Anti-oxidative and Anti-hyperglycemia Effects of Dung Beetle Extracts on the High Fat Diet SD Rats (고지방식이로 유발한 흰쥐에서 쇠똥구리 추출물의 항산화 효과 및 혈당강하에 미치는 영향)

  • Dung beetle (Catharsius molossus, CA) is a well-known group of insects thanks to their exploitation of animal feces, a behavioral trait with a global impact on earth′s ecosystems. This study was conducted to investigate the effect of CA extract on a high-fat diet in SD rats. Male rats were divided into 5 groups. Animals were fed on a high-fat diet for seven weeks before and dung beetle extract for a month during the administration. Weight gain was decreased in ethanol extract from CA group. Administration of CA extract reduced the organ weight of testis and kidney, and adipose tissue weight. Lipid oxidative stress was evaluated measuring malondialdehyde level in liver. There were no significant differences in groups. Protein oxidative stress was evaluated measuring protein carbonyl content in blood. The protein carbonyl in blood was significantly decreased in ethanol and acetone extracted dung beetle groups (p<0.05). Meanwhile, the protein carbonyl in hepatocyte was not significant among the groups. Fibronectin and laminin by using D-HUVEC cell in vitro were measured by ELISA assay. There was significance in CA extract. The level of IL-10, IL-1β, VEGF, eNOS was evaluated by ELISA. There was significance in IL-10 compared to control (p<0.05). SOD and GPx tended to increase by CA extract. Furthermore, CAT was increased significantly by CA extract (p<0.05). After administration of CA extracts the composition of saturated fatty acid in adipose tissue tend to decrease, while unsaturated fatty acid increases. In conclusion, dung beetle had anti-hyperglycemia effects of oxidative stress and antioxidant activity.

  • Objective: A series of experiment were conducted to evaluate the effects of replacing a part of soybean meal (SBM) at 6% of broiler diets with fermented soybean meal (FSBM) obtained by single or two-stage fermentation by measuring growth performance, antioxidant activity in the jejunum and distal intestinal microflora. Methods: Soybean meal samples were prepared by single-stage fermentation using Bacillus velezensis (Bv) (FSBMB), or Lactobacillus spp. (as commercial control) (FSBML). Additional SBM sample was prepared by two-stage fermentation using Bv and subsequently using Lactobacillus brevis ATCC 367 (Lb) (FSBMB+L). Enzyme activity, chemical composition, trichloroethanoic acid-nitrogen solubility index (TCA-NSI) and antioxidant activity were measured. Then, in an in vivo study, 320 Ross308 broilers were divided into four groups with ad libitum supply of feed and water. Four groups were fed either a corn-soybean meal diet (SBM), or one of fermented SBM diets (FSBMB+L, FSBMB, and FSBML). Growth, serum characteristics, microflora, and the mRNA expression of selected genes were measured. Results: Compared to SBM, FSBMB+L contained lower galacto-oligosaccharide, allergic protein, and trypsin inhibitor, and higher TCA-NSI by about three times (p<0.05). Reducing power and 1,1-diphenyl-2-picrylhydrazyl free radical scavenging ability correlated positively with the TCA-NSI content in FSBM. Growth performances were not significantly different among four groups. In jejunum of 35-day-old broilers, partial replacement of SBM by FSBMB+L increased the activity of superoxide dismutase and catalase (CAT), and the FSBMB group had the highest catalase activity (p<0.05). Partial replacement of SBM by FSBM increased relative mRNA expressions of nuclear factor erythroid 2-related factor 2 (Nrf2), heme oxygenase-1 (HO-1), and peptide transporter 1 (PepT1) (p<0.05); however, FSBMB+L increased CAT mRNA level to 5 times of the control (p<0.05). Conclusion: Using Bv- and Lb-processed SBM through two-stage fermentation to partially replace 6% of diets will improve the gut's antioxidant activity under commercial breeding in broilers.

  • In paddy rice farming, water management is a critical task. To suppress weed emergence during the early stages of growth, fields are deeply flooded, and after transplantation, the water level is reduced to promote rooting and stimulate stem generation. Later, water is drained to prevent the production of sterile tillers. The adequacy of water supply is influenced by various factors such as field location, irrigation channels, soil conditions, and weather, requiring farmers to frequently check water levels and control the ingress and egress of water. This effort increases if the fields are scattered in remote locations. Automated irrigation systems have been considered to reduce labor and improve productivity. However, the net income from rice production in 2022 was about KRW 320,000/10a on average, making it financially unfeasible to implement high-cost devices or construct new infrastructure. This study focused on developing an IoT-Based irrigation valve that can be easily integrated into existing agricultural infrastructure without additional construction. The research was carried out in three main areas: Firstly, an irrigation valve was designed for quick and easy installation on existing agricultural pipes. Secondly, a power circuit was developed to connect a low-power Cat M1 communication modem with an Arduino Nano board for remote operation. Thirdly, a cloud-based platform was used to set up a server and database environment and create a web interface that users can easily access.

Effect of Angelica keiskei Koidzumi Extract on Alcohol-Induced Hepatotoxicity In Vitro and In Vivo (In Vitro 및 In Vivo 알코올 유도 간 손상에 대한 신선초 추출물의 효과)

  • We investigated the hepatoprotective effects of Angelica keiskei Koidzumi extract (AK) in HepG2-overexpressing cytochrome P4502E1 (CYP2E1) and C57BL/6J mice. In HepG2 cells expressing CYP2E1, cell viability and catalase activity in the ethanol-AK co-treated group significantly increased compared to those in the ethanol-treated group. In the in vivo study with C57BL/6J mice, the AK-supplemented group with ethanol liquid diet showed significantly reduced hepatic markers, including serum aspartate aminotransferase, alanine aminotransferase, and ${\gamma}$-glutamyl transferase, compared to the ethanol group without AK supplementation. AK supplementation (20 mg/kg BW/d) also significantly attenuated reactive oxygen species generation and malondialdehyde level. Notably, a low dose of AK supplementation (20 mg/kg BW/d) suppressed expression of hepatic CYP2E1 and inhibited CYP2E1 enzyme activity. These data indicate that a low dose of AK supplementation could restrain alcohol-induced hepatic damage mediated by CYP2E1.

Prevalence of Feline Panleukopenia Virus in Stray and Household Cats in Seoul, Korea (한국의 서울에 사는 길 고양이와 집 고양이에서 고양이 범백혈구감소증 바이러스의 유병률)

  • We investigated the prevalence of feline panleukopenia virus (FPV) in stray and household cats in different regions of Seoul, Republic of Korea. Blood samples were collected from a total of 200 cats (100 stray cats and 100 household cats) and examined by polymerase chain reaction (PCR). The overall prevalence of FPV was 2%. Among test-positive cats, 3% (3/100) were stray cats and 1% (1/100) was a household cat. The incidence of FPV was higher in juvenile cats (< 1 year, 1.5%) than in adult cats (> 1-year-old, 0.5%). The FPV-positive rates of healthy infected cats and sick cats were 1.9% (3/156) and 2.2% (1/44), respectively. We found the positive rate of vaccinated and unvaccinated cats to be 1.3% (1/77) and 2.4% (3/123), respectively. Unlike antibody tests, FPV antigen tests detected current infections in stray and household cats. Therefore, these tests can help in disease diagnosis and treatment. To our knowledge, our study is the first to survey the prevalence of FPV in different cat populations across Seoul. We found a high prevalence of FPV infection in stray and juvenile cats. Therefore, proper vaccination and surveillance are important to prevent FPV outbreaks.

Effects of Ixeris dentata ext. on Lowering Lipid and Anti-oxidation (씀바귀의 지질강하 및 항산화효과)

  • To investigate effects of Ixeris dentata EtOH ext. on lowering lipid levels and anti-oxidation activity, hyperlipidemic rats were treated with Ixeris dentata EtOH ext. and lipid levels and anti-oxdation activity were measured. The Ixeris dentata EtOH ext. groups showed low concentration of plasma FFA, plasma triglyceride, plasma total cholesterol, and plasma LDL-cholesterol compared to control group. However, concentration of plasma HDLcholesterol was not significantly different among all the treatment groups. The Ixeris dentata EtOH ext. groups showed lower level of liver total cholesterol, liver triglyceride, plasma TBARS, and liver TBARS than those of control group. The Ixeris dentata EtOH ext. groups also showed higher level of GSH-Px activity, SOD activity, and CAT activity than those of control group. Moreover, the Ext. showed lower level of TNF-${\alpha}$, Apo-B, Apo-E, and leptin expression than those of control group. The results in this study shows that the Ixeris dentata EtOH ext. have positive effect in lowering lipid level, and anti-oxidative activity.

A Study on Analysis and Control of Circumvent Connection to the Private Network of Corporation (기업 사설 네트워크 우회 접속 분석 및 통제 대책 연구)

  • A company's private network protected by a firewall and NAT(Network Address Translation) is not accessible directly through an external internet. However, as Reverse Connection technology used by NetCat extends to the technologies such as SSH Tunnel or HTTP Tunnel, now anyone can easily access a private network of corporation protected by a firewall and NAT. Furthermore, while these kinds of technologies are commercially stretching out to various services such as a remote control and HTTP Tunnel, security managers in a company or general users are confused under the circumstances of inner or outer regulation which is not allowed to access to an internal system with a remote control. What is more serious is to make a covert channel invading a company's private network through a malicious code and all that technologies. By the way, what matters is that a given security system such as a firewall cannot shield from these perceived dangers. So, we analyze the indirect access of technological methods and the status quo about a company's internal network and find a solution to get rid of the related dangers.

Enhancement of the Thermostability of a Fibrinolytic Enzyme from Bacillus amyloliquefaciens CH51 (Bacillus amyloliquefaciens CH51이 생산하는 혈전용해효소의 열안정성 개선)

  • AprE51 from Bacillus amyloliquefaciens CH51 is a 27 kDa subtilisin-like protease with fibrinolytic activity. AprE51-6 showing increased catalytic activity was produced previously. To enhance the thermostability of AprE51-6, 2 residues, Gly-166 and Asn-218 based on B. subtilis subtilisin E were mutated by site-directed mutagenesis. The results of the mutational analysis showed that substitution of arginine for Gly-166 (AprE51-7) increased the fibrinolytic activity 1.8-fold. An N218S mutant (AprE51-8) also increased the fibrinolytic activity up to 4.5-fold in a fibrin plate assay. Purified AprE51-7 and AprE51-8 mutants had a 1.9- and a 2.5-fold higher $k_{cat}$, respectively, and a 2.1-1.9-fold lower $K_m$, respectively. This resulted in a 3.8- and a 4.7-fold increase in catalytic efficiency ($k_{cat}/K_m$), respectively, relative to that of wild-type AprE51. AprE51-8 had a broader pH range than AprE51-6 and nattokinase, especially at an alkaline pH value. In addition, AprE51-8 showed higher thermostability than AprE51-6 at $60^{\circ}C$. The half-lives of AprE51-7 and AprE51-8 at $50^{\circ}C$ were 21.5 and 27.3 min, respectively, which are 2.0 and 2.6 times longer, respectively, than that of the wild-type AprE51.

Operating Characteristics of a 0.25 MW Methanation Pilot Plant with Isothermal Reactor and Adiabatic Reactor (등온반응기와 단열반응기 조합으로 구성된 0.25 MW급 메탄합성 파일롯 공정 운전특성)

  • In this study, we analyzed the operational characteristics of a 0.25 MW methanation pilot plant. Isothermal reactor controled the heat released from methanation reaction by saturated water in shell side. Methanation process consisting of isothermal reactor and adiabatic reactor had advantages with no recycle compressor and more less reactors compared with methanation process with only adiabatic reactors. In case that $H_2$/CO ratio of syngas was under 3, carbon deposition occurred on catalyst in tube side of isothermal reactor and the pressure of reactors increased. In case that $H_2$/CO ratio was maintained around 3, no carbon deposition on catalyst in tube side of isothermal reactor was found by monitoring the differential pressure of reactors and by measuring the differential pressure of several of tubes filled with catalyst before and after operating. It was shown that CO conversion and $CH_4$selectivity were over 99, 97%, respectively, and the maximum $CH_4$productivity was $695ml/h{\cdot}g-cat$.

Inhibitory Effects of Panax ginseng C. A. Mayer Treated with High Temperature and High Pressure on Oxidative Stress (산화적 스트레스에 대한 고온고압처리 인삼의 억제 효과)

  • Reactive oxygen species (ROS) are produced by oxidative stresses which cause various chronic diseases such as diabetes and obesity. Ginseng (Panax ginseng C.A. Mayer) has been reported to contain various biological activities such as anti-cancer, anti-diabetic, neuroprotective, radioprotective, anti-amnestic and anti-aging effects. In this study, we investigated the effects of Panax ginseng, treated with high temperatures and high pressures, on oxidative stress in C2C12 myoblasts and 3T3-L1 adipocytes. Oxidative stress was induced in the C2C12 cells through the introduction of $H_2O_2$ (1 mM), and cells were then treated with various ginseng preparations: dried white ginseng (DG), steamed ginseng (SG) and high temperature and high pressure treated ginseng (HG). In addition, 3T3-L1 preadipocytes were treated with various ginsengs for up to 8 days following standard induction of differentiation. Our results show that HG treatment significantly protected oxidative stress in both cell lines and enhanced gene expression of antioxidant enzymes. Therefore, in this study, we investigated the protective effects of ginseng on the oxidative stress of adipocytes and muscle cells.
