THE EFFECT OF PULSED ELECTROMAGNETIC FIELDS ON ${\beta}$-TCP GRAFT IN RABBIT CRANIAL BONE DEFECT (가토 두개골 결손부에 이식된 ${\beta}$-TCP의 골치유과정에서 맥동전자기장의 영향에 관한 연구)

  • The purpose of this research was to investigate whether pulsed electromagnetic field (PEMF) stimulation applied to the rabbit cranial defects grafted with ${\beta}$-tricalcium phosphate (${\beta}$-TCP) could affect the new bone formation. With 16 New Zealand white rabbits under the same condition, bilateral calvarial bone defects were formed around the sagittal suture line. The defect on the left side was grafted with ${\beta}$-TCP, while on the right side was grafted by harvested autogenous bone. PEMF was applied to 8 rabbits for 8 hours per day. The bony specimen were divided into 3 groups, the group 1 was autogenous bone grafted specimen, the group 2 was ${\beta}$-TCP grafted with PEMF, and the group 3 was ${\beta}$-TCP grafted without PEMF. We investigated the bone regeneration & growth factor expression at 2, 4, 6, and 8 weeks. As a result, BMP 2 was expressed in the group 1 from 2 weeks, the group 2 from 4 weeks, and the group 3 from 6 weeks. BMP 4 was expressed in the group 1 from 2 weeks, in the group 2 and the group 3 from 4 weeks. 4. There was no significant difference in expression pattern of BMP 7, PDGF, VEGF, and TGF-${\beta}$1 during grafted bone regeneration in group 1, 2, and 3. According to our results, PEMF stimulation could be effective on the new bome formation in animal study, and have a feasibility of clinical use.

Preparation and Compressive Strength of Hydroxyapatite/Gelatin Composite (Hydroxyapatite/Gelatin복합체의 제조 및 압축강도)

  • Hydroxyapatite (HAp)/Gelatin (GEL) homogeneous composites of four different composition ratio were prepared by the co-precipitation process with synthetic HAp and GEL as a binder, HAP/GEL composites were molding by cold isostatic pressing and were sintering by various condition in air. Crystallinity and structure of sintered HAp/GEL composites were investigated by XRD and FTIR. Also, the compressive strength and the fracture surface of sintered specimens were measured by UTM and SEM. HAp/GEL composites showed a phase transformation to partially ${\alpha}$, ${\beta}$-tricalcium phosphate at the sintered condition of 1200$^{\circ}C$ for 3 h. The porosity of sintered body was in the range of 1.2-30.2%. The compressive strength of the sintered specimens was in the range of 16.2-60.1㎫, and its strength of sintered HAp/GEL comosites was higher than expected when the porosity was considered.

The pore size and porosity of sintered body were showed the range of 0.2∼3.0 $\mu\textrm{m}$ and 0.49∼13.43%, respectively. The compressive strength of sintered specimens were appeared the range of 36.6∼58.2 MPa. From these results, the sintered HAp/PAA comosites can be accounted for the microporous HAp having a good compressive strength due to homogeneous pore morphology.

  • Purpose: Previous clinical investigations of sinus floor augmentation have demonstrated repneumatization of grafted sinus. The aim of this study is to evaluate and compare the height changes of sinus floor after grafting with deprotenized bovine bone mineral (DBBM) and beta tricalcium phosphate (TCP). Materials and methods: 34 sinuses in 28 patients were augmented with 100% DBBM or 100% TCP through lateral approaches. Sinusgraft height was measured before, immediately after, and 6 months after bone graft with panoramic radiography. Result: After 6 months, the decreases of graft heights were 14.53% for DBBM group and 15.15% for TCP group. There was no statistically significant difference. Discussion and Conclusion: Long-term stability of sinus-graft height represents an important factor for implant success. After the uses of DBBM and TCP for maxillary sinus floor augmentations, acceptable graft height maintenances were observed.

The Influence of β-TCP Content on the Preparation of Biodegradable β-TCP/PLGA Composites Using Microwave Energy (마이크로파에 의한 생분해성 β-TCP/PLGA 복합체의 제조시 β-TCP 첨가량에 따른 영향)

  • Biodegradable $\beta$-tricalcium phosphate ( $\beta$-TCP)/poly(lactide-co-glycolide) (PLGA) composites were synthesized by in-situ polymerization with microwave energy. The influence of the $\beta$-TCP content in $\beta$-TCP/PLGA composites on the molecular weight, crystallinity, microstructure and mechanical properties was investigated. As the molecular weight of composites decreased, the $\beta$-TCP content increased up to 10 wt.%, while the excess addition of the $\beta$-TCP content above 10 wt.% the molecular weight increased with increasing of the $\beta$-TCP content. This behavior would be due to the superheating effect or nonthermal effect induced by microwave energy. It was found that the bending strength and Young's modulus of the $\beta$-TCP/PLGA composites was proportional to the molecular weight of PLGA. The bending strength of the $\beta$-TCP/PLGA composites ranged from 18 to 38 MPa, while Young's modulus was in the range from 2 to 6 GPa.

Bioactivity behavior of biphasic calcium phosphate powders prepared by co-precipitation method (공침법으로 합성된 biphasic calcium phosphate 분말의 생체활성 거동)

  • The co-precipitation technique has been applied to synthesize biphasic calcium phosphate (BCP). $Ca(NO_3)_2{\cdot}4H_2O$ and $(NH_4)_2HPO_4$ as the starting materials was used. X-ray diffraction (XRD) and Fourier transformed infrared (FT-IR) spectroscopy were used to characterize the structure of as-synthesized and calcined BCP powders. After immersion in Hanks' Balanced Salt Solution (HBSS), for 1 week a precipitation started to be formed with individual small granules on the specimen surface. An MTT assay indicated that BCP powders have no cytotoxic effects on MG-63 cells, and that they have good biocompatibility.

  • The co-precipitation technique has been applied to synthesize Biphasic Calcium Phosphate (BCP), Mg-BCP and Si-BCP. X-ray diffraction (XRD) and Fourier transformed infrared (FTIR) spectroscopy were used to characterize the structure of synthesized BCP, Mg-BCP and Si-BCP powders. The results have shown that BCP and substitution of magnesium and silicon in the calcium deficient apatites revealed the formation of biphasic mixtures of Hydroxyapatite (HAp)/${\beta}$-Tricalcium phosphate (${\beta}$-TCP) ratios after heating at $1000^{\circ}C$. Ionic substituted BCP is able to develop a new apatite phase on the surface in contact with physiological fluids faster than BCP does. An MTT assay indicated that BCP, Mg-BCP, and Si-BCP powders had no cytotoxic effects on MG-63 cells, and that they have good biocompatibility.

Late-term healing in an augmented sinus with different ratios of biphasic calcium phosphate: a pilot study using a rabbit sinus model

  • Purpose: The aim of this pilot study was to determine the osteoconductivity and dimensional stability of augmented sinuses using different ratios of biphasic calcium phosphate (BCP) in a rabbit sinus model. Methods: Each sinus of New Zealand white rabbits (2.5-3.5 kg) was assigned to one of two groups: BCP with a hydroxyapatite to ${\beta}$-tricalcium phosphate (HA:${\beta}$-TCP) ratio of 70:30 (group TCP30) and BCP with an HA:${\beta}$-TCP ratio of 30:70 (group TCP70). After preparing a window in the antral wall of a sinus, the Schneiderian membrane was elevated, and the applicable material was grafted. A fluorochrome calcein green was injected five days before euthanizing the animals at four months post-surgery. The specimens were analyzed histologically, histomorphometrically, and by using micro-computed tomography (micro-CT). Results: Micro-CT analysis revealed that the total augmented volume and the new bone volume did not differ significantly between the two groups whereas the resorption of materials was greater in the TCP70 group. The trabecular thickness, number, and separation also did not differ significantly between the two groups. Histomorphometrically, the areas of total augmentation, new bone, and residual material, as well as the ratio of new-bone-material contact did not differ significantly between the groups. Histologically, the residual particles were more scattered in the TCP70 group than in the TCP30 group. The fluorescence of the calcein green did not differ notably between the two groups. Conclusions: The osteoconductivity and dimensional stability of the two BCPs with different ratios tested in this study were comparable after four months of healing. Therefore, we conclude that both BCPs show promise as a bone substitute for sinus augmentation.

Effects of various toothpastes on remineralization of white spot lesions

  • Objective: The purpose of this in vitro study was to examine the effects of fluoridated, casein phosphopeptide-amorphous calcium phosphate complex (CPP-ACP)-containing, and functionalized ${\beta}$-tricalcium phosphate (fTCP)-containing toothpastes on remineralization of white spot lesions (WSLs) by using Quantitative light-induced fluorescence (QLF-D) Biluminator$^{TM}$ 2. Methods: Forty-eight premolars, extracted for orthodontic reasons from 12 patients, with artificially induced WSLs were randomly and equally assigned to four treatment groups: fluoride (1,000 ppm), CPP-ACP, fTCP (with sodium fluoride), and control (deionized water) groups. Specimens were treated twice daily for 2 weeks and stored in saliva solution (1:1 mixture of artificial and human stimulated saliva) otherwise. QLF-D Biluminator$^{TM}$ 2 was used to measure changes in fluorescence, indicating alterations in the mineral contents of the WSLs, immediately before and after the 2 weeks of treatment. Results: Fluorescence greatly increased in the fTCP and CPP-ACP groups compared with the fluoride and control groups, which did not show significant differences. Conclusions: fTCP- and CPP-ACP-containing toothpastes seem to be more effective in reducing WSLs than 1,000-ppm fluoride-containing toothpastes.

Improved Injection Behavior with the Addition of Granulated β-Tricalcium Phosphate in Brushite Bone Cement

  • To improve the injection behavior of brushite cement, dense ${\beta}-Ca_3(PO_4)_2$ (${\beta}-TCP$) granules were added to the starting material. The spherical ${\beta}-TCP$ granules prepared by spray-drying and subsequent sintering at $1000{\sim}1200^{\circ}C$ accounted for fractions of from 0.5 to 0.7 of the total ${\beta}-TCP$. The injection behavior was evaluated by measuring the injected mass divided by the loaded mass of paste in the syringe pump. The injected amount was increased with the increase in the fraction and sintering temperature of ${\beta}-TCP$ granules, except at $1200^{\circ}C$. The increase in the fraction of ${\beta}-TCP$ and its sintering temperature resulted in a decrease in the plastic limit, which is the volume of water required to liquefy the compact. The rest water could be utilized in the cement with the reduced plastic limit for improved injectability. The amounts of rest water assigned for powdery phase were estimated, and correlated with the injectability of paste.
