In this study, enzymes were investigated as an antistaling agent for a Korean rice cake. Thermograms by a DSC demonstrated that the gelatinization-onset temperature of the Korean rice cake was at its lowest temperature of 71.1$^{\circ}C$ with the GP (glucoamylase+pullulanase) treatment, followed by $\beta$-amylase and $\alpha$-amylase. The gelatinization peak temperature of the Korean rice cake with enzyme treatment was relatively lower compared to the control. Furthermore, the Korean rice cake with GP treatment showed the lowest peak temperature. Melting enthalpy of the Korean rice cake increased with the enzyme treatment, with $\beta$-amylase, followed by $\alpha$-amylase and GP. Melting enthalpy of the Korean rice cake with GP treatment was significantly lower compared to the $\beta$- and $\alpha$-amylase treatment. Recrystallinity in the case of GP treatment was also significantly lower than control. The range of Avrami exponent (n) was 0.90 ∼ 1.20 and the time constant of retrogradation (1/k) of the Korean rice cake crystalline decreased in the following order: GP, $\beta$-, $\alpha$ -amylase and control. Textural characteristics of the Korean rice cake with enzyme treatment differed greatly from that of control. The L* values of all the Korean rice cakes made without $\beta$-amylase decreased and the a* values were significantly different at p<0.05. The GP treatment altered the b* value toward blue color, whereas $\beta$-and $\alpha$-amylase changed to the direction to yellow color. In sensory evaluation, the Korean rice cake with enzyme treatment showed higher evaluation compared to control.
A Baczllus circdans KCTC3004 $\alpha$-amylase gene contained in a recombinant plasmid pAL850 was transferred into a new shuttle vector plasmid pALSIlI by ligating linearlzed DNAs of pUC19 and pUB110. B. subtilis RM125 and B. megatenurn ATCC14945 transfonned with pALS111 produced the $\alpha$-amylase substantially Most of the enzyme was produced during the exponential growth period. The maxiinurn activities of the $\alpha$-amylase produced by the Bucillus transformants were compared with that of the B. circulans gene donor strain. The B. subtilis RM125(pALS111) enzyme showed the actlvicy 95 times higher than that of the gene donor cells, followed by the B, nzegaterium ATCC14945(pALSlll) enzyme with activity 34 limes higher than that of the gene donor cells. While E coli secreted about 10% of the produced enzyme, B. subtilis excreted the enzyme inlo the medium wholly and B. megaterirun about 98% ofthe total product. The plasmid pALSI11 was quite stable inB. nzegaterium (92%), inoderately stable in B. subtilis (76%), but was unstable in E. coli (38%). The SDS-PAGE and zymogram of this enzyme produced in E. coli(pALS111), B. subtilis( pALS111) or B. megateril~m (pALS111) indicated a molecular weight of 55,000. The enzymes overproduced in three different host cells hydrolyzed starch to produce mainly maltoaiose and mallooligosaccharides.
Objective: This study investigated the effects of different dietary energy sources on early postmortem muscle metabolism of finishing pigs. Methods: Seventy-two barrow ($Duroc{\times}Landrace{\times}Yorkshire$, DLY) pigs ($65.0{\pm}2.0kg$) were allotted to three iso-energetic and iso-nitrogenous diets: A (44.1% starch, 5.9% crude fat, and 12.6% neutral detergent fibre [NDF]), B (37.6% starch, 9.5% crude fat, and 15.4% NDF) or C (30.9% starch, 14.3% crude fat, and 17.8% NDF). After the duration of 28-day feeding experiment, 24 pigs (eight per treatment) were slaughtered and the M. longissimus lumborum (LL) samples at 45 min postmortem were collected. Results: Compared with diet A, diet C resulted in greater adenosine triphosphate and decreased phosphocreatine (PCr) concentrations, greater activity of creatine kinase and reduced percentage bound activities of hexokinase (HK), and pyruvate kinase (PK) in LL muscles (p<0.05). Moreover, diet C decreased the phosphor-AKT level and increased the hydroxy-hypoxia-inducible $factor-1{\alpha}$ ($HIF-1{\alpha}$) level, as well as decreased the bound protein expressions of HK II, PKM2, and lactate dehydrogenase A (p<0.05). Conclusion: Diet C with the lowest level of starch and the highest levels of fat and NDF could enhance the PCr utilization and attenuate glycolysis early postmortem in LL muscle of finishing pigs.
Rice seeds of 4 cultivars including Whachung and Nampung, of the non-waxy rice cultivars, and Shinsunchalbyeo and Whachungchabyeo, of the waxy rice cultivars, were germinated at $27^{\circ}C$ for 3 days to compare the changes in some physico-chemical properties of the starch granules and the starch-hydrolysing enzyme activities during germination, respectively. With the starch granules, the amount of long glucose chains from amylose molecules were reduced in the non-waxy rices, while the chain length increased in the waxy rices. In the distribution profile of the glucose chain length from amylose molecules, we could observed that the chain length with DP (degree of polymerization) ranged 33 to 66 increased with the decreasing rate of the chain length with that above 130, regardless of the waxiness of rices. In addition, we observed that in contrast to a increase in chain length with DP ranged 14 to 33, the amount of short chains from A chain fraction decreased. Germination induced slight reduction in the polymerization rate of starch granules, and decrease in both initiation and termination temperatures for the gelatinization. ${\alpha}-Amylase$ activity of rices germinated for 3 days found to he higher than that of malt. Especially, the activity of Shinsunchalbyeo was revealed to be highest, about two fold higher than that of malt. In contrast, ${\beta}-amylase$ of the waxy rice found to be considerably less active than malt, although the waxy showed prevalent activity as compared to the non-waxy rices.
The strain BY-445 which produces new inhihitors of ${\alpha}$-amylase was isolated from a soil sample and identified. The aerial hyphae of this strain develope in the from of open spirals. The spore chain of BY-445 strain appears in spiral shape with spiny surface. Melanoid and soulble pigments were not observed. Gelatin was liquefied, and skin milk and starch was also hydrolyzed. The isolate contained LL-diaminopimelic acid in its cell wall hydrolysate. The content of fatty acid 16:iso, 15:0 anteiso and 16:0 was 25:30, 16.19 and 13.16%, respectively. BY-445 strain was closely related to Streptomyces violaceusinger but it was different from this strain in some cultural and physiological characteristics. This strain was, therefore, designated as Streptomyces sp. BY 445.
A material with the same high performance liquid chromatography (HPLC) retention profile as authentic ascorbic acid 2-Ο-$\alpha$-glucoside (AA-2G) was detected in kimchi. This material was identified as AA-2G by testing its susceptibility to $\alpha$-glucosidase hydrolysis, the HPLC profile, and through the elementary analysis. Among several strains of bacteria isolated from fermented kimchi, four strains could produce cydodextrin glucanotransferase (CGTase) which catalyzes the transglucosylation reaction of ascorbic acid. By using starch as the glycosyl donor, AA-2G was produced as the major product through this reaction.

This study was carried out to produce the oligosaccharides(G2~G7) directly from the culture broth of Bacillus cereous IAM1072. The maximal production of oligosaccharides was obtained after cultivation at $30^{\circ}C$ for 24hrs in the mixture of 12% soluble starch, 1.5% peptone and 0.25% $K_2HPO_4$ adjusted to pH 7.0. Among the oligosaccharides produced $G_5$ oligosaccharide was purified. After methylation, the result of GC analysis was suggested to be oligosaccharide consisting of 5 glucose unit with ${\alpha}-1,4-glucosidic$ bond.
The objective of this experiment was to study the influence of feed moisture content on the degree of enzymatic hydrolysis of com starch in a twin screw extruder and the saccharification yield of the dried extrudate. The feed moisture content was set at 25, 30, and 35% and ${\alpha}$-amylase solution was directly injected into the feed section at a barrel temperature of $95^{\circ}C$ and screw speed of 250 rpm. Amyloglucosidase was used for the saccharification of the dried extrudate at a concentration of 0.055%(w/w). Expansion ratio and swelling factor of extrudates decreased with increasing the feed moisture content. Addition of ${\alpha}$-amylase during extrusion process raised reducing sugar content of extrudates which also increased with the feed moisture content. The saccharification yield of dried extrudate was higher for the extrudate with lower feed moisture content.
Apparent digestibility coefficients (ADCs) of dry matter, crude protein, crude lipid, nitrogen-free extract, and energy in selected carbohydrate sources including wheat flour (WF), ${\alpha}-potato$ starch (PS), ${\alpha}-corn$ starch (CS), Na alginate (AL), dextrin (DEX), and carboxymethyl cellulose (CMC) were determined for olive flounder. The olive flounder averaging $150{\pm}8.0g$ were held in 300-L tanks at a density of 30 fish per tank. Chromic oxide was used as the inert marker. Feces were collected from the flounder by a fecal collector attached to a fish rearing tank. Apparent dry matter and energy digestibilities of flounder fed WF, PS, CS, and DEX diets were significantly higher than those of fish fed AL and CMC diets. Apparent crude protein digestibility coefficients of flounder fed PS and CS diets were significantly higher than those of fish fed AL, DEX, and CMC diets. Apparent crude lipid and nitrogen-free extract digestibility coefficients of flounder fed PS and DEX diets were significantly higher than those of fish fed WF, CS, AL, and CMC diets. The present findings indicate that PS and DEX could be effectively used as dietary carbohydrate energy compared to WF, CS, AL, and CMC for olive flounder.
A factorial feeding trial of different levels of three carbohydrate sources (wheat flour, $\alpha$-potato starch, and sodium alginate) was conducted to determine proper sources and levels of dietary carbohydrate for juvenile sea cucumbers. Three replicate groups of juvenile sea cucumbers (770 mg average weight) were fed the experimental diets once a day for 10 weeks. After feeding trial, survival of sea cucumbers was not significantly different between diets. The body weight of sea cucumbers was significantly (P<0.01) affected by both source and level of dietary carbohydrate, tending to decrease as dietary carbohydrate level increased. Body weight of sea cucumbers fed a 25% wheat flour diet was the highest but was not significantly different from that of sea cucumbers fed diet containing 10% wheat flour with 15% sodium alginate (P>0.05). No significant differences were observed in contents of moisture, crude protein, crude lipid and ash of sea cucumbers among dietary treatments. These results suggest that sea cucumbers utilize wheat flour and sodium alginate more efficiently than they do $\alpha$-potato starch, and that a formulated diet containing 43-60% carbohydrate may be suitable for juvenile sea cucumber culture.
