We recently reported a Philyra pisum lectin (PPL) that exerts mitogenic effects on human lymphocytes, and its molecular characterization. The present study provides a more detailed characterization of PPL based on the results from a monosaccharide analysis indicating that PPL is a glycoprotein, and circular dichroism spectra revealing its estimated ${\alpha}$-helix, ${\beta}$-sheet, ${\beta}$-turn, and random coil contents to be 14.0%, 39.6%, 15.8%, and 30.6%, respectively. These contents are quite similar to those of deglycosylated PPL, indicating that glycans do not affect its intact structure. The binding properties to different pathogen-associated molecular patterns were investigated with hemagglutination inhibition assays using lipoteichoic acid from Gram-positive bacteria, lipopolysaccharide from Gram-negative bacteria, and both mannan and ${\beta}$-1,3-glucan from fungi. PPL binds to lipoteichoic acids and mannan, but not to lipopolysaccharides or ${\beta}$-1,3-glucan. PPL exerted no significant antiproliferative effects against human breast or bladder cancer cells. These results indicate that PPL is a glycoprotein with a lipoteichoic acid or mannan-binding specificity and which contains low and high proportions of ${\alpha}$-helix and ${\beta}$-structures, respectively. These properties are inherent to the innate immune system of P. pisum and indicate that PPL could be involved in signal transmission into Gram-positive bacteria or fungi.
Near Infrared Reflectance Spectroscopy (NIRS) has been used as a tool for the rapid, accurate and nondestructive assay of small grain and forage quality analysis. The objective of this study was to establish the rapid, easy and accurate analysis method for major components of covered barley using NIRS system. NIRS used in this study was filter type instrument, Neotec 102. To obtain a useful calibration equation, standard regression between the data was analyzed by chemical analysis and by NIRS method. Standard errors of prediction (SEP) and simple correlations for unknown samples were calculated using obtained equation. SEPs for starch, $\beta$-glucan, protein and ash contents were 2.75%, 0.64%, 0.26% and 0.19%, respectively. The simple correlations for starch, $\beta$-glucan, protein and ash contents were 0.932, 0.588, 0.984 and 0.867, respectively. It was concluded that the NIRS method would be applicabl for the rapid determination of starch, protein and ash contents in barley grains.

The purpose of this study was to investigate the antioxidant activity and β-glucan content of extracts extracted by varying the temperature at 30, 55 and 80℃ after hot air drying or freeze drying of Pleurotus ostreatus and Pleurotus eryngii. For the analysis antioxidant activity of each mushroom, β-glucan, total phenol, flavonoid contents, and DPPH·ABTS+·Nitrite assay were measured. Also, the β-glucan content, total flavonoid content and ABTS+ scavenging activity increased with freeze drying rather than hot air drying, and increased with increasing extraction temperature in both mushrooms. However, the total phenol and nitrite scavenging activity increased with hot air drying rather than freeze drying, and decreased with increasing temperature in both mushrooms. DPPH scavenging activity was not significant in both mushrooms, but decreased with increasing extraction temperature. Pearson's correlations between total flavonoid content and antioxidant activities were r=0.719~0.753 (p<0.01). As a result, the β-glucan content, total flavonoid content, and ABTS radical cation scavenging activity were highest during freeze drying and extraction at 80℃. And the highest total phenol content, DPPH radical scavenging activity and nitrite scavenging activity were obtained during hot air drying and extraction at 30℃.
Chemical properties of spray-dried powders separated based on molecular weight from crude protein binding polysaccharide (CP-SD) of Agraricus blazei were examined. Contents of ${\beta}$-glucan in SD-1, SD-2 and SD-3 were 18.67%, 48.24%, and 37.15% respectively, and SD-2 (10-150 kDa) showed the highest molecular weight. Obtained ${\beta}$-glucans were not pure glucan, but was determined to be an acidic proteo-heteroglycan with a large amount of glucose (74.46-80.05%), galactose (8.91-15.2%), and mannose (4.9-5.46%). Composition of their amino acids was mainly aspartic and glutamic acids. FT-IR spectrum revealed SD-1, SD-2 and SD-3 were structures of ${\beta}$-1,3-glucans and ${\alpha}$-1,6-glucans at 890 and 930 $cm^{-1}$, respectively, signals of ${\alpha}$-1,6-glucans for CP-SD was not found. Useful CP-SD was recovered from A. blazei for preparation of three powder types as food materials.
Biopolymer is a versatile material used in food processing, medicine, construction, and soil reinforcement. 𝛽-glucan is one of the biopolymers that improves the soil water content and ion adsorption in a drought or toxic metal contaminated land for plant survival. We analyzed drought stress damage reduction in sweet potatoes (Ipomoea batatas L. cv. Sodammi) by measuring the growth and major protein expression and activity under 𝛽-glucan soil amendment. The result showed that sweet potato leaf length and width were not affected by drought stress for 14 days, but sweet potatoes grown in 𝛽-glucan-amended soil showed an effect in preventing wilting caused by drought in phenotypic changes. Under drought stress, sweet potato leaves did not show any changes in electrolyte leakage, but the relative water content was higher in sweet potatoes grown in 𝛽-glucan-amended soil than in normal soil. 𝛽-glucan soil amendment increased the expression of plasma membrane (PM) H+-ATPase, but it decreased the aquaporin PIP2 (plasma membrane intrinsic protein 2) in sweet potatoes under drought stress. Moreover, water maintenance affected the PM H+-ATPase activity, which contributed to tolerance under drought stress. These results indicate that 𝛽-glucan soil amendment improves the soil water content during drought and affects the water supply in sweet potatoes. Consequently, 𝛽-glucan is a potential material for maintaining soil water contents, and analysis of the major PM proteins is one of the indicators for evaluating the biopolymer effect on plant survival under drought stress.
This study was performed to investigate effect of dietary fibers in rice and barley on glucose, lipid and cadmium (Cd) metabolism in the rat. Fifty-six male Spague-Dawley rats weighing 244.6$\pm$2.7g were blocked into eight groups according to body weight and raised for four weeks with diets containing 0 or 0.04%(w/w) CdCl2 and four different carbohydrate sources, starch, rice flour, barley flour and mixture of rice and barley flour(7 : 3, w/w). Total dietary fibers and $\beta$-glucan contents of barley were about three times higher than those of rice (10.75% vs. 3.94%, 3.11% vs. 1.06%, respectively). Food intake, weight gain, food efficiency ratio, liver and kidney weights were lower in Cd exposed groups, and barley group among Cd exposed animals showed highest weight gain, food efficiency ratio and organ weights. Fasting serum glucose levels were not significantly different among groups, Serum cholesterol level was lowest in Cd exposed barley group. Serum HDL-cholesterol level was higher in none-Cd exposed starch and barley groups, and HDL-cholesterol : total cholesterol ratios were higher in none-Cd rice and mixed flour groups than other groups. Liver total lipid and triglyceride levels were lowest in barley groups regardless of Cd administration. Fecal total lipid, cholesterol and triglyceride excretions were high in barley and mixed flour groups. Liver Cd concentrations were low in Cd exposed barley and mixed flour groups. In Cd exposed barley group, fecal weight and Cd excretion were highest and Cd retention ratio was lowest among groups. Small intestine metallothionein(MT) concentration was highest in Cd exposed rice group, and kidney MT concentration was highest in Cd exposed barley group. In conclusion, cereals showed different effects on lipid and Cd metabolism that might be mediated by dietary fibers in cereals. Especially $\beta$-glucan-rich barley group showed greatest lipid and Cd lowering effects by increasing fecal lipids and Cd excretions.

"Hae-Songi" mushroom is a kind of Hypsizigus marmoreus, one of the edible mushrooms. Powder and hot water extracts of the mushroom fruit-body were investigated for their proximate composition, amino acid contents, ${\beta}-glucan$ contents, total phenolic contents and antioxidant activity. The measured antioxidant activity included free radical scavenging activity against DPPH, reducing power $Fe^{2+}$ chelating ability and SOD activity. Mushroom extracts exhibited in vitro antioxidant activity. This mushroom contained high protein (29%, total amino acid contents 204.86 mg/g), free amino acids (46.53 mg/g) and ${\beta}-glucan$(0.11%). At a concentration of 1% extracts solutions (w/v) according to different extraction times, DPPH free radical-scavenging activities were found to exhibit $89%{\sim}92%$ inhibition. Positive correlations $(R^2=0.9901{\sim}0.7424)$ were found between total phenolic content in the mushroom hot water extracts and their antioxidant activity. In this study, it is demonstrated that "Hae-Songi" mushroom may possess potential for use as a health food, due to theirantioxidant capacity.
This study was conducted to investigate the physiological activities and nutrition contents of Gonji No.10 and Solhyang, the strains of Neolentinus lepideus. In nutritional contents of two strains of N. lepideus, the content of free sugars and sugar alcohols and total polyphenol were more in Gonji No.10 than Solhyang. The total contents of amino acids and ${\beta}$-glucan in Solhyang are more than those in Gonji No.10. Solhyang also showed higher DPPH radical scavenging activity than Gonji No.10. In particular, hot water extract of Solhyang has higher inhibitory activity of ${\alpha}$-amylase and ${\alpha}$-amyloglucosidase while Gonji No.10 has higher anti-inflammatiory effect than Solhyang. But, they have similar effects on nitrite scavenging activity.
This study was conducted to investigate the characteristics of polysaccharides isolated from the fruit body and cultured mycelia of Phellinus linteus IY001. All fractions were extracted by hot water, followed by ethanol precipitation (F-THE and M-HE) or ultrafiltration (M-HU) (F-TH, F-THE; fruit body, M-HE, M-HU; cultured mycelia). Among these fractions, F-TH fraction was obtained at the highest yields of 6.83% and yield of F-THE was at the level 2.79%. The carbohydrates of these fractions was found to be a heteroglucan composed of glucose, galactose, mannose, fructose, ribose and xylose by analysis of gas chromatography. The total carbohydrate contents of M-HE and M-HU fractions were 99.2%, and 86.0% respectively. The glucose content of M-HE, M-HU and F-THE ranged from 54 to 84.8% of the total monosaccharide. Amino acid pattern showed that all fractions contained a large amount of aspartic acid, glycine, glutamic acid, alanine. Serine and threonine were found to be involved in the linkage, O-linked type. These fractions, except F. TH, contained polysaccharides with the molecular weights of 12 kD and showed the characteristics of IR absorption for ${\beta}-glucosides$ at $890\;cm^{-1}$.
Objectives : The purpose of this study was to investigate extracts from mixed culture of Oriental medicines and cereal medium and mycelia of Trichloma matsutake and Cordyceps militaris by fermentation to develop new material for pharmaceutical products and medicinal food, Methods : To evaluate physiological activities of OCM extracts, we examined antioxidant activity(total polyphenol contents, electronic donating ability, SOD-like activity), ${\beta}$-glucan contents, nitric oxide production and cytotoxicity by MTT assay. Results : Total polyphenol contents of fermented OCM(UF) and non-fermented OCM(UM) extracts were more than 40% UM and UF of DPPH radical scavenging activity was 25.67%, 23.43% respectively. Total polyphenol content of non-fermented extract (UME) was 12.57%, while that of fermented extract(UFE) was 7.05%. SOD like activity showed UM 85.35%, UF 76.18%, UME 58.42%, UFE 72.21%. UME, and UFE 31.43%, ${\beta}$-glucan contents of UME and UFE were more than 40%. NO productions of UME, and UFE showed a LPS dose dependent tendency. Cytotoxicity on Raw 264.7 cell showed more than 90% viability. Inhibitory effect of UFE on HT1080 cell growth was higher than UME. Conclusions : These results showed that extracts from mixed culture of mushroom mycelium and OCM have physiological activities which can be used in pharmaceutical products and medicinal food.
