해양 유래 Pseudomonas aeruginosa BYK-2(KCTC 18012P)가 생산하는 Biosurfactant의 구조분석

  • The Pseudomonas aeruginosa BYK-2(KCTC 18012p) produced three kinds of glycolipids on olive oil as a substrate and purified two types of major glycolipids(Rf=0.48, BS-1; Rf=0.65, BS-2) using silica gel chromatography, TLC, HPLC, etc. From the analysis of the chemical structure, the glycolipid of BS-1 was estimated as rhamnolipid($2-O-{\alpha}-L-rhamnopyranosyl- {\alpha}-L-rhamnopyranosyl-{\beta}-hydroxyldecanoyl-{\beta}-hydroxydecanoic$ acid; M.W. 650) and BS-2 was detected as rhamnolipid methyl ester($2-O-{\alpha}-L-rhamnopyranosyl-{\alpha}-L-rhamnopyranosyl-{\beta}-hydroxyldecanoyl-{\beta}-hydroxydecanoic$ acid methyl ester; M.W. 664) by FT-IR, FAB Mass spectrometry, $^1H-NMR$, $^{13}C$ FT-NMR, DEPT, 2D-NMR (TOCSY, RELAY, NOESY, HSQC, HMBC). In particular, It was found that a marine bacterium Pseudomonas aeruginosa BYK-2(KCTC 18012P) remarkably produced rhamnolipid and rhamnolipid methyl ester simultaneously.

Synthesis of Novel D-Glucose-derived Benzyl and Alkyl 1,2,3-Triazoles as Potential Antifungal and Antibacterial Agents

  • A series of novel glucose derived benzyl and alkyl 1,2,3-triazoles and their hydrochlorides have been synthesized via Cu(I)-catalyzed 1,3-dipolar cycloaddition. All the new compounds were characterized by MS, IR and NMR spectra. The DEPT, APT, $^1H$-$^1H$ and $^1H-^{13}C$ 2D NMR spectra for some compounds were also recorded. These compounds were evaluated for their in vitro antibacterial activities against Staphylococcus aureus ATCC 29213, Bacillus subtilis, Bacillus proteus, Pseudomonas aeruginosa, Escherichia coli ATCC 25922, and antifungal activities against Candida albicans and Aspergillus fumigatus. The bioactive data revealed that (3R,4S,5S,6S)-2-(hydroxymethyl)-6-methoxy-4,5-bis((1-octyl-1H-1,2,3-triazol-4-yl)methoxy)-tetrahydro-2H-pyran-3-ol 8a exhibited excellent antifungal activity against A. fumigatus with an MIC value of 0.055 mM compared to Fluconazole. It also showed broad inhibitory efficacy against tested bacterial strains with MIC values ranging from 0.049 mM to 0.39 mM.

Terpenoids from Citrus unshiu Peels and Their Effects on NO Production

  • Two new compounds, 3-methyl-but-2-en-1-yl-1-O-β-xylopyranosyl-(1"→2')-O-β-glucopyranoside (1) and 1-O-β-glucopyranosyl-6-hydroxy-2-methyl-hep-2-enoic acid (2), along with sixteen known terpenoids were isolated from the peels of Citrus unshiu Markov. Their structures were elucidated based on extensive NMR analyses (1H NMR, 13C NMR, DEPT, COSY, HMQC, and HMBC) and high-resolution mass spectrometry. In addition, all isolates (1 - 18) were tested their effects on nitric oxide (NO) production in RAW264.7 cells. Limonin (15) showed to inhibit LPS-induced NO production in a concentration-dependent manner without cytotoxicity.

  • Radical copolymerization behavior of 4'-vinylbenzo-15-crown-5, a vinyl monomer having a pendant 15-membared crown ether unit (VCE) with di(ethylene glycol) ethyl ether acrylate (DEGEEA) was carried out in toluene solution using 2,2-azobisisobutyronitrile (AIBN) as an initiator. The copolymers were characterized by means of FT-IR, $^1{H-NMR}$, and $^{13}{C-NMR}$ . The reactivity ratio of VCE and DEGEEA, determined by Fineman-Ross and Kelen-Tudos method, gave values 0.55 for VCE, and 0.11 for DEGEEA respectively.

  • Dimethylamine complexes of palladium(Ⅱ) [(PCP)Pd(NHMe2)](OTf) (PCP = 2,6-(R2PCH2)2C6H3); R = Ph (1), R = Cy (2)), have been prepared from the reaction of (PCP)Pd(OTf) and dimethylamine. The complex 1 is stable both in solution and in the solid state, while 2 is stable only in solution in the presence of dimethylamine although the formation of 2 in solution is quantitative by NMR Spectroscopy. A solution NMR spectroscopic study shows that dimethylamine favors over carbon monoxide in the coordination sphere of palladium(Ⅱ) having rigid terdentate ligands. The complexes 1 and 2 in chlorinated solvents undergo a chlorine abstract reaction yielding Pd(2,6-(R2PCH2)2C6H3)Cl in the presence of a base such as dimethylamine and DABCO (diazabicyclooctane), in which a transient dimethylamido palladium(Ⅱ) species likely involves.

Quantitative Analysis of Reaction Products from Glucose and Xylose in Acidic Aqueous Medium by 1H-NMR Spectroscopic Method (산성 수용액 조건에서 포도당과 자일로스 반응 산물의 1H-NMR 분광분석을 이용한 정량 분석)

  • Reaction of glucose and xylose to secondary hydrolysis of concentrated acid hydrolysis was quantitatively analyzed by $^1H$-NMR spectroscopic method. Anomeric hydrogen, furan and formic acid peaks were selected for quantitative analysis. The glucose was converted to the formic acid and the levulinic acid via the 5-hydroxymethylfurfural (HMF) but the xylose was converted to the fufural, which further degraded to the formic acid. The conversion to furans was slower for the glucose than the xylose. But the 5-HMF formed from the glucose was unstable in acidic aqueous medium, resulted in fast conversion to the levulinic acid and the formic acid. The furfural was relatively stable than 5-HMF at acidic aqueous medium.

Kinetic Study of Xylan Hydrolysis and Decomposition in Concentrated Sulfuric Acid Hydrolysis Process by $^1H$-NMR Spectroscopy ($^1H$-NMR에 의한 Xylan의 황산가수분해 과정에서 나타나는 반응 동력학 연구)

  • Proton-NMR spectroscopic method was applied to kinetic study of concentrated sulfuric acid hydrolysis reaction, especially focused on 2nd step of acid hydrolysis with deferent reaction time and temperature as main variables. Commercial xylan extracted from beech wood was used as model compound. In concentrated acid hydrolysis, xylan was converted to xylose, which is unstable in 2nd hydrolysis condition, which decomposed to furfural or other reaction products. Without neutralization steps, proton-NMR spectroscopic analysis method was valid for analysis of not only monosaccharide (xylose) but also other reaction products (furfural and formic acid) in acid hydrolyzates from concentrated acid hydrolysis of xylan, which was the main advantages of this analytical method. Higher temperature and longer reaction time at 2nd step acid hydrolysis led to less xylose concentration in xylan acid hydrolyzate, especially at $120^{\circ}C$ and 120 min, which meant hydrolyzed xylose was converted to furfural or other reaction products. Loss of xylose was not match with furfural formation, which meant part of furfural was degraded to other undetected compounds. Formation of formic acid was unexpected from acidic dehydration of pentose, which might come from the glucuronic acid at the side chain of xylan.

Spirostane-type steroidal saponin from Allium hookeri roots with mushroom tyrosinase inhibitory activity

  • Allium hookeri (Liliaceae) has been received the increasing attention as a bioactive resource due to its potent biological activities including anti-oxidant, anti-obesity, anti-microbial and lipid-regulating activities. The beneficial effects of A. hookeri are known contributed from the high content of organosulfur compounds in A. hookeri. Though a variety of articles demonstrated that A. hookeri contains 'saponin' as a bioactive constituent, the scientific evidence to prove it was limited. In the present study, we have attempted to identify saponin contained in A. hookeri through chromatographic isolation and NMR spectroscopic methods. As a result, a spirostane-type steroidal saponin (1) has been successfully isolated from the methanolic extract of A. hookeri roots. The structure of 1 was elucidated by extensive 1D and 2D spectroscopic methods including 1H-NMR, 13C-NMR, 1H-1H COSY, HSQC, HMBC and NOESY; identified as (3β, 22R, 25S)-spirost-5-en-3yl O-6-deoxy-α-L-mannopyranosyl-(1→4)-O-6-deoxy-α-L-mannopyranosyl-(1→4)-O-[6-deoxy-α-L-mannopyranosyl-(1→2)]-β-D-gluco pyranoside. 1 showed the significant inhibitory activity on mushroom tyrosinase with IC50 values of 248.7 μM while the inhibition on alpha-glucosidase was not significant.

Chemical Structure Analysis of Non-ionic Monomer Contrast Agents Using 1H-NMR Spectroscopy (1H-NMR Spectroscopy를 이용한 Non-ionic Monomer 조영제의 화학적 구조 분석)

  • Studies on the side effects of contrast agents are being discussed based on various cases, but studies analyzing the chemical structure of the underlying contrast agents are difficult to understand as the manufacturers have not disclosed them. Therefore, in this study, the chemical structure of the contrast medium was analyzed using 1H-NMR spectrometer for Omnipaque contrast medium prepared from Iohexol, which is a nonionic iodide contrast medium, Xenetix contrast medium from Iobitridol, and Iomeron contrast medium from Iomeprol. As a result, it was found that the Omnipaque contrast medium of Iohexol had 6 carboxyl groups, 3 carbonyl groups, 4 amine groups, 1 methyl group, and 2 cyano groups. It was found that the Xenetix contrast medium of the iobitridol formulation had 6 carboxyl groups, 3 carbonyl groups, 2 amine groups, and 4 cyano groups. It was found that the Iomeron contrast agent of the Iomeprol formulation had 5 carboxyl groups, 3 carbonyl groups, 4 amine groups, 1 methyl group, and 2 cyano groups. As shown in this study, the chemical structure of the non-ionic monomer contrast agent increases its affinity with water by binding a number of hydroxyl groups (OH) to the carboxyl group. It is necessary to accurately identify each of these factors and analyze the physical and chemical changes of the contrast medium according to various environmental factors.

Constituents of the Herb of Isodon excisus var. coreanus

  • The studies were carried out to evaluate the constituents in the aerial part of Isodon excisus var. coreanus (Labiatae). From the aqueous fraction of methanol extract, compound I (${\alpha}$-[[3-(3, 4-dihydroxyphenyl)-1-oxo-2-propenyl]oxy]-3,4-dihydroxy-benzenepropanoic acid), compound II (9-methyl-dihydroferulic acid-4-O-.betha.-D-glucopyranosyl $(1{\rightarrow}2)$-${\alpha}$-L- rhamnopyranosyl (1.rarw.4)-.betha.-D-glucopyranoside), compound III (ent-7.alpha., 11${\alpha}$,15.betha.-trihydroxy-kaur-16-en-1-O-.betha.-D-glucopyranoside) and compound IV ($2{\alpha}$,3${\beta}$,$7{\alpha}$,23-tetrahydroxy-olean-12 -en-28-oic acid 28-O-${\beta}$-D-glucopyranoside) were isolated and identified on the basis of their physicochemical and spectroscopic evidences[IR, FAB(-)MS,$^{1}H-NMR,$$^{13}C-NMR,$$ HMQC$$^{1}H-^{1}H $COSY and HMBC (Heteronuclear Multiple Bond Connectivity)]. Especially, New compounds II and III were named Isodonin A and Isodonin B respectively.
