Embryogenic callus culture of Tribulus terrestris L. a potential source of harmaline, harmine and diosgenin

  • In the present study, a simple one medium formulation protocol for callus culture, somatic embryogenesis and in vitro production of ${\beta}-carboline$ alkaloids and diosgenin in Tribulus terrestris L. was developed. Extensive callus induction and proliferation was obtained in leaf explant on Murashige and Skoog (MS) medium supplemented with $5.0{\mu}M$ 6 benzyl adenine (BA) and $2.5{\mu}M$ ${\alpha}-naphthaleneacetic$ acid (NAA). The embryogenic callus was maintained on subculture to fresh parental medium at 4-week intervals over a period of 28 months. The frequency of embryo formation was at a maximum ($18.1{\pm}0.9$ per g of callus) on MS medium containing $5.0{\mu}M$ BA and $2.5{\mu}M$ NAA together with $75mg\;1^{-1}$ casein hydrolysate. Globular embryo developed into torpedo stage embryo under the influence of starvation. The accumulation of ${\beta}-carboline$ alkaloids (harmaline and harmine) and steroidal saponin (diosgenin) in non-embryogenic and embryogenic callus culture derived from leaf explant was compared with root, leaf, stem, and fruit of the mother plant. The embryogenic callus accumulated equivalent amounts of harmaline ($66.4{\pm}0.5{\mu}g/g$ dry weight), harmine ($82.7{\pm}0.6{\mu}g/g$ dry weight), and diosgenin ($170.7{\pm}1.0{\mu}g/g$ dry weight) to that of the fruit of T. terrestris. The embryogenic callus culture of this species might offer a potential source for production of important pharmaceuticals.

이상환경하(異常環境下)의 영양문제(營養問題) 연구(硏究)(제1보) -진동하(振動下)의 영양문제(營養問題)- (Studies on the Nutrition under Abnormal Environment(I) -Nutrition under Vibration-)

  • On the assumption that the supplementation of certain nutrients or foods to the rice diet (low protein, low fat, and low vitamins) may decrease, to some extent, the degree of suffering from abnormal environments, such as vibration, noises, gases, dusts, smog etc. a series of experiments were started. As the first report the nutrition under vibration was studied in this experiment. Sixty (60) young growing male rats weighing about 65 grams were used, grouping to five (5) groups, twelve (12) rats each group. They were fed on the following five (5) experimental diets: rice diet (basal diet), rice diet+casein, rice diet+vitamins, rice $diet+{\alpha}-tocopherol$, and rice diet+ginseng powder (see the tables 1 and 2) for the period of 14 weeks experiment. During the experiment period the half number of the rats of each group were exposed to the three (3) hours vibration every day. The protective effect of each diet against the vibration may be summarized as follows. 1. The growth of rice diet group was impaired significantly under vibration, However, those of other groups (protein-supplemented, vitamin-supplemented, ${\alpha}-tocopherol-supplemented$ and ginseng-supplemented groups) were impaired much less compared with rice diet group. 2. The feed efficiency of the rice diet group was decreased significantly under vibration. It is estimated that the biological availability of nutrients was impaired under this environment. On the other hand, the feed efficiencies of protein supplemented, of vitamin supplemented, and of ginseng supplemented groups were not decreased under vibration, statistically. 3. There is tendency that the food spillages of vibration groups are higher than those of non-vibration groups. Especially it seems true in the case of rice diet group. The food spillage may be, to some extent, related with mental nervousness of animals. From the point that the food spillage of ginseng supplemented group is significantly lower than those of other groups it is thought ginseng acts some good role in protecting nervous system from suffering from vibration. 4. In all groups except protein supplemented group, liver fat of vibration group tends to be higher than that of non-vibration group. 5. It shows that, in general, the serum alkaline phosphatase activity of the vibration group is significantly higher than that of the non-vibration group. It seems that there may be, to some extent, corelation between the amount of liver fat and serum alkaline phosphatase activity. 6. There is tendency that, in rice diet group, the organs of vibration group are smaller than those of non-vibration group, especially lung is so. It is thought that this may be due to the poor growth of whole body size in vibration group.

Fibrin 용해 균주의 분리 및 특성 (Isolation and Characterization of a Bacterium with a Fibrinolytic Activity)

  • 혈전 용해효소를 생산하는 미생물을 탐색하기 위하여 장류 및 토양사료로부터 미생물을 분리하여 그 중 fibrin 용해활성이 높고 기질특이성이 우수한 S7-l6 균주를 선발하였다. 선발된 미생물은 Gram 양성의 간균으로 운동성이 있고, catalase 양성이었으며 포자를 형성하는 것으로 관찰되어 BClcilius sp. S7-16으로 명명하였다. $\alpha$cill$\mu$ sp S7-16의 효소생산 조건을 검토한 결과,0.5% (w/v) polypeptone, 0.5% (w/v) yeast extract, 0.3% (w/v) NaCI, 0.l%(w/v) $KH_2P0_4$, 0.3% (w/v) $KH_2P0_4$, 0.01% (w/v) $MgSO_4.7H_20$(pH7.0)의 배지조성하에서 $35^{\circ}C$애서 180 rpm으로 24시간 회전진탕배양하였을 때 fibrin 용해효소 생산이 최대를 보였으며, 이때 casein 용해활성은 나타나지 않았다. Fibrin 용해효소의 효소적 특성을 조사하기 위해 이 조건하에서 얻어진 배양상동액에 20~60% ammomum sulfate 분획침전을 행하여 조효소액을 조제하였으며, 얻어진 조효소액은 pH4~11의 범위에서 안정하였으며, $50^{\circ}C$까지 비교적 활성이 유지되는 열안 정성을 보여주었다. 조효소액의 최적 pH와 온도는 각각 7.5, $40^{\circ}C$부근에서 최적활성을 보였다.

Identification and Characterization of Lactobacillus salivarius subsp. salivarius from Korean Feces

  • This study was conducted to isolate lactobacilli having probiotic characteristics to be used as health adjuncts with fermented milk products. Acid tolerant strains were selected in Lactobacilli MRS broth adjusted to pH 4.0 from 80 healthy persons (infants, children and adults). And bile tolerant strains were examined in Lactobacilli MRS broth in which 1.0% bile salt was added. By estimation above characteristics, the strains No. 27, which was isolated from adult feces, was selected and identified as Lactobacillus salivarius subsp. salivarius based on carbohydrate fermentation and 16S rDNA sequencing. It was used as a probiotic strain in fermented milk products. The pH of fermented milk decreased from pH 6.7 to 5.0 and titratable acidity increased from 0.3% to 1.0% by L. salivarius subsp. salivarius (isolation strain 20, 35, and 37), when incubated for 36 h at $37^{\circ}C$. The number of viable cell counts of fermented milk was maximized at this incubation condition. The SDS-PAGE evidenced no significant change of casein but distinct changes of whey protein were observed by isolated L. salivarius subsp. salivarius for titratable acidity being incubated by $0.9{\sim}1.0%$ at $37^{\circ}C$. All of the strains produced 83.43 to 131.96 mM of lactic acid and 5.39 to 26.85 mM of isobutyric acid in fermented products. The in vitro culture experiment was performed to evaluate ability to reduce cholesterol levels and antimicrobial activity in the growth medium. The selected L. salivarius subsp. salivarius reduced $23{\sim}38%$ of cholesterol content in lactobacilli MRS broth during bacterial growth for 24 hours at $37^{\circ}C$. All of the isolated L. salivarius subsp. salivarius had an excellent antibacterial activity with $15{\sim}25$ mm of inhibition zone to E. coli KCTC1039, S. enteritidis KCCM3313, S. typhimurium M-15, and S. typhimurium KCCM40253 when its pH had not been adjusted. Also, all of the isolated L. salivarius subsp. salivarius had partial inhibition zone to E. coli KCTC1039, E. coli KCTC0115 and S. enteritidis KCCM3313 when it had been adjusted to pH 5.7. The selected strains were determined to have resistances of twelve antibiotic. Strains 27 and 35 among the L. salivarius subsp. salivarius showed the highest resistance to the antibiotics. Purified ${\alpha}$-galactosidase was obtained by DEAE-Sephadex A-50 ion exchange chromatography, Mono-Q ion exchange chromatography and HPLC column chromatography from L. salivarius subsp. salivarius 27. The specific activity of the purified enzyme was 8,994 units/mg protein, representing an 17.09 folds purification of the original cell crude extract. The molecular weight of enzyme was identified about 53,000 dalton by 12% SDS-PAGE. Optimal temperature and pH for activity of this enzyme were $40^{\circ}C$ and 7.0 respectively. The enzyme was found to be stable between 25 and $50^{\circ}C$. ${\alpha}$-galactosidase activity was lost rapidly below pH 5.0 and above pH 9.0. This enzyme was liberated galactose from melibiose, raffinose, and stachyose, and also the hydrolysis rate of substrate was compound by HPLC. These results indicated that some of the L. salivarius subsp. salivarius (strain 27 and 35) are considered as effective probiotic strains with a potential for industrial applications, but the further study is needed to establish their use as probiotics in vivo.

사삼주를 이용한 아펜젤러 치즈의 품질 특성 (Quality Properties of Appenzeller Cheese Containing Sasam(Codonopsis lanceolate) Wine)

  • 본 연구에서는 사삼주를 자연 치즈에 접목시킴으로써 사삼주의 약리효과와 고유의 향미가 부여된 한국형 기능성 치즈개발을 위해 사삼주 첨가 아펜젤러 치즈(Appenzeller cheese)를 제조하여 사삼주 첨가가 치즈의 전반적인 품질 변화와 관능성에 미치는 영향을 검토하였다. 사삼주의 첨가가 숙성 중 유산균의 증식과 산 생성에 큰 영향을 미치지는 않았으나, 사삼주 6.0% 첨가구가 다소 높게 나타났고. 숙성 기간에 따라 $\alpha_{s1}$-casein의 분해가 일어나고 사삼주의 첨가량이 증가할수록 분해도가 높았으며, 숙성 촉진의 효과를 기대해 볼 수 있었다. 치즈의 총 질소 화합물(NPN, NCN, WSN)의 변화에서는 숙성 기간이 경과함에 따라 대체적으로 대조구와 첨가구 모두 증가하였다. 사삼주 첨가에 따른 분해도는 숙성 초에는 차이가 거의 없었으나, 숙성 15주에는 사삼주 첨가량이 많을수록 단백질의 분해도가 더 높은 것으로 나타났다. 사삼주 치즈의 조사포닌 함량을 측정한 결과, 4.0% 첨가구에서 비교적 높은 사포닌 회수율을 보였고, TBA 값은 숙성 초기보다 숙성 이후가 대조구보다 첨가구에서 다소 낮은 값을 보이는 것을 알 수 있었다. 소비자 기호도에서는 치즈의 향미와 맛을 제외한 외관과 조직감에서 첨가구가 다소 높은 값을 보였다. 이상의 결과를 종합하면 사삼주의 첨가가 치즈 제조상의 특성에는 큰 영향을 미치지 않았으나 첨가 수준에 따른 단백질 분해도와 일반성분의 차이를 볼 수 있었으며, 사삼주를 첨가함으로써 사포닌이 치즈 내 잔존함을 확인할 수 있었기에 이를 이용한 한국형 기능성 자연 치즈 개발이 가능할 것으로 기대되었다.

어류 알로부터 Protease Inhibitors의 단백질 용해도 차이에 의한 분획 특성 (Fractionation and Characterization of Protease Inhibitors from Fish Eggs Based on Protein Solubility)

  • A protease inhibitor was fractionated from fish eggs using methods based on protein solubility. Fractionation efficiency was evaluated with regard to percent recovery and total inhibitory activity (U). The fractionation of protease inhibitor (PI) from egg extracts of skipjack tuna (ST, Katsuwonus pelamis), yellowfin tuna (YT, Thunnus albacores), and Alaska pollock (AP, Theragra chalcogramma) was performed by precipitation with cold acetone or ammonium sulfate (AS). Fractions exhibiting the strongest inhibitory activity contained 20-40% (v/v) cold acetone or 40-60% saturated AS fractions. AS fractionation was more effective in isolating PI than was precipitation with acetone. The total inhibitory activity and percent recovery of fraction obtained with AS 40-60% toward trypsin and $N{\alpha}$-benzoyl-L-arginine-p-nitroanilide (BAPNA) were 4,976 U and 24.2% for ST, 3,331 U and 38.1% for YT, and 4,750 U and 43.8% for AP, respectively. In comparisons against six commercial proteases, 40-80% AS fractions, made by combining the 40-60% and 60-80% AS fractions from fish egg extract, exhibited the strongest inhibition of trypsin when using a casein substrate. These results suggest that fish eggs act as serine protease inhibitors and may be useful for protease inhibition in foodstuffs.

Sodium Caseinate 가수분해물의 Angiotensin-I Converting Enzyme 저해효과에 관한 연구 (Effect of Sodium Caseinate Hydrolysates on Angiotensin-I Converting Enzyme Inhibition Activity)

  • 본 연구는 유단백질 유래 카제인염을 상업용 단백질가수분해 효소로 처리하여 효소의 종류와 가수분해 시간에 따른 ACE 저해효과를 살펴보고 주요 성인병인 고혈압의 예방을 위한 혈압강하 효과가 높은 가수분해물을 제조하고자 수행하였다. 카제인염을 6종의 단백질 분해효소로 기질대 효소비 1000:1로 첨가하여 8시간 가수분해했을 때 가수분해도는 2.54-4.25%로 나타났고, 다시 가수분해물을 기질대 효소비 500:1로 처리하여 4시간 동안 2차 가수분해하였을 때 4.30-5.22%의 가수분해가 일어났다. 효소별 가수분해물의 ACE 저해효과는 가수분해가 진행됨에 따라 $IC_{50}$의 수치는 감소하였고 저해율을 증가하였다. 1차 가수 분해시 8시간 가수분해한 가수분해물의 $IC_{50}$ 수치는 Protamex 처리군이 $516{\mu}g/mL$로 가장 낮았고 Flavourzyme이 $866{\mu}g/mL$로 가장 높았다. 1차 가수분해물을 Flavourzyme로 4시간 2차 가수분해를 하였을 때 1차 가수분해물 보다 $IC_{50}$ 수치가 감소되어 ACE저해 효과가 증가하였으며 Neutrase로 처리하였을 때 $282{\mu}g/mL$로 가장 낮았고 Protease M이 $570{\mu}g/mL$로 가장 효과가 적었다. 가장 효과가 좋은 Neutrase 2차 가수분해물을 소화효소인 pepsin, trypsin, ${\alpha}$-chymotrypsin으로 가수분해 하였을 경우 $IC_{50}$ 수치는 $597{\mu}g/mL$로 1차 가수분해물보다 저해효과가 유의적으로 감소되었다(p<0.05). Neutrase 2차 가수분해물을 MW 10,000로 한외여과하였을 때 MW 10,000 미만 permeate의 $IC_{50}$ 수치는 $273{\mu}g/mL$로 저해효과는 유의차가 없었으나, 10,000 이상의 retentate는 $IC_{50}$ 수치가 $635{\mu}g/mL$로 유의적 수준에서 저해효과가 감소하였다(p<0.05). 이에 효소의 종류와 가수분해 시간의 조합에 의한 ACE 저해활성을 측정하고 분리공정을 최적화하기 위한 추가 연구를 수행한다면 주요 유단백질인 카제인 유래 기능성 식품의 산업적 대량생산이 가능할 것으로 사료된다.

인체 급성백혈병 Jurkat T 세포에 있어서 L-canavanine에 의해 유도되는 세포자살기전에 미치는 단백질 티로신 키나아제 p56lck의 저해 효과 (A Natural L-Arginine Analog, L-Canavanine-Induced Apoptosis is Suppressed by Protein Tyrosine Kinase p56lck in Human Acute Leukemia Jurkat T Cells)

  • L-arginine 구조유사체인 L-canavanine의 인체 급성백혈병 Jurkat T 세포에 대한 apoptosis 유도활성이 단백질 티로신키나아제 $p56^{lck}$에 어떻게 조절되는지를 규명하기 위해 $p56^{lck}$를 발현하는 Jurkat T 세포주 E6.1과 $p56^{lck}$-결손 Jurkat T 세포주 JCaM1.6에 있어서 L-canavanine의 세포독성, L-canavanine에 의해 유도되는 apoptotic DNA fragmentation 및 apoptotic sub-$G_1$ peak를 비교하여 본 바, $p56^{lck}$-negative JCaM1.6 세포가 $p56^{lck}$-positive E6.1 세포에 비해 L-canavanine의 apoptotis 유도활성에 훨씬 더 민감한 것으로 나타났다. 이러한 $p56^{lck}$-negative JCaM1.6 세포의 민감성은 JCaM1.6 세포에 $p56^{lck}$ 유전자를 transfection시켜 발현시키면 현저히 감소되었다. L-Canavanine에 의해 유도되는 apoptosis관련 현상들을 $p56^{lck}$-stable transfectant인 JCaM1.6/lck 세포와 empty vector-transfectant 인 $p56^{lck}$-negaive JCaM1.6/vector 세포에서 Western blot analysis로 비교한 결과, L-canavanine에 의해 유도되는 mitochondrial membrane potential (${\Delta\Psi}m$)의 감소, caspase-9, -8, -7 및 -3의 활성화, 그리고 PARP 및 $PLC{\gamma}$-1의 분해가 JCaM1.6/vector 세포에 비해 JCaM1.6/lck 세포에서 더 약하게 나타났다. JCaM1.6/lck 세포를 2.5 mM L-canavanine으로 처리한 다음 세포 내 $p56^{lck}$ kinase 활성의 변화를 $\alpha$-casein을 기질로 하여 시간 별로 측정한 결과, L-canavanine의 처리 후 15분만에 $p56^{lck}$ kinase의 활성이 약 1.6배 증가되었으며 이후 6시간 동안은 약 1.3~1.4 배정도 증가된 수준으로 kinase 활성이 유지되는 것으로 확인되었다. L-Canavanine에 의한 apoptosis의 개시에 Fas/FasL 상호작용이 관련되는지를 규명하기 위해 FADD-negative Jurkat T 세포주 I2.1, caspase-8-negative Jurkat T 세포주 I9.2 및 wild-type Jurkat T 세포주 A3에 대한 L-canavanine의 세포독성을 비교한 결과, A3와 I2.1 세포의 경우는 L-canavanine의 세포독성이 동일하게 나타났고, 특히 caspase-8가 결손된 I9.2 세포의 경우는 L-canavanine의 세포독성에 대한 민감성이 A3와 I2.1 세포에 비해 단지 미약하게만 완화되는 것으로 나타나, L-canavanine의한 apoptosis에는 Fas/FasL 상호작용이 관련되어 있지 않으며, 또한 caspase-8의 역할이 필수적이지 않음을 시사하였다. Jurkat T 세포에 있어서 L-canavanie에 의해 유도되는 sub-$G_1$ peak 및 caspases 활성화에 미치는 pan-caspase inhibitor (z-VAD-fmk), caspase-9 inhibitor (z-LEHD-fmk), caspase-3 inhibitor (z-DEVD-fmk), caspase-4 inhibitor (z-LEVD-fmk) 및 caspase-12 inhibitor (z-ATAD-fmk)의 영향을 조사한 결과, L-canavanie에 의한 apoptosis는 ${\Delta\Psi}m$의 감소, caspase-9 및 caspase -3의 활성화에 뒤따른 caspase-8 및 caspase-7의 활성화, 그리고 PARP의 분해의 순서로 유도되는 것으로 나타났으며, 아울러 caspase-9의 활성화와 함께 caspase-12의 활성화가 L-canavanine 처리에 따른 caspase-3의 활성화에 요구되는 것으로 확인되었다. 결론적으로, L-canavanine 처리에 의한 Jurkat T 세포의 apoptosis는 ${\Delta\Psi}m$ 감소, caspase-9, caspase-3 및 caspase-7의 활성화에 의해 유도되며, 이들 apoptosis 현상들은 $p56^{lck}$에 의해 negative regulation되었다.
