Thermal Inactivation Parameters of Peroxidase in Flammulina velutipes and Lyophyllum ulmarium (팽이 및 만가닥버섯에서 추출한 peroxidase의 열 불활성화 특성)

  • Peroxidase was used as a standard enzyme to determine optimum blanching conditions of Flammulina velutipes and Lyophyllum ulmarium. Crude peroxidase extracted from raw mushrooms had maximum activity at $10{\sim}15^{\circ}C$ and pH 5.5 (50 mM, potassium phosphate buffer) using substrates of $H_2O_2$ and p-Phenylendiamine. Thermal inactivation of the crude peroxidase followed the first-order kinetics. The activation energy and z value of the crude peroxidase for F. velutipes were 59.58 kcal/mol and $9.0^{\circ}C$, whereas were 43.05 kcal/mol and $12.4^{\circ}C$ for L. ulmarium, respectively. On the basis of thermal kinetics parameters obtained, the optimum blanching conditions for F. velutipes and L. ulmarium were 1 min at $70^{\circ}C$ and 5 min at $80^{\circ}C$, respectively. Activation energies and z values of peroxidases extracted from heat-treated mushrooms were 7.97 and 6.55 kcal/mol, and $59.8^{\circ}C\;and\;74.1^{\circ}C$ for F. velutipes and L. ulmarium, respectively.

Effects of Gintonin-enriched fraction on the gene expression of six lysophosphatidic receptor subtypes

  • Background: Gintonin, isolated from ginseng, acts as a ginseng-derived lysophosphatidic acid (LPA) receptor ligand and elicits the [Ca2+]i transient through six LPA receptor subtypes (LPARSs). However, the long-term effects of gintonin-enriched fraction (GEF) on the gene expression of six LPARSs remain unknown. We examined changes in the gene expression of six LPA receptors in the mouse whole brain, heart, lungs, liver, kidneys, spleen, small intestine, colon, and testis after long-term oral GEF administration. Methods: C57BL/6 mice were divided into two groups: control vehicle and GEF (100 mg/kg, p.o.). After 21-day saline or GEF treatment, total RNA was extracted from nine mouse organs. Quantitative-real-time PCR (qRT-PCR) and western blot were performed to quantify changes in the gene and protein expression of the six LPARSs, respectively. Results: qRT-PCR analysis before GEF treatment revealed that the LPA6 RS was predominant in all organs except the small intestine. The LPA2 RS was most abundant in the small intestine. Long-term GEF administration differentially regulated the six LPARSs. Upon GEF treatment, the LPA6 RS significantly increased in the liver, small intestine, colon, and testis but decreased in the whole brain, heart, lungs, and kidneys. Western blot analysis of the LPA6 RS confirmed the differential effects of GEF on LPA6 receptor protein levels in the whole brain, liver, small intestine, and testis. Conclusion: The LPA6 receptor was predominantly expressed in all nine organs examined; long-term oral GEF administration differentially regulated LPA3, LPA4, and LPA6 receptors in the whole brain, heart, lungs, liver, kidneys, small intestine, and testis.

A FA Intersectional Hybrid Lily 'Orange Crown' with Orange Petals (오렌지색 FA 아속간 잡종나리 '오렌지 크라운' 육성)

  • A FA (Lilium formolongi $\times$ Lilium asiatic hybrid) intersectional hybrid lily cultivar 'Orange Crown' was developed in 2007 at National Institute of Horticultural and Herbal Science (NIHHS), Rural Development Administration (RDA) of Korea. The cross and immature embryo rescue was conducted between female parents 'Migreen' (Lilium formolongi 'Raizan' $\times$ Lilium asiatic hybrid 'A61') and male parent 'A01-187' (L. asiatic 'A96-28' $\times$ 'Sanzio') by cut style pollination method at Suwon in 2001. It was preliminarily selected as 'FA04-27' in 2004. Multiplication, bulb production and characteristic tests were conducted from 2004 to 2007. The evaluation of characteristics and preference were surveyed at a lily flower show of NIHHS at Taean in 2007. 'Orange Crown' flowers in the late of June and grows more than 183 cm stem length. Flowers bloom upward-facing, a little spotted and orange (RHS, O24A) petals. The pollen of 'Orange Crown' was sterile.

  • This study was designed to evaluate the characteristics, land use and genesis of well adapted paddy soils. They were mostly classified as the Haplaquepts in the U.S.D.A soil taxonomy and as the Grey soils in the Japanese soil classification system. The proportion of these soils in the total acreage of paddy lands was thirty three percents. The fifty four percents in average of these soils were distributed on the local valley and fans on gentle slopes developed from granite, granite gneiss and shale parent materials. The rests were on the fluvio-marine deposits and alluvial deposits. The soils were characterized with prominant development of gleized horizons and clayey or fine loamy textured category. The available soil depth and the ground water level were relatively deep. The base saturation percent were high with weak acidic pH. The potential productivity of these soils was high.

  • This study was carried out to investigate the supplying capacity of various forms(Water soluble, exchangeable, and available) of potassium in soils of different site quality by using surface soils from the Japanese cedar(Cryptomeria japonica) forests in Cheju island. Major results can be summerized as follows. Using continuous leaching methods with 0.01N-HCl, accumulated amounts of available potassium from surface soils of site upper and site low were about 0.6me/100g and 0.4me/100g, respectively. The release ratio of available potassium in the first $1{\ell}$ leaching of 0.01N-HCl to the total available potassium leached with $6{\ell}$ of 0.01N-HCl was over 80% for both sites, and this suggester that surface soil of both sites had the weak potassium adsorptivity as the typical volcanic ash soil. The ratio of the exchangeable potassium to the water soluble potassium was 1 : 1 for both sites. The ratio of the exchangeable potassium to the available potassium was 1 : 1 for the site low but the smaller ratio value for the site upper, indicating that the potassium supplying power of the site upper was greater than that of the site low. Available potassium was highly correlated with exchangeable Ca($0.83^{**}$) and Mg($0.84^{**}$).

  • The study was intended to analyze the interaction effects of paraquat and oxytluorfen as an orchard herbicide-mixture. Data were prepared from the former report of authors. The algebraic expression for the actions of paraquat and oxyfluorfen on the control percentages of peach orchard weeds, and their interactions were determined from the multiple regression polynomial and plotted in three-dimensional graphs. As a result of treatments by combination of paraquat and oxyfluorfen on the field which was dominated by perennial weeds, the most effective interactions were detected at combination rates of $245\;gHa^{-1}$ paraquat and $470-705\;gHa^{-1}$ oxyfluorfen. However, to develope the long-term weeding-efficacies, the combination rates of paraquat are expected to raise up to $500-700\;gHa^{-1}$, and oxyfluorfen to fit at lower limits of rates, respectively.

Determination of Nitrogen Content in Rice Tissue Using Near Infrared Spectroscopy

  • The rice plant is one of the important staple crops in Korea. The high yield with low cost in rice is required the soil fertility and the development of new precise method of fertilizer application by nutritional diagnosis. Now, in Korea, the nitrogen application system for the rice plant is composed of the basal fertilization, fertilization at tillering stage and fertilization at panicle stage, which the nitrogen fertilization at panicle stage amount to about 30 percent in the total amount. Thus, this experiment carried out to the development of the system that can measure the nitrogen content in the rice plant at panicle stage rapidly with the near infrared spectroscopy, and to predict the appropriate quantity of the nitrogen fertilization at panicle stage based on calibration model for test of nitrogen content in rice plant. The samples were collected from 48 varieties in 4 regions which are mainly cultivated in the southern part of Korea. And then, it collected by classifying into the leaf, the whole plant and the stem since 7 days before the nitrogen fertilization at panicle stage. The ranges of the nitrogen contents were 1.6∼4.0%, 1.7∼3.0% and 1.4∼2.7% in the leaf, the whole plant and the stem, respectively. In the calibration models created by each part of the plant under the Multiple Linear Regression(MLR) method, the calibration model for the leaf recorded the relatively high accuracy. The mutual crossing test on unknown samples were carried out using Partial Least Square(PLS) calibration model. That is, the nitrogen content in the stem was tested by calibration model made by the leaf model and that of stem was tested by calibration model made by whole plant sample. When unknown leaf sample was tested by calibration model made by all sample that collected from each part in rice plant such as leaf, stem and whole plant, it recorded the highest accuracy. As a result, to test the nitrogen content in the rice plant at panicle stage, the nitrogen content in the leaf shall be tested by the calibration model composed of the leaf, the stem and the whole plant. In future, to estimated the amount of nitrogen fertilization at panicle stage for rice plant , it will be calculated based on regression model between rice yield and nitrogen content of leaf measured by calibration model made by mixed sample including leaf, stem and whole plant.

  • If there are problems for us to sleep, we are faced with fatigue and dizziness in the day. Snoring and OSAS (obstructive sleep apnea syndrome) during sleeping are the main cause of sleep disorder. Treatments through surgical method and sleep splint can be performed to treat snoring and OSAS. Relapse of snoring and OSAS is common after treatment by surgical method. But, Recently sleep splint is frequently applied to treat snoring and OSAS with surgical treatment, because it is convenient and conservative. Sleep splint treat snoring and OSAS by ensuring airway through nose. As first step of fabrication occlusal bite is gained at a point that patient get feeling of increased nasal breathing in supined position, and next, the bite is transfered to sleep splint. This study surveyed the effect of sleep splint by questionnaire to the out-patients (the Dept. of Oral and Maxillofacial Surgery, Chonbuk National University Hospital) weared sleep splint and their partners, secondarily measured airflow through nose by aerophone II after wearing sleep splint and finally evaluated the effect of treatment of snoring and OSAS by sleep splint. The obtained result were as follows; 1. Though 'sleep splint' couldn't eliminate fundamental problems of snoring, it could improve the symptoms when patients were selected could using the 'Nakagawa's respiration method'. 2. Patients who used the sleep splint could breathe stably when patients are sleeping stably. Wearing a 'sleep splint' improved airflow by expanding the upper airway. 3. Even though sleep splint can be made with variable materials, the patients expressed the most satisfaction on the splint with '0.75mm hard shell'. 4. The 'Herbst' may allows the mandible to move the TMJ to relax. Nevertheless, some patients experienced a discomfort or irritation. 5. In Snoring and OSAS cases, it is recommended that patients should first explore non-surgical options prior to choosing a surgical treatment.

A Study on Properites of PV Solar cell AZO thin films post-annealing by RTP technique (RTP 공정을 통한 태양전지용 AZO 박막의 후열처리 특성연구)

  • In this paper, ZnO:Al thin films with c-axis preferred orientation were prepared on Soda lime glass substrates by RF magnetron sputtering technique. AZO thin film were prepared in order to clarify optimum conditions for growth of the thin film depending upon process, and then by changing a number of deposition conditions and substrate temperature conditions variously, structural and electrical characteristics were measured. For the manufacture of the AZO were vapor-deposited in the named order. It is well-known that post-annealing is an important method to improve crystal quality. For the annealing process, the dislocation nd other defects arise in the material and adsorption/decomposition occurs. The XRD patterns of the AZO films deposited with grey theory prediction design, annealed in a vacuum ambient($2.0{\times}10-3$Torr)at temperatures of 200, 300, 400 and $500^{\circ}C$ for a period of 30min. The diffraction patterns of all the films show the AZO films had a hexagonal wurtzite structure with a preferential orientation along the c-axis perpendicular to the substrate surface. As can be seen, the (002)peak intensities of the AZO films became more intense and sharper when the annealing temperature increased. On the other hand, When the annealing temperature was $500^{\circ}C$ the peak intensity decreased. The surface morphologies and surface toughness of films were examined by atomic force microscopy(AFM, XE-100, PSIA). Electrical resistivity, Gall mobility and carrier concentration were measured by Hall effect measuring system (HL5500PC, Accent optical Technology, USA). The optical absorption spectra of films in the ultraviolet-visibleinfrared( UV-Vis-IR) region were recorder by the UV spectrophotometer(U-3501, Hitachi, Japan). The resistivity, carrier concentration, and Hall mobility of ZnS deposited on glass substrate as a function of post-annealing.

  • Biological nitrogen fixation is an important process for academic and industrial aspects. This review will briefly compare industrial and biological nitrogen fixation and cover the characteristics of biological nitrogen fixation studied in Azotobacter vinelandii. Various organisms can carry out biological nitrogen fixation and recently the researches on the reaction mechanism were concentrated on the free-living microorganism, A. vinelandii. Nitrogen fixation, which transforms atmospheric $N_2$ into ammonia, is chemically a reduction reaction requiring electron donation. Nitrogenase, the biological nitrgen fixer, accepts electrons from biological electron donors, and transfers them to the active site, FeMo-cofactor, through $Fe_4S_4$ cluster in Fe protein and P-cluster in MoFe protein. The electron transport and the proton transport are very important processes in the nitrogenase catalysis to understand its reaction mechanism, and the interactions between FeMo-cofactor and nitrogen molecule are at the center of biological nitrogen fixation mechanism. Spectroscopic studies including protein X-ray crystallography, EPR and $M{\ddot{o}}ssbauer$, biochemical approaches including substrate and inhibitor interactions as well as site-directed mutation study, and chemical approach to synthesize the FeMo-cofactor model compounds were used for biological nitrogen fixation study. Recent research results from these area were presented, and finally, a new nitrogenase reaction mechanism will be proposed based on the various research results.
