열처리로 야기되는 우유 유청 단백질의 반응 (Heat-Induced Reaction of Bovine Whey Proteins)

  • $\alpha$-La(holo-, apo form)과 $\alpha$-La과 $\beta$-Lg의 혼합물의 열 처리 시 변화의 정도를 DSC를 실시하여 비교하였다. Holo-$\alpha$-La의 열 변성 정점 온도(Td)는 apo-$\alpha$-La에 비해 높게 나타남으로써 열에 의한 변성에 더 안정함을 알 수 있었다. 두 종류 $\alpha$-La의 혼합물의 경우 Td는 일치하지 않았으며 ago-$\alpha$-La이 holo-$\alpha$-La보다 더 낮게 나타났는데 이는 apo-$\alpha$-La이열에 더 불안정함을 의미한다. $\alpha$-La과 $\beta$-Lg의 혼합물의 경우 hole-$\alpha$-La과 $\beta$-Lg의 혼합물의 변성 온도는 holo-$\alpha$-La 하나만을 열처리 했을 때보다 변성 온도가 더 낮아져서, 열에 의한 변성이 $\beta$-Lg의 유리 SH기에 의해 촉진되었음을 유추할 수 있었고, ago-$\alpha$-La과 $\beta$-Lg의 혼합물의 경우 변성 온도는 apo-$\alpha$-La하나만을 열처리 했을 때보다 변성 온도가 2배 가까이 상승하면서, 열 처리에 더 안정해짐을 보여주었다. $\beta$-Lg의 열 변성 양상은 pH 6.8의 조건에서 살펴보았을 때 가열 초기 단계부터 다소불안정한 peak를 보였으며 $\alpha$-La과 혼합되었을 때 변성을 더 촉진시키면서 열에 불안정해짐을 시사하였다.

$\alpha$-Lactalbumin의 암세포 증식 저해효과에 관한 연구 (Antitumor Activity of $\alpha$-Lactalbumin on the Tumor Cells)

  • 상업적으로 생산 판매되고 있는 여러 가지 유청 단백질을 이용하여 각종 암세포에 대한 세포독성을 조사한 결과, IgG, BSA, $\beta$-LG는 세포독성이 거의 없었으나, $\alpha$-LA가 강한 세포독성을 갖고 있음이 확인되었다. MKN45, HeLa, WiDr, 그리고 A498의 경우, $\alpha$-LA(1mg/ml)에 의해 세포성장이 95% 이상 억제되는 것으로 나타났다. HPLC를 이용하여 $\alpha$-LA에 혼입되어 있는 미량성분을 제거한 후, 세포독성을 조사한 결과, $\alpha$-LA이 나타내었던 세포독성이 대부분 없어지는 것으로 나타났다. 또한 $\alpha$-LA을 trypsin으로 부분 가수분해한 후, 각종 암세포에 대한 성장 억제효과를 검토한 결과, 암세포에 대한 독성을 상실하는 것으로 나타났으며, $\alpha$-LA를 EDTA로 처리할 경우, $\alpha$-LA의 세포독성이 대부분 상실되는 것으로 나타났다.

Electrophoretic Behaviors of α-Lactalbumin and β-Lactoglobulin Mixtures Caused by Heat Treatment

  • In order to study the reaction behaviors of bovine $\alpha$-lactalbumin ($\alpha$-La), $\beta$-lactoglobulin ($\beta$-Lg), and their mixtures during heat treatment, samples were analyzed using native-polyacrylamide gel electrophoresis (Native-PAGE), sodium dodecylsulfate (SDS)-PAGE, and two-dimensional (2-D)-PAGE. The electrophoresis demonstrated that the loss of native-$\alpha$-La increased as temperature increased, and that the loss of apo-$\alpha$-La was slightly higher than that of holo-$\alpha$-La. The tests also showed that during heat treatment, a mixture of $\alpha$-La and $\beta$-Lg was less stable than $\alpha$-La alone. As such, it was assumed that $\beta$-Lg induced holo-$\alpha$-La to be less stable than apo-$\alpha$-La during heat treatment. The reaction behavior of $\alpha$-La (holo-, apo-form) during heat treatment showed similar patterns in the 2-D-PAGE electropherogram, but the mixture of $\alpha$-La and $\beta$-Lg created new bands. In particular, the results showed a greater loss of native $\alpha$-La in the holo-$\alpha$-La and $\beta$-Lg mixture than in the apo-$\alpha$-La and $\beta$-Lg mixture. Thus, it can be concluded that the holo-$\alpha$-La and $\beta$-Lg mixture was more intensively affected by heat treatment than other samples, and that free sulphydryl groups took part in the heat-induced denaturation.

올레산과 미생물 유래 트랜스글루타미나제를 첨가한 우유 알파-락트알부민의 암세포사멸 효과 (Effects of Bovine α-Lactalbumin Added with Oleic Acid and Microbial Transglutaminase on Cancer Cell Apoptosis)

  • 본 연구에서는 기능적 특성이 우수한 우유 단백질 소재를 개발하여 식품산업에 응용하고자 우유 ${\alpha}$-La의 영양생리학적 특성에 대하여 분석하였다. 우유 ${\alpha}$-La의 기능성을 향상 시키려는 시도로써 MTGase의 첨가가 이 단백질의 이화학적 및 면역학적 특성에 미치는 영향을 조사하고, 수종의 암세포주의 생육에 대한 억제 효과를 탐색하였으며, ${\alpha}$-La의 구조적 상태에 따라 항암활성의 관련성을 검토하였다. ${\alpha}$-La에 반응한 MTGase에 의해 생성된 중합체는 인체 내 가수분해효소에 의해 분해가 용이한 것으로 확인되었다. 위암 세포주에 ${\alpha}$-La을 10 mg/mL 농도로 처리한 경우 높은 세포독성을 나타냈다. ${\alpha}$-La과 올레산의 복합체인 BAMLET은 간암 세포주에서 ${\alpha}$-La에 비해 더 강한 활성을 보였다. BAMLET은 apo 상태로 상당량의 이차구조를 지니고 있지만, 소수성 중심부는 느슨하게 형성된 MG와 같은 형태를 지니고 있을 것으로 추정되었다. NMR 분석 결과, BAMLET은 1.2 ppm과 0.8 ppm에서 지방산 보조인자인 올레산과 상호 작용하는 것으로 분석되었다.

우우 유청으로부터 분리한 $\alpha$-락트알부민의 기능적 특성 (Functional Properties of $\alpha$-Lactalbumin Separated from Bovine Whey)

  • This study was performed to obtain a large quantity of $\alpha$-lactalbumin ($\alpha$-LA) from milk by an improved separation and purification method. Functional properties-solubility viscosity, emulsifying activity, foamability surface hydrophobicity and gelation-of the purified $\alpha$-LA were investigated, $\alpha$-LA was purified in a large quantity by DEAE-Sephacel chromatography using 0.15M NaCl in 20mM Tris-HCl buffer(pH 7.2), as an eluent. The yield and purity of the purified $\alpha$-LA were 23.6%, 92.5%, respect-ively. The solubility viscosity and emulsifying activity of the purified $\alpha$-LA were 92.2$\pm$2% 3.46$\pm$0.19 cP and 345$\pm$5.0m^2$-/g respecively. The foamability of $\alpha$ -LA was 762 after 5min whipping which was lower than that of WPC and showed decreasing tendency with whipping time. The relative surface hydrophobicity of the $\alpha$-LA was formed when a 10% $\alpha$-LA solution containing 100mM NaCl and 20 mM $CaCl_2$ was heated at 92$^{\circ}C$for 40min. The $\alpha$-LA gel showed 31.5 as hardness and showed low springiness and cohesiveness.

  • In order to study the reaction behavior of bovine holo- and apo-$\alpha$-lactalbumin ($\alpha$-La) during heat treatment at 65~10$0^{\circ}C$, the samples were analysed by first (ID)-and second-dimensional (2D) native-polyacrylamide gel electrophoresis (Native-PAGE) and sodium dodecylsulfate (SDS)-PAGE. When bolo-$\alpha$-La or apo- $\alpha$ -La were heated, they formed non-native, monomers, dimers and trimers. The apo-$\alpha$-La was more heat-sensitive than holo-$\alpha$-La. The monomers seemed to have the same composition as the native $\alpha$-La, but many of the disulfide bonds could be non-native.

Induction of ER Stress-Mediated Apoptosis by ${\alpha}$-Lipoic Acid in A549 Cell Lines

  • Background: ${\alpha}$-Lipoic acid (${\alpha}$-LA) has been studied as an anticancer agent as well as a therapeutic agent for diabetes and obesity. We performed this study to evaluate the anticancer effects and mechanisms of ${\alpha}$-LA in a lung cancer cell line, A549. Materials and Methods: ${\alpha}$-LA-induced apoptosis of A549 cells was detected by fluorescence-activated cell sorting analysis and a DNA fragmentation assay. Expression of apoptosis-related genes was analyzed by western blot and reverse transcription.polymerase chain reaction analyses. Results: ${\alpha}$-LA induced apoptosis and DNA fragmentation in A549 cells in a dose- and time-dependent manner. ${\alpha}$-LA increased caspase activity and the degradation of poly (ADP-ribose) polymerase. It induced expression of endoplasmic reticulum (ER) stress-related genes, such as glucose-regulated protein 78, C/EBP-homologous protein, and the short form of X-box binding protein-1, and decreased expression of the anti-apoptotic protein, X-linked inhibitor of apoptosis protein. Reactive oxygen species (ROS) production was induced by ${\alpha}$-LA, and the antioxidant N-acetyl-L-cysteine decreased the ${\alpha}$-LA-induced increase in expression of apoptosis and ER stress-related proteins. Conclusion: ${\alpha}$-LA induced ER stress-mediated apoptosis in A549 cells via ROS. ${\alpha}$-LA may therefore be clinically useful for treating lung cancer.

  • Alumina ceramics was reinforced by in-situ formation of La-${\beta}$-aluminate in ${\alpha}$-alumina matrix. The powder mixture of which composition is (100-12x)Al2O3+x(La2O3+11Al2O3) was prepared for the formation of La-${\beta}$-aluminate in ${\alpha}$-alumina matrix. The amount of La-${\beta}$-aluminate in the matrix was controlled by varing x which is number of moles. The dense composite was produced by sintering at 1600$^{\circ}C$ in air or hot-pressing at 1550$^{\circ}C$ under 30 MPa. Bending strength and fracture toughness were increased, resulting from the grain growth inhibition and the crack deflection and crack bridging mechanism when La-${\beta}$-aluminate was produced in ${\alpha}$-alumina matrix.

Alcalase에 의한 유청단백질 가수분해물의 항원성 저감 효과 (Reduction in antigenesity of whey protein by alcalase)

  • The aim of this study was to produce enzymatic hydrolysis of ${\alpha}$-LA, ${\beta}$-LG and BSA with alcalase for the possible application of hypoallergenic foods toward cow's milk allergenic infant. The molecular weights of most of the peptides in hydrolysates from ${\alpha}$-LA, ${\beta}$-LG and BSA by alcalase were below 3,000 dalton. Antigenesity of ${\alpha}$-LA, ${\beta}$-LG and BSA hydrolysates to rabbit anti-${\alpha}$-LA antiserum, ${\beta}$-LG antiserum and BSA antiserum were remarkably decreased by more than $10^{-3}$ at 20% inhibitionrate. Antigenesity of polyvalent antigenic peptide in ${\alpha}$-LA, ${\beta}$-LG and BSA hydrolysates to specific rabbit anti-${\alpha}$-LA antiserum, ${\beta}$-LG antiserum and BSA antiserum was determined by PCS test using guina-pig. Hydrolysates of ${\alpha}$-LA, ${\beta}$-LG and BSA with less than 3,000 dalton did not show polyvalent antigenic reaction against rabbit antiserum. Hydrolysates of ${\alpha}$-LA, ${\beta}$-LG and BSA could be a source for the manufacturing of hypoallergenic food.

  • The concept of interval-valued (${\alpha},{\beta}$)-fuzzy ideals, interval-valued (${\alpha},{\beta}$)-fuzzy generalized bi-ideals are introduced in LA-semigroups, using the ideas of belonging and quasi-coincidence of an interval-valued fuzzy point with an interval-valued fuzzy set and some related properties are investigated. We define the lower and upper parts of interval-valued fuzzy subsets of an LA-semigroup. Regular LA-semigroups are characterized by the properties of the lower part of interval-valued (${\in},{\in}{\vee}q$)-fuzzy left ideals, interval-valued (${\in},{\in}{\vee}q$)-fuzzy quasi-ideals and interval-valued (${\in},{\in}{\vee}q$)-fuzzy generalized bi-ideals.
