천연우라늄 오염에 관한 방사선/능 측정기술 연구 (A Study on the Natural Uranium Contamination Measuring Technology)

  • 본 연구는 우라늄변환시설의 해체과정에서 발생되는 콘크리트 및 구성부품에 대한 알파분광과 감마분광에 대한 방사선계측방법의 타당성을 입증하고자 한다. 우라늄변환시설내의 구성부품 및 내부벽면은 천연우라늄 물질로 오염되어있다. 스테인레스 스틸 파이프와 벽면의 콘크리트의 일부에 대하여 시료를 채취하고 알파분석과 감마분석을 수행하였다. 천연우라늄 시료(AUC)의 측정에서 0.01 Bq/g 이상에서는 알파선 측정결과와 감마선 측정결과가 잘 일치하지만, 0.005 Bq/g의 낮은 농도에서는 감마선 측정결과가 상대적으로 높게 평가된다. 변환공정의 천연우라늄인 $^{238}U$$^{214}Pb$, $^{214}Bi$$^{234}Th$, $^{234m}Pa$을 비교 측정하여 그 농도를 구할 수 있다. 우라늄변환시설의 벽면은 대부분 우라늄으로 오염되어있다. 우라늄변환시설 해체과정에서 발생되는 배경방사능 준위의 폐기물을 감마분광법을 이용하여 계측하여 보수적인 평가 자료로 활용할 수 있다.

고전압 방전 플라즈마에 의해 합성한 질화탄소 박막의 열처리 효과 (Effect of Annealing on Carbon Nitride Films Prepared by High Voltage Discharge Plasma)

  • I have investigated the effects of annealing on a polymeric $\alpha-C_3N_{4.2}$ at high pressure and temperature in the presence of seeds of crystalline carbon nitride films prepared by a high voltage discharge plasma. The samples were evaluated by x-ray photoelectron spectroscopy (XPS), infrared spectroscopy, Auger electron spectroscopy and x-ray diffraction(XRD). Notably, XPS studies of the film composition before and after annealing demonstrate that the nitrogen composition in $\alpha-C_3N_{4.2}$ material initially containing more than 58% nitrogen decreases during the annealing process and reaches a common, stable composition of ~43%. XPS analysis also shows that the nitrogen composition in the annealed films without polymeric $\alpha-C_3N_{4.2}$ was reduced from 35% to 17%. Furthermore the concentration of the sp$^3$bonded phase increased with the increment of the annealing temperature.

Signal Generation Due to Alpha Particle in Hydrogenated Amorphous Silicon Radiation Detectors

  • The hydrogenated amorphous silicon (a-Si : H) holds good promise for radiation detection from its inherent merits over crystalline counterpart. For the application to alpha spectroscopy, the induced charge collection in a-Si : H pin detector diodes ons simulated based on a relevant non-uniform charge generation model. The simulation was peformed for the initial energy and the range of incident alpha particles, detector thickness and the operational parameters such as the applied reverse bias voltage and shaping time. From the simulation, the total charge collection was strongly affected by hole collection as expected. To get a reasonable signal generation, therefore, the hole collection should be seriously considered for detector operational parameters such as shaping time and reverse voltage etc. For the spectroscopy of alpha particle from common alpha sources, the amorphous silicon should have about 70${\mu}{\textrm}{m}$ thickness.

Determination of Alpha Defect Center in the Nature Using EPR Spectroscopy

  • Natural alpha radiation produced a stable defect center to certain minerals. Electron Paramagnetic Resonance(EPR) spectroscopy is a powerful tool f3r quantifying this defect center. EPR method has been applied to trace alpha-radiation effect around the uranium ore deposit. The results show that EPR technique can be used to measure rapidly and nondestructively the defect center produced by natural alpha radiation. In general, a good correlation was achieved between defect center concentration and actinide elements(U, Th). These results imply that the concentration of defect center is dependent on the alpha radiation dose over long time scale.

Recent advances in NMR-based structural characterization of αB-crystallin and its potential role in human diseases

  • ${\alpha}B$-crystallin (${\alpha}BC$) is a member of a small heat-shock protein (sHSP) superfamily and plays a predominant role in cellular protein homeostasis network by rescuing misfolded proteins from irreversible aggregation. ${\alpha}BC$ assembles into dynamic and polydisperse high molecular weight complexes containing 12 to 48 monomers; this variable stereochemistry of ${\alpha}BC$ has been linked to quaternary subunit exchange and its chaperone activity. The chaperone activity of ${\alpha}BC$ poses great potential as therapeutic agents for various neurodegenerative diseases. In this mini-review, we briefly outline the recent advancement in structural characterization of ${\alpha}BCs$ and its potential role to inhibit protein misfolding and aggregation in various human diseases. In particular, nuclear magnetic resonance (NMR) spectroscopy and its complimentary techniques have contributed much to elucidate highly-dynamic nature of ${\alpha}BCs$, among which notable advancements are discussed in detail. We highlight the importance of resolving the structural details of various ${\alpha}BC$ oligomers, their quaternary dynamics, and structural heterogeneity.

Structure studies of Pulmonary Surfactant Protein B(SP-B(3,4)) by NMR Spectroscopy and Molecular Modeling

  • Synthetic pulmonary surfactants consisting of a mixture of phospholipids with synthetic peptides based on human surfactant-associated protein SP-B were prepared. These surfactants were analyzed f3r their secondary structures by circular dichroism (CD) spectroscopy and NMR spectroscopy. Two synthetic peptides (SP-B(3), SP-B(4)) combined with the phospholipid mixture displayed significant surfactant properties. The CD spectra showed that the u-helical propensities of the peptides in DPC micelles. In the NMR spectroscopy, the tertiary structures of SP-B(3) show that it has $\alpha$-helical structure from Gln5 to Arg13 in DPC micelle and SP-B(4) show that they have $\alpha$-helical structure from Gln5 to Leu12 in DPC micelle. Based on these structures, truncated peptides originated from SP-B protein, can be designed as effective synthetic surfactants for clinical use.

Backbone Resonance Assignment of a Proteolysis-Resistant Fragment in the Oxygen-Dependent Degradation Domain of the Hypoxia Inducible Factor 1α

  • Hypoxia-inducible factor $1{\alpha}$ ($HIF1{\alpha}$) is a transcription factor that plays a key role in the adaptation of cells to low oxygen stress and oxygen homeostasis. The oxygen-dependent degradation (ODD) domain of $HIF1{\alpha}$ responsible for the negative regulation of $HIF1{\alpha}$ in normoxia is intrinsically unfolded. Here, we carried out the backbone $^1H$, $^{15}N$, and $^{13}C$ resonance assignment of a proteolysis-resistant fragment (residues 404-477) in the $HIF1{\alpha}$ ODD domain using NMR spectroscopy. About 98% (344/352) of all the $^1HN$, $^{15}N$, $^{13}C{\alpha}$, $^{13}C{\beta}$, and $^{13}CO$ resonances were unambiguously assigned. The results will be useful for further investigation of the structural and dynamic states of the $HIF1{\alpha}$ ODD domain and its interaction with binding partners.

  • Rhizopus nigricans produces 11.alpha.-hydroxyprogesternoe with a unidentified byproduct, which is hardly separated. Results of chromatography, IR and NMR spectroscopy identified the byproduct to be 11.alpha.-hydroxy-allopregnane-3,20-dione. R. nigricans was found to transform progesternoe into a monoform intermediate, 11.alpha.-hydroxyprogesterone, from which 11.alpha.-hydroxy-allopregnane-3,20-dione and 6.betha., 11.alpha. - dihydroxyprogesterone were formed respectively by 5.alpha.-reduction and 6.betha.-hydroxylation.

  • The chiral stationary phase derived from (+) (18-crown-6)-2.3, 11.12-tetracarboxylic acid (18-C-6- TA) as a chiral selector has been employed for resolution of several $\alpha$-amino acids in HPLC. In a quest for the origin of chiral recognition of $\alpha$-amino acids in the presence of 18-C-6- T, A, as a chiral selector, these interactions responsible for the differential affinities shown toward enantioners were investigated by NNR spectroscopy.
