Study on Anti-obesity and Hypoglycemic Effects of Lycium chinense Mill Extracts (구기자 추출물의 항비만 및 혈당강하 효과)

  • This study was designed to evaluate the anti-obesity and hypoglycemic effects of Gugija (Lycium chinense Mill) extracts in 3T3-L1 adipocytes. We investigated the $\alpha$-amylase and $\alpha$-glucosidase inhibitory activities of extracts from Gugija. Gugija was extracted by 70% EtOH and 80% MeOH and aqueous, respectively. A single oral dose of Gugija extract inhibited the increase of blood glucose levels significantly at 0, 30, 60, 90 and 120 min and decreased incremental response areas under the glycemic response curve. These results suggest that Gugija 70% EtOH extracts may delay carbohydrate digestion and reduce postprandial hyperglycemia. In addition, triglyceride content in 3T3-L1 adipocytes decreased at higher concentrations of Gugija 70% EtOH extract. Free fatty acid content in 3T3-L1 adipocytes was increased at higher concentrations of Gugija 70% EtOH extract. Also, glucose transporter 4 (GLUT4), the key insulin signaling pathway transcription factor, was remarkably increased by the Gugija 70% EtOH extract when compared to those of control cells in protein expression levels. Therefore, Gugija can be developed as an effective anti-obesity and hypoglycemic agent.

Characterization of Barley ${\alpha}$-Amylase Chimeric Enzymes Expressed in Pichia pastoris (Pichia pastoris에서 발현된 보리 알파아밀라제 Chimera 효소들의 특성)

  • Two different ${\alpha}$-amylase isozymes (AMY1 and AMY2) found in barley malt share up to 80% of amino acid sequence identity with each other, but their enzymatic properties differ remarkably. AMY1 shows the highest activity at low concentration of calcium ion, while AMY2 is highly active at high calcium concentration. Meanwhile, BASI (Barley ${\alpha}$-Amylase/Subtilisin Inhibitor) protein specifically inhibits only AMY2. In the present study, three separate regions in AMY genes (I, II, and III) were assigned on the basis of restriction enzyme sites and four kinds of chimeric amylases have been obtained by swapping a part of regions with each other. Each chimera gene was successfully over-expressed in Pichia pastoris. From the results of enzymatic characterization, both AMY211 and AMY122 showed the mixed or intermediate type of calcium-dependent activity between AMY1 and 2. Meanwhile, only AMY221 chimera could be significantly inhibited by BASI protein. As a result, it can be proposed that some amino acid residues in the region I and II, except region III, of barley ${\alpha}$-amylases play very important roles in calcium-dependency and interaction with BASI.

Hypoglycemic Effects of a Medicinal Herb Mixture Prepared through the Traditional Antidiabetic Prescription (당뇨 처방에 근거한 생약재 복합물의 혈당강하 효과)

  • For the purpose of investigating the in vitro antidiabetic activity of a medicinal herb mixture prepared through traditional antidiabetic prescription, the study analyzed the existence of insulin-similar components and examined ${\alpha}$-amylase and ${\alpha}$-glucosidase inhibition activity. As a result of arranging the medicinal herb mixture extracts over the 3T3-L1 fibroblast in the concentration of $10{\mu}g/mL$, which confirmed that it included much of insulin sensitizer components as 151.7% in the differentiation of 3T3-L1 fibroblast. The inhibition activity against ${\alpha}$-amylase of the medicinal herb mixture extracts as hypoglycemic agent were 38.4, 31.5 and 16.6% in the concentration of 10.0, 1.0 and 0.1 mg/mL, respectively. The inhibition activity against ${\alpha}$-glucosidase of the medicinal herb mixture extracts were 81.3, 35.8 and 26.7% in the concentration of 10.0, 1.0 and 0.1 mg/mL, respectively. The inhibition activity against ${\alpha}$-glucosidase in the ethyl acetate fractions of the water and 80% ethanol extracts were 66.9% and 55.1%, respectively, the highest levels in the various solvent extracts.

Antioxidative Effect and Digestive Enzyme Inhibition of Grape Seed Extract (GSE) (포도씨 추출물의 항산화 효과 및 소화효소 저해 효과)

  • The purpose of this study is to investigate the antioxidative activity and digestive enzyme inhibition of grape seed extract (GSE). The GSE was tested for its effect on various antioxidative potentials (scavenging activities of DPPH radical, superoxide anion radical and hydroxyl radical) and inhibitory effect of various digestive enzymes (trypsin, $\alpha$-chymotrypsin, $\alpha$-amylase, $\beta$-glucosidase and lipase). DPPH radical scavenging activity ($SC_{50}$, 50% scavenging concentration) of GSE was 4.76${\pm}$0.27 ppm while those of positive controls (EGCG and vitamin C) were 2.22${\pm}$0.12 ppm and 9.50${\pm}$0.72 ppm, respectively. $SC_{50}$ value of GSE against superoxide anion radical and hydroxyl radical were 3.82${\pm}$0.07 ppm and 803.23${\pm}$27.16 ppm, respectively. In addition, $IC_{50}$ values of GSE against trypsin, $\alpha$-chymotrypsin, $\alpha$-amylase, $\alpha$-glucosidase and lipase were 2.17${\pm}$0.59 ppm, 7.46${\pm}$1.25 ppm, 18.25${\pm}$3.54 ppm, 12.30${\pm}$1.12 ppm, and 653.23${\pm}$79.34 ppm, respectively. These results suggest that GSE may be useful for the prevention or treatment of obesity.

Purification of ${\alpha}-Amylase$ Inhibitor from White Kidney Bean(Phaseolus vulgaris) (White Kidney bean(Phaseolus vulgaris)로부터 ${\alpha}-Amylase$ 저해제의 분리.정제)

  • White kidney bean, Phaseolus vulgaris, contains proteinaceous inhibitors of ${\alpha}-amylase$. Two inhibitors have been purified by conventional protein fractionation methods such as ethanol precipitation, ammonium sulfate fractionation, DEAE-Sephadex ion exchange chromatography and Sephadex G-100 gel chromatography. The inhibitors were purified as I-1 and I-2 based on their elution order from the DEAE-Sephadex column. The overall purification ratio were about 15.0 and 14.8 for I-1 and I-2, respectively. The molecular weights of purified ${\alpha}-amylase$ inhibitors were 50,000 and 45,000 determined by Sephadex G-100 gel filtration and SDS-polyacrylamide gel electrophoresis. They contain $17.6{\sim}17%$ of carbohydrate, $70{\sim}80%$ of protein. The carbohydrates were composed of glucose : xylose : mannose : N-acetylglucosamine (5 : 3 : 50 : 42).

Inhibitory Effects of Angiotensin Converting Enzyme and α-Glucosidase, and Alcohol Metabolizing Activity of Fermented Omija (Schizandra chinensis Baillon) Beverage (오미자 발효음료의 알코올 분해능과 Angiotensin Converting Enzyme 및 α-Glucosidase 저해효과)

  • The nutraceutical role of fermented omija (Schizandra chinensis) beverage (FOB) was determined through the analysis of fibrinolytic and alcohol metabolizing activities, nitrite scavenging activity, and angiotensin converting enzyme and $\alpha$-glucosidase inhibitory effects. Firstly, FOB increased fibrinolytic activity in a dose-dependent manner and indicated angiotensin converting enzyme inhibitory activity of 94.8% at 20% FOB (0.6 mg/mL). In addition, the inhibitory activities of FOB on $\alpha$-amylase and $\alpha$-glucosidase were determined to be 100% at 100% FOB (3 mg/mL) and 49% at 60% FOB (1.8 mg/mL), respectively. Nitrite scavenging activity of FOB was about 96.1%, 72.3%, and 68.3% on pH 1.2, 3.0, and 6.0 at 100% FOB, respectively. To determine influence of FOB on alcohol metabolism, the generating activities of reduced-nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) were measured. Facilitating rate of ADH activity was 70.3% at 50% FOB, but ALDH activity was not affected. These results revealed that FOB has a strong alcohol metabolizing activity, and fibrinolytic and nitrite scavenging activities and exhibits the angiotensin converting enzyme, $\alpha$-amylase, and $\alpha$-glucosidase inhibitory activities.

  • These experiments were conducted to investigate the enzymatic characteristics of the purified $\alpha$-amylase (F-2A) of Bacillus circulans F-2 and the digestion rate of various starches. 1. The molecular weight was estimated to be 93000 by SDS-polyacrylamide disc gel electrophoresis. The isoelectric point was about pH 5.0. The optimum pH for the enzyme action was 6.0-6.5 and the stable pH ranged pH 5.5-12.0. The optimum temperature was 6$0^{\circ}C$, and the purified $\alpha$-amylase was stable below 4$0^{\circ}C$. 2. The purified $\alpha$-amylase was activated by Mn$^{++}$ and Co$^{++}$, whereas it was inhibited by Ag$^{+}$, HT$^{++}$, Cu$^{++}$ and Pb$^{++}$. 3. The purified $\alpha$-amylase is considered to have no sulfhydryl residue essential for its catalytic activity. 4. Michaelis constant (Km) was 1.704 mg/$m\ell$. Activation energy between 25-4$0^{\circ}C$ was 12.297 Kcal/mole, and between 40-6$0^{\circ}C$, it was 7.831 Kcal/mole. 5. The hydrolysis product from soluble starch, amylose and amylopectin in the early stage of hydrolysis was G$_{6}$, and as hydrolysis proceeds, G$_4$and G$_2$appeared. 6. Products from each oligosaccarides are as follows: G$_4$longrightarrow G$_2$+ G$_2$,G$_3$ +G$_1$,G$_{5}$longrightarrow G$_4$+G$_1$,G$_{6}$longrightarrowG$_4$+ G$_2$,G$_{7}$ G$_4$,G$_{8}$longrightarrow G$_4$+G$_4$, 7. On raw potato starch, raw sago starch and raw yam starch, the purified enzyme exhibited a remarkably high digestion rate than Porcine pancreatic amylase and Streptococcus bovis amylase.

Antioxidant and anti-diabetic effects of Ixeris strigosa extract (선씀바귀 추출물의 항산화 및 항당뇨 효과)

  • Purpose: Ixeris strigosa (IS) is a perennial plant that commonly grows in meadows. The leaves and roots of IS have been used in medicine as a sedative. This study evaluated the antioxidant and carbohydrate-digestive-enzyme inhibitory effects of IS to determine its potential as an essential antioxidant and glycemic inhibitor for type 2 diabetics. Methods: The antioxidative and α-amylase and α-glucosidase inhibitory activities were examined using the water extracts (ISW), ethanol extracts (ISE), and solvent fractions from IS. The antioxidative activities were measured using in vitro methods by measuring the 1,1-diphenyl-2-picrylhydrazyl and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid radical scavenging activity. Results: Investigations of the total polyphenol, flavonoid content, in vitro antioxidant activity, and α-amylase and α-glucosidase inhibitory activities of the IS extract showed that the ISE had higher total phenolic and flavonoid contents than the ISW, as well as high antioxidant activity. The ethanolic extracts of IS (70%) had an α-amylase inhibitory activity of 78.55%. The ethyl acetate fraction (90.56%) showed higher α-glucosidase inhibitory activity than the positive control, acarbose (83.01%). Conclusion: Among the ISE fractions, the ethyl acetate and butanol fractions showed the best digestive enzyme inhibitory activity. Moreover, the antioxidant activity of the extract and the carbohydrate, α-amylase, and α-glucosidase inhibitory effects showed a stronger correlation with the total phenol and flavonoid contents compared to the ISW. As a result, the antioxidant and digestive enzyme inhibitory activities of high ISE are due to the phenolic compounds, particularly the flavonoid compounds. Therefore, ethyl acetate and butanol fractions of the 70% ethanol extract are excellent anti-diabetic functional materials.

Biological Activity of Omija (Schizandra chinensis Baillon) Extracts (오미자(Schizandra chinensis Baillon) 추출물의 생리활성)

  • Extracts from Schizandra chinensis Baillon (Korean name: Omija) were tested for antioxidant and their inhibitory activities of ${\alpha}-amylase$ and ${\alpha}-glucosidase$. Total contents of phenolics were found as 4.35 mg/g (water extract)${\sim}$6.35 mg/g (60% ethanol extract). Electron donating ability (EDA), ABTS [2,2'-azinobis(3-ethyl-benzothiaznoline-6-sulfonic acid)] radical decolorization, antioxidant protection factor (PF) and thiobarbituric acid reactive substance (TBARS) were measured for the antioxidative activity of the extracts from S. chinensis. The water extract were determined as 97.5% at 200 ${\mu}g/ml$ while the activity of 60% ethanol extract were 96.2% at 200 ${\mu}g/ml$ in EDA. The 60% ethanol extract showed higher antioxidant activity than water extract when evaluated by ABTS radical decolorization, antioxidant PF and TBARS. ${\alpha}-Amylase$ inhibitory activity of water extract was similar with that of 60% EtOH extract. ${\alpha}-glucosidase$ inhibitory activities of water extract (97.4%) was higher than that of 60% ethanol extract (84.5%) at 200 ${\mu}g$/ml. The water extract from S. chinensis did not show an antimicrobial activity against Helicobacter pylori, but the 60% ethanol extract showed high antimicrobial activities such as 23 ${\pm}$ 1.6 mm of clear zone in 200 ${\mu}g/ml$ of phenolics. The result suggest that the water and 60% ethanol extract from S. chinensis will be useful as natural antioxidants and functional foods.

Screening of Biological Activities of Ethanol Extracts from Several Varieties of Endives (품종별 치콘 추출물의 생리활성 탐색)

  • We investigated the biological activities of ethanol extracts obtained from five endive cultivars. Free radical-scavenging activities of extracts from Redoria, Halifax, Vintor, Kibora, and Metafora cultivars were 94.2%, 60.8%, 66.6%, 70.0% and 67.4%, respectively. The extract from Redoria showed the highest activity among all the tested samples including BHT (Butylated hydroxyanisole) and vitamine C, known as antioxidant. Endive extracts slightly (17.5 - 38.6%) inhibited $\alpha$-glucosidase activity. However, there is no inhibitory activity against $\alpha$-amylase. In terms of proteolytic activity, all endive extracts showed strong activity than pancreatin (used as a positive control), and the extract of Redoria also exhibited stronger activity than other extracts as well as free radical scavenging activity. These results indicate that endive, especially the Redoria cultivar, can be used as a natural resource for development of antioxidants or materials aiding digestion.
