Solution Conformations of the Substrates and Inhibitor of Hepatitis C Virus NS3 Protease

  • Hepatitis C virus (HCV) has been known to be an enveloped virus with a positive strand RNA genome and the major agent of the vast majority of transfusion associated cases of hepatitis. For viral replication, HCV structural proteins are first processed by host cell signal peptidases and NS2/NS3 site of the nonstructural protein is cleaved by a zinc-dependent protease NS2 with N-terminal NS3. The four remaining junctions are cleaved by a separate NS3 protease. The solution conformations of NS4B/5A, NS5A/5B substrates and NS5A/5B inhibitor have been determined by two-dimensional nuclear magnetic resonance (NMR) spectroscopy. NMR data suggested that the both NS5A/5B substrate and inhibitor appeared to have a folded tum-like conformation not only between P1 and P6 position but also C-terminal region, whereas the NS4B/5A substrate exhibited mostly extended conformation. In addition, we have found that the conformation of the NS5A/5B inhibitor slightly differs from that of NS5A/5B substrate peptide, suggesting different binding mode for NS3 protease. These findings will be of importance for designing efficient inhibitor to suppress HCV processing.

USE OF MOLD INHIBITOR FOR FEED STORAGE AND IMPROVED CHICK PERFORMANCE

  • Two experiments were conducted to evaluate the effect of mold inhibitor in the ration which had two different protein levels (18% and 12%) and two different particle sizes (80 or 40% of the particles in the ration less than 1.19 mm). The experimental diets with ave. 12.7% moisture which were treated at the level of 0.1% mold inhibitor were stored under 85% humidity and at $29{\pm}1^{\circ}C$ for 10 to 40 days. In experiment 1, after 40 days of storage the $CO_2$ production in the feed treated with mold inhibitor was higher (p < 0.01) than when 40% of the ration's panicle size was 1.19 mm. Aflatoxin production in the experimental diet with mold inhibitor was affected (p<0.05) by the levels of protein and the different particle size ranges after 40 days storage. The interaction of protein levels and particle size ranges on the anatoxin and $CO_2$ production was significant (p<0.05) at 40 days storage. In experiment 2, there was a decrease in total body weight gain and total feed intake observed in chicks fed the untreated diet of 18% protein with 40% of the particles in the ration less than 1.19 mm stored for 40 days. Feed conversion was depressed (p<0.05) in the chicks fed the untreated diets of both particle sizes. Particle size X types of feed interaction in feed conversion was significant (p<0.05).

Synthesis of 2-(4-Hydroxyphenyl)benzofurans and Their Application to $\beta$-Amyloid Aggregation Inhibitor

  • The facile synthesis of a series of 2-(4-hydroxyphenyl)benzofurans (4a-e) is described. The one-pot reaction of 4-substituted phenols with the chloride 1 in the presence of zinc chloride afforded 3-methylthio-2-(4-acetoxyphenyl)benzofurans (2a-e). The compounds 4a-e were obtained from the hydrolysis of 2a-e followed by the desulfurization of the resulting 3-methylthio-2-(4-hydroxyphenyl)benzofurans (3a-e). 5-Methyl-3-p-toluoyl-2 -[4-(3-diethylaminopropoxy)phenyl]benzofuran (7), a $\beta$-amyloid aggregation inhibitor, was synthesized by three steps starting from 4a.

Characterization of an Elastase Inhibitor Produced by Streptomyces lavendulae SMF11

  • An elastase inhibitor, SMFEI02, was isolated from culture broth of Streptomyces lavendulae SMF11. The inhibitor was purified by ultrafiltration followed by XAD-7 column and Dowex-1 anion-exchange chromatographies, and preparative HPLC. The molecular formula was determined to be $C_{14}H_{16}N_2O_2$ (MW244) by HRFAB-MS analysis. The inhibitor was identified to be a diketopiperazine cyclo(S-Phe-S-Pro) by the optical rotation value and MNR spectral data, and showed inhibitory activities for trypsin, chymotrypsin, cathepsin B, and papain as well as elastase with the Ki values ranging from 1.78mM to $2.86{\;}\mu\textrm{m}$. The inhibition showed a competitive mode for elastase, chymotrypsin, and cathepsin B, whereas it showed a noncompetitive mode for trypsin and papain.

Valistatin (3-Amino-2-Hydroxy-4-Phenylbutanoyl-Valyl-Valine), a New Aminopeptidase M Inhibitor, Produced by Streptomyces sp. SL20209

  • Valistatin, a new inhibitor of aminopeptidase M(AP-M) was discovered in the culture broth of Streptomyces sp. SL20209 isolated from a soil sample. The inhibitor was purified by extraction with n-butanol and the various column chromatographies, and then isolated as whitish powder. The $^1 H-and ^1 H, ^1 H-COSY$ NMR studies, amino acid analysis, and fragmentation patterns by FAB-MS suggested the presence of one 3-amino-2-hydroxy-4-phenylbutanoic acid and two valine residues in the inhibitor. Thus, the structure of valistatin was determined as 3-amino-2-hydroxy-4-phenylbutanoyl-valyl-valine. Valistatin has the molecular formular $C_20H_31N_3 O_5$ (MW 394), and its $IC_50$ value against hog kidney AP-M was determined to be 3.12 $mu g/ml$.

Garcinexanthone G, a Selective Butyrylcholinesterase Inhibitor from the Stem Bark of Garcinia atroviridis

  • The present study was undertaken to investigate the isolated compounds from the stem bark of Garcinia atroviridis as potential cholinesterase inhibitors and the ligand-enzyme interactions of selected bioactive compounds in silico. The in vitro cholinesterase results showed that quercetin (3) was the most active AChE inhibitor ($12.65{\pm}1.57{\mu}g/ml$) while garcinexanthone G (6) was the most active BChE inhibitor ($18.86{\pm}2.41{\mu}g/ml$). It is noteworthy to note that compound 6 was a selective inhibitor with the selectivity index of 11.82. Molecular insight from docking interaction further substantiate that orientation of compound 6 in the catalytic site which enhanced its binding affinity as compared to other xanthones. The nature of protein-ligand interactions of compound 6 is mainly hydrogen bonding, and the hydroxyl group of compound 6 at C-10 is vital in BChE inhibition activity. Therefore, compound 6 is a notable lead for further drug design and development of BChE selective inhibitor.

Ginger Extract as Green Corrosion Inhibitor for Steel in Sulfide Polluted Salt Water

  • Extract of ginger has been evaluated as a green inhibitor for the corrosion of steel in sulfide polluted NaCl solution using potentiodynamic polarization, electrochemical impedance spectroscopy (EIS) and electrochemical frequency modulation (EFM) techniques. Potentiodynamic polarization measurements showed that this extract acts as a mixed type inhibitor but mainly inhibits the cathodic reaction. The inhibition efficiency was found to increase with inhibitor concentration reaching to approximately 83.9% using 250 ppm of ginger. Nyquist plots show a single capacitive loop in uninhibited and inhibited solutions. From EFM the causality factors are very close to theoretical values which indicate that the measured data are of good quality. The adsorption process of the studied extract on steel surface obeys Temkin adsorption isotherm. The results obtained from the different electrochemical techniques were in good agreement which prove the validity of these tools in measurement of corrosion rate. Ginger extract has no effect on Escherichia Coli and can be applied safely on waste water treatment plants.

Roles of Opioid Receptor Subtype in the Spinal Antinociception of Selective Cyclooxygenase 2 Inhibitor

  • Background: Selective inhibitors of cycloosygenase (COX)-2 are commonly used analgesics in various pain conditions. Although their actions are largely thought to be mediated by the blockade of prostaglandin (PG) biosynthesis, evidences suggesting endogenous opioid peptide link in spinal antinociception of COX inhibitor have been reported. We investigated the roles of opioid receptor subtypes in the spinal antionociception of selective COX-2 inhibitor. Methods: To examine the antionociception of a selective COX-2 inhibitor, DUP-697 was delivered through an intrathecal catheter, 10 minutes before the formalin test in male Sprague-Dawley rats. Then, the effect of intrathecal pretreatment with CTOP, naltrindole and GNTI, which are ${\mu}$, $\delta$, and k opioid receptor antagonist, respectively, on the analgesia induced by DUP-697 was assessed. Results: Intrathecal DUP-697 reduced the flinching response evoked by formalin injection during phase 1 and 2 Naltrindole and GNTI attenuated the antinociceptive effect of intrathecal DUP-697 during both phases of the formalin test, CTOP reversed the antinociception of DUP-697 during phase 2, but not during phase 1, Conclusions: Intrathecal DUP-697, a selective COX-2 inhibitor, effectively relieved inflammatory pain in rats. The $\delta$ and $\kappa$ opioid receptors are involved in the activity of COX-2 inhibitor on the facilitated state as well as acute pain at the spinal level, whereas the ${\mu}$ opioid receptor is related only to facilitated pain.

Trichoderma sp. MR-93 균주가 생산하는 Isocyanide 계열의 Melanin 생성 저해물질 (Production of the Isocyanide Inhibitor of Melanin Biosynthesis by Trichoderma sp. MR-93)

  • During the screening of inhibitors of melanin biosynthesis from microbial secondary metabolites, a fungal strain MR-93 which was capable of producing high level of an inhibitor was selected from plant leaf. Based on taxonomic studies, the fungus could be classified as a strain of Trichoderma sp.. The active compound (MR-93D) was purified from the culture broth by Diaion HP-20 column chromatography, ethylacetate extraction, Sephadex LH-20 column chromatography and HPLC. The inhibitor was identified as 4-hydroxy-8-isocyano-l-oxaspiro[4-4]cyclonon-8-en-2- one by spectroscopic methods of UV, $^{1}$H-NMR, ESIMS and IR. MR-93D showed a strong tyrosinase inhibitory activity with 0.03 $\mu$g/m of IC$_{50}$ value. It also inhibited melanin biosynthesis with 35 mm inhibition zone at 30 $\mu$g/paper disc in Streptomyces bikiniensis, a bacterium used as an indicator organism in this work.
