일본 나나키다 하구 갯벌에 있어서 탄소 질소 안정동위원소를 이용해 먹이 연쇄의 공간적 가변성에 대해 조사했다. 서로 다른 특징을 갖고 있는 갯벌에서 잠재적인 유기물(육상식물, 해양 입자성 유기물, 저서 부착 미세조류 및 하구 입자성 유기물), 퇴적유기물 및 저서 무척추 동물(Nuttallia olivacea and Nereidae)에 대한 샘플링을 행했다. 본 연구의 목적은 좁은 공간적 가변성에 따른 Nuttallia olivacea와 Nereidae의 먹이원을 결정하는 것이다. 육상식물(${\delta}^{13}C=-26.6{\pm}0.76$, ${\delta}^{15}N=2.7{\pm}0.31$) 과 하구 입자성 유기물(${\delta}^{13}C=-25.5{\pm}0.13$, ${\delta}^{15}N=5.2{\pm}0.46$)은 저서 부착 미소조류${\delta}^{13}C=-16.3$, ${\delta}^{15}N=6.2$)와 해양 입자성 유기물(${\delta}^{13}C=-19.6{\pm}0.08$, ${\delta}^{15}N=8.9{\pm}1.70$)의 탄소 질소 안정동위원소비 보다 낮았다. 퇴적물의 탄소 안정동위원소 비는 -27.4~-22.8‰ 나타냈으며, 하구에서 하천 방향으로 갈수록 낮은 탄소 안정동위 원소비를 나타냈다. 저서 무척추 동물의 탄소 질소 안정동위원소비는 각각 -22.8~-18.4‰, 8.1~11.9‰ 범위를 나타냈다. 이러한 결과와 더불어 혼합 모델을 이용해 저서 무척추 동물의 먹이원의 기여율을 추정한 결과 해양 입자성유기물과 저서 부착 미세조류의 기여율은 높았지만, 육상식물과 하구 입자성 유기물의 기여율은 비교적 낮았다. 이러한 저서 무척추 동물의 먹이 기여는 각 장소마다 계절 및 물리적 환경 요소의 영향을 받는다고 사료된다.
이 연구에서는 곰소만 조간대의 현생 해양환경, 2종 (Elphidium etigoense와 Ammonia beccarii tepida)의 유공충의 산소 및 탄소 동위원소 성분비와의 관계, 그리 고 유공충 종간의 차이 등을 연구하였다. E. etigoense의 $\delta$/SUP 18/O값은 -1.11$\textperthousand$에 서 +0.61$\textperthousand$이고 $\delta$/SUP 13/C값은 -2.04$\textperthousand$에서 -1.40$\textperthousand$이다. 상부, 중부, 하부 조간대 사이의 차이는 없었으며 유공충 2종간에는 아주 낮은 측정값을 보여주는 시료 한 개를 제외하면 평균$\delta$/SUP 18/O 값의 차는 0.20%이고 평균 $\delta$/SUP 18/O값의 차는 1.63$\textperthousand$이 었다.
후기 플라이스토신 및 홀로신 시기 동아시아 여름 몬순의 변화를 밝히기 위해 지화학적 분석 방법(탄소동원원소(${\delta}^{13}C$), 탄질율(C/N), 대자율, 입도 분석 등)을 활용하여 서해안 염하구 습지 퇴적물을 분석하였다. 연구 결과에 따르면 한반도에서 여름 몬순은 7,700-7,800년 전에 정점이었고 7,400년 전 경부터 약화되기 시작한다. 그리고 마지막 빙하 최성기 중 24,000-24,500년 전 경에는 몬순이 상대적으로 약했고 18,500-19,500년 전 경에는 상대적으로 강했다. 염하구 습지 퇴적물의 지화학적 분석 자료는 과거 여름 몬순의 변동을 밝힐 수 있는 새로운 고환경 자료로서 가치를 갖는다. 특히 고환경 자료가 부족한 한국 학계에 큰 도움이 될 것으로 사료된다.
본 논문에서는 퇴적유기물에 있어서, 퇴적유기물의 조성 및 세균 먹이원 이용에 대한 변화를 알아보기 위하여, 안정동위원소와 지방산의 농도를 이용 하였다. 이를 위해 실험에 이용된 퇴적물은 가모 석호(Gamo Lagoon)에서, 잠재적 유기물(육상 식물, 해양 입자성 유기물, 저서 부착 미소조류, 하천 입자성 유기물)은 나나키타 하구(Nanakita estuary)에서 시료를 채취하였다. 채취된 시료인 퇴적물, 잠재적 유기물 그리고 세균에 대해서 안정동위원소 및 지방산의 변화를 조사하였다. 이러한 조사 결과, 각각의 잠재적 유기물에 있어서 ${\delta}^{13}C$와 ${\delta}^{15}N$은 육상 식물(-26.6‰과 3.6‰), 하천 입자성 유기물(-25.5‰와 8.9‰), 저서 부착 미소조류(-16.3‰과 6.2‰), 해양 입자성 유기물(-20.3‰과 10.3‰)으로 나타났으며, 퇴적 유기물의 안정동위원소비는 -20.7에서 -19.3‰를 나타났다. 또한 세균의 탄소 질소 안정동위원소비는 각각 -20.8에서 -18.6‰로, 6.5에서 8.6‰로의 변화를 나타냈다. 결국 퇴적유기물에 있어서 세균은 다양한 유기물이 혼합된 상태에서 우선적으로 분해하기 쉬운 저서 부착 미소조류와 해양 입자성 유기물을 탄소원으로 이용하고, 순차적으로 분해하기 어려운 육상 식물 유래의 유기물을 탄소원으로 이용하는 것으로 나타났다.
Natural abundances of stable isotopes of nitrogen and carbon (${\delta}^{15}N$ and ${\delta}^{13}C$) are being widely used to study N and C cycle processes in plant and soil systems. Variations in ${\delta}^{15}N$ of the soil and the plant reflect the potentially variable isotope signature of the external N sources and the isotope fractionation during the N cycle process. $N_2$ fixation and N fertilizer supply the nitrogen, whose ${\delta}^{15}N$ is close to 0%o, whereas the compost as. an organic input generally provides the nitrogen enriched in $^{15}N$ compared to the atmospheric $N_2$. The isotope fractionation during the N cycle process decreases the ${\delta}^{15}N$ of the substrate and increases the ${\delta}^{15}N$ of the product. N transformations such as N mineralization, nitrification, denitrification, assimilation, and the $NH_3$ volatilization have a specific isotope fractionation factor (${\alpha}$) for each N process. Variation in the ${\delta}^{13}C$ of plants reflects the photosynthetic type of plant, which affects the isotope fractionation during photosynthesis. The ${\delta}^{13}C$ of C3 plant is significantly lower than, whereas the ${\delta}^{13}C$ of C4 plant is similar to that of the atmospheric $CO_2$. Variation in the isotope fractionation of carbon and nitrogen can be observed under different environmental conditions. The effect of environmental factors on the stomatal conductance and the carboxylation rate affects the carbon isotope fractionation during photosynthesis. Changes in the environmental factors such as temperature and salt concentration affect the nitrogen isotope fractionation during the N cycle processes; however, the mechanism of variation in the nitrogen isotope fractionation has not been studied as much as that in the carbon isotope fractionation. Isotope fractionation factors of carbon and nitrogen could be the integrated factors for interpreting the effects of the environmental factors on plants and soils.
Korean mariculture Undaria pinnatifida was collected during the months of January, February, March, and December of 2010, as well as from January of 2011 to investigate the changes in the carbon and nitrogen stable isotope ratios (${\delta}^{13}C$ and ${\delta}^{15}N$) and heavy metal with respect to it growth and to identify the factors that influence such changes. The blades of U. pinnatifida showed ${\delta}^{13}C$ and ${\delta}^{15}N$ in the range (mean) of -13.11 to -19.42‰ (-16.93‰) and 2.99 to 7.57‰ (4.71‰), respectively. Among samples with the same grow-out period, those that weighed more tended to have higher ${\delta}^{13}C$ suggesting a close association between the carbon isotope ratio and growth rate of U. pinnatifida. Indeed, we found a very high positive linear correlation between the monthly average ${\delta}^{13}C$ and the absolute growth rate in weight ($r^2=0.89$). Nitrogen isotope ratio tended to be relatively lower when nitrogen content in the blade was higher, probably due to the strengthening of isotope fractionation stemming from plenty of nitrogen in the surrounding environment. In fact, a negative linear correlation was observed with the nitrate concentration in the nearby seawaters ($r^2=0.83$). Concentrations of Cu, Cd, Pb, Cr, Hg, and Fe in the blades showed a rapid decrease in their concentration per unit weight in the more mature U. pinnatifida. Specifically, compared to adult samples, Cu, Hg, and Pb were concentrated by 30, 55, and 73 folds, respectively, in the young blades. Therefore, U. pinnatifida tissue ${\delta}^{13}C$ is as an indirect indicator of its growth rate, while ${\delta}^{15}N$ values and heavy metal concentrations serve as tracers that reflect the environmental characteristics.
Fatty acid compositions of the seed oils of P. schinseng, A. continentalis and A. sessiliflorus, were analyzed by gas chromatography (GC) equipped with a capillary column. A large unusual peak was observed just before the peak corresponding to oleic acid $(cis-9-C_{18:1})$. This unknown fatty acid was isolated by silver ion chromatography and then derivatized into the picolinyl ester. The mass spectrum of the picolinyl ester showed molecular ion at m/z=373 with other diagnostic ions such as m/z=178, 218, 232, 246, 274, 288, 302 and 344. Characteristic absorption peaks at $720\;cm^{-1}$, $1640\;cm^{-1}$ and $3010\;cm^{-1}$ in IR spectrum indicated the presence of cis-configurational double bond in the molecule. The $^1H-NMR$ spectrum of this acid gave two quintets centered at ${\delta}1.638$ (2H, C-3) and ${\delta}1.377$ (2H, C-4), and two multiplets centered at ${\delta}2.022{\sim}2.047$ (2H, C-5) and ${\delta}2.000{\sim}2.022$ (2H, C-8), and multiplet signals of olefinic protons centered at ${\delta}5.3015{\sim}5.3426$ (C-6, J=9.5 Hz) and ${\delta}\;5.3465{\sim}5.3877$ (C-7, J=9.5 Hz). The $^13C-NMR$ spectrum showed 18 carbon resonance signals including an overlapped signal at ${\delta}29.7002$ for C-12 and ${\delta}29.6520$ for C-13 (or they can be reversed), and other highly resolved signals at ${\delta}33.950$, ${\delta}24.558$, ${\delta}26.773$ and ${\delta}27.205$ due to C-2, C-3, C-5 and C-8 of a ${\Delta}^6-octadecenoic$ acid, respectively. From analysis results this unknown fatty acid could be identified as cis-6-octadecenoic acid. The seed oils of P. schinseng and A. sessiliflorus contained petroselinic acid (59.7%, 56.0%), oleic acid (18.3%, 6.1%) and linoleic acid (16.2%, 30.4%) with small amount of palmitic acid (3.0%, 3.1%) while the seed oil of A. continentalis comprised mainly oleic acid (30.2%), petroselinic acid (29.0%), linoleic acid (24.1%) and palmitic acid (13.1%).
All the triacylglycerols including the molecular species having ${\Delta}^5$-unsaturated fatty acids from the seeds of Pinus Koraiensis, were split into a mixture of diacylglycerols by a Grignard reagent prepared with allyl bromide without arousing acyl chains of a glycerol moiety to migration, and were also easily partially hydrolyzed to diacylglycerols by pancreatic lipase. (S)-(+)-(1-naphthyl)ethyl urethane(NEU) derivatives of the diacylglycerol mixture derived from the triacylglycerols were fractionated into sn-1, 3-, sn-1, 2- and sn-2, 3-DG-NEU by silica-HPLC and the fatty acid composition of these fractions was analysed. $C_{18:1{\omega}9}$ is distributed evenly in the three positions of TG with $C_{18:2{\omega}6}$ mainly located in sn-2 position, while ${\Delta}^5$-unsaturated fatty acids such as ${\Delta}^{5.9}-C_{18:2}$, ${\Delta}^{5.9.12}-C_{18:3}$ and ${\Delta}^{5.11.14}-C_{20:3}$ are exclusively present in the sn-3 position. These results could be confirmed by $^{13}C$-NMR spectroscopy : the signals at $^{\delta}$173.231 ppm and $^{\delta}$172.811 ppm of the carbonyl carbon of acyl moieties indicate the presence of saturated acids and/or $C_{18:1{\omega}9}$ (oleic acid) in the ${\alpha}({\alpha}')$- or ${\beta}$- positions, and $C_{18:2{\omega}6}$ including $C_{18:1{\omega}9}$ in the ${\beta}$-position, respectively. In addition, the resonance at $^{\delta}$173.044 ppm suggested a location of ${\Delta}^5$-unsaturated fatty acid moiety in the ${\alpha}({\alpha}')$-position.
It is known that ${\delta}^{13}C_{DIC}$ (carbon-13 isotope of dissolved inorganic carbonate (DIC) ions) of water increases when dissolved $CO_2$ degases. However, ${\delta}^{13}C_{DIC}$ could decrease when the pH of water is lower than 5.5 at the early stage of degassing. Laboratory experiments were performed to observe the changes of ${\delta}^{13}C_{DIC}$ as $CO_2$ degassed from three different artificial $CO_2$-rich waters (ACWs) in which the initial pH was 4.9, 5.4, and 6.4, respectively. The pH, alkalinity and ${\delta}^{13}C_{DIC}$ were measured until 240 hours after degassing began and those data were compared with kinetic isotope fractionation calculations. Furthermore, same experiment was conducted with the natural $CO_2$-rich water (pH 4.9) from Daepyeong, Sejong City. As a result of experiments, we could observe the decrease of DIC and increase of pH as the degassing progressed. ACW with an initial pH of 6.4, ${\delta}^{13}C_{DIC}$ kept increasing but, in cases where the initial pH was lower than 5.5, ${\delta}^{13}C_{DIC}$ decreased until 6 hours. After 6 hours ${\delta}^{13}C_{DIC}$ increased within all cases because the $CO_2$ degassing caused pH increase and subsequently the ratio of $HCO_3{^-}$ in solution. In the early stage of $CO_2$ degassing, the laboratory measurements were well matched with the calculations, but after about 48 hours, the experiment results were deviated from the calculations, probably due to the equilibrium interaction with the atmosphere and precipitation of carbonates. The result of this study may be not applicable to all natural environments because the pressure and $CO_2$ concentration in headspace of reaction vessels was not maintained constant as well as the temperature. Nevertheless, this study provides fundamental knowledge on the ${\delta}^{13}C_{DIC}$ evolution during $CO_2$ degassing, and therefore it can be utilized in the studies about carbonated water with low pH and the monitoring of geologic carbon sequestration.
${\delta}^{13}C$ and ${\delta}^{18}O$ values were determined for the Paleozoic limestones (Great Linestone Series) from the Taebaegsan region and the age-unknown limestones (Janggun Formation) from the Janggun mine, Korea. Limestones of the Great Limestone Series exhibit a range of carbon isotopic composition from -4.5 +1.3‰ with a mean ${\delta}^{13}C$ value of -1.1‰, relative to the PDB standard, and of oxygen isotpic composition from +8.8 to +23.3‰ with a mean ${\delta}^{18}O$ value of +16.0‰, relative to the SMOW, falling into the normal marine limestone range according to Keith and Weber (1964), and Degens and Epstein(1964). Carbon isotopic composition of limestones of the Great Limestone Series becomes progressively lighter from Pungchon limestone of middle Cambrian age to mid-Ordovician Maggol limestone, possibly due to change in depositional environment in the Taebaegsan basin. Variation in isotopic composition of limestones from Hwajeol to Dumugal formation offers the possibility or deposition in shallow sea environment, in which fresh waters were added in several stages. Janggun limestone of unknown age may be corelated with the Paleozoic limestones of Great Limestone Series as infered from the istopic composition ranging from -2.8 to + 0.7‰ of ${\delta}^{13}C$ and +13.4 to +22.4‰ of ${\delta}^{18}O$.
