An extracellular $\beta$-glucosidase produced by Monascus purpureus NRRL1992 in submerged cultivation was purified by acetone precipitation, gel filtration, and hydrophobic interaction chromatography, resulting in a purification factor of 92-fold. A $2^2$ central-composite design (CCD) was performed to find the best temperature and pH conditions for enzyme activity. Maximum activity was observed in a wide range of temperature and pH values, with optimal conditions set at $50^{\circ}C$ and pH 5.5. The $\beta$-glucosidase showed moderate thermostability, was inhibited by $HgCl_2$, $K_2Cr_O_4$, and $K_2Cr_2O_7$, whereas other reagents including $\beta$-mercaptoethanol, SDS, and EDTA showed no effect. Activity was slightly stimulated by low concentrations of ethanol and methanol. Hydrolysis of p-nitrophenyl-$\beta$-D-glucopyranoside (pNPG), cellobiose, salicin, n-octyl-$\beta$-D-glucopyranoside, and maltose indicates that the $\beta$-glucosidase has broad substrate specificity. Apparently, glucosyl residues were removed from the nonreducing end of p-nitrophenyl-$\beta$-D-cellobiose. $\beta$-Glucosidase affinity and hydrolytic efficiency were higher for pNPG, followed by maltose and cellobiose. Glucose and cellobiose competitively inhibited pNPG hydrolysis.
Diazotized chitin (CHITN) was synthesized reacting with NaN3 and HCl to alkaline hydrolyzed chitin for the support of immobilized enzyme. Immobilized $\beta$-glucosidase on diazotized chitin(CHITN-Gase) was produced reacting with glutaraldehyds as bifunctional reagent. CHITN-Gase activities were determined reacting with p-nitrophenol-$\beta$-D-glucopyranoside in plug flow reactor as a reference. Optimum temperature, optimum pH, reaction constant and deactivation rate were determined with variation of flow rate and H/D. The particle size of immobilized enzyme in the best was, 35 mesh (CHITN35-Gase). The optimum conditions of immobilized enzyme were $70^{\circ}C$ in temperature and 5.0 in pH. Diameter and flow rate of plug flow reactor in the best was 8.5mm in diameter and 0.8ml/min in flow rate. Reaction constant was mainly influenced by electrostatic force. The best glucose hydrolizing activities of CHITN3 5-Gase was 3.34$\times$10-5 M/1. while that of native-$\beta$-glucosidase was 2.44$\times$10-5 M/1.
Fro the purpose of producing glouse from cellobiose or oligo saccharide and obtaining genetic information of beta-1,4-glucosidase gene, alpha beta-1,4-glucosidase gene of Pseudomonas sp. LBC505, potent cellulase complex and xylanase producing strain, was cloned in Esherichia coli and Bacillus subtilis into pUC19 and pBD64, respectively. Recombinant plasmid pGL1 contained 1.2kb EcoRI fragment was isolated from transformants forming blue color around colony on LB agar plate containing 20 ng/ml of 5-bromo-4-chloro-3-indolyl-beta-D-glucopyranoside(X-glu) and ampicillin.
중고압 하에서 $\beta$-glucosidase효소반응을 물리화학적 관점에서 연구하였다. 모델 기질 (p-nitrophenyl-${\beta}$-D-glucopyranoside)에 대한 $\beta$-glucosidase 효소의 작용에 대한 압력 효과를 실험 하였다. 즉, 압력 조건(25MPa, 50 MPa, 75 MPa, 100 MPa)과 시간 (10분, 60분, 1시간, 6시간, 24시간, 40시간)의 처리 조건에서 효소 활성도를 분광학적인 표준방법에 따라 측정하였다. 효소-기질 반응의 단계를 크게 kinetic 구간과 평형 구간으로 구분하여 물리화학적 모델을 적용하여, 정 역반응속도 상수, 평형상수, 압력에 의한 부피 감소 등을 산출하였다. 대기압에서 100MPa까지 압력이 증가할수록 효소-기질 반응의 생성물이 더 많이 형성되었으며 전형적인 kinetic 구간과 평형 구간이 나타났다. 압력, 시간, 생성물농도 등의 데이터로부터 kinetic 구간과 평형에서의 생성물 예측 모델을 완성하였다. 결론적으로 중고압 처리에 의하여 효소-기질 반응이 촉진됨을 알 수 있었고, 임의의 압력 및 시간 조건에 따른 생성물의 농도를 예측할 수 있게 되었다.
Cellulomonas sp.YE-5가 생산하는 cellulase를 분리, 정제하여 효소의 특성을 알아보았다. Avicelase, CMCase, $\beta$-glucosidase의 반응 최적온도은 각각 40, 45, $40^{\circ}C$이었고, 반은 최적 pH는 5.5, 6.0 그리고 6.0이었다. 효소의 열안정성은 30~$70^{\circ}C$에서 6시간 처리하였을 때 avicelase와 $\beta$-glucosidase는 $50^{\circ}C$ 이상에서 거의 실활하였고, CMCase는 $50^{\circ}C$에서 약 40%의 활성을 유지하였다. 효소의 안정성에 미치는 pH의 영향은 $25^{\circ}C$에서 24시간 처리하였을 때 avicelase와 CMCase는 PH 5.0~9.0 사이에서 안정하였으며, $\beta$-glucosidase는 pH 5.0~8.0 사이에서 안정하였다.
Chitin을 35 mesh로 분쇄하여 강산과 강알카리로 처리하여 CHITA와 CHITB를 만들고 여기에 glutaraldehyde를 작용시켜 $\beta$-glucosidase를 고정하여 CHITA-Gase 및 CHITB-Gase를 제조하였다. 이 두 종류의 고정화 효소를 기질 p-nitropheol-$\beta$-D-gliucopyranoside(PNG)과 회분식 반응기, 연속 흐름 반응기 및 플러그 흐름 반응기에서 반응시켜 최적 온도, 최적 pH, 반응 속도 상수, 물질 전달 계수, 효율 인자 및 효소 불활성화 속도 등을 구하여 반응기별 효율을 검토하였다. 반응 최적 온도는 세가지의 반응기 모두 5$0^{\circ}C$였으며, 최적 pH는 플러그 흐름 반응기에서는 Nat-Gase와 같이 pH5.0이었고 회분식 반응기 및 연속 흐름 반응기에서는 최적 pH의 이동이 일어나 pH6.0으로 이었다. 반응기의 최적 조건에서 km값은 회분식 반응기에서 CHITB-Gase$1.725$\times$10^-^5M/1$가 CHITA-Gase($1.725$\times$10^-^5M/1$)보다 작았으며, 연속 흐름 반응기 및 플러그 흐름 반응기에서는 유속 증가에 따라 Km'치가 감소하는 경향을 보였고, CHITB-Gase가 CHITB-Gase보다 더 작았다. $V^m^a^x'$값은 회분식 반응기, 연속 흐름 반응기, 플러그 흐름 반응기에서 모두 CHITA-Gase가 CHITB-Gase보다 높은 것으로 나타났다. 그리고, 물질전달계수 및 효율인자, 효소 불활성화 속도등의 값은 환경은 CHITB-Gase의 것이 나은 것으로 나타났다. 이들의 결과에서 CHITA-Gase 및 CHITB-Gase는 기질과의 반응 환경이 좋으므로 chitind은 $\beta$-glucosidase의 좋은 지지체라고 판단되어 공업적 응용이 기대된다.
대두의 발효를 통하여 생리활성을 가지고 있는 이소플라본 aglycone 함량을 높이기 위한 ${\beta}$-glucosidase와 대두에 다량 함유되어 있는 stachyose, rafinose와 같은 난소화성 oligosaccharides를 분해하기 위해 ${\alpha}$-galactosidase 효소 분비 미생물을 김치로부터 ${\alpha}$-galactosidase와 ${\beta}$-glucosidase를 생산하는 미생물을 탐색하였다. 탐색과정을 위해서 선별한 미생물을 16S rDNA sequencing 동정한 결과, Weissella cibaria 동정되어 Weissella cibaria K-M1-4로 명명하였다. Weissella cibaria K-Ml-4를 대두 액체배지에서 18시간동안 배양한 후, 생산한 효소는 배양액을 에탄을 침전, DEAE sepharose, sephacryl S-100HR column chromatography 통하여 ${\alpha}$-galactosidase의 경우, 정제도 5.3배, 수율 3.5% 그리고 ${\beta}$-glucosidase의 경우, 정제도 4.4배, 수율 2.9%로 정제되었다. ${\alpha}$-Galactosidase 효소특성은 $60^{\circ}C$에서 최대 활성을 나타내었으며, $80^{\circ}C$에서 30분 처리시 43% 잔존활성을 보였다. pH 8.0에서 최대 활성을 나타내었으며, pH 5.0-9.0에서 안정하였다. 금속이온에 대한 영향에서 $Fe^{2+}$과 $Cu^{2+}$을 첨가하였을 때 효소 활성이 증가하였다. p-Nitrophenyl-${\alpha}$-D-galacto-pyranoside (PNPG) 기질에 대한 Km은 0.98 mM이었고, Vmax는 $1.81{\mu}$mole/min 이었다. ${\beta}$-Glucosidase 효소 특성은 $50^{\circ}C$에서 최대 활성을 나타내었으며, $80^{\circ}C$에서 30분 처리시 46% 잔존활성을 보였다. pH 7.0에서 최대 활성을 나타내었으며, pH 5.0-9.0에서 안정하였다. 금속이온에 대한 영향에서 $Fe^{2+},\;Co^{2+},\;Cu^{2+}$을 첨가하였을 때 효소 활성이 증가하였다. p-Nitrophenyl-${\beta}$-D-gluco-pyranoside (PNPG)에 대한 Km값은 1.24mM이었고, Vmax는 $6.81{\mu}$mole/min 이었다.

A novel ${\beta}-glucosidase$ gene, bglA, was isolated from uncultured soil bacteria and characterized. Using genomic libraries constructed from soil DNA, a gene encoding a protein that hydrolyzes a fluorogenic analog of cellulose, 4-methylumbelliferyl ${\beta}-D-cellobioside$ (MUC), was isolated using a microtiter plate assay. The gene, bglA, was sequenced using a shotgun approach, and expressed in E. coli. The deduced 55-kDa amino acid sequence for bglA showed a 56% identity with the family 1 glycosyl hydrolase Chloroflexus aurantiacus. BglA included two conserved family 1 glycosyl hydrolase regions. When using $p-nitrophenyl-{\beta}-D-glucoside$ (pNPG) as the substrate, the maximum activity of the purified ${\beta}-glucosidase$ exhibited at pH 6.5 and $55^{\circ}C$, and was enhanced in the presence of $Mn^{2+}$. The $K_m\;and\;V_{max}$ values for the purified enzyme with pNPG were 0.16 mM and $19.10{\mu}mol/min$, respectively. The purified BglA enzyme hydrolyzed both pNPG and $p-nitrophenyl-{\beta}-D-fucoside$. The enzyme also exhibited substantial glycosyl hydrolase activities with natural glycosyl substrates, such as sophorose, cellobiose, cellotriose, cellotetraose, and cellopentaose, yet low hydrolytic activities with gentiobiose, salicin, and arbutin. Moreover, BglA was able to convert the major ginsenoside $Rb_1$ into the pharmaceutically active minor ginsenoside Rd within 24 h.

The ${\beta}-glucosidase$ from olive fruit is of particular interest compared to the ones from other sources because it has shown to have high specifity to convert the oleuropein into dialdehydes, which have antibacterial activity and are of high interest for their application in the food and pharmaceutical fields. The enzyme is not yet commercially available and advanced clean and safe technologies for its purification able to maintain the functional stability are foreseen. The purification of this protein from fruit extracts has been already tempted by electrophoresis but either enzyme deactivation or high background with unclear profiles occurred. In this work, fruit extracts obtained from the ripening stage that showed the highest enzyme activity have been processed by diafiltration and ultrafiltration. Asymmetric membranes made of polyamide or polysulphone having 50 and 30 kDa molecular weight cut-off, respectively, were tested for the diafiltration process. Ultrafiltration membranes made of polyethersulfone with 4 kDa molecular weight cut-off were used to concentrate the dia-filtered permeate solutions. The efficiency of the separation processes was evaluated byenzyme activity tests using the hydrolysis of p-D-nitrophenyl-${\beta}$-D-glucopyranoside (pNPGlc) as reaction model. Qualitative and quantitative electrophoresis were applied to analyze the composition of protein solution before and after the membrane separation; in addition dot blot and western blot analyses were applied to verify the presence of ${\beta}-glucosidase$ in the processed fractions. The overall results showed that the ${\beta}-glucosidase$ functional stability was preserved during the membrane operations and the removal of 20 kDa proteins allowed to increase the specific activity of the enzyme of about 52% compared to the one present in the initial fruit extract.
The roots of Codonopsis lanceolata afforded tangshenoside I(1) and $\beta$-adenosine (2) as $\alpha$-glucosidase inhibitors. Their structures were unambiguously determined by 1D and 2D NMR data including HMQC and HMBC experiments. Compounds 1 and 2 exhibited weak $\alpha$-glucosidase inhibitory activities in vitro with $IC_{50}$ of 1.4 and 9.3 mM, respectively.
