Generation and Characterization of Monoclonal Antibodies against Human Interferon-lambda1

  • Background: Members belonging to the interferon-lambda (IFN-${\lambda}$) family exert protective action against viral infection; however, the mechanisms of their action have remained elusive. To study IFN-${\lambda}$ biology, such as endocytosis of IFN-${\lambda}$, we produced monoclonal antibodies (Abs) against human IFN-${\lambda}$ and examined their usefulness. Methods: We purified recombinant human IFN-${\lambda}$1 expressed in Escherichia coli by using affinity columns. Then, we generated hybridoma cells by fusing myeloma cells with splenocytes from IFN-${\lambda}$1-immunized mice. For evaluating the neutralizing activity of the monoclonal Abs against IFN-${\lambda}$1, we performed RT-PCR for the MxA transcript. In order to study the binding activity of IFN-${\lambda}$ and the monoclonal Ab complex on HepG2 cells, we labeled the monoclonal Ab with rhodamine and determined the fluorescence intensity. Results: Four hybridoma clones secreting Abs specific to IFN-${\lambda}$1 were generated and designated as HL1, HL2, HL3, and HL4. All the Abs reacted with IFN-${\lambda}$1 in the denatured form as well as in the native form. Abs produced by HL1, HL3, and HL4 did not neutralize the induction of the MxA gene by IFN-${\lambda}$1. We also demonstrated the binding of the HL1 monoclonal anbitody and IFN-${\lambda}$ complex on HepG2 cells. Conclusion: Monoclonal Abs against IFN-${\lambda}$1 were produced. These Abs can be used to study the cellular binding and internalization of IFN-${\lambda}$.

Electric Conductance of Dilute Solutions of Lithium, Sodium and Potassium Chloride in Isopropanol_Water Mixtures (이소프로판올-물 混合溶媒中의 리튬, 나트륨 및 칼륨 鹽化沕 溶液의 電導度)

  • The conductances of dilute solutions of LiCl, NaCl and KCl in a series of isopropanol-water mixtures were determined at $30^{\circ}C$. The values of equivalent conductance agreed well with Debye-Huckel-Onsager equation and the limiting equivalent conductance was greatly reduced as the isopropanol content of the solvent was increased in accord with predictions based on solvent viscosity and dielectric properties. Also, the limiting equivalent conductance increased in sequence ${\Lambda}_{0,LiCl} < {\Lambda}_{0,NaCl} < {\Lambda}_{0,KCl} in 0.0, 0.1 and 0.2 mole fraction isopropanol, but {\Lambda}_{0,LiCl} < {\Lambda}_{0,KCl} < {\Lambda}_{0,NaCl} in 0.3 mole fraction ispropanol. The maximum Walden product, {\Lambda}_{0{\eta}0}$ was found in 0.1 mole fraction isopropanol for all electrolytes.

The characteristic of diffraction efficiency depending on Ag doping on film of amorphous AsGeSeS (비정질 As-Ge-Se-S 박막에서의 Ag 도핑에 따른 회절효율 특성 연구)

  • This paper shows the characteristic of diffraction efficiency on film of amorphous AsGeSeS according to Ag doping. We compare amorhous AsGeSeS with the characteristics of AsGeSeS/Ag (10nm) and AsGeSeS/Ag (20m). We made film such as $\lambda$, $\lambda/2$, $\lambda/4$, $\lambda/8on$ the basis of 633nm, which is wavelength of recording laser(He-Ne). The highest diffraction efficiency on AsGeSeS film is $\lambda/4$, followed by $\lambda/2$, $\lambda$, $\lambda/8$. In the case of Ag 10nm, it is followed by $\lambda/4$, $\lambda/2$, $\lambda$, $\lambda/8$. On the occasion of Ag 20nm, it seems that the highest is $\lambda/2$, followed by $\lambda$, $\lambda/4$, $\lambda/8$. In other words, it appears that the highest point on AsGeSeS(158nm) single layer is 0.09%, the one of AsGeSeS(158nm)/Ag(10nm) is 1.6%, and the point of AsGeSeS(316nm)/Ag(20nm) is3.2%. Comparing the highest point in each case, we conclude that there is a distinctive increase in diffraction efficiency according the effect on Ag doping.

The Analysis of Light Emissions on Ar DC Glow Discharge under the Atmosphere Pressure (대기압 Ar 직류 글로우 방전에서 인가전압의 파형특성에 따른 광원효율 분석)

  • We developed a one-dimensional Ar fluid model running in DC-type applied voltage with a sine and a pulse waveform at the atmosphere pressure. We investigated the light emissions and efficiencies of ${\lambda}_{128nm}$, ${\lambda}_{727nm}$, ${\lambda}_{912nm}$ and ${\lambda}_{966nm}$ from the Ar excited particles. From the results, the light emission of ${\lambda}_{128nm}$ and ${\lambda}_{727nm}$ in the applied voltage with a sine waveform were almost two times as in DC voltage type. The shorter the switching time of applied voltage was, the more the light emissions of ${\lambda}_{128nm}$ and ${\lambda}_{727nm}$ were. We discussed the power consumption and Joule heating by charged particles.

  • The space of periodic Boehmians with $\Delta$-convergence is a complete topological algebra which is not locally convex. A family of Boehmians $\{T_\lambda\}$ such that $T_0$ is the identity and $T_{\lambda_1+\lambda_2}=T_\lambda_1*T_\lambda_2$ for all real numbers $\lambda_1$ and $\lambda_2$ is called a one-parameter group of Boehmians. We show that if $\{T_\lambda\}$ is strongly continuous at zero, then $\{T_\lambda\}$ has an exponential representation. We also obtain some results concerning the infinitesimal generator for $\{T_\lambda\}$.

Some Notes on the Fourier Series of an Almost Periodic Weakly Stationary Process

  • In my former paper [3] I defined an almost periodicity of weakly sationary random processes (a.p.w.s.p.) and presented some basic results of it. In this paper I shall present some notes on the Fourier series of an a.p.w.s.p., resulting from [3]. All the conditions at the introduction of [3] are assumed to hold without repreating them here. The essential facts are as follows : The weakly stationary process $X(t,\omega), t\in(-\infty,\infty), \omega\in\Omega$, defined on a probability space $(\Omega,a,P)$, has a spectral representation $$X(t,\omega)=\int_{-\infty}^{infty}{e^{it\lambda\xi}(d\lambda,\omega)},$$ where $\xi(\lambda)$ is a random measure. Then, the continuous covariance $\rho(\mu) = E(X(t+u) X(t))$ has the form $$\rho(u)=\int_{-\infty}^{infty}{e^{iu\lambda}F(d\lambda)},$$ $E$\mid$\xi(\lambda+0)-\xi(\lambda-0)$\mid$^2 = F(\lambda+0) - F(\lambda-0) \lambda\in(-\infty,\infty)$, assumimg that $\rho(u)$ is a uniformly almost periodic function.

Analysis of Immunoglobulin λ Light Chain Repertoire in Systemic Lupus Erythematosus (루푸스 환자의 면역글로불린 λ 경쇄 레파토리 분석)

  • Background: Immunoglobulin (Ig) light chain repertoire has been implicated as a critical determinant in regulation of autoreactive B cells and production of pathogenic anti-DNA antibodies in systemic lupus erythematosus (SLE). We analyzed the impact of Ig ${\lambda}$ chain repertoire on development of autoimmunity in patients with SLE. Methods: We obtained genomic DNA from individual peripheral CD19+ B cells of 3 untreated active SLE patients, and amplified $V{\lambda}$ rearrangements from each single cell by polymerase chain reaction. Results: A total number of 208 $V{\lambda}J{\lambda}$ rearrangements were analyzed. Analyzed sequences included 158 productive rearrangements and 50 nonproductive rearrangements. The differences in $V{\lambda}$ gene usage in the productive and nonproductive repertoire of SLE patients were found compared to the non-autoimmune individuals. $V{\lambda}$ gene, 9A was significantly overrepresented in nonproducative repertoire of SLE patients (P=0.016). In the productive repertoire, $V{\lambda}$ genes, 3L and 1E were found more often in the SLE patients (P=0.001, P=0.043). When the productive and the nonproductive repertoires were compared, 9A was found significantly less in the productive repertoire in the SLE patients (P=0.000). There were no significant differences in the $J{\lambda}$ gene usage between SLE patients and non-autoimmune individuals, but $J{\lambda}2/3$ gene was the most frequently used in SLE, whereas $J{\lambda}7$ gene was the most frequently used in the normal subjects. In the productive SLE $V{\lambda}$ repertoire, 9.4% of the total sequences employed identical CDR3. It was particularly striking to find 7 identical versions of the 1G-$J{\lambda}2/3$ $V{\lambda}J{\lambda}$ rearrangements from one patient and 3 of the same sequence from another patient. Notably, identical $V{\lambda}$ junctions in the SLE patients utilized significantly more homologous joining compared to $V{\lambda}$ junctions of the normal adults (P=0.044). Conclusion: These data demonstrate regulation of ${\lambda}$ light chain expression in the SLE patients by selection of unique $V{\lambda}$ genes. Also, biased selection and clonal expansion of particular $V{\lambda}$ rearrangements are apparent in the SLE ${\lambda}$ repertoire.

ON A CLASS OF CONSTACYCLIC CODES OF LENGTH 2ps OVER $\frac{\mathbb{F}_{p^m}[u]}{{\langle}u^a{\rangle}}$

  • The aim of this paper is to study the class of ${\Lambda}$-constacyclic codes of length $2p^s$ over the finite commutative chain ring ${\mathcal{R}}_a=\frac{{\mathbb{F}_{p^m}}[u]}{{\langle}u^a{\rangle}}={\mathbb{F}}_{p^m}+u{\mathbb{F}}_{p^m}+{\cdots}+u^{a-1}{\mathbb{F}}_{p^m}$, for all units ${\Lambda}$ of ${\mathcal{R}}_a$ that have the form ${\Lambda}={\Lambda}_0+u{\Lambda}_1+{\cdots}+u^{a-1}{\Lambda}_{a-1}$, where ${\Lambda}_0,{\Lambda}_1,{\cdots},{\Lambda}_{a-1}{\in}{\mathbb{F}}_{p^m}$, ${\Lambda}_0{\neq}0$, ${\Lambda}_1{\neq}0$. The algebraic structure of all ${\Lambda}$-constacyclic codes of length $2p^s$ over ${\mathcal{R}}_a$ and their duals are established. As an application, this structure is used to determine the Rosenbloom-Tsfasman (RT) distance and weight distributions of all such codes. Among such constacyclic codes, the unique MDS code with respect to the RT distance is obtained.
