Physico-chemical Changes of Commercial Ssamjang during Storage (공장식 쌈장의 저장기간에 따른 이화학적 성분변화)

  • Physico-chemical properties of ssamjang prepared by industrial process were investigated. Overall experiments were planned by central composite design for five independent variables, kochujang mash aging period $(X_{1})$, doenjang aging period $(X_{2})$, doenjang content $(X_{3})$, sterilization temperature $(X_{4})$, and storage temperature $(X_{5})$. Storage period had no consistent effect on moisture content of ssamjang. Doenjang having longer aging period showed lower moisture content than that having shorter aging period. Titratable acidity and pH of ssamjang gradually increased and decreased with storage period, respectively, with pH of ssamjang significantly affected by aging period of doenjang and kochujang mashes, and sterilization and storage temperatures. Amino nitrogen contents of ssamjang increased during storage and were more affected by sterilization temperature than by aging period and content of doenjang, and storage temperature. Crude protein content of ssamjang irregularly changed during storage, and was slightly affected by content of doenjang.

Overexpression of Thermoalkalophilic Lipase from Bacillus stearothermophilus L1 in Saccharomyces cerevisiae

  • An expression vector system was developed for the secretory production of recombinant Bacillus stearothermophilus L1 lipase in Saccharomyces cerevisiae. The mature L1 lipase gene was fused to ${\alpha}-amylase$ signal sequence from Aspergillus oryzae for the effective secretion into the culture broth and the expression was controlled under GAL10 (the gene coding UDP-galactose epimerase of S. cerevisiae) promoter. S. cerevisiae harboring the resulting plasmid successfully secreted L1 lipase into the culture broth. To examine an optimum condition for L1 lipase expression in the fed-batch culture, L1 lipase expression was induced at three different growth phases (early, mid, and late-exponential growth phases). Maximum product on of L1 lipase (1,254,000 U/l, corresponding to 0.65/1) was found when the culture was induced at an early growth phase. Secreted recombinant L1 lipase was purified only through CM-Sepharose chromatography, and the purified enzyme showed 1,963 U/mg of specific activity and thermoalkalophilic properties similar to those reported for the enzyme expressed in Escherichia coli.

Antipathogenic Activity of Bacillus amyloliquefaciens Isolated from Korean Traditional Rice Wine (막걸리로부터 분리된 Bacillus amyloliquefaciens 균주의 항균 활성)

  • The presence of bacterial strains showing antagonistic activity to common pathogens found in a variety of fermented foods in Korea was explored. A bacterium inhibiting the growth of pathogens such as Aspergillus terreus (KCTC6178), A. flavus (KCTC6984), Staphylococcus aureus (KCCM12214), Escherichia coli O157:H7 (KCCM40406), Bacillus cereus (KCTC1012), Cryptococcus neoformans (ATCC208821), Salmonella typhimurium (ATCC19430), and Listeria monocytogenes (KCTC3569) was isolated from Makgeolli, a Korean traditional rice wine. The strain showing high antipathogenic activity was identified as B. amyloliquefaciens based on the nucleotide sequence of the 16S ribosomal RNA gene. Compared with B. amyloliquefaciens KCTC1660, whose genome has been sequenced, the isolate exhibited significantly low activities of starch-degrading enzymes and high resistance to high temperature and low pH.

Effects of Enzyme Supplementation on Growth, Intestinal Content Viscosity, and Digestive Enzyme Activities in Growing Pigs Fed Rough Rice-based Diet

  • The purpose of the present study was to investigate the effects of exogenous non-starch polysaccharides (NSP) enzymes on performance, intestinal content viscosity and digestive enzyme activities of growing pigs fed a rough rice-based diet. A total of 60 crossbred barrows with an initial body weight of 35.16 kg (SD = 0.82) were blocked by body weight and randomly assigned to two treatments with three replications. Each group was fed the diet based on rice with or without exogenous NSP enzymes (2 g/kg of diet). During the 70 days of the feeding trial, all pigs were given free access to feed and water. At the end of the feeding trial, six pigs from each treatment were randomly selected and slaughtered to collect intestinal digesta, intestinal mucosa, and pancreas. The addition of NSP enzymes improved average daily gain (p<0.05) and feed:gain (p<0.05), and decreased viscosity of digesta in the jejunum (p<0.001) and ileum (p<0.01) of pigs. The supplementation of NSP enzymes increased activities of protease (p<0.01), trypsin (p<0.01) and ${\alpha}$-amylase (p<0.05) in duodenal contents. However, digestive enzymes in the pancreas, jejunal and ileal mucosa were unaffected by the supplemental NSP enzymes (p>0.10). The results indicate that the addition of NSP enzymes to rough rice-based diets improved performance of pigs, reduced viscosity and increased digestive activity in the small intestine.

Dietary Intakes and Status of Folate in Koean Women of Child-bearing Potential (가임기 여성의 엽산 섭취량 및 엽산영양상태)

  • We examined the folate intakes and assessed folate nutritional status of Korean women with childbearing potential. A total of 91 healthy women aged between 15 and 49 participated. They were divided into three groups by their age : A(15-24 yrs), B(25-34 yrs) and C(35-49 yrs). Folate intakes were determined by direct analysis. The foods consumed for 24 hours were collected proportionally and assessed folate. Their blood drawn in fasting state were analyzed folate levels. Folate contents of food homogenate, plasma and erythrocyte were determined a microbiological method using Lactobacillus. casei (ATCC 7469). Prior to the micro-assay, the food homogenate were treated with alpha-amylase, protease and folate conjugase. Mean daily folate intake of the total subjects was 145.8$\mu\textrm{g}$/d and in each group of A, B, and C was 114.0$\mu\textrm{g}$/d, 141.6$\mu\textrm{g}$/d, and 164.6$\mu\textrm{g}$/d, respectively. That of group C was significantly higher than that of group A(p<0.05). However, those of all the groups were lower than compared to the Korean Recommened Dietary Allowances(RDA) for folate. Especially the subjects in the group A consumed folate least that was below the half of the Korean RDA. The mean energy intake of all subjects was 1638㎉/d and those in each group of A, B, and C did not meet the Korean RDA for energy. The energy intake were significantly correlated with folate intakes(r=0.5050, p<0.001). Mean plasma and erythrocyte folate concentrations of total subjects were 6.9ng/mL and 266.3ng/mL, respectively. None were found to be deficient both in plasma(<3ng/mL)and erythrocyte (<140ng/mL) folate levels. There was only one subject who had red blood cell folate level below 157ng/mL concentration. These results show that folate status of the Korean women of reproductive age is not much bad. But it should be better that letting them improve their folate status by increasing energy intake, choosing high folate foods.

Enzymeaticial Analysis and Soluble Proteins Assays on Radioprotective Effects of Cordyceps militaris (번데기 동충하초 추출물의 방사선 방호효과에 대한 효소 활성도 및 수용성 단백질 양상 분석)

  • Effect of single pre-administration of Cordyceps militaris(Cm) extract on the survival ratio, body weight and organ weight changes and blood cell counts after whole-body ${\gamma}-irradiation$ were investigated. The single pre-administration of Cm extract at 24 hrs before ${\gamma}-irradiation$ increased the 40-day survival ratio of irradiated mice from 60.1% to 71.4%. The administration of Cm extract completely prevented weight reductions of spleen and thymus produced by ${\gamma}-irradiation$ (P<0.01, P<0.05). Similar but somewhat less radioprotective effect was also found In the testis of the Cm treated mice. The administration of Cm extract retarded the reduction of both leukocyte and lymphocyte counts occured during the first 7 days and accelerated the recovery of the counts thereafter. The exrtract also acclerated the recovery of the erythrocyte counts occured after the day 21th. SDS-polyacrylamide gel electrophoresis of the soluble proteins extracted from various organs did not reveal differences to any extent in all groups except in the livers of the irradiated and extract treated groups, in which some proteins were missing or less present. Also, the result of general intra and extra mycelial enzyme assays with Cm, extramycelial enzyme activity was relatively higher than the intramycelial enzyme, Cm appeared to indicate that ${\alpha}-amylase$ was the highest among the enzymes and gluosidase and chitinase were followed. Since the spleen, thymus and testis have been well known as radiosensitive organs, the protective action of Cm extract on irradiated mice may be responsible for its enhancing recovery of these organs. Although the exact mechanism in protective effect of Cm extract on irradiated mice is not clear yet, the present study is the first report regarding the Cm which was tested and found to be a potential radioprotective agent.

BIOCHEMICAL CHARACTERISTICS OF SALIVA TO BE LINKED TO THE MULTIPLE CARIES IN CHILDREN (소아의 다발성 치아우식증과 연관된 타액의 생화학적 특성)

  • Saliva is obviously potential medium to protect the dental caries by not only physical clearing effect, but aggregating action of protein with bacteria. Nevertheless, we still do not understand how the dental caries occur and what brings the individual difference in caries prevalence. In the regards of dental caries prevalence, we hypothesized that the composition of salivary protein might be different from caries susceptible group to caries resistant group. The purposes of this experiment were focused on the molecular analysis of salivary proteins from the subjects who were involved in multiple caries. Electrophoretic analysis was done on the whole saliva collected from the children with and without multiple caries. We found 86.2% of subjects with multiple caries has approximately 120 KDa protein band while 30.4% in the healthy subjects. And the concentration of the total protein on the subjects with multiple caries is significantly higher than that of the healthy group. However, it turned out that the difference of the salivary composition does not affect the bacterial adhesion to hydroxyapatite bead. With regards of enzymes in saliva, the activity of ${\alpha}-amylase$ and lactate dehydrogenase does not have any significant difference between both groups. However, the concentrations of $Na^+\;and\;Cl^-$ in saliva from multiple caries group is higher than that of the control group. Taken all together, it may be concluded that 120 KDa protein in saliva may be associated with the process of dental caries, also the high concentration of protein and $Na^+,\;Cl^-$ in saliva may be linked to dental caries development as a cofactors.

Expression of Fungal Phytase on the Cell Surface of Saccharomyces cerevisiae

  • Phytase improves the bioavailability of phytate phosphorus in plant foods to humans and animals, and reduces the phosphorus pollution of animal waste. We have engineered the cell surface of the yeast. Saccharomyces cerevisiae, by anchoring active fungal phytase on its cell wall, in order to apply it as a dietary supplement containing bioconversional functions in animal foods and a whole cell bio-catalyst for the treatment of waste. The phytase gene (phyA) of Aspergillus niger with a signal peptide of rice amylase 1A (Ramy1A) was fused with the gene encoding the C-terminal half (320 amino acid residues from the C-terminus) of yeast ${\alpha}-agglutinin$, a protein which is involved in mating and is covalently anchored to the cell wall. The resulting fusion construct was introduced into S. cerevisiae and expressed under the control of the constitutive glyceraldehydes-3-phosphate dehydrogenase (GPD) promoter. Phytase plate assay revealed that the surface-engineered cell exhibited a catalytically active opaque zone which was restricted to the margin of the colony. Additionally, the phytase activity was detected in the cell fraction, but was not detected in the culture medium when it was grown in liquid. These results indicate that the phytase was successfully anchored to the cell surface of yeast and was displayed as its active form. The amount of recombinant phytase on the surface of yeast cells was estimated to be 16,000 molecules per cell.

  • Wheat is one of important food crop in the world. However, wheat has some negative things that are allergen. Wheat includes four different allergen kwon as alpha-, beta- and gamma gliadin and amylase inhibitor. Recently, people are interesting to gluten free food or low gliadin food. To study about wheat allergies and research trend is important for future research like as wheat breeding and allergy reduction technology. In this study, we analyzed that the reported on the research field of wheat allergy have been reported until now, and was to provide direction for future research. The analysis of 235 episodes of major papers published from 2007 to 2016. The last 5 years from 2012, wheat allergy appears increasing number of related papers. The ratio of national papers 13% and 60% were published by the United States and other European countries, respectively. The fields of wheat allergen-related preclinical technology and wheat allergen related to genome research for discovery technology were represented high rate by 58% and 26%, respectively. In the case of Korea, significantly account genetic and breeding areas. Recently, however, the research of glutenin protein which is closely linked to wheat allergy is in progress. So, we expected to increase wheat allergy of the research is in the future in Korea.

Purification and Characterization of Complement System Activating Polysaccharide from the Bark of Kalopanax pictus N. (음나무 수피로부터 보체계 활성화 다당의 정제 및 특성)

  • It was observed that the hot-water extract of the bark of Kalopanax pictus N. had the highest anti-complementary activity among the 11 kinds of forest materials. Methanol-and ethanol-soluble portions had low anti-complementary activities, but crude polysaccharide. HKP-0 had a high activity of 80%. HKP-0 contained 54.8% of total sugar and 27.9% of protein. The neutral sugars of HKP-0 consisted of mainly arabinose, galactose and glucose. HKP-4 fraction obtained by cetavlon treatment of HKP-0 showed the highest anti-complementary activity of 90%. The activity was not changed by pronase digestion bu decreased greatly by periodate oxidation. HKP-4 consisted of mainly arabinose and glucose with molar ratio of 1.0 : 22.4, HKP-4-I, an unabsorbed fraction from HKP-4 on DEAE Sepharose CL-6B column showed higher yield and activity than those of absorbed fractions. HKP-4-I was homogeneous, and its molecular weight was about 25,000. HKP-4-I contained 84.0% of neutral sugar and consisted of arabinose and glucose with molar ratio of 1.0 : 11.2. The anti-complementary activity of HKP-4-I was not decreased by the treatment of polymyxin B, and the polysaccharide activated both classical and alternative pathway in complement system. Void volume fraction obtained from HKP-4-I hydrolyzed with ${\alpha}$-amylase on Sephadex G-25 column only had a high anti-complementary activity.
