Radiolabeling of nanoparticle for enhanced molecular imaging

  • The combination of nanoparticle with radioisotope could give the in vivo information with high sensitivity and specificity. However, radioisotope labeling of nanoparticle is very difficult and radioisotopes have different physicochemical properties, so the radioisotope selection of nanoparticle should be carefully considered. $^{18}F$ was first option to be considered for labeling of nanoparticle. For the labeling of $^{18}F$ with nanoparticle, Prosthetic group is widely used. Iodine, another radioactive halogen, is often used. Since radioiodine isotopes are various, they can be used for different imaging technique or therapy in the same labeling procedures. $^{99m}Tc$ can easily be obtained as pertechnatate ($^{99m}{TcO_4}^-$) by commercial generator. Ionic $^{68}Ga$ (III) in dilute HCl solution is also obtained by generator system, but $^{68}Ga$ can be substituted for $^{67}Ga$ because of the short half-life (67.8 min). $^{64}Cu$ emits not only positron but also ${\beta}-particle$. Therefore $^{64}Cu$ can be used for imaging and therapy at the same time. These radioactive metals can be labeled with nanoparticle using the bifunctional chelator. $^{89}Zr$ has longer half-life (78.4 h) and is used for the longer imaging time. Unlike different metals, $^{89}Zr$ should use the other chelate such as DFO, 3,4,3-(LI-1,2-HOPO) or DFOB.

Factors Influencing for the Compliance of Preventing Bloodborne Infection for Operating Room Nurses (수술실 간호사의 혈행성 감염 예방지침 수행 영향요인)

  • Purpose: The purpose of this study was to examine factors influencing for the compliance of guidelines of preventing bloodborne infection for operating room nurses. Method: Data was collected through questionnaires from March 16 to April 12, 2005. The subjects of this study were consisted of 208 operating room nurses, who had been working at 4 university hospitals and 10 general hospitals in Busan. The Instrument for the compliance of guidelines for preventing bloodborne infection was 24-item questionnaire. It was revised based on the Standard Precaution of CDC and Bloodborne Pathogen Standards of OSHA. In addition, to find out the knowledge level of AIDS, hepatitis B and hepatitis C, 15-item questionnaire was used, which had been made by researcher based on Kim(1999) et, al. Results: The affecting factors of compliance for preventing bloodborne infection were work skill(${\beta}=.27$), operating room education on infection prevention(${\beta}=.25$), knowledge on infection(${\beta}=.19$), readiness of using personal protective equipment(${\beta}=.18$), and posting a notice of infected surgery patients(${\beta}=.16$). Theses 5 factors account for 28.5% on the compliance of guidelines for preventing bloodborne infection for operating room nurses. Conclusion: According to the results, to increase the compliance level of operating room nurses, personal efforts should be made to improve work skills and infection knowledge. In addition to organizational efforts on practical education and management of environment were required.

Scaling pain and related factors in adults (성인의 스케일링 통증과 관련요인)

  • Objectives: The purpose of this study was to review depression and anxiety associated with pain during scaling procedures, and to establish measures for reducing scaling pain encompassing not only physical factors but also psychological aspects of patients. This study also attempts to reduce anxiety through proper patient education prior to scaling procedures. Methods: In Seoul, and Gyeonggi area from July 26, 2017 to August 19, 2017, there were 327 copies of questionnaire data collected, excluding 23 questionnaires with insufficient information such as missing entries. The following inductions were made based on data collected. Results: There are positive correlations between pain and depression, dental anxiety, trait anxiety, and state anxiety. Especially, stronger correlation is present between pain and dental anxiety. Depression (${\beta}=0.439$, p<0.001) is the most influential factor associated with pain. Next is dental anxiety (${\beta}=0.292$, p<0.001). Higher the depression and dental anxiety tend to increase pain over scaling procedure. This model is with adjusted explanatory power of 28.2%. Conclusions: The result demonstrates that there is a correlation between scaling pain and depression, dental anxiety, trait anxiety, and state anxiety. Especially, depression and dental anxiety were prominent factors in affecting scaling pain. Therefore, considering aforementioned-findings, a dental hygienist's positive attitude may help in reducing the pain of the patient during scaling procedures, by affecting the psychological state of the patient and reduce the anxiety through proper education prior to scaling procedures.

Central Neural Pathways Projecting to the Acupoints B62 and K6 Using the Neural Tracer (신경추적자를 이용한 신맥과 조해에서 투사되는 신경원의 표지부위에 대한 연구)

  • The purpose of this morphological studies was to investigate the central neural pathway projecting to the acupoints $B_{62}$ and $K_6$ using the neuroanatomical method following injection of transsynaptic neurotropic virus, pseudorabies virus(PRV-Ba and PRV-Ga) into the $B_{62}$ and $K_6$. After survival times of 96 hours following injection into the twenty rats with PRV-Ba(Bartha strain) and PRV-Ga(Bartha strain, ${\beta}$-galacidodase insertion). They were perfused, and their spinal cord and brain were frozen sectioned($30{\mu}m$). These sections were stained by X-gal histochemical and PRV immunohistochemical staining method, and observed with light microscope. The results were as follows : 1. In spinal cord, overlaped PRV-Ba and PRV-Ga labeled neurons projecting to the $B_{62}$ and $K_6$ were founded in thoracic, lumbar and sacral spinal segments. In thoracic spinal segments, Densely labeled areas were founded in lamina IV, V, VII(intermediolateral nucleus) and X areas. In lumbar segemnts, labeled areas were founded in lamina II, IV, V and X areas. In sacral spinal segments, labeled areas were founded in lamina IV, V and VI areas. 2. In brain, overlaped PRV-Ba and PRV-Ga labeled neurons projecting to the $B_{62}$ and $K_6$ were founded in the $A_1$ noradrenalin cells/$C_1$ adrenalin cells/caudoventrolateral reticular nucleus, rostroventrolateral reticular nuclens, nucleus tractus solitarius, area postrema, raphe obscurus nucleus, raphe paltidus nucleus, raphe magnus nucleus, lateral paragigantoceltular nucleus, lateral rcticular nucleus, gigantocellular nucleus, locus coeruleus, subcoeruleus nucleus, motor trigeminal nucleus, Kolliker-Fuse nucleus, $A_5$ cell group, central gray matter, oculomotor nerve, paraventricular hypothalamic nucleus, median eminence, amygdaloid nucleus, frontal cortex, forelimb area, hindlimb area, 1, 2 areas of parietal cortex and granular and agranular cortex. This results were suggest that overlaped PRV-Ba and PRV-Ga labeled areas projecting to the $B_{62}$ and $K_6$ may be related to the emotional relay pathway in the central autonomic center.

Effects of Sound Stress on Physiological Processes of the American Leafminer, Liriomyza trifolii, and Proteomic Analysis (스트레스 음파 처리에 따른 아메리카잎굴파리(Liriomyza trifolii)의 생리 변화와 프로테오믹 분석)

  • This study investigated the adverse effects of sound treatment on physiological processes of the American leafminer, Liriomyza trifolii, during several developmental stages. Larval feeding activity was analyzed by measuring feeding tunnel length. It was significantly suppressed by sound treatment (5,000 Hz, 95 dB). Sound treatment delayed the pupal period at 315 - 5,000 Hz and prevented adult emergence at 1,000 - 5,000 Hz. Female oviposition was also inhibited by the stress sound treatments. However, phototactic adult movement was not affected by sound treatment. Pupae treated with 5,000 Hz showed marked changes in protein patterns analyzed by two dimensional electrophoresis. MALDI-TOF analysis of specific protein spots indicated that trafficking protein particle complex I, triosephosphate isomerase, hypothetical protein TcasGA2_TC013388, polycystin-2, paraneoplastic neuronal antigen MA1, and tropomyosin I (isoform M) were predicted in the control insects and disappeared in the insects treated with sound. By contrast, DOCK9, cytoskeletal keratin II, and F0F1-ATP synthase beta subunit were predicted only in the sound-treated insects. Furthermore, stress sound significantly increased the susceptibility of L. trifolii to insecticides. These results suggest that physiological processes of L. trifolii are altered by sound stress, which may be exploited to develop a novel physical control tactic against L. trifolii.

A Feasibility Study of a SiPM Based Intraoperative Beta Imaging/Gamma Probe using the Depth of Interaction Measurement (실리콘 광증폭기와 반응깊이 측정방법을 이용한 수술용 베타 영상/감마 프로브 가능성 연구)

  • Radiopharmaceutical agents for positron emission tomography (PET), such as $^{18}F$-FDG and $^{68}Ga$, have been used not only for whole-body PET imaging but also for intraoperative radionuclide-guided surgery due to their quantitative and sensitive imaging characteristics. Current intraoperative probes detect gamma or beta particles, but not both of them. Gamma probes have low sensitivities since a collimator has to be used to reduce backgrounds. Positron probes have a high tumor-to-background ratio, but they have a 1-2 mm depth limitation from the body surface. Most of current intraoperative probes produce only audible sounds proportional to count rates without providing tumor images. This research aims to detect both positrons and annihilation photons from $^{18}F$ using plastic scintillators and a GAGG scintillation crystal attached to silicon photomultiplier (SiPM). The depth-of-interaction (DOI) along the plastic scintillator can be used to obtain the 2-D images of tumors near the body surface. The front and rear part of the intraoperative probe consists of $4{\times}1$ plastic scintillators ($2.9{\times}2.0{\times}12.0mm^3$) for positron detection and a Ce:GAGG scintillation crystal ($12.0{\times}12.0{\times}9.0mm^3$) for annihilation photon detection, respectively. The DOI resolution of $4.4{\pm}1.6mm$ along the plastic scintillator was obtained by using the 3M enhanced specular reflector (ESR) with rectangular holes between the plastic scintillators, which showed the feasibility of a 2-D image pixel size of $2.9{\times}4.4mm^2$ (X-direction ${\times}$ Y-direction).

The Effects of Emotion and EEG of People with Mental Illness by Vocational Rehabilitation Program Based on Horticultural Therapy (원예치료 기반 직업재활 프로그램이 정신장애인의 정서 및 뇌파에 미치는 영향)

  • The purpose of this study was to investigate the effects of emotion and EEG of people with mental illness by vocational rehabilitation program based on horticultural therapy. The subjects of the study were three people with mental illness who were training in a vocational rehabilitation team at a health center located in G city, Gyeongsangbuk-do. The result of this study, positive affect increased and negative affect decreased. The result of EEG inspection was increased 17.27% on pre test to 21.39% on post test for Theta wave and 39.66% on pre test to 49.02% on post test for Alpha wave in case of subject A. Also Beta wave was decreased 27.53% on pre test to 17.54% on post test. Subject B increased Alpha wave 29.70% on pre to 31.82% on post. Subject C increased Alpha wave 26.30% on pre to 49.42% on post. Beta wave decreased 29.85% on pre to 16.13% on post. Theta wave, when creatively idea or thought occurs, alpha wave when safety and focusing was increased and beta wave when stress, tension and anxiety was decreased. As a result of profits analysis, subject A spent cost a total of 101,700 won for material and achieve profits of 203,400 won by setting a sales price. Therefore, gain the 101,700 won of net revenue. Subject B spent cost a total of 81,000 won for material and achieve profits of 162,000 won by setting a sales price. Therefore, gain the 81,000 won of net revenue. Subject C spent cost a total of 56,200 won for material and achieve profits of 112,400 won by setting a sales price. Therefore, gain the 56,200 won of net revenue.

Effects of the Molecular Weight and Type of Chitosans on Shelf-life of Makkulli (막걸리의 저장성에 미치는 분자량별 및 형태별 키토산의 영향)

  • Effects of the molecular weight and type of chitosans on shelf-life of Makkulli were evaluated during 18 days of storage at $25^{\circ}C$. Two types of chitosans were studied: ${\alpha}$-chitosans with 11 different molecular weights (water-soluble, Mw = 1, 8, 22, 43, 67 and 616 kDa; acid-soluble, Mw = 282, 440, 746, 1,110 and 2,025 kDa) and ${\beta}$-chitosan (acid-soluble, Mw = 577 kDa). Acid-soluble chitosans were applied as a form of chitosan-ascorbate. All chitosans were added to Makkulli at 0.002% concentration, the optimum concentration established in a preliminary test. Among 12 chitosans, the ${\alpha}$-chitosans with 22 and 440 kDa exhibited stronger antimicrobial effects than did other ${\alpha}$- and ${\beta}$-chitosans. The results for pH, acidity, alcohol concentration, viable cell counts, and sensory evaluation suggested that addition of ${\alpha}$-chitosans with 22 and 440 kDa increased the shelf-life of Makkulli by almost 1 week at $25^{\circ}C$ compared with that of control (without chitosan) and other chitosan-added groups. Extension of Makkulli shelf-life by 1 week is fairly significant in view of the magnitude of the total amount of Makkulli produced in Korea.

Inhibitory Effects of Water Extracts of Eucommiae Cortex and Psoraleae Semen Alone and in Combination on Osteoclast Differentiation and Bone

  • Objectives : The purpose of this study was to evaluate the effects of water extracts of Eucommiae cortex (EC), Psoraleae semen (PS), and their combination on receptor activator of nuclear factor-kappa-B ligand (RANKL)-induced osteoclast differentiation. Methods : We assayed the protein expression levels of nuclear factor of activated T-cells, cytoplasmic 1 (NFATc1), c-Fos, mitogen-activated protein kinases (MAPKs), and ${\beta}-actin$ in cell lysates using western blotting. Similarly, mRNA expression levels of NFATc1, c-Fos, tartrateresistant acid phosphate (TRAP), and glyceraldehyde-3-phosphate dehydrogenase, spermatogeni (GAPDHS) from bone marrow macrophages (BMMs) were analyzed using reverse transcription-polymerase chain reaction (RT-PCR). Furthermore, we determined the anti-osteoporotic effects of the water extracts of EC, PS, and their combination in a lipopolysaccharide (LPS)-induced bone-loss mouse model. Results : The in vitro data revealed showed that the combination of EC and PS extract showed a more remarkable inhibition of osteoclast differentiation than each herb did alone. The combination downregulated the induction of c-Fos, NFATc1, and TRAP by suppressing the phosphorylation of p38 and c-Jun N-terminal kinases (JNKs) and inhibiting nuclear factor kappa-light-chain-enhancer of activated B cells ($NF-{\kappa}B$). Lastly, the in vivo data showed that PS reduced the LPS-induced bone erosion. Conclusion : The result of this study suggests that EC and PS could be potential therapeutic agents for bone loss diseases such as osteoporosis.
