDNA와 각 primer를 이용하여 RT (reverse transcriptase)-PCR을 수행하였다. 각 CDNA clone의 염기서열을 작성하여 분석한 결과, 이들은 안토시아닌 생합성에 관여하는 phenylalanine ammonia lyase (PAL), 4-cummarate CoA ligase (4CL), chalcone synthase (CHS), chalcone isomerase (CHI), flavanone-3-hydroxylase (F3H), dihydroflavonol 4-reductase (DFR), anthocyanidine synthase (ANS) 그리고 UDP-glucose:flavonoid-3-O-glucosyltransferase (UFGT) 효소에 해당되었다. Northern blot 분석 결과, 이들 유전자는 과실 발달과정에서 시기적으로 조절되었다. 특히 PAL을 제외한 모든 유전자는 과실에서만 주로 발현되었다. PAL, DFR 그리고 ANS유전자는 과실 초기 발달 단계인 개화 후 10일에 검출된 후 감소하다가, 22일에 다시 증가하기 시작하여 34일에 최대가 되었다. 한편, 다른 유전자들은 초기에는 발현되지 않다가, 안토시아닌이 축적되기 시작하는 개화 후 $22{\sim}30$일에 처음으로 검출되었다. 본 연구를 통해, 딸기 과실 발달과정에서 안토시아닌 생합성 과정에 관여하는 여러 유전자가 과실 숙기에 함께 조절되는 현상을 알 수 있다. 이러한 연구 결과는 안토시아닌 합성과정을 제어하는 조절 유전자가 존재한다는 것을 시사한다. 그리고 딸기의 안토시아닌 생합성 유전자의 발현패턴을 크게 두 가지로 나눌 수 있는 것으로 보아, 딸기의 안토시아닌 생합성에는 적어도 두 가지 서로 다른 조절 기작이 관여하여 색소 발달 과정을 제어할 것으로 보인다.

미생물에 의한 살어성물질의 생성 및 그 작용 (Production and Action of Microbial Piscicidal Substance)

  • 진균류인 Aspergilus flavus의 대사생성물인 aflatoxin이 발암성물질이카는 것이 알려진 이래 비병원성 미생물이라고 ale어왔던 여러가지 미생물의 독성물질 생성여부에 관한 연구가 많이 이루어져 우리 식생활에 미생물독 즉 mycotoxin에 대한 문제는 더욱 중요하게 되였다. 본 실험에서는 토양에서 분리한 방선균속 균주가 생성하는 살어성물질의 기본적인 성질은 pH 3~7사이는 안정하였으나 PH 8이상에서는 매우 불안정하였으며 열에 대해서도 비교적 불안정한 물질이었으며 어종과 어체중에 따른 독성의 비교는 체중이 증가함에 따라 생존시간이 길었지만 버들붕어와 미꾸라지가 타어종에 비해서 본 독성물질에 대한 저항성이 매우 켰다. 일반적으로 미생물독의 생물에 대한 작용$^{(24~26)}$은 hemolytic necrotizing, proteolytic, neurotoxic, cardiotoxic, hepatotoxic, nephrotoxic 등을 들 수 있으나 대부분의 독성물질은 상기 독작용 가운데 한 두 가지로서 그 주인이 된다. 본 물질의 잉어에 대한 각 조직의 succinic dehydrogenase 활성에 대한 조해작용을 조사한 결과 근육, 심장, 뇌 조직에 강한 조해현상을 나타내었다. 본 독성물질생성에 대한 금속 ion의 영향을 조사한 결과 $Cu^{2+}$, $Pb^{2+}$의 첨가가 크게 관계한다는 것을 알았으며 기타 본물질성장에 carbohydrate, amino acid, vitamin이 미치는 영향을 조사해 보고자 하며 어류이외의 타 온혈동물의 조직에 본독성물질을 작용시켜 그 결과를 알고자 한다. 한편 본 물질이 잉어의 각 조직에 대한 succinic dehydrogenase 활성에 미치는 정도 및 본 물질생성에 미치는 금속염의 영향을 볼 때 송$^{19~22}$ 등이 분이한 물질과는 차이가 있음을 알았다.소되었다. Alkaline protease는 배양 24시간에서 48시간까지는 0.1%구와 0.5%구가 대조구보다 효소력이 높았고 그 이상의 인삼엑기스가 첨가된 처리구보다 효소력이 낮았다. 그러나 48시간 이후부터는 효소력은 인삼엑기스 첨가량 순으로 감소를 나타내었다. period of May 13th-June 4th 1976. The study data was collected by the direct interview method based on the questionnaire which the investigator made out. The study result was analyzed by percentage, t - test. The findings can be summarized as follows: 1. General characteristics of nurses doing duty on puerperal ward: a. Nurses′average age was 24.8 years old. b. 84.6% had educational background of 4 years of college. c. 69.2% had a religion. d. 53.8% were married. e. 53.8% had clinical experience of 1 year -3 years. f, 61.5% did duty on puerperal ward during 1 year -3 years. g. 46.2% desired to do duty on obstetric ? gynecologic ward. 2. General characteristics of the women who were studied during their puerperal period: a. Women′s average age was 26.4 years old. b. 79.5% had educational background above high school.

Effects of Organic or Inorganic Acid Supplementation on Growth Performance, Nutrient Digestibility and White Blood Cell Counts in Weanling Pigs

  • Four experiments were conducted to investigate the effect of organic or inorganic acid supplementation on the growth performance, nutrient digestibility, intestinal measurements and white blood cell counts of weanling pigs. In growth trial (Exp I), a total of 100 crossbred pigs ({$Landrace{\times}Yorkshire$}${\times}$Duroc), weaned at $23{\pm}2$ days of age and $7.25{\pm}0.10kg$ average initial body weight (BW), were allotted to 5 treatments by body weight and sex in a randomized complete block (RCB) design. Three different organic acids (fumaric [FUA], formic [FOA] or lactic acid [LAA]) and one inorganic acid (hydrochloric acid [SHA]) were supplemented to each treatment diet. Each treatment had 5 replicates with 4 pigs per pen. During 0-3 wk, average daily gain (ADG), average daily feed intake (ADFI) and feed efficiency (G/F ratio) were not significantly different among treatments. However, pigs fed LAA or SHA diet showed improved ADG by 15 or 13% respectively and 12% greater ADFI in both treatments compared to CON diets. Moreover, compared to organic acid treatments, better ADG (p = 0.07) and ADFI (p = 0.09) were observed in SHA diet compared to pigs that were fed the diet containing organic acids (FUA, FOA or LAA). However, during 4-5 wk, no differences in ADG, ADFI and G/F ratio were observed among treatments. Overall, ADG, ADFI and G/F ratio were not affected by acidifier supplementation. Although it showed no significant difference, pigs fed LAA or SHA diets showed numerically higher ADG and ADFI than pigs fed other treatments. In metabolic trial (Exp II), 15 pigs were used to evaluate the effect of acidifier supplementation on nutrient digestibility. The digestibility of dry matter (DM), crude protein (CP), crude fat (CF), crude ash (CA), calcium (Ca) and phosphorus (P) was not improved by acidifier supplementation. Although the amount of fecal-N excretion was not different among treatments, that of urinary-N excretion was reduced in acidsupplemented treatments compared to CON group (p = 0.12). Subsequently, N retention was improved in acid-supplemented groups (p = 0.17). In anatomical trial (Exp III), the pH and $Cl^-$ concentrations of digesta in gastrointestinal (GI) tracts were not affected by acidifier supplementation. No detrimental effect of intestinal and lingual (taste bud) morphology was observed by acidifier supplementation particularly in inorganic acid treatment. In white blood cell assay (Exp IV), 45 pigs were used for measuring white blood cell (WBC) counts. In all pigs after LPS injection, WBC counts had slightly declined at 2 h and kept elevating at 8 h, then returned to baseline by 24 h after injection of lipopolysaccharide (LPS). However, overall WBC counts were not affected by acidifier supplementation. In conclusion, there was no difference between organic and inorganic acidifier supplementation in weanling pigs' diet, however inorganic acidifier might have a beneficial effect on growth performance and N utilization with lower supplementation levels. Furthermore, inorganic acidifier had no negative effect on intestinal measurements and white blood cell counts in weanling pigs. These results suggested that inorganic acidifier might be a good alternative to organic acidifiers in weanling pigs.

Electrochemical Characteristics of Nanotubular Ti-25Nb-xZr Ternary Alloys for Dental Implant Materials

  • Purpose: The purpose of this study was to investigate the electrochemical characteristics of nanotubular Ti-25Nb-xZr ternary alloys for dental implant materials. Materials and Methods: Ti-25Nb-xZr alloys with different Zr contents (0, 3, 7, and 15 wt.%) were manufactured using commercially pure titanium (CP-Ti), niobium (Nb), and zirconium (Zr) (99.95 wt.% purity). The alloys were prepared by arc melting in argon (Ar) atmosphere. The Ti-25Nb-xZr alloys were homogenized in Ar atmosphere at $1,000^{\circ}C$ for 12 hours followed by quenching into ice water. The microstructure of the Ti-25Nb-xZr alloys was examined by a field emission scanning electron microscope. The phases in the alloys were identified by an X-ray diffractometer. The chemical composition of the nanotube-formed surfaces was determined by energy-dispersive X-ray spectroscopy. Self-organized $TiO_2$ was prepared by electrochemical oxidation of the samples in a $1.0M\;H_3PO_4+0.8wt.%$ NaF electrolyte. The anodization potential was 30 V and time was 1 hour by DC supplier. Surface wettability was evaluated for both the metallographically polished and nanotube-formed surfaces using a contact-angle goniometer. The corrosion properties of the specimens were investigated using a 0.9 wt.% aqueous solution of NaCl at $36^{\circ}C{\pm}5^{\circ}C$ using a potentiodynamic polarization test. Result: Needle-like structure of Ti-25Nb-xZr alloys was transform to equiaxed structure as Zr content increased. Nanotube formed on Ti-25Nb-xZr alloys show two sizes of nanotube structure. The diameters of the large tubes decreased and small tubes increased as Zr content increased. The lower contact angles for nanotube formed Ti-25NbxZr alloys surfaces showed compare to non-nanotube formed surface. The corrosion resistance of alloy increased as Zr content increased, and nanotube formed surface showed longer the passive regions compared to non-treatment surface. Conclusion: It is confirmed that corrosion resistance of alloy increased as Zr content increased, and nanotube formed surface has longer passive region compared to without treatment surface.

아미노산 후처치의 이종 심낭보철편 석회화 완화 효과 (Mitigation of Calcification in Bovine Pericardial Bioprosthesis after Amino Acids Posttreatment)

  • Glutaraldehyde (GA)에 보존한 이종보철편들은 생체 이식 후 석회화 변성을 보이는 문제점을 나타내므로 이 단점을 보완하기 위해 연구자는 GA 용액에 Mg염을 첨가 칼슘의 반응을 막아 석회화를 완화하며 더불어 chitosan 및 glutamate 등의 아미노산으로 후처치하여 석회화 완화를 극대화하고자 하였다. 대상 및 방법: 우심낭을 정방형 조각으로 만들어 40조각의 우심낭편은 대조군으로 MgC1$_2$6$H_2O$를 4g/L 첨가한 0.625% GA 용액에 보존하고(1군), 40조각은 이 GA 용액에 보관하다 이식 24시간 전 2% chitosan 용액에 옮겨 보존(2군), 또 다른 40조각 역시 같은 방법으로 8% glutamate 용액에 후처치(3군)하여, 이들을 40마리의 백서 복부 피하에 각각 한 조각씩 이식하여 1개월, 2개월, 3개월 및 4개월째에 적출, spectophotometry로 침착한 칼슘을 정량하였다. 결과: 이식 후 1개월 및 2개월에는 세균간의 석회화 정도에 차이가 없었으나. 3개월째에는 1군 5.45$\pm$0.42 mg/g, 2군 3.22$\pm$1.31 mg/g, 3군 4.20$\pm$0.55 mg/g의 통계적 차이를 보여, 2군에서 유의한 석회화 완화 효과가 관찰되고 있고 (p<0.05), 4개월째에는 1군 6.01$\pm$1.21 mg/g, 2군 3.78$\pm$1.82 mg/g, 3군 3.92$\pm$0.92 mg/g으로 2, 3군에서 유의한 석회화 완화 효과를 보여주고 있다(p<0.05). 결론: 즉 2% chitosan 후처치로 3개월의 시간이 경과함에 따라 피하에 이식된 보철편의 석회화가 완화되었으며 8% glutamate는 4개월에서 그 완화효과를 보여주고 있다.다. 현대의 실정 에 맞는 과학적 제조방법을 이용하여 산업화한다면 재래식으로 인해 걸리는 시간, 노력 에 걸맞은 가격, 품질 등을 유지해 나갈 수 있으리라 본다.는 음식 10위를 조사한 결과 선호음식의 경우 저체중군과 정상군의 경우 순위는 달랐지만 10위에 선별된 음식들이 주로 과 (수박, 귤, 딸기, 복숭아, 바나나, 포도, 사과)과 음료수(사이다, 환타) 등 열량밀도가 낮고 수분함량이 높은 식품인 반면 과체중군은 햄버거, 돈까스, 튀김 만두, 샌드위치, 쇠고기 및 돼지고기 구이 등이 선호음식 10위내에 포함되어 정상군이나 저체중군보다 고지방.고단백 음식들의 기호도가 과체중군에서 높음을 확인하였다., 단맛을 좋아하는 사람이 싫어하는 사람보다 열량과 당질섭취량은 유의 적으로 적게, 지방 섭취량은 유의적으로 높았다. 짠맛을 좋아하는 사람은 나트륨 섭취량이 7890 mg으로 짠맛을 싫어하는 사람 5579 mg보다 유의적 (p < 0.05)으로 많은 양을 섭취하였다.고, Ca : P 중량비가 각각 2.63$\pm$1.99, 1.79$\pm$0.39, 2.80$\pm$0.53, 8.35$\pm$12.87이었으며, Ca : Mg 중량비는 각각 11.86$\pm$540, 9.29$\pm$1.34, 9.09$\pm$2.09, 32.50$\pm$41.35이었다. 칼슘과 마그네슘의 흡수에 영향을 미치는 P : Mg 분자량비는 곡류가공품이 4.11$\pm$1.54, 라면류가 4.17$\pm$0.67, 레토르트식품은 2.58$\pm$0.45, 음료류는 2.59$\pm$2.50으로 나타났다. 위의 연구결과에서 볼 때 시료의 93.0%가 실제측정값이 표시량의 80% 이상이어야

Isolation, Expression Pattern, Polymorphism and Association Analysis of Porcine TIAF1 Gene

  • TIAF1 is a TGF-${\beta}$1-induced anti-apoptotic factor that plays a critical role in blocking TNF (tumor necrosis factor) cytotoxicity in mouse fibroblasts and participates in TGF-${\beta}$-mediated growth regulation. In this study, we obtained the full-length cDNA sequence of the porcine TIAF1 gene. Real-time PCR further revealed that the TIAF1 gene was expressed at the highest level in liver and kidney with prominent expressions detected in uterus, and lower levels detected in heart, spleen, lung, stomach, small intestine, skeletal muscle and fat of Large White pigs. Sequence analysis indicated that a 6 base-pair deletion mutation existed in the exon of the TIAF1 gene between Meishan and Large White pigs. This mutation induced deletion of Gln and Val amino acids. PCR-RFLP was used to detect the polymorphism in 394 pigs of a "Large White${\times}$Meishan" $F_{2}$ resource population and four purebred pig populations. The frequencies of the A allele (with a 6 bp deletion) were dominant in Chinese Meishan and Bamei pigs, and the frequencies of the B allele (no 6 bp deletion) were dominant in Large White and Landrace pigs. Association analyses revealed that the deletion mutation had highly significant associations (p<0.01) with meat marbling score of the thorax-waist longissimus dorsi (LD) muscle (MM1) and intramuscular fat percentage (IMF), and significant associations (p<0.05) with carcass length (CL). The results presented here supply evidence that the 6 bp deletion mutation in the TIAF1 gene affects porcine meat quality and provides useful information for further porcine breeding.

  • 고대에는 단청이나 고분벽화, 사찰벽화, 불화 등 문화재에 다양한 색상을 구현하고자 무기안료 또는 유기안료를 많이 사용하였다. 이 연구는 통도사 영산전내 벽체에 그려진 사찰벽화의 안료를 휴대용X선형광분석기로 정성분석하여 사용안료의 종류를 밝히고자 하였다. 성분분석 결과와 기존 고대 벽화 안료의 분석 결과들을 종합해 볼 때, 영산전 벽화에 사용된 적색은 주사와 석간주로 추정되며, 백색은 연백이 사용되었고, 일부 호분이나 백악, 백토 등의 사용 가능성도 있다. 녹색은 2가지가 사용되었는데 주로 바탕칠에 사용된 밝은 녹색은 녹토이고, 광배부분 등의 진한녹색은 석록이나 동록, 녹염동광으로 판단된다. 황색과 흑색은 각각 황토와 먹이 사용된 것으로 보이며, 육색과 분홍 등 원색이 아닌 안료들은 적색에 백색안료를 혼합하거나, 진적색에 황색안료를 혼합하는 것과 같이 2가지 이상의 안료를 적절하게 섞어서 사용하였을 것으로 추정된다. 결론적으로 성분분석으로만 판단하였을 때 통도사 영산전에 사용된 안료는 모두 고대에 주로 이용되었던 무기 안료임을 알 수 있었다. 다만, 문화재임으로 안료의 일부를 채취하여 결정구조분석 등의 방법으로 광물을 동정한 것이 아니므로 특정 원료 광물을 명확하게 판단하기는 어려웠다.
