PCR-based Prevalence of Feline Vector-borne Pathogens in Yangju and Gwacheon Cities, South Korea

  • The objective of this polymerase chain reaction (PCR)-based research was to determine the prevalence of vector-borne pathogens in stray cats in Yangju and Gwacheon cities, South Korea. Total 50 stray cats were sampled for this PCR-based survey; 33 samples and 17 samples were collected from Yangju and Gwacheon cities, respectively. Total positive presence rates were 6%, 6% and 24% for hemotropic mycoplasmas, Rickettsia spp. and Babesia spp., respectively in this study. Babesia spp. was the predominant pathogen present in the stray cats of both cities followed by hemotropic mycoplasmas and Rickettsia spp. It is recommended that a large-scale study of the prevalence of infectious agents among stray cats should be undertaken in all regions of South Korea.

  • There are now increasing evidences that free radicals and reactive oxygen species are involved in a variety of pathological events. Reactive Oxygen Species (ROS) are produced during normal cellular function. ROS lead to lipid peroxidation, massive protein oxdiation and degradation. Under normal conditions, antioxidant are substnaces that either directly or indirectly protect cell against adverse effect of ROS. several biologically important compound include ${\beta}$-carotene, taruine and flavonoids reported have antioxidant function. The various antioxidant either scavange superoxide and free radicals or stimulate the detoxification mechanisms within cells resulting in increased detoxification of free radicals formation and thus in prevention of many pathophysiologic processes. This study carried out to investigate the antioxidant activity of flavonoids, myricetin with other antioxidants, ${\beta}$-carotene and taurine on B16Fl0. In order to investigate the efficacy of antioxidant activity, we measured cell viability, antioxidant enzyme activity (SOD, GPX, CAT) and intracellular reactive oxygen intermediate (ROI). In this results, we show that these flavonoids with other antioxidant substrates are increased antioxidant activity level.

A family with Townes-Brocks syndrome with congenital hypothyroidism and a novel mutation of the $SALL1$ gene

  • Townes-Brocks syndrome (TBS) is a rare autosomal dominant congenital disorder caused by mutations in the $SALL1$ gene. Its signs and symptoms overlap with other genetic syndromes, including VACTERL association, Pendred syndrome, Baller-Gerold syndrome, and cat eye syndrome. Structural vertebral abnormalities, hypoplasia of the thumb, and radial bone abnormalities, which are not usually associated with TBS, help in the differential diagnosis of these syndromes. We report the case of a family whose members were diagnosed with TBS with congenital hypothyroidism and had a novel $SALL1$ gene mutation.

Action of Asparagus racemosus Against Streptozotocin-Induced Oxidative Stress

  • Antidiabetic treatment with tubers of Asparagus racemosus (Liliaceae) extract (l00 and 250 mg/kg body weight) for three weeks showed significant reduction in thiobarbituric acid reactive substances (TBARS) and glutathione reductase (GSH-R) in both liver and kidney. The treatment with A. racemosus significantly altered the glutathione (GSH) and GSH-R to be comparable with the control group. A. racemosus and glibenclamide treated rats showed decreased lipid peroxidation that is associated with increased activity of superoxide dismutase (SOD) and Catalase (CAT). The ability of A. racemosus on tissue lipid per oxidation and antioxidant status in diabetic animals has not been studied before. The result of this study thus shows that though, A. racemosus possesses moderate antidiabetic activity, but it exhibits potent antioxidant potential in diabetic conditions.

Effect of lipopelysaccharide administration on the production of serum ${\gamma}$-globulin in stressed domestic cats (스트레스 상태의 고양이에서 lipopolysaccharide 투여가 혈중 감마글로불린 생산에 미치는 영향)

  • The activated immune systems with lipopolysaccharide(LPS) were evaluates by ${\gamma}$- globulin levels in domestic cats. Especially, cats were in a stress condition, which was unfamiliar environmental change. Effect of continuous unfamiliar environmental stressor was a diminution of production of serum total globulin auld ${\gamma}$- globulin on feline infectious peritonitis(FIP) vaccination. The ${\gamma}$-globulin production on FIP vaccination more increased in LPS treated 77ups than non-LPS treated group. Also. the ${\gamma}$-globulin production on FIP vaccination was maintained in LPS treated group under stress condition. These results imply that that the ${\gamma}$-globulin can be produced under a stress condition by mitogen like as LPS.

Over-expression of Cu/ZnSOD Increases Cadmium Tolerance in Arabidopsis thaliana

  • Over-expression of a copper/zinc superoxide dismutase (Cu/ZnSOD) resulted in substantially increased tolerance to cadmium exposure in Arabidopsis thaliana. Lower lipid peroxidation and $H_2O_2$ accumulation and the higher activities of $H_2O_2$ scavenging enzymes, including catalase (CAT) and ascorbate peroxidase (APX) in transformants (CuZnSOD-tr) compared to untransformed controls (wt) indicated that oxidative stress was the key factor in cadmium tolerance. Although progressive reductions in the dark-adapted photochemical efficiency (Fv/Fm) and quantum efficiency yield were observed with increasing cadmium levels, the chlorophyll fluorescence parameters were less marked in CuZnSOD-tr than in wi. These observations indicate that oxidative stress in the photosynthetic apparatus is a principal cause of Cd-induced phytotoxicity, and that Cu/ZnSOD plays a critical role in protection against Cd-induced oxidative stress.

Antioxidant and Liver-protective Effects of Caesalpinia sappan (소목 추출물의 항산화 및 간보호작용)

  • The heartwood of Caeralpinia sappan L. (Leguminosae) has been used to activate blood flow, remove blood stasis, reduce swelling and relieve pain in Korean folk medicine. In this study. the antioxidant effects of this crude drug and its hepatoprotective activity on CCl$_4$-induced liver injury in rats were evaluated. The levels of antioxidant enzymes, superoxide dismutase (SOD) and catalase (CAT) reduced by CCl$_4$treatment, were recovered by this crude drug. It is suggested that Caesalpinia sappan L. have antioxidant effect. The increased levels of glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) by CCl$_4$were also recovered by treatment with this crude drug. These results can be attributed to the agent's antioxidant and membrane - stabilizing actions.

Change of Blood Cortisol Concentration in Domestic Cats by Various Stress Types (스트레스 유형에 따른 고양이 혈중 Cortisol농도의 변화)

  • The present study was performed to evaluate circadian rhythm of blood nortisol concentration and change of blood cortisol concentration by various stress type in domestic cats. The ELISA kit for cortisol measurement was evaluated on the effective assay range, day to day variation, within-run variation and accuracy. The results were 1-1,000 ng/ ml, 0.7-5.9%, 0.9-4.5% and 98%, respectively. The circadian rhythm of serum cortisol concentration is presented in cats. Change of serum cortisol concentration was also examined with several stressors, stimulation with hamster as a prey, beep electronic sounds, vehicle transportation and unfamiliar environment. Unfamiliar environment showed most remarkable change in serum cortisol level.

Protective Effect of Sachungwhan against CCl4-induced Hepatotoxicity

  • Sachungwhan reduced hepatotoxicity induced by carbon tetrachloride($CCl_4$). Improved liver function was observed by measuring the activities of aspartate aminotransferase(AST), alanine aminotransferase(ALT), alkaline phosphatase(ALP), blood urea nitrogen(BUN), creatinine(CRE), total cholesterol(TCHO), triglyceride(TG), low density lipoprotein cholesterol(LDL-CHO), high density lipoprotein cholesterol(HDL-CHO), total protein(TP), albumin(ALB) and total bilirubin(BIL) in serum. Hepatic parameters monitored were levels of cholesterol(CHO), triglyceride(TG), malondialdehyde(MDA), content of cytochrome P450(CYP), level of glutathione(GSH), and activities of NADPH-CYP reductase, superoxide dismutase(SOD), catalase(CAT), glutathione S-transferase(GST), glutathione reductase(GR), glutathione peroxidase(GPx). The histopathological examination showed that the treatment of Sachungwhan relieved the ballooning degeneration of hepatocytes which had been generated by $CCl_4$. The results suggested that hepatoprotective effects of Sachungwhan possibly are due to their promising antioxidative activity.

  • We synthesized catechol-conjugated chitosan (CCC) to study its usefulness as a construction material additive in cement mortar. The degree of catechol conju-gation (DOCcat) of the synthesized CCC was determined to be approximately14% by UV-vis and 1H NMR spectroscopy. Furthermore, the hydroxyl and amine groups in CCC could play a crucial role in hydrogen bonding, metal coordination, and cross-linking processes via interaction with adducts from cement mortar. In this study, we observedanimprovement in the compressive strength and absorption rate, suggesting that CCC is a promising candidateforhigh-performance cement mortar.
