Primary productivity was determined by using $^{13}C$ tracer according to different cell size of phytoplank-ton through in situ incubation experiments in upper stream of the Lake Shihwa. The average concentration of chlorophyll a was 14 ${\mu}g\;L^{-1}$ demonstrating an eutrophic water. The ratio of POC/Chl-a was lower than 30, reflecting that the origin of organic matter might be mainly phytoplankton. The primary productivity was 93.9 mgC m^{-2}\;d^{-1}$ at St. 1, which was about 40-fold lower than the average value of the lake (3,972 mgC m^{-2}\;d^{-1}$) determined by Choi et al. (1997) before opening of gate but it was higher than the average primary productivity (3.98 mgC m^{-2}\;d^{-1}$) reported by KOWACO in 1993 before constructing dam. The fractionated size (20 ${\sim}$ 53 ${\mu}m$) of phytoplankton community account for 51% of total primary productivity, indicating the highest assimilation rate. This study suggest that $^{13}C$ tracer methodology should be applied as a useful approach for the water ecological research in the future.
The in situ incubation experiment was carried out using $^{13}C$ tracer to determine primary productivity and fatty acid production rate in the shallow, eutrophic Shingu reservoir on 4th July. Particulate organic matter (POM) accounted for 76% in suspended particulate matter (SPM), and average concentration of chlorophyll-${\alpha}$ was $89{\mu}g\;L^{-1}$ in the euphotic layer. Total amount of chlorophyll-${\alpha}$ (Chl-${alpha}$), primary productivity and Chl-${alpha}$ specific productivity in euphotic layer were 112 mg Chl-${alpha}m^{-2}$, 3.53 g C $m^{-2}\;d^{-1}$, 32mg C mg Chl-${\alpha}^{-1}\;d^{-1}$, respectively. The fatty acid composition in newly produced organic matter and suspended organic matter didn't show any significant difference, demonstrating that autochthonous organic matter should be a major source of POM pool. In addition, the fatty acids of bacterial origin were increased through extracellular release of newly photosynthesized DOC, and closely coupled with bacterial assimilation. This result suggests that organic carbon should be actively cycled through the microbial loop in Shingu reservoir in summer.
While a fairly large amount of organic matter is produced daily via phytoplankton photosynthesis in Lake Paldang, South Korea, knowledge of the role of algal-derived organic matter (OM) as a refractory OM source is not adequate. To understand the contribution of algal-derived OM to the refractory pool, biodegradation experiment and $KMnO_4$ oxidation experiment were conducted for 60 days using $^{13}C$ and $^{15}N$ labeled natural phytoplankton assemblage. The assemblage was collected from Lake Paldang on May 20, 2010. The photosynthetically produced total organic carbon ($TO^{13}C$), particulate organic carbon ($PO^{13}C$), and particulate nitrogen ($P^{15}N$) remained at 26%, 20%, and 17% of the initial concentrations, respectively, in the form of non-biodegradable organic matter. In addition, 12% and 38% of $PO^{13}C$ remained after $KMnO_4$ treatment on Day 0 and 60, respectively. These results indicate that photosynthetic products could be an important source of refractory organic matter after microbial degradation. Moreover, the microbially transformed algal-derived OM could contribute to the oxidation rate of the chemical oxygen demand.

Myocardial blood low (MBF) in human can be noninvasively quantified using dynamic N-13 ammonia PET and two-compartment tracer kinetic model. In this study, factor analysis was used to extract the "pure" blood-pool time-activity curves (TACs) and to generate actor images. ive human N-13 ammonia PET dynamic studies were obtained. Three actors and their corresponding actor images were extracted rom each study. The accuracy of MBF estimated by the actor analysis (FA/FA MBF) was examined by comparing to the values estimated using the conventional ROI method (ROI/ROI MBF). MBF obtained by the actor analysis linearly correlated with MBF obtained by the ROI method (slope=0.98, r=0.91). Input unctions obtained by the two methods agreed well. In conclusion, MBF can be measured accurately and noninvasively with dynamic N-13 ammonia PET imaging and actor analysis. This method is simple and acurate and can measure MBF without blood sampling, ROI drawing nor spillover correction.
The study was designed to establish a more nearly quantitative method for assessing the marginal leakage of dental restorations. 27 Class V cavities with $45^{\circ}$ bevel joint were prepared and classified into 2 groups. One group was filled with Scotchbond and silux. The other group was filled with glass ionomer cement, Scotchbond and silux. After finishing, all specimens were subjected manually to 100 thermal cycles at $0^{\circ}C$ and $100^{\circ}C$ water-bath. They were soaked in a samarium nitrate solution for 3 hours, irradiated with flux of $6{\times}10^{12}$ neutrons/$cm^2$/sec for 11 hours, cooled for 200 hours, counted with the HPGE detector and the tracer uptake was determined by comparison with a standard of samarium (10 ${\mu}g$). The following results were obtained. 1. Both of the two groups showed a considerable amounts of marginal leakage. 2. The group filled without glass ionomer cement base showed more marginal leakage than the group filled with glass ionomer cement base. 3. Neutron Activation Analysis produced a good quantitative method to measure the marginal leakage and samarium was appropriate to measure the marginal leakage of resin restorations using neutron activation analysis.
Stable isotope tracers were first applied to evaluate the Microcystis cell assimilation efficiency of bivalves, since the past identification method has been limited to tracking the changes of each chl-a, clearity, and nutrient. The filter-feeders (Sinanodonta woodiana and Unio douglasiae) were assessed under the condition of cyanobacteria (Microcystis aeruginosa) blooms through an in mesocosm experiment using $^{13}C$ and $^{15}N$ dual isotope tracers. chl-a concentration in the treatment mesocosm was dramatically decreased after the beginning of the second day, ranging from 116 to $66{\mu}g\;L^{-1}$. In addition, the incorporated $^{13}C$ and $^{15}N$ atom % in the S. woodiana bivalve showed higher values than U. douglasiae bivalves. The results demonstrate that U. douglasiae has less capacity to assimilate toxic cyanobacteria derived from diet. Our results therefore also indicate that S. woodiana can eliminate the toxin more rapidly than U. douglasiae, having a larger detoxification capacity.
The study was designed to establish quantitative method for assessing the marginal leakage of dental restorations. 18 Class V cavities with $45^{\circ}$ bevel joint were prepared and replicas of these teeth were made with polyethylene wax. and classified with three groups. First group was filled with Scotch bond and silux. Second group was filled with glass ionomer cement:scotchbond/silux. Third group was filled with Dentin-Adhesit/Heliosit. After finishing, all specimens were subjected manually to 100 thermal cycles at $0^{\circ}C$ and $100^{\circ}C$ Samarium nitrate solution, irradiated with flux of $6{\times}12^{12}$ neutrons/$cm^2$/sec for 11 hours, woled for 200 hours, counted with the HpGe detector and the tracer uptake was determined by comparison with a standard of samarium ($10{\mu}g$). The following results were obtained. 1) The group filled with glass ionomer cement base showed least marginal leakage. 2) The group filled with Dentin-Adhesit/Heliosit showed less marginal leakage than the group filled with scotchbond/silux.
The established methods in the residue analysis of endosulfan require an extensive sample clean-up prior to quantification by relatively complex equipment. A radioimmunoassay(RIA) provides a simple procedure with theoretically higher sensitivity and specificity necessitating only a minimum of sample clean-up. Endosulfan-specific antibodies were developed in rabbits by using a bovine serum albumin(BSA) conjugate wherein the alcohol form of endosulfan was multiply bound to the protein via succinylation. Produced antibodies showed the high titers to endosulfan-BSA(1 : 32,000). An RIA method was developed in water and carp by using $^{14}C-labeled$ endosulfan as a tracer. The lowest detection amount of endosulfan was 1 ng in the liver, kidneys, gut and water samples, and 3 ng in the whole body sample of carp without any clean-up, corresponding to 0.1 ppb of endosulfan.
The fractional synthesis rates(FSR) were measured with 2l-wk and 3l-wk-old broiler breeder pullets and hens to investigate the effect of sexual maturity on FSR. The FSR were obtained from chicken tissues and blood samples using High-Performance Liquid Chromatography/Mass Spectrometry(HPLC/MS). A L-l-13C, 15N -leucine saline solution was infused by bolus injection as a tracer into broiler breeder pullets in the experiment. A rapid HPLC/MS method was developed to measure the isotopic enrichments of leucine in plasma, tissue samples, and eggs. The enrichments of stable isotope leucine incorporated into protein and the enrichments of the stable isotope free leucine were measured in liver, breast muscle and blood samples. Two sets of experiments were conducted. In experiment one, 2l-wk-old, sexually immature broiler breeder pullets were divided into groups of three and blood samples were collected at 20 or 30 min intervals until 1.5 h from initial injection. The pullets were sacrificed in groups of three at varying time intervals for 7 h after injection. The liver, breast muscle and blood samples were removed for analysis. The FSR were estimated to be 8.7l%/day for liver, 4.06%/day for breast muscle, and 5.08%/day for blood samples in 30 minutes after injection from the enrichment ratios. In experiment two, sexually matured 3l-wk-old broiler breeder hens were assorted into groups of three and blood samples were obtained at 20 or 30 min intervals for 2 h. The FSR for blood samples were determined. The broiler breeder hens were sacrificed in groups of three at various time intervals until 7 h after injection and liver, breast muscle and blood samples were removed for analysis. The FSR were calculated to be 5.96%/day for liver. Eggs were collected from five chickens daily for 10 days after large bolus injection. The average of total enrichments of stable isotope in egg albumin was increased by 0.064% at 4 days after injection and was back to normal in 7 days.
Purpose In a cyclosporine experiment using a robotic liquid handing system has found a deviation of its standard curve and low reproducibility of patients's results. The difference of the test is that methanol is mixed with samples and the extractions are used for the test. Therefore, we assumed that the abnormal test results came from using methanol and conducted this test. In a manual of a robotic liquid handling system mentions that we can choose several setting parameters depending on the viscosity of the liquids being used, the size of the sampling tips and the motor speeds that you elect to use but there's no exact order. This study was undertaken to confirm pipetting ability depending on types of liquids and investigate proper setting parameters for the optimum dispensing ability. Materials and Methods 4types of liquids(water, serum, methanol, PEG 6000(25%)) and $TSH^{125}I$ tracer(515 kBq) are used to confirm pipetting ability. 29 specimens for Cyclosporine test are used to compare results. Prepare 8 plastic tubes for each of the liquids and with multi pipette $400{\mu}l$ of each liquid is dispensed to 8 tubes and $100{\mu}l$ of $TSH^{125}I$ tracer are dispensed to all of the tubes. From the prepared samples, $100{\mu}l$ of liquids are dispensed using a robotic liquid handing system, counted and calculated its CV(%) depending on types of liquids. And then by adjusting several setting parameters(air gap, dispense time, delay time) the change of the CV(%)are calcutated and finds optimum setting parameters. 29 specimens are tested with 3 methods. The first(A) is manual method and the second(B) is used robotic liquid handling system with existing parameters. The third(C) is used robotic liquid handling system with adjusted parameters. Pipetting ability depending on types of liquids is assessed with CV(%). On the basis of (A), patients's test results are compared (A)and(B), (A)and(C) and they are assessed with %RE(%Relative error) and %Diff(%Difference). Results The CV(%) of the CPM depending on liquid types were water 0.88, serum 0.95, methanol 10.22 and PEG 0.68. As expected dispensing of methanol using a liquid handling system was the problem and others were good. The methanol's dispensing were conducted by adjusting several setting parameters. When transport air gap 0 was adjusted to 2 and 5, CV(%) were 20.16, 12.54 and when system air gap 0 was adjusted to 2 and 5, CV(%) were 8.94, 1.36. When adjusted to system air gap 2, transport air gap 2 was 12.96 and adjusted to system air gap 5, Transport air gap 5 was 1.33. When dispense speed was adjusted 300 to 100, CV(%) was 13.32 and when dispense delay was adjusted 200 to 100 was 13.55. When compared (B) to (A), the result increased 99.44% and %RE was 93.59%. When compared (C-system air gap was adjusted 0 to 5) to (A), the result increased 6.75% and %RE was 5.10%. Conclusion Adjusting speed and delay time of aspiration and dispense was meaningless but changing system air gap was effective. By adjusting several parameters proper value was found and it affected the practical result of the experiment. To optimize the system active efforts are needed through the test and in case of dispensing new types of liquids proper test is required to check the liquid is suitable for using the equipment.
