A Novel Prosapogenin from the Methanolyzate of Melandrium Crude Saponins

  • Two compounds were isolated from the methanolyzate of the butanol-soluble fraction obtained from the whole plants of Melandrium firmum (Caryophyllaceae) and identified as $3-{\beta}-D-glucuronopyranosylmelandrigenin$ methyl ester and $2{\beta},\;21{\beta}-dihydroxy-16,\;23-dioxo-28-norolean-13(18)-ene$.

  • Two new diterpenoids, orientalin A (1), and B (2), have been isolated together with six known compounds, kirenol (3), $ent-16{\beta},17-dihydroxykauran-19-oic$ acid (4), $ent-16{\beta},17-dihydroxykauran-19-oic$ $acid-16{\beta},l7-acetonide$ (5), $3,7-dimethylquercetin$ (6), ${\beta}-sitosterol$ (7), and daucosterol (8) from the ethanol extract of Siegesbeckia orientalis (Compositae). Their chemical structures have been elucidated as $ent-15-acetoxy-2{\alpha},16,19-trihydroxypimar-8(14)-ene$ (1), $ent-16-acetoxy-2{\alpha},15,19-trihydroxypimar-8(14)-ene$ (2), respectively, on the basis of chemical and spectral evidence.

  • Approximations for the null distribution of a test statistic arising in multivariate analysis to test homogeneity of variances and a mixture of two beta distributions by making use of a product of beta baseline density function and a polynomial adjustment, so called beta-polynomial density approximant, are discussed. Explicit representations of density and distribution approximants of interest in each case can easily be obtained. Beta-polynomial density approximants produce good approximation over the entire range of the test statistic and also accommodate even the bimodal distribution using an artificial example of a mixture of two beta distributions.

  • Activity-guided fractionation of the n-hexane and ${CHCl_3}-soluble$ fractions of Sindora sumatrana using a bioassay based on the inhibition of lipopolysaccharide (LPS)-induced nitric oxide (NO) production by inducible nitric oxide synthase (iNOS) in murine macrophage RAW 264.7 cells led to the isolation of the known compound, $(+)-7{\beta}-acetoxy-15,16-epoxy-3$, 13(16), 14-clero-datriene-18-oic acid (2) as an active constituent. In addition, a new trans-clerodane diterpenoid, (+)-2-oxokolavenic acid (1), together with six known compounds, (+)-3, 13-clerodadiene-16,15-olide-18-oic acid (3), $(+)-7{\beta}-acetoxy-3$,13-clerodadiene-16,15-olide-18-oic acid (4), $(+)-7{\beta}-acetoxy-16-hydroxy-3$,13-clerodadiene-16, 15-olide-18-oic acid (5), ${\beta}-caryophyllene$ oxide (6), $clovane-2{\beta},9{\beta}-diol (7),{\;}and{\;}caryolane-1,9{\beta}-diol$ (8) were isolated and found to be inactive. The structure of compound 1 was determined using physical and spectroscopic methods such as 1D and 2D-NMR experiments. The known compounds 2-8 were identified by the spectroscopic data and by comparison with the published values. Of eight isolates (1-8), only compound 2 exhibited an iNOS inhibitory activity with $IC_{50}$/ value of $51.6{\;}\mu\textrm{m}M$.

Effect of Kenpaullone, a Specific Inhibitor of GSK3${\beta}$, on Melanin Synthesis in B16 Melanoma and Human Melanocytes (GSK3${\beta}$의 선택적 저해제인 Kenpaullone의 B16 멜라노마 및 인간 멜라노사이트에서의 영향)

  • Effects of Kenpaullone, a specific inhibitor of GSK3${\beta}$, on melanin synthesis in B16 melanoma cells and human melanocytes were investigated. Kenpaullone showed a melanogenesis stimulation activity in a concentrationdependent manner in murine B16 melanoma cells and human melanocytes without any significant effects on cell proliferation. Tyrosinase activity was increased 48 h after treatment of B16 cells with Kenpaullone. The protein expression level of tyrosinase was dose-dependently enhanced after the treatment with Kenpaullone. At the same time, the expression level of tyrosinase mRNA was also increased after addition of Kenpaullone. The stimulatory effect of Kenpaullone mainly resulted from increased expression of tyrosinase. These findings suggest that the application of GSK3${\beta}$ inhibitors may be a potential therapeutic agent for the treatment of hypopigmentation disorder.

Immunostimulatory Effects of ${\beta}$-glucan Purified from Paenibacillus polymyxa JB115 on Mouse Splenocytes

  • We investigated the effects of ${\beta}$-glucan purified from Paenibacillus polymyxa JB115 on the viability and proliferation of splenocytes. Splenocytes play a critical role in host immunity. MTT assays and trypan blue exclusion tests revealed that ${\beta}$-glucan significantly promoted the viability and proliferation of splenocytes over a range of concentrations. However, there was no specific subset change. ${\beta}$-glucan protected splenocytes from cytokine withdrawal-induced spontaneous cell death. For further mechanistic studies, ELISA assay revealed that ${\beta}$-glucan enhanced the expression of anti-apoptotic molecules and interleukin 7 (IL-7), a cytokine critical for lymphocyte survival. We also investigated the IL-2 dependency of ${\beta}$-glucan-treated splenocytes to determine if treated cells could still undergo clonal expansion. In flow cytometric analysis, ${\beta}$-glucan induced increased levels of the activation marker CD25 on the surface of splenocytes and ${\beta}$-glucan-treated splenocytes showed higher proliferation rates in response to IL-2 treatment. This study demonstrates that ${\beta}$-glucan can enhance the survival of splenocytes and provides valuable information to broaden the use of ${\beta}$-glucan in research fields.

  • We consider distribution, reliability and moment of ratio in two independent beta random variables X and Y, and reliability and $K^{th}$ moment of ratio are represented by a mathematical generalized hypergeometric function. We introduce an approximate maximum likelihood estimate(AML) of reliability and right-tail probability in the beta distribution.

Antimetastatic Effect of Proteoglycan Isolated from the Mycelium of Ganoderma lucidum IY009 in vitro and in vivo (영지버섯 균사체 (Ganoderma lucidum IY009)로부터 추출한 단백다당체의 전이암 억제 효과)

  • $\beta$-Immunan, a proteoglycan, was isolated from the mycelium of Canoderma lucidum which belongs to a medicinal mushroom. The effects of $\beta$-Immunan on cell-cell and cell-matrix interactions mediated by carbohydrate-recognition and the mechanism responsible for the inhibition of experimental metastasis of Bl6-F10 and B16/BL6 murine melanoma were studied. The results showed that $\beta$-Immunan inhibited Bl6-Fl melanoma cell's adhesion to laminin and asialofetuin-induced homotypic aggregation and reduced invasion against Bl6-F10 murine melanoma cells through matrigel in vivo assay. When $\beta$-Immunan was intraperitoneally administrated to C57B/6 mice bearing B16/BL6 murine melanoma cells, it was decreased the number of pulmonary metastatic colony by the dose dependent manner ranging from 20 to 100 mg/kg/day. The results indirectly indicate that clinical treatment with $\beta$-Immunan might be expected to exhibit anti-metastatic effect. In the pulmonary metastasis, the number of pulmonary metastatic colony of melanoma when $\beta$-Immunan was intraperitoneally administrated to C57BL/6 mice bearing B16/BL6 murine melanoma cells by intravenous injection were decreased by the dose dependent manner ranging from 20 to 100 mg/kg/day.

The Correlation between TGF-beta 1 Blood Levels and the Formation of Bullae in Patients with Spontaneous Pneumothorax (자연 기흉 환자의 혈액 내 TGF-beta 1 Ligand 양과 폐 기포 형성과의 연관관계에 대한 연구)

  • Background: The overexpression of transforming growth factor-beta 1 receptor II (TGF-${\beta}1$RII) and transforming growth factor-beta 1 (TGF-${\beta}1$) ligand may be involved in the formation of a bulla. In this study, we tested if serum TGF-${\beta}1$ ligand levels correlated with the expression level of TGF-${\beta}1$RII and TGF-${\beta}1$ in bullous tissues from patients with spontaneous pneumothorax. Material and Method: Bullous lung tissues and blood samples were obtained from 19 patients with spontaneous pneumothorax, 18 males and 1 female, aged 17 to 35 years old. The bullous tissues were obtained by video-assisted thoracic surgery (VATS), fixed in formalin, embedded in paraffin, and cut into $5{\sim}6{\mu}m$ thick slices. Sections were immunohistochemically stained with primary antibodies against TGF-${\beta}1$ or TGF-${\beta}1$RII, and serum levels of TGF-${\beta}1$ in patients and normal controls was measured by enzyme-linked immunosorbent assay (ELISA). Result: Of the 19 patients, 16 were TGF-${\beta}1$ positive and 10 were TGF-${\beta}1$RII positive. Among the 16 TGF-${\beta}1$ positives, 9 were also TGF-${\beta}1RII$ positive. As seen previously, strong immunohistochemical staining of TGF-${\beta}1$RII and TGF-${\beta}$ was detected in the boundary region between the bullous and normal lung tissues. Average TGF-${\beta}1$ blood levels of both TGF-${\beta}1$ and TGF-${\beta}1$RII positive patients was $38.36{\pm}16.2ng/mL$, and that of five controls was $54.06{\pm}15ng/mL$. Conclusion: These results suggest that overexpression of TGF-${\beta}1$ and TGF-${\beta}1$RII expression may be involved in the formation of bullae. TGF-${\beta}1$ blood levels in patients with primary spontaneous pneumothorax is lower than normal people, suggesting that the high level of local TGF-${\beta}1$ expression in the bullous tissue region, but not in the whole blood, may contribute more in the formation of bullae.

Molecular Cloning, Overexpression, and Enzymatic Characterization of Glycosyl Hydrolase Family 16 ${\beta}$-Agarase from Marine Bacterium Saccharophagus sp. AG21 in Escherichia coli

  • An agar-degrading bacterium was isolated from red seaweed (Gelidium amansii) on a natural seawater agar plate, and identified as Saccharophagus sp. AG21. The ${\beta}$-agarase gene from Saccharophagus sp. AG21 (agy1) was screened by long and accurate (LA)-PCR. The predicted sequence has a 1,908 bp open reading frame encoding 636 amino acids (aa), and includes a glycosyl hydrolase family 16 (GH16) ${\beta}$-agarase module and two carbohydrate binding modules of family 6 (CBM6). The deduced aa sequence showed 93.7% and 84.9% similarity to ${\beta}$-agarase of Saccharophagus degradans and Microbulbifer agarilyticus, respectively. The mature agy1 was cloned and overexpressed as a His-tagged recombinant ${\beta}$-agarase (rAgy1) in Escherichia coli, and had a predicted molecular mass of 69 kDa and an isoelectric point of 4.5. rAgy1 showed optimum activity at $55^{\circ}C$ and pH 7.6, and had a specific activity of 85 U/mg. The rAgy1 activity was enhanced by $FeSO_4$ (40%), KCl (34%), and NaCl (34%), compared with the control. The newly identified rAgy1 is a ${\beta}$-agarase, which acts to degrade agarose to neoagarotetraose (NA4) and neoagarohexaose (NA6) and may be useful for applications in the cosmetics, food, bioethanol, and reagent industries.
