When extracting semi-volatile components of herbal medicines using hot water vapor, some substances may react with water vapor or oxygen, and some volatile substances may be lost, when using an organic solvent extraction method has the disadvantage that it may contain a non-volatile material and residual organic solvent. In addition, it is inefficient to separate semi-volatile substances from herbal medicines into each single component and conduct biological activity research for each component to determine the effective ingredient, and some components may be lost in the separation process. In this study, semi-volatile substances evaporated under two pressure-reduced conditions in Chinese herbal medicines such as Cnidii Rhizoma, Aucklandiae Radix and Amomum Fructus were separated by cooling with liquid nitrogen. Those were analyzed by gas chromatography-mass spectrometry (GC-MS) to identify the components, and this method may be used to study biological activities at the cellular level. The substances separated under reduced pressure, essential oil obtained by simultaneous distillation extraction (SDE) method and substances by using solid phase micro-extraction (SPME) from Cnidii Rhizoma, Aucklandiae Radix and Amomum Fructus were analyzed by GC-MS. In the case of Cnidii Rhizoma and Aucklandiae Radix, there were some differences among the essential oil components obtained by SDE and those identified by low temperature capture (CT) and SPME method, these were believed to be produced by some volatiles reacting with water or oxygen at the boiling point temperature of water.
The main purpose of this study was to examine the correlation between the consumption of red ginseng-based 'SSR' for 30 days and the reduction in human fatigue, blood component changes, and immune cell activity in 35 human subjects. 'SSR' is composed of zinc oxide, folic acid, and D-α-tocopherol with red ginseng as the main component. According to the protocol criteria of the study, 35 subjects who understood the purpose of the study and signed an informed consent form were selected. The fatigue survey was conducted through a questionnaire, and after taking 'SSR', a decreased tendency of physical, mental, and neurosensory fatigue was observed. In hematological analysis, no significant changes were observed in the levels of WBC, RBC, and hemoglobin; however, AST (SGOT) and ALT (SGPT) levels were statistically significantly decreased. In immunological analysis, it was observed that the proliferative effect of T cells (CD3+CD4+) was greater than that of NK cells (CD16+CD56+). The collected data were subjected to t-test analysis using the SPSS 25.0 statistical program. The result from this study proposes that 'SSR' can be used as a functional food material as it reduces human fatigue and enhances immune function.
차량 실내 공기질 대응 프로세스는 두 단계의 시험 방법으로 이루어져 있다. 먼저 모듈 부품평가를 통해 차량 실내 VOCs의 주요 원인을 찾고, 절단 시험편을 이용하여 VOCs가 주로 방출되는 소재를 찾는다. 만약 VOCs가 주로 방출되는 소재를 확인 시, 소재 개선을 통해 차량과 모듈 부품에서 방출되는 VOCs를 줄일 수 있다. 하지만 기존의 절단 시험편을 이용한 시험법은 VOCs 원인을 찾는데는 절단면에서의 방출 문제가 있기 때문에, 구성 부품 단위의 평가방법 적용이 필요하다. ISO 12219-5 시험 방법은 부품 절단없이 평가를 하기 때문에 소재 및 표면적의 영향이 함께 고려되어, 모듈 부품에서 구성 부품의 VOCs 영향도 확인이 가능하였다. 트림과 콘솔의 구성부품 평가를 통해 모듈내의 주요 원인 부품을 확인할 수 있었다. 또한 ISO 12219-5 모듈 부품 시험법과 구성 부품 시험법간의 정량적 상관관계 규명을 통해 품질관리를 위한 관리값을 설정할 수 있었다.
Reconstruction and separation of explosive-contaminated overlapping fingerprints constitutes an analytical challenge of high significance in forensic sciences. Laser-induced breakdown spectroscopy (LIBS) allows real-time chemical mapping by detecting the light emissions from laser-induced plasma and can offer powerful means of fingerprint classification based on the chemical components of the sample. During recent years LIBS has been studied one of the spectroscopic techniques with larger capability for forensic sciences. However, despite of the great sensitivity, LIBS suffers from a limited detection due to difficulties in reconstruction of overlapping fingerprints. Here, the authors propose a simple, yet effective, method of using chemical mapping to separate and reconstruct the explosive-contaminated, overlapping fingerprints. A Q-switched Nd:YAG laser system (1064 nm), which allows the laser beam diameter and the area of the ablated crater to be controlled, was used to analyze the chemical compositions of eight samples of explosive-contaminated fingerprints (featuring two sample explosive and four individuals) via the LIBS. Then, the chemical validations were further performed by applying the Raman spectroscopy. The results were subjected to principal component and partial least-squares multivariate analyses, and showed the classification of contaminated fingerprints at higher than 91% accuracy. Robustness and sensitivity tests indicate that the novel method used here is effective for separating and reconstructing the overlapping fingerprints with explosive trace.
Alcoholic and nonalcoholic steaohepatitis is a leading form of chronic liver disease with few biomakers ad treatment options currently available. a progressive disease of NAFLD may lead to fibrosis, cirrhosis, and hepatocellular carcinoma. Recently, we extracted HIMH0021, which is an active flavonoid component in the Acer tegmentosum extract, has been shown to protect against liver damage caused by hepatic dysfunction. Therefore, in this study, we aimed to investigate whether HIMH0021 could regulate steatohepatitis and liver fibrosis during alcoholic or nonalcoholic metabolic process. HIMH0021, which was isolated from the active methanol extract of A. tegmentosum, inhibited alcohol-induced steatosis and attenuated the serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) during hepatocellular alcohol metabolism, both of which promote lipogenesis as well as liver inflammation. Treatment with HIMH0021 conferred protection against lipogenesis and liver injury, inhibited the expression of cytochrome P4502E1, and increased serum adiponectin levels in the mice subjected to chronic-plus-binge feeding. Furthermore, in hepatocytes, HIMH0021 activated fatty acid oxidation by activating pAMPK, which comprises pACC and CPT1a. These findings suggested that HIMH0021 could be used to target a TNFα-related pathway for treating patients with alcoholic hepatitis.
Common carp (Cyprinus carpio) (Weight 23.05 ± 6.98 g, Length 12.65 ± 1.38 cm) were exposed to waterborne nitrite at 0, 50, 100, 200, 400 and 800 mg NO2-/L for 96 hours. The lethal concentration 50 (LC50) of C. carpio exposed to waterborne nitrite was 398.6 mg NO2-/L. Hemoglobin, hematocrit and RBC count were significantly decreased by waterborne nitrite exposure. The MCV (mean mean corpuscular volume) (µl), MCH (mean corpuscular hemoglobin) (pg) and MCHC (mean corpuscular hemoglobin concentration) (%) were significantly increased. The inorganic component, plasma calcium, was significantly increased, and the organic components such as plasma cholesterol and total protein were significantly increased showing a similar tendency with calcium. In enzymatic components, the AST and ALT were also significantly increased by nitrite exposure. The results of this study indicate that exposure to nitrite can affect the survival and hematological physiology of C. carpio.
화재 현장에서 완전 연소하지 않은 잔류물의 성분을 규명하는 것은 화재 원인을 추적하는 중요한 단서가 된다. 화재 현장에서 많이 발견되는 플라스틱 연소 잔류물의 종류를 규명하기 위하여 폴리에틸렌 (Polyethylene, PE) 수지와 Acrylonitrile butadiene styrene (ABS) 수지의 연소 후 잔류물에 관한 연구를 진행 하였다. 콘 칼로리미터 (Cone calorimeter)를 사용하여 플라스틱 시료를 200, 300, 350, 400, $500^{\circ}C$로 각각 3분 동안 연소시킨 후 변화된 무게와 연소생성물을 얻었다. 각 온도에서 얻어진 연소생성물을 동결 건조 후 분쇄한 분말 시료를 FT-near infrared (NIR) spectrometer로 스펙트럼을 측정 하였다. 폴리에틸렌 수지 시료를 연소시켰을 때 $350^{\circ}C$까지는 무게의 변화가 없었으나 $400^{\circ}C$ 이상에서는 급격한 무게의 변화를 측정 할 수 있었다. 각 온도에서 얻어진 폴리에틸렌 수지 및 ABS 수지 시료의 FT-NIR 측정 스펙트럼을 주성분 분석(Principle component Analysis, PCA)를 통해서 연소 잔유물이 폴리에틸렌과 ABS 지임을 확인할 수 있었다. 따라서 근적외선 분광법으로 화재현장에서 발견되는 불연소 플라스틱의 종류를 신속하게 확인할 수 있었다.
1,1,1,2-수소불화탄소(HFC-134a)는 에어컨에 주로 사용되는 냉매로, 최근 온실가스로 규제되어 정제를 통한 재사용 방법이 권장되고 있다. 폐냉매의 재사용 기준 평가를 위해서는 폐냉매에 존재하는 미량 성분의 정량분석이 매우 중요하다. 본 연구에서는 표준 물질이 없어서 정량화하기 어려웠던 C, H, Cl, F가 포함된 미량 성분들을 GC/AED (gas chromatograph-atomic emission detector)를 이용하여 정량분석하였다. 이를 위하여 GC/MSD (mass selective detector)를 통한 정성분석을 선행하였다. 또한 성분의 원자 수와 비례하여 반응하는 AED의 특성을 조사하기 위하여, 탄화수소 혼합 표준물질을 이용하여 선형성을 확인하였다. 시료 중 C, H, Cl, F가 포함된 미량 성분의 정성 분석 결과, 주성분인 HFC-134a와 유사 냉매류들을 포함한 총 15 개의 성분이 검출되었다. MSD 결과를 토대로 AED를 이용한 미량 성분들을 정량 분석한 결과, 한 시료는 $CHClF_2$ 성분($45438.38{\mu}mol/mol$), 또 다른 시료는 $C_2H_2ClF_3$ 성분($1311.47{\mu}mol/mol$)이 가장 높은 몰분율을 나타냈다. 본 연구에서는 이 분석법을 기반으로 하여, 표준 물질이 존재하지 않아 정량화하기 어려운 복합 성분들의 정성 및 정량 분석의 확장 적용이 가능할 것으로 보인다.
졸-겔법에 의한 코디어라이트계 전구체(precursor) 제조 시 마그네슘 에톡사이드를 사용하면 황금색을 띠는 불투명한 전구체 용액이 되고, 조핵제인 5%-$Zr(OC_3H_7)_4$ 첨가 시에는 투명한 코디어라이트를 얻을 수 있다. 적외선 흡수 분광 분석에서 $SiO_2$ 성분은 $1045cm^{-1}$에서 흡수 피크를 확인 할 수 있었으며, 동시에 $1140cm^{-1}$과 $940cm^{-1}$의 두 개 피크 분리는 열처리 한 겔체인 $SiO_4$ 신축 진동이고, $Al_2O_3$ 성분은 $580cm^{-1}$과 $680cm^{-1}$의 흡수 피크로 보아 $Al_2O_6$ 망목 구조이고, MgO 성분은 $575cm^{-1}$에서 MgO 신축 진동 임을 알 수 있었다. X-선 회절 분석에서 코디어라이트계 전구체 : 물 몰비 1:5의 경우 반응 소결 온도 $1000^{\circ}C$ 이상에서는 ${\mu}$-cordierite 결정상이 나타남을 알 수 있었고, 코디어라이트계 전구체 : 암모니아 몰비 1:5 건조 겔체의 경우 $1050^{\circ}C$에서는 ${\mu}$-cordierite와 ${\alpha}$-cordierite가 공존 하다가 $1100^{\circ}C$에서는 ${\alpha}$-cordierite만이 존재함을 알 수 있었다.
본 시험은 오리 사료 내 난황항체제제 첨가 시 생산성, 혈액 특성, 장내 미생물 및 면역글로불린에 미치는 영향을 조사하기 위하여 수행하였다. 1일령 오리(Cherry valley) 총 300수를 공시하여 5처리 3반복, 반복당 20수씩 공시하여 6주간 시험을 실시하였다. 처리구는 항생제를 첨가하지 않은 처리구(NC, negative control), 항생제 처리구(PC, positive control), 난황항체제제 0.1% 첨가구(T1), 난황항체제제 0.5% 첨가구(T2) 그리고 난황항체제제 1.0% 첨가구(T3)로 나누어 실험을 실시하였다. 종료 시 체중은 난황항체제제 첨가구와 항생제첨가구가무항생제처리구보다 유의적으로 높았고(P<0.05), 증체량 역시 난황항체제제 첨가구와 항생제 첨가구가 무항생제 처리구에 비해 높게 나타났다(P<0.05). 하지만 전 기간에 대한 사료 섭취량과 사료 요구율은 처리구간에 통계적인 유의차가 나타나지 않았다. 6주간 사양 T실험 후 혈액 생화학 및 혈구 분석 결과, 난황항체제제 0.1, 1.0% 첨가구에서는 total cholesterol 함량이 무항생제 처리구에 비해 다소 감소하는 경향을 보였으나, 통계적인 유의차는 없었다. 또한 AST와 ALT 함량 역시 처리구 간에 유의적인 차이를 보이지 않았다. 오리의 맹장 내 유산균 수는 난황항체제제 0.1와 0.5% 급여 시 다른 처리구에 비해 가장 높았으며(P<0.05), 대장균 수는 난황항체제제 수준이 증가함에 따라 감소하는 경향을 보였으나, 통계적 유의차는 없었다. 또한 혈청 내 면역글로불린 함량에도 처리구간에 유의적인 차이는 나타나지 않았다. 결과적으로 오리 사료 내 난황항체첨가 시 오리의 생산성을 개선시키고, 장내 미생물 균총에 효과적인 것으로 나타났으나, 아직까지 국내에서는 난황항체제제 첨가에 따른 사양 시험과 관련된 연구가 부족하다는 점을 감안할 때 향후 이에 대한 많은 연구가 필요할 것으로 사료된다.
