Regulation of Nek6 Functions by Its SUMOylation on the $K^{252}$ Residue

  • Nek6 belongs to NIMA1 (never in mitosis, gene A) related kinase, which was originally identified in Aspergillus nidulans as a serine/threonine kinase critical for cell cycle progression. We noticed that the putative SUMOylation site is localized on the $K^{252}$ residue in $^{251}FKsD^{254}$ of Nek6, based on the consensus sequence ${\Phi}KxE$; where ${\Phi}$ represents L, I, V or F and x is any amino acid. We observed that the Nek6 SUMO mutant (K252R) has decreased protein kinase activity, nuclear speckle localization and protein stability, compared with that of the Nek6 wild type. However, the Nek6 SUMO mutant increased the cell survival rate of COS-1 cells as determined by FACS analysis. Therefore, our data suggest that SUMOylation on the $K^{252}$ residue of Nek6 is required for its normal functions, such as proper nuclear localization, kinase activity and protein stability, to control cell cycle.

Study on Creep Life Prediction by Initial Strain Method for Friction Welded Joints of Heat Resisting Steels (내열강 마찰용접재의 ISM에 의한 크리프 수명예측에 관한 연구)

  • In this paper, the real-time prediction of high temperature creep life was carried out for the friction welded joints of dissimilar heat resisting steels (SUH3-SUH35). various life prediction method such as LMP (Larson_miller Parameter) and ISM (initial strain method) were applied. The creep behaviors of those steels and the welds under static load were examined by ISM combined with LMP at 500, 600 and $700^{\circ}C$, and the relationship between these two methods was investigated. A real-time creep lie (tr, hr) prediction equation by initial strain (${\varepsilon}_0$, %) under any creep stress ($\sigma$, MPa) at any high temperature (T, K) was developed as follows: $t_r={\alpha}{\varepsilon}_0^{\beta}{\sigma}^{-1}$ where, ${\phi}=16: {\alpha}=10^{51.412-0.104T+5.375{\times}10^5T^2}$, $ {\beta}=-83.989+0.180T-9.957{\times}10^{-5}T^2,{\phi}=20:$ ${\alpha}=10^{69.910-0.146T+7.744{\times}10^{-5}T^2$, ${\beta}=-51.442+0.105T-5.595{\times}10^{-5}T^2$ for SUH3-SUH35 friction weld of =16mm and 20mm, respectively.

  • This paper deals with the design and development of an Injection-Locking Coupled Oscillators(ILCO), which functions like phase-shifter in the Active Intergrated Phased Array Antenna(AIPAA). This linear array 2-element ILCO consists of two Injection Locking Hair-pin Resonator Oscillators(ILHRO) and an unilateral amplifier. The first and second elements of the ILCO have same frequency tuning range but locking bandwidths of 11.5MHz and 14MHz respectively. A phase shift of .DELTA..PHI.=158.4.deg.(-78.0.deg. to 80.4.deg.) could be obtained inthe second element of ILCO when the first elementof the ILCO was in the reference locking mode(.DELTA..PHI.=0.deg.). When the ILCO is applied to the AIPAA, the predicted beam scanning angle value will be 38.4.deg.. Each ILCO gives good frequency stability and lower AM, FM, and PM noise charactheristics in the mutual coupling lockingmode. The ILCO can not only play a part as the phase shifter for the AIPAA but it can also be usedas the power combining device in the mm-wave frequency range and as a part of a T/R MMIC module.

A Study on Attitude angle control of Quadruped Walking Robot (4족 보행로봇의 자세각 제어에 관한 연구)

  • In this paper, we used the quadruped walking robot Titan-VIII in order to carry out this simulation of sway compensation trajectory. The attitude angle ${\phi}_r$ and ${\phi}_p$ is obtained from 3-D motion sensor that is attached at the center of robot body and the attitude control carried out at every 10[ms] for stable gait of robot. Duty factor, that is fixed at 0.5. When we change period T into 1.5, 2.0, 3.0[sec] each and moving distance per period is changed into 0.2, 0.3(m), we sim띠ate several walking experiment of robot. and then we analyze the experiment results if there are any difference between the imaginary ZMP and actual ZMP of robot and the stable gait of robot is realized.

Comparative study of various buffer layers on IBAD- MgO template (IBAD-MgO 기판 위 다양한 완충층들의 비교 연구)

  • On highly-textured IBAD-MgO templates, we have tried to find proper buffer layers among various candidate materials, including $LaMnO_3$ (LMO), $La_2Zr_2O_7$ (LAO), $LaAlO_3$ (LAO), $LaGaO_3$ (LGO), $NdGaO_3$ (NGO), and $BaZrO_3$ (BZO). All buffer layers were deposited on the IBAD-MgO templates by KrF pulsed laser deposition(PLD). LAO layer showed an armorphous phase. LZO, LGO, and NGO layers showed polycrystalline growth. Only LMO and BZO layers exhibited c-axis oriented biaxially textured films. Optimally processed LMO buffer layer at deposition temperature of $750^{\circ}C$ and $PO_2$ of 100mTorr exhibited ${\triangle}{\phi}$ value of ${\sim}-5.2^{\circ}$ and RMS roughness of 5.6nm. Interestingly, BZO buffer layers with ${\triangle}{\phi}$ values of ${\sim}-6^{\circ}$ could be routinely produced over a wide PLD processing condition.

  • In this work we obtain conditions for nonspecial line bundles on general ${\kappa}$-gonal curves failing to be normally generated. Let L be a nonspecial very ample line bundle on a general ${\kappa}$-gonal curve X with ${\kappa}{\geq}4$ and $deg\mathcal{L}{\geq}{\frac{3}{2}}g+{\frac{g-2}{{\kappa}}}+1$. If L fails to be normally generated, then L is isomorphic to $\mathcal{K}_X-(ng^1_{\kappa}+B)+R$ for some $n{\geq}1$, B and R satisfying (1) $h^0(R)=h^0(B)=1$, (2) $n+3{\leq}degR{\leq}2n+2$, (3) $deg(R{\cap}F){\leq}1$ for any $F{\in}g^1_k $. Its converse also holds under some additional restrictions. As a corollary, a very ample line bundle $\mathcal{L}{\simeq}\mathcal{K}_X-g^0_d+{\xi}^0_e$ is normally generated if $g^0_d{\in}X^{(d)}$ and ${\xi}^0_e{\in}X^{(e)}$ satisfy $d{\leq}{\frac{g}{2}}-{\frac{g-2}{\kappa}}-3$, supp$(g^0_d{\cap}{\xi}^0_e)={\phi}$ and deg$(g^0_d{\cap}F){\leq}{\kappa}-2$ for any $F{\in}g^1_k$.

Assessment of Heavy Metal Effects on the Freshwater Microalga, Chlorella vulgaris, by Chlorophyll Fluorescence Analysis (엽록소형광분석을 이용한 담수산 클로렐라(Chlorella vulgaris)에 미치는 중금속의 영향 평가)

  • The response of the freshwater microalga, Chlorella vulgaris, to heavy metal stress was examined based on chlorophyll fluorescence analysis to assess the toxic effects of heavy metals in freshwater ecosystems. When toxic effects were analyzed using regular chlorophyll fluorescence analysis, photosystem II activity($F_v/F_m$) decreased significantly when exposed to $Cu^{2+}$ and $Hg^{2+}$ for 12 h, and decreased in the order of $Hg^{2+}>Cu^{2+}>Cd^{2+}>Ni^{2+}$ when exposed for 24h. The effective photochemical quantum yield(${\phi}{\prime}_{PSII}$), chlorophyll fluorescence decrease ratio($R_{Fd}$), minimal fluorescence yield($F_o$), and non-photochemical quenching(NPQ), but not photochemical quenching(qP), responded sensitively to $Hg^{2+}$, $Cu^{2+}$, and $Cd^{2+}$. These results suggest that $F_v/F_m$, as well as ${\phi}{\prime}_{PSII}$, $R_{Fd}$, $F_o$, and NPQ could be used to assess the effects of heavy metal ions in freshwater ecosystems. However, because many types of heavy metal ions and toxic compounds co-occur under natural conditions, it is difficult to assess heavy metal toxicity in freshwater ecosystems. When Chlorella was exposed to heavy metal ions for 12 or 24h, $F_v/F_m$ and maximal fluorescence yield($F_m$) changed in response to $Hg^{2+}$ and $Cu^{2+}$ based on image analysis. However, assessing quantitatively the toxic effects of several heavy metal ions is challenging.

Reproductive Biology and Population Dynamics of Luciogobius guttatus (Pisces: Gobiidae) in the Southwestern of Korea (서해 중부 해역에 출현하는 미끈망둑, Luciogobius guttatus (Gobiidae)의 생식생태와 개체군 동태)

  • Reproductive biology and population dynamics of Luciogobius guttatus were investigated. Higher gonadosomatic index (GSI) was observed from March~May. The fecundity ranged form 241 eggs at 3.1~4.0 cm (SL) to 716 eggs at 5.1~5.7 cm (SL). The relationship between length (SL) and fecundity (F) was $F=11.242SL^{2.464}$ ($R^2=0.65$) and fecundity increased with length. We estimated the SL at 50% group maturity as 2.6 cm. The spawning period was March~May. The von Bertalanffy growth function (VBGF) estimates were: $L_{\infty}=63.00mm$ TL; $K=0.85yr^{-1}$. The structure of recruitment pattern obtained by the FiSAT II program indicated one normally distributed group. The growth performance index (${\phi}$) was calculated with 3.53. Natural mortality rate (M) was $1.00yr^{-1}$.

The modulation of TRPV4 channel activity through its Ser 824 residue phosphorylation by SGK1

  • With the consensus sequence information of the serum glucocorticoid-induced protein kinase-1 (SGK1) phosphorylation site {R-X-R-X-X-(S/T)$\Phi$; where $\Phi$ is any hydrophobic amino acid}, we noticed that the transient receptor potential vanilloid 4 (TRPV4) cation channel, a member of the TRP vanilloid subfamily, harbors the putative SGK1 phosphorylation site (on its Ser 824). We have demonstrated that TRPV4 is an SGK1 authentic substrate protein, with the phosphorylation on the Ser 824 of TRPV4 by SGK1. Further, using TRPV4 mutants (S824A and S824D), we noted that the modification of the Ser 824 activates its $Ca^{2+}$ entry, and sensitizes the TRPV4 channel to 4-$\alpha$-phorbol 12,13-didecanoate (4-${\alpha}PDD$) or heat, simultaneously enhancing its active state. Additionally, we determined that the modification of the Ser 824 controls both its plasma membrane localization and its protein interactions with calmodulin. Thus, we have proposed herein that phosphorylation on the Ser 824 of TRPV4 is one of the control points for the regulation of its functions.
