α-amylase and α-glucosidase inhibition effects of Korean edible brown, green, and red seaweed extracts

  • The control of intestinal α-amylase and α-glucosidase is an effective therapeutic strategy for prevention of post-prandial hyperglycemia associated with diabetes mellitus. The objective of this study was to evaluate the anti-diabetes activities of Korean edible seaweed against α-amylase and α-glucosidase, two carbolytic enzymes involved in serum glucose regulation. Of the 41 species initially screened, Cladophora wrightiana var. minor, Eisenia bicyclis, Ecklonia cava, Ishige foliacea, and Ishige okamurae exhibited the strongest inhibitory activities from brown seaweeds. Asparagopsis taxiformis showed the strongest inhibitory effects from red seaweeds. The results of this study suggest that the crude brown seaweed extracts (C. wrightiana var. minor, E. bicyclis, E. cava, I. foliacea, and I. okamurae) and crude red seaweed extracts (A. taxiformis) may have beneficial effects suppressing the rise in postprandial hyperglycemia through the inhibition of α-amylase and α-glucosidase.

Production and Characterization of a New ${\alpha}$-Glucosidase Inhibitory Peptide from Aspergillus oryzae N159-1

  • An ${\alpha}$-glucosidase inhibitor was developed from Aspergillus oryzae N159-1, which was screened from traditional fermented Korean foods. The intracellular concentration of the inhibitor reached its highest level when the fungus was cultured in tryptic soy broth medium at $27^{\circ}C$ for five days. The inhibitor was purified using a series of purification steps involving ultrafiltration, Sephadex G-25 gel permeation chromatography, strong cation exchange solid phase extraction, reverse-phase high performance liquid chromatography, and size exclusion chromatography. The final yield of the purification was 1.9%. Results of the liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis indicated that the purified ${\alpha}$-glucosidase inhibitor was a tri-peptide, Pro-Phe-Pro, with the molecular weight of 360.1 Da. The IC50 value of the peptide against ${\alpha}$-glucosidase activity was 3.1 mg/mL. Using Lineweaver-Burk plot analysis, the inhibition pattern indicated that the inhibitor acts as a mixed type inhibitor.

Antioxidant and Biological Activity in the Leaves of Adzuki Bean (Vigna angularis L.)

  • The adzuki bean (Vigna angularis L.) is a red-grained legume that has a number of essential nutrients and is used in traditional dishes in Asia. Adzuki bean industrial by-products are also a potential low-cost source of some unique bioactive polyphenols. Hence, here, the authors aimed to perform a comparative study of the phytochemical profiles of the leaves and seeds of the adzuki bean and compare their antioxidant, ${\alpha}$-glucosidase inhibition, and tyrosinase inhibition activity. The authors assessed antioxidant activity by DPPH, ABTS, FRAP, PR, TPC, and SOD assays, which showed wide variation, respectively. From the relative antioxidant capacity index results, 10 adzuki bean landraces were selected to compare for phytochemicals and bioactivity using leaf and seed extracts. Antioxidant, ${\alpha}$-glucosidase inhibition, and tyrosinase inhibition activity in the leaf extracts were higher than in the seed extracts, and there were more flavonols and isoflavones in the leaf extracts than in the seed extracts. This study demonstrated that adzuki bean leaf extracts could be a new natural antioxidant or antidiabetic agent and a skin whitener and can also be used in industrial applications.

반응표면분석법에 의한 메밀(Fagopyrum esculentum M.) 새싹 기능성분의 추출 조건 최적화 (Optimization of Extraction Conditions to Obtain Functional Components from Buckwheat (Fagopyrum esculentum M.) Sprouts, using Response Surface Methodology)

  • 본 실험은 메밀새싹의 추출조건에 따른 수율, 총 페놀, $\alpha$-glucosidase 저해활성능, 및 ACE 저해활성능에 대해 반응표면 분석법을 이용하여 추출조건을 최적화하였다. 중심합성계획에 따라 추출온도($0{\sim}100^{\circ}C$), 추출시간(0~12 hr) 및 에탄올 농도(0~100%)를 달리하였을 때 반응 표면 회귀식의 R2는 수율, 총 페놀, $\alpha$-glucosidase 저해활성능 및 ACE 저해활성능에서 각각 0.9461(p<0.001), 0.8875(p<0.005), 0.9186(p<0.005) 및 0.8667(p<0.005)로 나타내었으며, 추출조건별 총 페놀 함량, $\alpha$-glucosidase 저해활성능 및 ACE 저해활성능에 대한 반응표면을 superimposing하여 얻은 최적 추출조건 범위는 추출온도 $0{\sim}70^{\circ}C$, 추출시간 2~8 hr 및 에탄올 농도 30~80%로 나타내었다. 최적 추출조건 범위내의 임의의 조건인 추출온도 $15^{\circ}C$, 추출시간 5 hr 및 에탄올 농도 50 %를 회귀식에 대입하여 얻은 예측값은 수율 15.18%, 총 페놀 함량 189.27 mg/100 g, $\alpha$-glucosidase 저해활성능 77.44 % 및 ACE 저해활성능 85.24%으로 예측되었으며, 실제 실험을 통해 얻어진 값은 수율 16.18%, 총 페놀 함량 175.57 mg/100 g, $\alpha$-glucosidase 저해활성능 79.17% 및 ACE 저해활성능 81.60%으로 매우 유사하게 나타났다.

  • We examined ethanol extracts prepared from the Korean marine algae belonging to the Sargassaceae family for their inhibitory effects on ${\alpha}$-glucosidase activity and free radicals in vitro. Among five marine algae, the extracts of Sargassum yezoense were found to possess strongly ${\alpha}$-glucosidase inhibition and free radicals scavenging activities. Two compounds were isolated via bioactivity guided isolation and tested for their effects on ${\alpha}$-glucosidase, DPPH, $ABTS^{+}$ and $Photochem^{(R)}$ analysis. Their chemical structures were elucidated by spectral analysis and direct comparison with authentic compounds; their structures were identified as sargaquinoic acid (1) and sargahydroquinoic acid (2). The inhibitory effects of compound 1 and 2 ($IC_{50}$ value:14.2 and 12.8 ${\mu}M$, respectively) on ${\alpha}$-glucosidase were more potent that of deoxynojirimycin as a positive control ($IC_{50}$ value:18.0 ${\mu}M$). All compounds displayed antioxidative activity which was measured by DPPH, $ABTS^{+}$ and $Photochem^{(R)}$ apparatus.

  • As an effort to find a new scaffold for ${\alpha}$-glucosidase inhibition, we have prepared total 11 phenylalkylated piperazine derivatives and tested their ${\alpha}$-glucosidase inhibitory activities. Compounds 8 ($IC_{50}=2.73{\pm}0.075mM$) possessing two 3-methoxyphenethyl group on 1,4-position of piperazine showed comparable potency to acarbose used as reference. But other compounds were inactive to ${\alpha}$-glucosidase. The result indicated that proper substituents on the piperazine can engender ${\alpha}$-glucosidase inhibitory activities on the piperazine derivatives.

약용식물 메탄올 추출물의 항산화 및 항당뇨 활성 (Anti-Oxidative and Anti-Diabetic Effects of Methanol Extracts from Medicinal Plants)

  • 약용식물 23종 메탄올 추출물의 폴리페놀 함량을 측정한 후 항산화 활성을 측정하고, 항당뇨 활성으로는 ${\alpha}$-amylase 저해 활성, ${\alpha}$-glucosidase 저해 활성을 검정하여 혈당강하 활성을 나타낼 수 있는 약용식물을 선발하고자 한다. 약용식물의 폴리페놀 함량을 분석한 결과 1.50~15.25 mg GAE/g의 다양한 함량 범위로 나타났으며 가장 높은 함량은 주목열매(15.25 mg GAE/g), 화살나무줄기(15.12 mg GAE/g), 두충잎(14.24 mg GAE/g)으로 3종이 나타났다. 60% 이상 DPPH 라디칼 소거능을 본 약용식물로는 두충 줄기수피(80.10%), 구기자나무 뿌리(64.25%), 화살나무 줄기(73.59%), 비수리(78.20%), 주목열매(70.52%), 뽕잎나무 잎줄기(67.81%) 부위 추출물로 6종이 선발되었다. ${\alpha}$-Glucosidase를 80% 이상 저해한 약용소재로는 두충 줄기심재, 두충 줄기수피, 화살나무 줄기, 마, 율무, 녹두 6종이 선발되었다. ${\alpha}$-Amylase를 80% 이상 저해한 약용식물로는 칡뿌리, 두충 줄기수피, 두충잎, 구기자 열매, 화살나무 잎줄기, 화살나무 줄기, 조릿대 지상부, 마, 뽕잎나무 잎줄기 추출물 등 9종이 선발되었다. 항산화 및 항당뇨 활성이 우수했던 약용식물들은 향후 새로운 의약품 개발 및 성인병 예방의 건강기능식품으로 개발하는 데 있어 좋은 기초자료로 활용될 것으로 기대된다.

  • The synthesis of cis- and trans-1,6-dihydroxypyrrolizidine starting from trans-4-hydroxy-L-proline and their evaluation as glycosidase inhibitors are reported. The cis-isomer was found to be a potent inhibitor against .alpha.-glucosidase and showed weak inhibitory effect against other glycosidases. The trans-isomer exhibited weak inhibitions of b-glucosidase and amylo-glucosidase and poor inhibition of other glycosidases.

Evaluation of Cytotoxicity, Carbohydrate, and Lipid Inhibitory Activity of Codonopsis lanceolata using Different Solvent Fractions

  • This study was conducted to evaluate the cytotoxicity and ${\alpha}-Amylase$, ${\alpha}-Glucosidase$, pancreatic lipase inhibition in vitro by different solvent fractions from the roots of Codonopsis lanceolata. The values of $IC_{50}$ against Calu-6 cell showed a high effect in n-hexane fraction ($10.13{\mu}g/mL$) whereas DW fraction exhibited the weakest inhibition on cell viability, having an $IC_{50}$ value of over $1,000{\mu}g/mL$. The values of $IC_{50}$ against HCT-116 cell showed the highest activity in n-BuOH fraction ($102.01{\mu}g/mL$), followed by n-hexane fraction ($145.85{\mu}g/mL$), methylene chloride fraction ($332.02{\mu}g/mL$), ethyl acetate fraction ($462.93{\mu}g/mL$) and DW fracion ($>1,000{\mu}g/mL$). ${\alpha}-Amylase$ inhibitory activity in methylene chloride fraction and ethyl acetate fraction was found to have a higher inhibitory effect with 24.5% and 25.6% than the other fractions. The highest ${\alpha}-Glucosidase$ inhibitory activity was observed from the ethyl acetate fraction extract, while the extract of DW fraction showed the lowest level of inhibitory activity at given experiment concentration. The pancreatic lipase inhibitory activity of C. lanceolata was found to have a higher the effect in ethyl acetate fraction. Inhibition of lipase activity of the ethyl acetate fraction and n-hexane fraction showed a relatively high, while the extract of DW fraction showed the lowest level at given experiment concentration. These results suggested that the roots of C. lanceolata may assist in the potential biological activity on carbohydrate, lipid Inhibitory activity and anticancer activity.

혈당강하 기능 오미자 소스의 이화학적 품질 및 생리활성 (Physicochemical Quality and Hypoglycemic Effect of Omija Sauce)

  • 본 연구에서는 오미자 농축액 첨가량(0~5%)에 따른 소스의 품질특성과 혈당강하능 및 관능적 특성에 미치는 영향에 대해 조사하였다. 오미자 농축액 첨가량이 증가할수록 수분함량, 총 페놀함량 및 환원당은 증가하였고 pH는 감소하였다. 소스의 이화학 시험 결과 당도, 환원당, 총 페놀함량, ${\alpha}$-amylase 저해율 및 ${\alpha}$-glucosidase 저해율 등은 오미자 농축액의 첨가량에 따라 증가하였다. 소스의 색도는 오미자 농축액 첨가량이 증가함에 따라 명도는 감소하였고, 적색도, 황색도 및 ${\Delta}E$ 값은 증가하였다. 관능적 특성은 소스만 섭취 하였을 때 4% 첨가한 오미자 소스에서 가장 높은 기호도를 나타내었고, 닭 가슴살과 함께 섭취하였을 때 5% 첨가한 오미자 소스의 기호도가 가장 높았다.
