Antioxidant Effects of the Methanol Extracts Obtained from Aerial Part and Rhizomes of Ferns Native to Korea (자생 양치식물 지상부와 근경 메탄올 추출물의 항산화 효과)

  • Phenolic compound contents and scavenging activity on DPPH and ABTS radicals in twelve fern species were studied by analysing ultrasonification extraction of dried aerial parts and rhizomes using methanol solvent. Total polyphenol content ranged from 2.97 to 140.1 mg per 1 g dried sample and 0.80 to $49.58\;mg{\cdot}g^{-1}$ fresh sample. Highest polyphenol content was obtained with aerial part of Davallia mariesii and Polystichum lepidocaulon. Total flavonoid content of dried sample was $2.56{\sim}34.91\;mg{\cdot}g^{-1}$ and fresh sample $0.71{\sim}11.49\;mg{\cdot}g^{-1}$. Higher flavonoids were obtained with dried aerial part of Dryopteris crassirhizoma, but with fresh aerial part of P. lepidocaulon. In general, total polyphenol content was higher in rhizomes, except in case of total flavonoid. Scavenging effects on DPPH and ABTS radicals was higher with rhizome extracts compared to aerial parts. The rhizome of Polystichum polyblepharum showed highest effects on both radicals. However, the amount of fresh sample for $RC_{50}$ with consideration of water content and extraction yield. P. lepidocaulon aerial part was more advantageous than P. polyblepharum rhizome. All the fern extracts except for the Coniogramme japonica extract demonstrated superior scavenging effects on ABTS radicals, being similar activity of ascorbic acid and BHT.

  • The aim of this study was to investigate the applicability of Chinese mung bean as a natural antioxidant agent. This study evaluated the phenolic compounds content and antioxidative activity of methanol extract from Chinese mung bean. Antioxidative activities were measured by in vitro models such as 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, and 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid(ABTS) radical scavenging activity. The contents of total phenolics and total flavonoids of Chinese mung bean extract were $174.83{\pm}2.90GAE\;mg/g$ and $68.87{\pm}2.84QE\;mg/g$, respectively. The antioxidative activities of Chinese mung bean extract were significantly increased in a dose dependent manner on DPPH radical scavenging and ABTS radical scavenging(p<0.05). The concentration of Chinese mung bean extract that reduces the free radical ABTS about 50%($IC_{50}$) was 2.85 mg/mL. These results suggest that Chinese mung bean may have great potential as a natural antioxidant source linked with health benefits.

Radical Scavenging Activity of Ethanol Extracts and Solvent Partitioned Fractions from Various Red Seaweeds (홍조류 에탄올 추출물 및 다양한 용매 분획물의 라디칼 소거능)

  • The EtOH extracts of red seaweeds (Symphyocladia latiuscula, Chondrus ocellatus and Carpopeltis affinis) and solvent partitioned fractions were investigated for their 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) and 1,1-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging effects and the total phenolic contents were correlated with ABTS and DPPH radical scavenging activities. The EtOH extracts and their solvent partitioned fractions exhibited strong ABTS and DPPH radical scavenging activities. Among the solvent partitioned fractions obtained from n-Hexane (HX), methylenchloride (MC), ethylacetate (EA), and buthanol (BuOH), the HX fraction from C. affinis showed higher radical scavenging activities than other fractions. Total phenolic contents showed significant correlation ($r^2$ = 0.709) with ABTS radical scavenging activity. The results of this study suggest that the strong radical scavenging activity of HX fraction from C. affinis is a promising natural antioxidant for healthcare products.

  • To determination of antioxidant substance homoorientin, from Phyllostachys bambusoides leaves, the ultrasonic extraction and HPLC on-line $ABTS^+$ screening method were empolyed. Also, the various experimental variables such as the frequency and time of ultrasonic system were investigated and homoorientin was extracted efficiently at the low frequency 35 kHz and the extraction time 60 min. The values were positive peak 1574.71 (relative area, 23.67%) and negative peak 6924.34 (relative area, 1.23%), respectively. This HPLC on-line $ABTS^+$ screening method was rapid and efficient to search for antioxidants from natural products. These results will provide a database for investigating the constituents of natural products and the resources of pharmaceutical and cosmetic products.

Rapid Identification of Radical Scavenging Compounds in Blueberry Extract by HPLC Coupled to an On-line ABTS Based Assay and HPLC-ESI/MS

  • This study employed high performance liquid chromatography (HPLC) coupled to an on-line $ABTS^+$ radical scavenging detection (RSD) system along with HPLC-electro spin impact/mass spectrometry (ESI/MS), to rapidly determine and identify antioxidant compounds occurring in blueberry extract. The extract was separated by HPLC, and then the radical scavenging activities of the separated compounds were evaluated by the on-line coupled $ABTS^+$-RSD system. The negative peaks of the $ABTS^+$-RSD system, which indicates the presence of antioxidant activity, were monitored by measuring the decrease in absorbance at 734 nm. The active components in the blueberry extract were identified by HPLC-ESI/MS using their MS spectra and retention times. According to the data acquired from the on-line HPLC-$ABTS^+$-based assay and HPLC-ESI/MS systems, the antioxidant compounds detected in the blueberry extract were identified as chlorogenic acid and 11 anthocyanins.

  • In this study, we have attempted to determine the optimum concentration of inducers responsible for efficient laccase production by the white-rot fungus, Trametes sp. Variations in laccase activity were investigated with changing concentrations of 2,5-xylidine, syringaldazine, ABTS, and guaiacol. Enhancement of peak laccase activity was achieved via the combination of 2,5-xylidine with ABTS, syringaldazine, or guaiacol, resulting in increases of up to 359, 313, and 340%, respectively, as compared to control values. Among the tested inducers, the addition of 0.1mM of ABTS coupled with 1.0mM of 2,5-xylidine in the medium after 24 h of cultivation proved optimal with regard to laccase enzyme production.

Isolation of Prenylated Isoflavonoids from Cudrania tricuspidata Fruits that Inhibit A2E Photooxidation

  • High-performance liquid chromatography coupled to an online $ABTS^+$-based assay (online HPLC-$ABTS^+$) system was used to determine the principal antioxidants in Cudrania tricuspidata fruits. Six prenylated isoflavonoids (1 - 6) were isolated from C. tricuspidata fruits according to the online HPLC-$ABTS^+$ system. The structures of isolated compounds, alpiniumisoflavone (1), 6,8-diprenylorobol (2), 6,8-diprenylgenistein (3), pomiferin (4), 4'-methylalpiniumisoflavone (5), and osajin (6) were identified by their retention time, UV spectra, ESI-MS, and NMR data. Among these compounds, 6,8-diprenylorobol (2) and pomiferin (4) reduced A2E photooxidation in a dose dependent manner.

  • Eight hydroxylated stilbene derivatives including resveratrol, desoxyrhapontigenin and piceatannol as potential radical scavenger and tyrosinase inhibitor are synthesized using optimized Wittig-Horner reaction for excellent trans-selectivity in good yields. Antioxidant activity was tested against ABTS radical and tyrosinase inhibitory activity was performed with L-tyrosine as the substrate based on previous procedure with some modification. In general, catecholic stilbenes showed stronger activity against ABTS radical and resorcinolic moiety showed stronger tyrosinase inhibitory activity. Synthetic piceatannol which containing both catecholic and resorcinolic moieties showed the strongest activity in both as ABTS radical scavenger and tyrosinase inhibitor with $IC_{50}$ values of 4.1 and $8.6\;{\mu}M$, respectively.

  • Acer tegmentosum was a traditional Korean herbal medicine showing various pharmacological activities. In this work, A. tegmentosum were extracted with boiling water and then successively partitioned with dichloromethane, ethyl acetate, n-butyl alcohol (n-BuOH), and water. Salidoside, the target compound, was purified in n-BuOH phase using a chromatography method. For the analysis of salidoside, TLC and LC-MS were used as well as on-line screening $HPLC-ABTS^+$ assay with three different wavelength of 254, 280, and 320 nm. Validation of Analytical Methods for Plasma Total Antioxidant Capacity by Comparing with Urinary 8-Isoprostane Level

  • Although several analytical methods for measuring total antioxidant capacity (TAC) have been applied to biological samples, there were often dissimilar results due to the different principles of methods applied. Thus, this study aimed to validate four conventional analytical methods for measuring plasma TAC, including the ABTS assay, DPPH assay, FRAP assay, and ORAC assay, by comparing with urinary 8-isoprostane concentration. In addition, TAC results were compared with antioxidant enzyme activities including superoxide dismutase (SOD) and glutathione peroxidase in erythrocyte, and catalase in plasma. Plasma TAC measure by ABTS assay was strongly correlated with the result by FRAP assay. Plasma TAC by FRAP and ORAC assays were negatively correlated with erythrocyte SOD activity. The agreement among the four TAC assay methods and 8-isoprostane was determined using 95% prediction limits of linear regression, expressed as the mean of 8-isoprostane ${\pm}95%$ prediction limits. The ABTS method better agreed with 8-isoprostane than the other methods, demonstrating narrow prediction of limits. Furthermore, only plasma TAC determined by the ABTS assay was inversely correlated with urinary 8-isoprostane (r = -0.35, p < 0.05). In summary, the ABTS assay would be an appropriate method to measure overall plasma antioxidant capacity and predict the body's antioxidant status.
