Identification of inhibitors against ROS1 targeting NSCLC by In- Silico approach

  • ROS1 (c-ros oncogene) is one of the gene with mutation in NSCLC (non-small cell lung cancer). The increased expression of ROS1 is leading to the increase proliferation of cell, cell migration and survival. Crizotinib and Entrectinib are the drugs that have been approved by FDA against ROS1 protein, but recently patients started to develop resistance against Crizotinib and there is a need of new drug that could act as an effective drug against ROS1 for NSCLC. In this study, we have performed virtual screening, where compounds are taken from Zinc 15 dataset and molecular docking was performed. The top compounds were taken based upon their binding affinity and their interactions with the residues. The compounds stability and chemical reactivity was also studied through Density Functional theory and their properties. Further study of these compounds could reveal the required information of ROS1-inhibitor complex and in the discovery of potent inhibitors.

착체중합법에 의한 $ZnWO_4$ 나노분말의 저온합성 (Low temperature synthesis of $ZnWO_4$ nanopowders using polymeric complex precursor)

  • 착체중합법을 사용하여 nano-size의 $ZnWO_4$ powder를 저온에서 합성하였다. 금속이온물질로서 zinc acetate와 tungstic acid를 사용하였으며 용매는 de-ionized water를 사용하였다. $300^{\circ}C$ 부터 $600^{\circ}C$의 온도 영역에서 하소한 분말에 대해 열분해 및 결정화 과정, 분말의 형상, 입도 변화 양상을 분석하였다. 일반적인 고상합성 시에 필요한 온도보다 현저히 낮은 온도인 $400^{\circ}C$에서 $ZnWO_4$상이 생성되었으며, $600^{\circ}C$에서 완전한 결정상을 얻을 수 있었다. 합성된 분말은 $400^{\circ}C$에서 원형과 silk-worm 형태가 혼합된 입자 형상을 나타내었고 $500^{\circ}C$에서 보다 균질한 양상을 나타내었다. 합성된 분말의 입자 크기는 $400^{\circ}C$~$600^{\circ}C$의 온도영역에서 17.62~24.71 nm 정도로 매우 미세하였으며, 하소 온도가 증가함에 따라 분말의 결정상과 입도가 증가하는 것을 확인하였다.

수용액, 에탄올-물 및 아세톤-물 혼합용매 내에서의 망간, 아연 및 코발트의 시트르산 착물 (Citrate Complexes of Manganese, Zinc and Cobalt in Aqueous, Ethanol-Water and Acetone-Water Solutions)

  • 양 이온 교환수지와 시트르산 이온($Cit^{3-}$)이 들어 있는 용액에서 방사성 $Mn^{++},\;Zn^{++}$$Co^{++}$이 수지와 용액 사이에 어떻게 분배되는가를 실온에서 조사함으로써 이들금속 이온의 시트르산 착물 형성에 관한 연구를 하였다. 용매로서 $H_2O$, 20%에탄올-$H_2O$ 및 20% 아세톤-$H_2O$를 사용하였다. 용액의 pHsms 7.0-7.4로 조절하였고 이온 강도는 약 0.10으로 유지하였다. 본 연구의 결과 $Mn^{++}$$Zn^{++}$은 어떤 용매계에서도 1:1 시트르산 착물[M Cit]$^-$을 형성하는 것을 알았다. 한편 $Co^{++}$은 수용액에서는 1:1 착물만을 형성하지만 위에 말한 혼합용매에서는 1:1 착물 [Co Cit]$^-$과 1:2착물 $[Co\;Cit_2]^{4-}$을 형성하는 것을 알았다. 그리고 이들 시트르산 착물이 수용액에서 보다 혼합용매 내에서 더욱 안정함을 알았다.

Distribution Characteristics of Dust and Heavy Metals in the Atmosphere Around the Steel Industrial Complex

  • In Dangjin, Chungcheongnam-do, there are not only power plants and large steel complexes, but also small and medium-sized air pollutant emission facilities. The dust generated by these facilities has a very small particle size and a large surface area due to condensation and physical and chemical reactions, and is discharged containing various harmful substances. Therefore, this study analyzed the distribution of particulate matter and heavy metal concentrations by particle size in the vicinity of the steel complex, residential area, and reference point using an eight-stage Cascade Impactor. Overall, the direct impact sites with a short distance from the steel complex had the highest concentration, followed by the indirect impact sites, and the non-impact sites had the lowest concentration, indicating that they are directly affected by the steel complex. The atmospheric dust concentration distribution showed a bimodal distribution with a minimum value around the 1.1 to 2.1 ㎛ particle diameter. However, during the yellow dust event, the maximum concentration was biased toward coarse particles. The proportion of PM2.5 in the dust tended to be higher in winter, while the ratio between PM2.5 and PM10 was relatively higher in spring. Regardless of the location of the impact point, heavy metals in the dust were dominated by iron and aluminum, followed by zinc, lead, and manganese.

Effect of Combined Exposure to EDTA and Zinc Pyrithione on Pyrithione Absorption in Rats

  • Zinc pyrithione (ZnPT) is a coordination complex of zinc and has been used widely as an anti-dandruff agent in shampoos. Many shampoos contain both ZnPT and EDTA, a chelating agent speculated to increase ZnPT absorption, thereby raising concerns about neurotoxicity. Here, we investigated the effect of EDTA on ZnPT absorption by direct comparison of ZnPT and pyrithione (PT) concentrations in shampoo formulations, and by pharmacokinetic analysis of ZnPT, PT, and 2-methanesulfonylpyridine (MSP), the main ZnPT metabolite, in rat plasma or urine following exposure to shampoo containing ZnPT alone or a combination of ZnPT and EDTA. Approximately 17.3% of ZnPT was converted to PT by the addition of EDTA in the shampoo formulation. Plasma ZnPT and PT concentrations were not measured up to 24 hr after treatment with shampoo containing 1% ZnPT or 1% ZnPT + 2% EDTA in all rats. However, PT amount in 24-hr urine sample, MSP concentration in plasma, and MSP amount in 24-hr urine sample were approximately 4-, 2.6-, and 2.7-fold higher, respectively, in the 1% ZnPT + 2% EDTA shampoo group than in the 1% ZnPT shampoo group. As confirmed by the formulation analysis and in vivo pharmacokinetic analysis, the exposure of ZnPT could be increased by the absorption of PT due to partial dissociation of ZnPT into PT.

Aflatoxin $B_1$ Charge-transfer Complex에 관(關)한 연구(硏究) -제1보(第一報) Benzene과의 Charge-transfer Complex- (Studies on the Charge-transfer Complex including Aflatoxin $B_1$ -Part I. Charge-transfer Complex with Benzene-)

  • Aflatoxin $B_1$은 전자수용성(電子受容性)을 높여주는 염화아연(鹽化亞鉛) 존재하(存在下)에서 전자공여성분자(電子供與性分子)인 Benzene과 Charge-transfer Complex를 만들며, 그 생성기구(生成機構)는 Aflatoxin $B_1$이 염화아연(鹽化亞鉛)과 일차적(一次的)으로 배위결합(配位結合)된 화합물(化合物)을 거쳐, 이것이 Benzene과 결합(結合)하여 착물(錯物)을 형성(形成)한다. 이 착물(錯物)의 안정도상수(安定度常數) 즉(卽) 평형상수(平衡常數) 0.198 l/mole이었다. 따라서 Aflatoxin $B_1$은 약(弱)한 전자수용체(電子受容體)이나, Benzene 보다 강(强)한 전자공여체(電子供與體)와는 염화아연(鹽化亞鉛)이 존재(存在)하지 않아도 Charge-transfer Complex를 만들 수 있다는 가능성(可能性)을 제시(提示)해 주는 것이며, Tryptophane, Histidine과 같은 강(强)한 전자공여체(電子供與體)를 함유(含有)한 단백질(蛋白質) 또는 Guanine, Adenine과 같은 전자공여체(電子供與體)를 함유(含有)한 DNA등(等)과의 Aflatoxin $B_1$의 결합(結合)은 그 결합(結合) Mechanism으로서 Charge-transfer Complex 형성(形成)으로 이루어진다는 추정(推定)을 할 수 있다.

Application of Synergistic Solvent Extraction by Formation of Ternary Complex for Determination of Trace Zn(II) in Water Samples

  • The application of a synergistic solvent extraction by the formation of ternary complex with pyrocatechol violet (PV) and benzalkonium chloride (BC) was studied for determination of trace Zn(II) in water samples. The pH of sample solution and the amount of PV and BC added were optimized for the formation of the stable complex, a proper solvent was selected for the effective extraction, and the concentration of nitric acid was fixed for the back extraction of the complex from the solvent. After the ionic strength of 100 mL sample solution was adjusted to 0.1 M by adding NaCl and the pH was fixed at 9 with a carbonate buffer, 1.0 mL of 2% PV solution was added to form Zn(II)-PV complex then the Zn(II)-PV/BC ternary complex was made by adding 1.0 mL of 10% BC solution. The ternary complex was extracted into 10 mL of MIBK. And the ternary complex was back-extracted with 10 mL of 1.0 mol/L nitric acid to determine Zn(II) by a flame atomic absorption spectrophotometer (flame-AAS). The interference of concomitant ions on the extraction of Zn(II) was investigated. This procedure was applied to the analysis of three real samples such as Dalbang-dam water, laboratory tap water and Jungnajin seawater. The recoveries of Zn(II) in spiked samples were 86.58-104.1%.

2-Oximino-1-indanone을 이용한 Pt(IV) ion의 정량에 관한 연구 (Spectrophotometric Determination of Platinum (IV) with 2-Oximino-1-indanone)

  • A method is described for the spectrophotometric determination of platinum (IV) with 2-oximino-1-indanone based on solvent extraction of Pt:2-oximino-1-indanone complex. The 2-oximino-1-indanone reacted with Pt(IV) to form a dark-orange complex which shows a characterisic maximum absorption at 342nm. The optimum PH for the platinum extraction lies between 5.4~8.0. Beer's law obeys up to 0.98-16.3ppm of platinum (IV) and the molar absorption coefficient is $1.06{\times}10^{-4}L.mol^{-1}.cm^{-1}$. The relative standard deviation of the method was $\times2.1%$. The composition of the complex is estimated to be Pt : In= 1 : 1, by the mole ratio method and ion exchange resin experiment. The optimum condition for the determination of platinum has been studied in detail. The 2-oximin-1-indanone is found to be a selectivereagent for the determination of platinum, since the synthesixed 2-oximino-1-indanone did not react with other metals such as cobalt, cadmium, copper, manganese nickel, iron, lead and zinc, to form the complex. In this studies, we have also clarified Sindhwani and Singh's spectrophotometric determination data of various metals with acenaphthenequinone monooxime (Talanta 20,248, 1973), whose results were not correct.

Preparation of a Fluoroionophore Based on Porphyrin-Crown Ether

  • A porphyrin compound containing a crown ether moiety (Por-Crown) and its zinc complex (ZnPor-Crown) have been prepared and the effect of the addition of alkali metal on their fluorescence has been investigated. As alkali metal cations were added, the absorption and fluorescence maxima did not change. However, the absorbance and intensity of fluorescence increased dramatically. Among the alkali metal cations tested, addition of K$^{+}$ and Cs$^{+}$ showed strongest enhancement of absorbance and fluorescence intensity of Por-Crown and ZnPor-Crown.own.
