Zika Virus on YouTube: An Analysis of English-language Video Content by Source

  • Objectives: The purpose of this study was to describe the source, length, number of views, and content of the most widely viewed Zika virus (ZIKV)-related YouTube videos. We hypothesized that ZIKV-related videos uploaded by different sources contained different content. Methods: The 100 most viewed English ZIKV-related videos were manually coded and analyzed statistically. Results: Among the 100 videos, there were 43 consumer-generated videos, 38 Internet-based news videos, 15 TV-based news videos, and 4 professional videos. Internet news sources captured over two-thirds of the total of 8 894 505 views. Compared with consumer-generated videos, Internet-based news videos were more likely to mention the impact of ZIKV on babies (odds ratio [OR], 6.25; 95% confidence interval [CI], 1.64 to 23.76), the number of cases in Latin America (OR, 5.63; 95% CI, 1.47 to 21.52); and ZIKV in Africa (OR, 2.56; 95% CI, 1.04 to 6.31). Compared with consumer-generated videos, TV-based news videos were more likely to express anxiety or fear of catching ZIKV (OR, 6.67; 95% CI, 1.36 to 32.70); to highlight fear of ZIKV among members of the public (OR, 7.45; 95% CI, 1.20 to 46.16); and to discuss avoiding pregnancy (OR, 3.88; 95% CI, 1.13 to 13.25). Conclusions: Public health agencies should establish a larger presence on YouTube to reach more people with evidence-based information about ZIKV.

Zika Virus Proteins NS2A and NS4A Are Major Antagonists that Reduce IFN-β Promoter Activity Induced by the MDA5/RIG-I Signaling Pathway

  • Zika virus (ZIKV) is a mosquito-transmitted, emerging Flavivirus that causes Guillain-$Barr{\acute{e}}$ syndrome and microcephaly in adults and fetuses, respectively. Since ZIKV was first isolated in 1947, severe outbreaks have occurred at various places worldwide, including Yap Island in 2007, French Polynesia in 2013, and Brazil in 2015. Although incidences of ZIKV infection and dissemination have drastically increased, the mechanisms underlying the pathogenesis of ZIKV have not been sufficiently studied. In addition, despite extensive research, the exact roles of individual ZIKV genes in the viral evasion of the host innate immune responses remain elusive. Besides, it is still possible that more than one ZIKV-encoded protein may negatively affect type I interferon (IFN) induction. Hence, in this study, we aimed to determine the modulations of the IFN promoter activity, induced by the MDA5/RIG-I signaling pathway, by over-expressing individual ZIKV genes. Our results show that two nonstructural proteins, NS2A and NS4A, significantly down-regulated the promoter activity of IFN-${\beta}$ by inhibiting multiple signaling molecules involved in the activation of IFN-${\beta}$. Interestingly, while NS2A suppressed both full-length and constitutively active RIG-I, NS4A had inhibitory activity only on full-length RIG-I. In addition, while NS2A inhibited all forms of IRF3 (full-length, regulatory domain-deficient, and constitutively active), NS4A could not inhibit constitutively active IRF3-5D. Taken together, our results showed that NS2A and NS4A play major roles as antagonists of MDA5/RIG-I-mediated IFN-${\beta}$ induction and more importantly, these two viral proteins seem to inhibit induction of the type I IFN responses in differential mechanisms. We believe this study expands our understanding regarding the mechanisms via which ZIKV controls the innate immune responses in cells and may pave the way to development of ZIKV-specific therapeutics.

CLIMEX-based Analysis of Potential Geographical Distribution of Aedes albopictus and Aedes aegypti in South Korea

  • Purpose: Aedes aegypti and Aedes albopictus are notorious disease vectors that spread various viruses including dengue, yellow fever, chikungunya, and Zika. Recent Zika virus outbreaks were carried by Ae. aegypti, raising awareness about the perils of its global distribution. Because Ae. albopictus is spread throughout South Korea and can carry the same viruses as Ae. aegypti, monitoring potential distributions of Ae. albopictus and Ae. aegypti is necessary. Methods: In this study, the potential distributions of Ae. albopictus and Ae. aegypti in South Korea were modeled using CLIMEX software, and changes in response to climate change were predicted. Results: The results indicated that the climatic suitability for Ae. albopictus was consistently high, while occurrence of Ae. aegypti was only predicted for Jeju Island in 2080. Conclusions: The results provide basic information for preventing the invasion of Ae. aegypti, and consequent interactions between Ae. aegypti and Ae. albopictus, which may cause severe outbreaks of dangerous diseases.

  • 플라비바이러스(Flavivirus)는 모기와 같은 곤충을 매개로 하여 인체에 감염된다고 잘 알려져 있다. 그 대표적인 예로 지카 바이러스(Zika virus), 뎅기 바이러스(Dengue virus), 황열 바이러스(Yellow fever virus), 일본 뇌염 바이러스(Japanese encephalitis virus) 등을 들 수 있다. 본 연구에서는 생물정보학을 기반으로 인체 감염 주요 플라비바이러스인 지카 바이러스, 뎅기 바이러스. 황열 바이러스, 일본 뇌염 바이러스의 총 4종류 플라비바이러스에 공통적으로 적용 가능한 펩타이드 백신 후보를 제시하고자 한다. 먼저 UniProt (The Universal Protein Resource)의 유전자 서열정보를 이용하여 4종류의 바이러스가 가진 단백질 중 백신으로써 적합한 단백질을 선정하였다. 선정된 단백질의 아미노산 서열정보를 바탕으로 IEDB (Immune Epitope Database And Analysis Resource)를 활용한 에피토프(epitope) 분석을 통해 에피토프로 작용하는 4 종류 바이러스의 공통적인 서열을 도출하였다.

One Health Perspectives on Emerging Public Health Threats

  • Antimicrobial resistance and emerging infectious diseases, including avian influenza, Ebola virus disease, and Zika virus disease have significantly affected humankind in recent years. In the premodern era, no distinction was made between animal and human medicine. However, as medical science developed, the gap between human and animal science grew deeper. Cooperation among human, animal, and environmental sciences to combat emerging public health threats has become an important issue under the One Health Initiative. Herein, we presented the history of One Health, reviewed current public health threats, and suggested opportunities for the field of public health through better understanding of the One Health paradigm.

Seasonal Prevalence of Mosquitoes Collected from Light Traps in Gyeongsangnam Province, Republic of Korea (2013-2014)

  • Adult mosquito surveillance was conducted from 2013 through 2014 at four cattle sheds, a wild bird refuge, and two residential areas located in Gyeongnam Province in the Republic of Korea. Adult mosquitoes were collected in black light traps from April 1, through November 30. Mosquito surveillance was conducted to figure out population densities of vector mosquitoes, possibly invaded mosquitoes and identify various virus infections at the selected sites. A total of 107,466 females comprising 14 species and 7 genera were collected from 2013 to 2014. The most common species collected were Culex tritaeniorhynchus Giles (63.8%), Anopheles sinensis s.l. (18.9%), Aedes vexans nipponii (Theobald) (7.7%), and Culex pipiens Coquillett (5.1%). Trap indices (TIs) varied widely for species over their range, due to geographical distribution and degree of association with rural and urban communities. The most collected An. sinensiss.l. and Cx. tritaeniorhynchus appeared at a cow shed in Hapcheon (TI 347.5) and a pigsty in Daejeo-1-dong, Busan (TI 1,040.8), respectively, due in part to their situation near breeding sites such as rice paddies. The bi-weekly population densities for mosquito species were variable for each of the years, apparently as a result of variable annual weather conditions. None of the mosquito species collected tested for the flavivirus including Japanese Encephalitis Virus, West Nile Virus, Dengue Virus, and Zika Virus infections by polymerase chain reaction (PCR) assay were positive.

  • 본 연구에서는 2016년 전 세계에 공포를 안겼던 지카 바이러스(Zika virus)와 최근 아열대성 기후의 확대로 인해 향후 대유행이 예상되는 뎅기 바이러스(Dengue virus)의 구별 진단 방법에 대해 논의하고자 한다. 두 바이러스는 모두 플라비바이러스 속(Flavivirus genus)에 해당되며, 동일한 단백질 도메인(domain)으로 구성되어 있다. 본 연구에서는 여러 도메인 중에서도 진단에 유리하다고 판단되는 비구조단백질 1(NS1, non-structural protein 1)을 선정하였으며, UniProt (Universal Protein Resource) 데이터베이스를 활용하여 지카 바이러스와 뎅기 바이러스의 비구조단백질 1의 아미노산 서열을 상호 비교 분석 하였다. 더 나아가 IEDB (Immune Epitope Database) 데이터베이스를 활용하여 비구조단백질 1의 아미노산 중, 진단에 용이한 에피토프로 예상되는 5개의 후보를 도출하였다. 최종적으로 후보군으로부터 지카 바이러스와 뎅기 바이러스를 구별하여 진단할 수 있다고 판단되는 에피토프(펩타이드)를 제안하였다.

Text Mining and Visualization of Papers Reviews Using R Language

  • Nowadays, people share and discuss scientific papers on social media such as the Web 2.0, big data, online forums, blogs, Twitter, Facebook and scholar community, etc. In addition to a variety of metrics such as numbers of citation, download, recommendation, etc., paper review text is also one of the effective resources for the study of scientific impact. The social media tools improve the research process: recording a series online scholarly behaviors. This paper aims to research the huge amount of paper reviews which have generated in the social media platforms to explore the implicit information about research papers. We implemented and shown the result of text mining on review texts using R language. And we found that Zika virus was the research hotspot and association research methods were widely used in 2016. We also mined the news review about one paper and derived the public opinion.

  • In the aftermath of a recent outbreak of Middle East respiratory syndrome (MERS), legal matters on the spread of epidemics have emerged as major social issues in Korea, with the awareness that rampant contagious diseases are disasters in society. Problems arose over damages arising from patients who contract infectious diseases, compensation for people under quarantine and for a partial, or entire, shutdown of hospitals, and information disclosure on infectious diseases. As epidemic-linked problems have never been socially disputed in Korea, the relevant legal issues appear to be studied comparatively less than other areas. As interests are heightened over legal issues on infectious illnesses amid the MERS scare, in-depth, further research on such legal issues should be conducted in an utmost effort to prevent and make preparations for epidemics which potentially become a social disaster, such as new Hong Kong flu, Ebola virus and Zika virus.

Monitoring and Distribution of Mosquitoes in the Downtown Areas and Migratory Bird Habitats in the Jeonbuk Region, 2021 (2021년 전북지역 도심과 철새서식처의 모기 감시와 분포)

  • This survey was performed to monitor the spread of specific mosquito-borne pathogens at Jeonbuk. The frequency of occurrence of mosquito borne pathogens including Japanese encephalitis virus, West Nile virus, Zika virus, and yellow fever virus was assessed by collecting mosquitoes twice a month from March to December 2021 from various areas in Jeonbuk. A total of 15,975 mosquitoes from 15 species and 7 genera were collected. The highest number of 9,116 mosquitoes (trap index: TI, 506.4) were collected in the Wanju cattle pen, followed by the habitat for migratory birds and the downtown area in Jeonju. In the Gunsan habitat for migratory birds, 3,217 mosquitoes (TI, 178.7) were collected in the reed fields, 356 (TI, 19.7) in the men's toilets, and 1,948 (TI, 108.2) in the women's toilets. In Jeonju, 677 mosquitoes (TI, 37.6) were collected in the Deokjin park, 358 (TI, 19.8) in the Deokjin-gu office, and 303 (TI, 16.8) at the Jeonbuk National University. The largest population of mosquitoes was collected in the men's toilets in Gunsan and the Deokjin Park in downtown Jeonju. The results of the RT-PCR confirmation to determine the pathogen infection of the collected mosquitoes were all negative. These results provide a basis for tackling integrated mosquito-borne diseases in the Jeonbuk region.
